Abstract Mapped monthly data products of surface ocean acidification indicators from 1998 to 2022 on a 0.25° by 0.25° spatial grid have been developed for eleven U.S. large marine ecosystems (LMEs). ...The data products were constructed using observations from the Surface Ocean CO 2 Atlas, co-located surface ocean properties, and two types of machine learning algorithms: Gaussian mixture models to organize LMEs into clusters of similar environmental variability and random forest regressions (RFRs) that were trained and applied within each cluster to spatiotemporally interpolate the observational data. The data products, called RFR-LMEs, have been averaged into regional timeseries to summarize the status of ocean acidification in U.S. coastal waters, showing a domain-wide carbon dioxide partial pressure increase of 1.4 ± 0.4 μatm yr −1 and pH decrease of 0.0014 ± 0.0004 yr −1 . RFR-LMEs have been evaluated via comparisons to discrete shipboard data, fixed timeseries, and other mapped surface ocean carbon chemistry data products. Regionally averaged timeseries of RFR-LME indicators are provided online through the NOAA National Marine Ecosystem Status web portal.
Hydrogel microcapsules have been used for decades to encapsulate cells and treat diseases ranging from neurodegenerative disorders to more systemic applications like Type I Diabetes. This cell ...encapsulation modality has been developed through more cumulative experiments than perhaps any other, owing to the relative ease of accessing the required materials, the commercial availability of droplet-generating instrumentation, and the mild microenvironment and unique permeability properties of hydrogels that are difficult to attain with alternative encapsulation systems employing thermoplastic materials. Because of their size and shape, microcapsules have an inherent advantage over macroencapsulation devices due to the more favorable surface area to volume ratio, which allows for greater efficiency in the amount of cellular cargo that is entrapped and enhanced nutrient exchange and efflux of secreted products. Unfortunately, with this significant positive benefit comes the caveat of difficult or impractical retrievability, highlighting the paradox that is particularly relevant as differentiated stem cell sources become more readily available. This chapter focuses on several techniques that can be used to evaluate the permeability and pore structure of hydrogel microcapsules, including a simplistic model for describing the diffusive behavior of alginate-polycation-alginate (APA) microcapsules with a liquid core, and an ancillary method to evaluate the ultrastructure of the APA membrane including morphometric analysis.
Within human physiology, systemic interactions couple physiological variables to maintain homeostasis. These interactions change according to health status and are modified by factors such as age and ...sex. For several physiological processes, sex-based distinctions in normal physiology are present and defined in isolation. However, new methodologies are indispensable to analyze system-wide properties and interactions with the objective of exploring differences between sexes. Here we propose a new method to construct complex inferential networks from a normalization using the clinical criteria for health of physiological variables, and the correlations between anthropometric and blood tests biomarkers of 198 healthy young participants (117 women, 81 men, from 18 to 27 years old). Physiological networks of men have less correlations, displayed higher modularity, higher small-world index, but were more vulnerable to directed attacks, whereas networks of women were more resilient. The networks of both men and women displayed sex-specific connections that are consistent with the literature. Additionally, we carried out a time-series study on heart rate variability (HRV) using Physionet’s Fantasia database. Autocorrelation of HRV, variance, and Poincare’s plots, as a measure of variability, are statistically significant higher in young men and statistically significant different from young women. These differences are attenuated in older men and women, that have similar HRV distributions. The network approach revealed differences in the association of variables related to glucose homeostasis, nitrogen balance, kidney function, and fat depots. The clusters of physiological variables and their roles within the network remained similar regardless of sex. Both methodologies show a higher number of associations between variables in the physiological system of women, implying redundant mechanisms of control and simultaneously showing that these systems display less variability in time than those of men, constituting a more resilient system.
There has been increasing interest in developing bioadhesive nanoparticles due to their great potential as carriers for therapeutics in oral drug delivery systems. Despite decades of research, such a ...system still has not been successfully implemented. This paper demonstrates the enormous potential of such engineered systems: the incorporation of a bioadhesive coating, poly(butadiene-maleic anhydride-co-L-DOPA) (PBMAD), to non-bioadhesive nanospheres resulted in an enhancement of particle uptake in the small intestine from 5.8±1.9% to 66.9±12.9%. Direct correlation was obtained between bulk tensile strength, in vitro binding to everted intestinal sacs and quantitative in vivo uptake; this data suggests that bulk properties of polymers can be used to predict bioadhesive properties of nano- and microparticles. The differential distribution of the nanospheres to various tissues following uptake suggests surface chemistry plays a significant role in their localization within the body. The results of these studies provide strong support for the use of bioadhesive polymers to enhance nano- and micro-particle uptake from the small intestine for oral drug delivery.

Objective
To identify distinct trajectories of toxicity in colorectal cancer (CRC) patients after adjuvant chemotherapy and its impact on quality of life (QoL) and psychological symptoms.
Methods
A ...prospective, multicenter study was conducted in 157 patients. A latent class analysis defined the unobserved latent constructs that can be predicted as symptom clusters, considering the intensity of four types of adverse events (AEs). Patients completed EORTC-QLQ-C30, BSI-18, PDRQ-9, and DRS scales.
Results
Ninety-six percent had some degree of toxicity, with grades 3–4 being the most common: neurotoxicity (7.2%), hematological (13.1%), digestive (5.2%), and skin toxicity (1.4%). Three distinct latent classes were identified (high 72.5%, mild 16.9%, and low 10.6% toxicity). Patients with high toxicity had the worst QoL scores and moderately high somatization and psychological distress scores.
Conclusions
Adjuvant chemotherapy for CRC was associated with frequent toxicity that negatively impacted QoL and psychological wellbeing.
A common strategy for calculating the direction and rate of carbon dioxide gas (CO2) exchange between the ocean and atmosphere relies on knowledge of the partial pressure of CO2 in surface seawater ...(pCO2(sw)), a quantity that is frequently observed by autonomous sensors on ships and moored buoys, albeit with significant spatial and temporal gaps. Here we present a monthly gridded data product of pCO2(sw) at 0.25∘ latitude by 0.25∘ longitude resolution in the northeastern Pacific Ocean, centered on the California Current System (CCS) and spanning all months from January 1998 to December 2020. The data product (RFR-CCS; Sharp et al., 2022; 10.5281/zenodo.5523389) was created using observations from the most recent (2021) version of the Surface Ocean CO2 Atlas (Bakker et al., 2016). These observations were fit against a variety of collocated and contemporaneous satellite- and model-derived surface variables using a random forest regression (RFR) model. We validate RFR-CCS in multiple ways, including direct comparisons with observations from sensors on moored buoys, and find that the data product effectively captures seasonal pCO2(sw) cycles at nearshore sites. This result is notable because global gridded pCO2(sw) products do not capture local variability effectively in this region, suggesting that RFR-CCS is a better option than regional extractions from global products to representpCO2(sw) in the CCS over the last 2 decades. Lessons learned from the construction of RFR-CCS provide insight into how global pCO2(sw) products could effectively characterize seasonal variability in nearshore coastal environments. We briefly review the physical and biological processes – acting across a variety of spatial and temporal scales – that are responsible for the latitudinal and nearshore-to-offshorepCO2(sw) gradients seen in the RFR-CCS reconstruction ofpCO2(sw). RFR-CCS will be valuable for the validation of high-resolution models, the attribution of spatiotemporal carbonate system variability to physical and biological drivers, and the quantification of multiyear trends and interannual variability of ocean acidification.
