Complex disease genetics has been revolutionised in recent years by the advent of genome-wide association (GWA) studies. The chronic inflammatory bowel diseases (IBDs), Crohn's disease and ulcerative ...colitis have seen notable successes culminating in the discovery of 99 published susceptibility loci/genes (71 Crohn's disease; 47 ulcerative colitis) to date. Approximately one-third of loci described confer susceptibility to both Crohn's disease and ulcerative colitis. Amongst these are multiple genes involved in IL23/Th17 signalling (IL23R, IL12B, JAK2, TYK2 and STAT3), IL10, IL1R2, REL, CARD9, NKX2.3, ICOSLG, PRDM1, SMAD3 and ORMDL3. The evolving genetic architecture of IBD has furthered our understanding of disease pathogenesis. For Crohn's disease, defective processing of intracellular bacteria has become a central theme, following gene discoveries in autophagy and innate immunity (associations with NOD2, IRGM, ATG16L1 are specific to Crohn's disease). Genetic evidence has also demonstrated the importance of barrier function to the development of ulcerative colitis (HNF4A, LAMB1, CDH1 and GNA12). However, when the data are analysed in more detail, deeper themes emerge including the shared susceptibility seen with other diseases. Many immune-mediated diseases overlap in this respect, paralleling the reported epidemiological evidence. However, in several cases the reported shared susceptibility appears at odds with the clinical picture. Examples include both type 1 and type 2 diabetes mellitus. In this review we will detail the presently available data on the genetic overlap between IBD and other diseases. The discussion will be informed by the epidemiological data in the published literature and the implications for pathogenesis and therapy will be outlined. This arena will move forwards very quickly in the next few years. Ultimately, we anticipate that these genetic insights will transform the landscape of common complex diseases such as IBD.

However, tropical ecosystems are also subject to pervasive and interacting stressors, such as deforestation, overfishing and climate change, and they are set within a socio-economic context that includes growing pressure from an increasingly globalized world, larger and more affluent tropical populations, and weak governance and response capacities. Concerted local, national and international actions are urgently required to prevent a collapse of tropical biodiversity.

Heat stress is a worldwide phenomenon that is associated with reduced animal productivity and welfare, particularly during the summer months. Animal responses to their thermal environment are extremely varied, however, it is clear that the thermal environment influences the health, productivity, and welfare of cattle. Whilst knowledge continues to be developed, managing livestock to reduce the negative impact of hot climatic conditions remains somewhat challenging. This review provides an overview of the impact of heat stress on production and reproduction in bovines.

Vasa previa (VP) remains a condition that is not screened for routinely, even though it has been possible to diagnose VP prenatally with ultrasound imaging for 35 years and its diagnosis before birth is highly effective in preventing associated perinatal morbidity and mortality1-10. In this Opinion, we discuss the controversies surrounding, and possible strategies for, screening for VP.
In an effort to ensure energy independence and exploit mineral resources, the governments of Amazonian countries are embarking on a major dam building drive on the basin’s rivers, with 191 dams ...finished and a further 246 planned or under construction. This rush to harvest the basin’s vast renewable energy capacity has come without proper consideration of the likely negative environmental externalities on the world’s most speciose freshwater and terrestrial biotas. Here we highlight the economic drivers for hydropower development and review the literature to summarise the impacts of dam building on Amazonian biodiversity. We identify both direct and indirect impacts through the anticipated loss, fragmentation and degradation of riparian habitats. We then propose a series of measures to assess, curb and mitigate the impacts of destructive dams on Amazonian biodiversity.
Tropical biotas provide excellent settings in which to explore mechanisms of evolutionary diversification, yet these processes remain poorly understood. Pioneering work on biodiversity patterns and ...diversification processes in other tropical regions has recently been complemented by studies in Madagascar. Here we review diversity models and diversification mechanisms proposed for the fauna of this island and the perspectives for testing them. Madagascar has a diverse biota that has evolved in isolation, and is characterised by regionally pronounced and locally steep environmental gradients, common patterns of microendemism across taxa and numerous evolutionary radiations. These characteristics establish Madagascar as a promising system for the study of pattern and process in species diversification.
