This Letter reports on a cavity haloscope search for dark matter axions in the Galactic halo in the mass range 2.81-3.31 μeV. This search utilizes the combination of a low-noise Josephson parametric ...amplifier and a large-cavity haloscope to achieve unprecedented sensitivity across this mass range. This search excludes the full range of axion-photon coupling values predicted in benchmark models of the invisible axion that solve the strong CP problem of quantum chromodynamics.
To report a pregnancy in a hermaphrodite and review of the literature.
Case report and literature review.
Clinical.
A patient with male-predominant mosaic karyotype 96% 46XY.
Removal of left ...ovotestis in combination with a supracervical hysterectomy and bilateral salpingo-oophorectomy.
Identification of published cases of pregnancy and hermaphroditism.
The current patient had previous pregnancy and a wedge biopsy of her left gonad, which demonstrated an ovotestis and an area suspicious for a gonadoblastoma. After delivery of her second pregnancy, the patient underwent a hysterectomy and bilateral salpingo-oophorectomy. The histopathologic evaluation following bilateral oophorectomy demonstrated a residual ovotestis without further evidence of a gonadoblastoma. Review of the literature identified 10 other cases of pregnancy in a hermaphrodite patient.
This is the 11th reported case of fertility in a true hermaphrodite and only the second report of a mosaic true hermaphrodite to demonstrate fertility. This is the only case of a pregnancy involving a male-predominant mosaic 96% 46XY and the only case to confirm the genetics of the offspring.
DiGIR1 is a group I-like cleavage ribozyme found as a structural domain within a nuclear twin-ribozyme group I intron. DiGIR1 catalyzes cleavage by branching at an Internal Processing Site (IPS) ...leading to formation of a lariat cap at the 5'-end of the 3'-cleavage product. The 3'-cleavage product is subsequently processed into an mRNA encoding a homing endonuclease. By analysis of combinations of 5'- and 3'-deletions, we identify a hairpin in the 5'-UTR of the mRNA (HEG P1) that is formed by conformational switching following cleavage. The formation of HEG P1 inhibits the reversal of the branching reaction, thus giving it directionality. Furthermore, the release of the mRNA is a consequence of branching rather than hydrolytic cleavage. A model is put forward that explains the release of the I-DirI mRNA with a lariat cap and a structured 5'-UTR as a direct consequence of the DiGIR1 branching reaction. The role of HEG P1 in GIR1 branching is reminiscent of that of hairpin P-1 in splicing of the Tetrahymena rRNA group I intron and illustrates a general principle in RNA-directed RNA processing.
Risk factors and infection rates of radical robotic procedures have yet to be described in gynecology. A practice improvement strategy using a solitary dose of vaginal metronidazole the night before ...surgery was initiated to determine if it decreased the risk of pelvic infection.
A retrospective chart review of robotic radical hysterectomies for gynecologic malignancy at our institution from April 2010 through April 2016 was performed. Demographic data, operative data, and data on use of metronidazole before surgery were collected. χ Statistical analysis, Student t test, and multivariate analysis were performed to analyze the data.
Ninety-four patients met the inclusion criteria, and 46 patients received vaginal metronidazole. Demographic and clinical factors were similar between the 2 groups. The pelvic infection rate was significantly higher in nonusers at 13% (6/46) compared with users at 0% (0/42) (P ≤ 0.05). The genitourinary infection rate was also significantly higher in nonusers at 20% as compared with users at 2.2% (P = 0.02). Operative risk factors found to be associated with pelvic infection included hospital length of stay, blood loss, and metronidazole use. Multivariate regression analysis determined that only vaginal metronidazole had a clinically significant reduction of pelvic and genitourinary infection.
A single dose of preoperative vaginal metronidazole reduces the risk of pelvic and genitourinary infection after robotic radical hysterectomy.
Schwab, W.C.; Baldwin, W.E.; Hapke, C.J.; Lentz, E.E.; Gayes, P.T.; Denny, J.F.; List, J.H., and Warner, J.C., 2013. Geologic evidence for onshore sediment transport from the inner continental shelf: ...Fire Island, New York. Sediment budget analyses along the south shore of Fire Island, New York, have been conducted and debated in the scientific and coastal engineering literature for decades. It is well documented that a primary component of sediment transport in this system is directed alongshore from E to W, but discrepancies in volumetric sediment budget calculations remain. An additional quantity of sand, averaging about 200,000 m3/y is required to explain the growth of the western segment of the barrier island, a prograding spit. Littoral sediment derived from updrift erosion of the coast, addition of beach nourishment fill, and onshore transport of inner continental shelf, shoreface sediments, or both have all been proposed as potential sources of the additional sediment needed to balance the sediment budget deficit. Analysis of high-resolution seafloor mapping data collected in 2011, including seismic reflection profiles and inteferometric sonar acoustic backscatter and swath bathymetry; comparison with seafloor mapping data collected in 1996–1997; and shoreline change analysis from 1933 to 2011 support previous suggestions that the inner-shelf Holocene sedimentary deposit is a likely source to resolve this sediment budget discrepancy.
Sea-level rise (SLR) is projected to increase dramatically with profound effects on tidal marshes, yet uncertainty stemming from underlying climate change scenarios, model specifications, and ...temporal scale is a major hurdle to conservation planning. We compared likely effects of SLR for 2030 and 2050 under static inundation and dynamic response model predictions for the northeastern USA, where tidal marshes experience elevated rates of SLR compared to global averages. Static inundation and dynamic response models of SLR, which differ in how they incorporate uncertainty associated with local processes and biophysical feedbacks, have historically been applied at different scales, and generally differ in spatial and temporal predictions of marsh vulnerability. We used population estimates for five tidal marsh bird species of conservation concern to predict patterns of population change for each SLR model and examined how uncertainty affects planning decisions for these species. Static inundation and dynamic response models differed markedly in their predictions for 2030, yet both models predicted with reasonable certainty that only 10–15% of tidal marsh in northeastern USA is likely (> 66% chance; as defined by the IPCC) to remain by 2050. Most (85–90%) of the marsh is predicted to be as likely as not (33–66% chance) to disappear, representing high potential for the loss of habitat for > 85% of current populations of four of the five bird species. We propose a planning approach using guidelines established by the IPCC to categorize uncertainty associated with marsh loss due to SLR and apply it to prioritize key sites for preservation.
Structures of mammalian carboxylesterases (CEs) reveal the presence of a ‘side door’ that is proposed to act as an alternative pore for the trafficking of substrates and products. p-Nitrobenzyl ...esterase (pnb CE) from Bacillus subtilis exhibits close structural homology and a similar side-door domain as mammalian CEs. We investigated the role of a specific ‘gate’ residue at the side door (i.e., Leu 362) during pnb CE-catalyzed hydrolysis of model esters, pesticides, and lipids. Recombinant pnb CE proteins containing mutations at position 362 demonstrated markedly lower k cat and k cat/K m values. The mutation with the most significant impact on catalysis was the L362R mutant (k cat/K m was 22-fold lower). Moreover, the ability of the L362R mutant to be inhibited by organophosphates (OP) was also lower. Investigation into the altered catalytic proficiency using pH-activity studies indicated that the catalytic triad of the mutant enzyme was preserved. Furthermore, viscosity variation and carbamate inhibition experiments indicated that rates of substrate association and acylation/deacylation were lower. Finally, recombinant CEs were found to possess lipolytic activity toward cholesteryl oleate and 2-arachidonylglycerol. In summary, the L362R mutant CE markedly slowed the rate of ester hydrolysis and was less sensitive to OP inhibition. The apparent causes of the diminished catalysis are discussed.
The μeV axion is a well-motivated extension to the standard model. The Axion Dark Matter eXperiment (ADMX) collaboration seeks to discover this particle by looking for the resonant conversion of ...dark-matter axions to microwave photons in a strong magnetic field. In this Letter, we report results from a pathfinder experiment, the ADMX "Sidecar," which is designed to pave the way for future, higher mass, searches. This testbed experiment lives inside of and operates in tandem with the main ADMX experiment. The Sidecar experiment excludes masses in three widely spaced frequency ranges (4202-4249, 5086-5799, and 7173-7203 MHz). In addition, Sidecar demonstrates the successful use of a piezoelectric actuator for cavity tuning. Finally, this publication is the first to report data measured using both the TM_{010} and TM_{020} modes.
