Abstract
Highly pathogenic avian influenza (HPAI) viruses of the A/Goose/Guangdong/1/1996 lineage (GsGd), which threaten the health of poultry, wildlife and humans, are spreading across Asia, Europe, ...Africa and North America but are currently absent from South America and Oceania. In December 2021, H5N1 HPAI viruses were detected in poultry and a free-living gull in St. John’s, Newfoundland and Labrador, Canada. Our phylogenetic analysis showed that these viruses were most closely related to HPAI GsGd viruses circulating in northwestern Europe in spring 2021. Our analysis of wild bird migration suggested that these viruses may have been carried across the Atlantic via Iceland, Greenland/Arctic or pelagic routes. The here documented incursion of HPAI GsGd viruses into North America raises concern for further virus spread across the Americas by wild bird migration.
Tobacco (Nicotiana tabacum) is an important plant model system that has played a key role in the early development of molecular plant biology. The tobacco genome is large and its characterisation ...challenging because it is an allotetraploid, likely arising from hybridisation between diploid N. sylvestris and N. tomentosiformis ancestors. A draft assembly was recently published for N. tabacum, but because of the aforementioned genome complexities it was of limited utility due to a high level of fragmentation.
Here we report an improved tobacco genome assembly, which, aided by the application of optical mapping, achieves an N
size of 2.17 Mb and enables anchoring of 64% of the genome to pseudomolecules; a significant increase from the previous value of 19%. We use this assembly to identify two homeologous genes that explain the differentiation of the burley tobacco market class, with potential for greater understanding of Nitrogen Utilization Efficiency and Nitrogen Use Efficiency in plants; an important trait for future sustainability of agricultural production.
Development of an improved genome assembly for N. tabacum enables what we believe to be the first successful map-based gene discovery for the species, and demonstrates the value of an improved assembly for future research in this model and commercially-important species.
Abstract
Difficulties with tracheal intubation commonly arise and impact patient safety. This systematic review evaluates whether videolaryngoscopes reduce intubation failure and complications ...compared with direct laryngoscopy in adults. We searched CENTRAL, MEDLINE, Embase and clinicaltrials.gov up to February 2015, and conducted forward and backward citation tracking. We included randomized controlled trials that compared adult patients undergoing laryngoscopy with videolaryngoscopy or Macintosh laryngoscopy. We did not primarily intend to compare individual videolaryngoscopes. Sixty-four studies (7044 participants) were included. Moderate quality evidence showed that videolaryngoscopy reduced failed intubations (Odds Ratio (OR) 0.35, 95% Confidence Interval (CI) 0.19-0.65) including in participants with anticipated difficult airways (OR 0.28, 95% CI 0.15-0.55). There was no evidence of reduction in hypoxia or mortality, but few studies reported these outcomes. Videolaryngoscopes reduced laryngeal/airway trauma (OR 0.68, 95% CI 0.48-0.96) and hoarseness (OR 0.57, 95% CI 0.36-0.88). Videolaryngoscopy increased easy laryngeal views (OR 6.77, 95% CI 4.17-10.98) and reduced difficult views (OR 0.18, 95% CI 0.13-0.27) and intubation difficulty, typically using an 'intubation difficulty score' (OR 7.13, 95% CI 3.12-16.31). Failed intubations were reduced with experienced operators (OR 0.32, 95% CI 0.13-0.75) but not with inexperienced users. We identified no difference in number of first attempts and incidence of sore throat. Heterogeneity around time for intubation data prevented meta-analysis. We found evidence of differential performance between different videolaryngoscope designs. Lack of data prevented analysis of impact of obesity or clinical location on failed intubation rates. Videolaryngoscopes may reduce the number of failed intubations, particularly among patients presenting with a difficult airway. They improve the glottic view and may reduce laryngeal/airway trauma. Currently, no evidence indicates that use of a videolaryngoscope reduces the number of intubation attempts or the incidence of hypoxia or respiratory complications, and no evidence indicates that use of a videolaryngoscope affects time required for intubation.
We propose a new dynamical method to connect equilibrium quantum phase transitions and quantum coherence using out-of-time-order correlations (OTOCs). Adopting the iconic Lipkin-Meshkov-Glick and ...transverse-field Ising models as illustrative examples, we show that an abrupt change in coherence and entanglement of the ground state across a quantum phase transition is observable in the spectrum of multiple quantum coherence intensities, which are a special type of OTOC. We also develop a robust protocol to obtain the relevant OTOCs using quasi-adiabatic quenches through the ground state phase diagram. Our scheme allows for the detection of OTOCs without time reversal of coherent dynamics, making it applicable and important for a broad range of current experiments where time reversal cannot be achieved by inverting the sign of the underlying Hamiltonian.
This paper presents results of testing carried out to assess the wear and RCF performance of laser clad rail. Stronger and harder materials can be laser clad on top of the working surfaces of ...standard (e.g. 260 grade) rail in order to improve wear and RCF life. A twin-disc method has been used to assess the suitability of various candidate cladding materials. The materials were clad on top of 260 grade rail discs and were tested against a disc of standard wheel material. Wear was measured by weighing the discs before and after each test. An Ellotest B1 differential eddy current crack detector was used to detect RCF cracks in the rail disc. Four clad materials were used namely, Hadfield, Stellite 6, Maraging and 316 Stainless Steels. In the tests carried out, wear was not always reduced with the cladding. It is assumed that these tests were not long enough for some of the materials to fully work harden and therefore wear rates would improve with a greater number of test cycles. However, all but the Stainless Steel showed that they did not deform under the cyclic loading applied and would offer a greatly enhanced RCF life.
•Tests have been done to determine the suitability laser cladding for repairing rails.•The wear and RCF performance of various candidate clads was measured.•All cladding materials show satisfactory levels of traction under wet conditions.•Stellite 6 shows wear rates in line with non-clad 260 grade rail.•Most claddings show RCF resistance at least 3 times greater than 260 grade rail.
Global-scale nitrogen budgets developed to quantify anthropogenic impacts on the nitrogen cycle do not explicitly consider nitrate stored in the vadose zone. Here we show that the vadose zone is an ...important store of nitrate that should be considered in future budgets for effective policymaking. Using estimates of groundwater depth and nitrate leaching for 1900-2000, we quantify the peak global storage of nitrate in the vadose zone as 605-1814 Teragrams (Tg). Estimates of nitrate storage are validated using basin-scale and national-scale estimates and observed groundwater nitrate data. Nitrate storage per unit area is greatest in North America, China and Europe where there are thick vadose zones and extensive historical agriculture. In these areas, long travel times in the vadose zone may delay the impact of changes in agricultural practices on groundwater quality. We argue that in these areas use of conventional nitrogen budget approaches is inappropriate.
The Nadir and Occultation for MArs Discovery (NOMAD) instrument on board ExoMars Trace Gas Orbiter measured a large increase in water vapor at altitudes in the range of 40–100 km during the 2018 ...global dust storm on Mars. Using a three-dimensional general circulation model, we examine the mechanism responsible for the enhancement of water vapor in the upper atmosphere. Experiments with different prescribed vertical profiles of dust show that when more dust is present higher in the atmosphere, the temperature increases, and the amount of water ascending over the tropics is not limited by saturation until reaching heights of 70–100 km. The warmer temperatures allow more water to ascend to the mesosphere. Photochemical simulations show a strong increase in high-altitude atomic hydrogen following the high-altitude water vapor increase by a few days.
Store-operated calcium channels (SOCs) are a nearly ubiquitous Ca(2+) entry pathway stimulated by numerous cell surface receptors via the reduction of Ca(2+) concentration in the ER. The discovery of ...STIM proteins as ER Ca(2+) sensors and Orai proteins as structural components of the Ca(2+) release-activated Ca(2+) (CRAC) channel, a prototypic SOC, opened the floodgates for exploring the molecular mechanism of this pathway and its functions. This review focuses on recent advances made possible by the use of STIM and Orai as molecular tools. I will describe our current understanding of the store-operated Ca(2+) entry mechanism and its emerging roles in physiology and disease, areas of uncertainty in which further progress is needed, and recent findings that are opening new directions for research in this rapidly growing field.
Ca(2+) entry into cells of the peripheral immune system occurs through highly Ca(2+)-selective channels known as CRAC (calcium release-activated calcium) channels. CRAC channels are a very ...well-characterized example of store-operated Ca(2+) channels, so designated because they open when the endoplasmic reticulum (ER) Ca(2+) store becomes depleted. Physiologically, Ca(2+) is released from the ER lumen into the cytoplasm when activated receptors couple to phospholipase C and trigger production of the second messenger inositol 1,4,5-trisphosphate (IP(3)). IP(3) binds to IP(3) receptors in the ER membrane and activates Ca(2+) release. The proteins STIM and ORAI were discovered through limited and genome-wide RNAi screens, respectively, performed in Drosophila cells and focused on identifying modulators of store-operated Ca(2+) entry. STIM1 and STIM2 sense the depletion of ER Ca(2+) stores, whereas ORAI1 is a pore subunit of the CRAC channel. In this review, we discuss selected aspects of Ca(2+) signaling in cells of the immune system, focusing on the roles of STIM and ORAI proteins in store-operated Ca(2+) entry.
Some railway managers and practitioners fear that introducing premium rail materials will have a detrimental effect on the wheels of trains that use the line. A review of relevant investigations ...across all scales in the laboratory, and in the field has been carried out. This showed that, as rail hardness increases, its wear, and overall system wear reduces. Wheel wear does increase with increasing rail hardness, but only for wheels running on rails that are softer than them. Similar trends were observed in all studies, so it seems that the fears were unfounded.
While the wear trends appear well characterised some issues have been identified. One relates to the varying work hardening capability of wheel and rail materials. Often only bulk hardness is quoted, but work hardening can increase material surface hardness by up to 2.5 times and make materials that were initially softer, harder than the opposing material. Another related issue is test length. It is essential that enough cycles are applied such that the materials reach steady state wear, i.e., the point at which work hardening has reached its limit. In previous work it is not always clear that steady state wear has been reached. Some gaps have been identified in the current knowledge base, the largest of which is the failure to determine which mechanisms lead to the wear trends seen.
Analysis of recent work on different clad layers on rail discs and premium rail materials allowed some of these gaps to be addressed. Results indicated that opposing wheel material hardened to the same level independent of rail hardness. Wheel wear is therefore stress driven under the conditions used, and dictated by the wheel material properties only. At higher slip levels relationships become less clear, but here temperature and therefore hot hardness is most influential and is as yet uncharacterised.
