National Day of Service (NDS) volunteering events have become common, yet little is known about how the design of such events affects volunteer satisfaction. This relationship is important because ...volunteer satisfaction ensures a strong volunteer base for special events and promotes sustained volunteerism. We explore how the design of NDS projects promotes volunteer job satisfaction. Our approach to the research question is informed by work design theory. Based on interview, participant observation, and focus group data from an NDS in the Netherlands, the findings suggest that nonprofit organizations can elicit volunteer job satisfaction by designing NDS projects that create a sense of added value, support productivity, and make volunteers feel comfortable. Designing NDS projects that incorporate task significance, symbolic social support, feedback from others, beneficiary contact, task identity, project preparation, physically demanding work, social support, and limited autonomy help to achieve these goals.

...-methyladenosine, we show that other modifications, in particular pseudouridine (Ψ) and 2'-O-methylation (Nm), also result in characteristic base-calling 'error' signatures in the nanopore data. Focusing on Ψ modification sites, we detected known and uncovered previously unreported Ψ sites in mRNAs, non-coding RNAs and rRNAs, including a Pus4-dependent Ψ modification in yeast mitochondrial rRNA. To explore the dynamics of pseudouridylation, we treated yeast cells with oxidative, cold and heat stresses and detected heat-sensitive Ψ-modified sites in small nuclear RNAs, small nucleolar RNAs and mRNAs. Finally, we developed a software, nanoRMS, that estimates per-site modification stoichiometries by identifying single-molecule reads with altered current intensity and trace profiles. This work demonstrates that Nm and Ψ RNA modifications can be detected in cellular RNAs and that their modification stoichiometry can be quantified by nanopore sequencing of native RNA.
Due to anthropogenic emissions and changes in land use, trees are now exposed to atmospheric levels of CO₂ that are unprecedented for 650,000 y Lüthi et al. (2008) Nature 453:379–382 (thousands of ...tree generations). Trees are expected to acclimate by modulating leaf–gas exchanges and alter water use efficiency which may result in forest productivity changes. Here, we present evidence of one of the strongest, nonlinear, and unequivocal postindustrial increases in intrinsic water use efficiency (iWUE) ever documented (+59%). A dual-isotope tree-ring analysis (δ
13C and δ
18O) covering 715 y of growth of North America’s oldest boreal trees (Thuja occidentalis L.) revealed an unprecedented increase in iWUE that was directly linked to elevated assimilation rates of CO₂ (A). However, limited nutrient availability, changes in carbon allocation strategies, and changes in stomatal density may have offset stem growth benefits awarded by the increased iWUE. Our results demonstrate that even in scenarios where a positive CO₂ fertilization effect is observed, other mechanisms may prevent trees from assimilating and storing supplementary anthropogenic emissions as above-ground biomass. In such cases, the sink capacity of forests in response to changing atmospheric conditions might be overestimated.
Obesity during pregnancy has a long-term effect on the health of the offspring including risk of developing the metabolic syndrome. Using a mouse model of maternal diet-induced obesity, we employed a ...genome-wide approach to investigate the microRNA (miRNA) and miRNA transcription profile in adipose tissue to understand mechanisms through which this occurs. Male offspring of diet-induced obese mothers, fed a control diet from weaning, showed no differences in body weight or adiposity at 8 weeks of age. However, offspring from the obese dams had up-regulated cytokine (Tnfα; P < .05) and chemokine (Ccl2 and Ccl7; P < .05) signaling in their adipose tissue. This was accompanied by reduced expression of miR-706, which we showed can directly regulate translation of the inflammatory proteins IL-33 (41% up-regulated; P < .05) and calcium/calmodulin-dependent protein kinase 1D (30% up-regulated; P < .01). We conclude that exposure to obesity during development primes an inflammatory environment in adipose tissue that is independent of offspring adiposity. Programming of adipose tissue miRNAs that regulate expression of inflammatory signaling molecules may be a contributing mechanism.

There is substantial evidence supporting a key role for chemoattractants in ...directed migration, however, less is known about how cell–cell contacts affect the migratory behavior of leukocytes in innate immunity. Here, we explore how cell–cell contacts can affect the directed migration of innate immune cells, including their role in attracting, repelling, or stopping cell motility. Further investigation of cell contact dynamics as guidance cues may yield new insights into the regulation of innate immunity.

Stress granules are macromolecular aggregates of mRNA and proteins assembling in response to stresses that promote the repression of protein synthesis. Most of the work characterizing ...stress granules has been done under acute stress conditions or during viral infection. Comparatively less work has been done to understand stress granule assembly during chronic stress, specifically regarding the composition and function of stress granules in this alternative context. Here, we describe key aspects of stress granule biology under acute stress, and how these stress granule hallmarks differ in the context of chronic stress conditions. We will provide perspective for future work aimed at further uncovering the form and function of both acute and chronic stress granules and discuss aspects of stress granule biology that have the potential to be exploited in human disease.
Individualized current-flow models are needed for precise targeting of brain structures using transcranial electrical or magnetic stimulation (TES/TMS). The same is true for current-source ...reconstruction in electroencephalography and magnetoencephalography (EEG/MEG). The first step in generating such models is to obtain an accurate segmentation of individual head anatomy, including not only brain but also cerebrospinal fluid (CSF), skull and soft tissues, with a field of view (FOV) that covers the whole head. Currently available automated segmentation tools only provide results for brain tissues, have a limited FOV, and do not guarantee continuity and smoothness of tissues, which is crucially important for accurate current-flow estimates. Here we present a tool that addresses these needs. It is based on a rigorous Bayesian inference framework that combines image intensity model, anatomical prior (atlas) and morphological constraints using Markov random fields (MRF). The method is evaluated on 20 simulated and 8 real head volumes acquired with magnetic resonance imaging (MRI) at 1 mm3 resolution. We find improved surface smoothness and continuity as compared to the segmentation algorithms currently implemented in Statistical Parametric Mapping (SPM). With this tool, accurate and morphologically correct modeling of the whole-head anatomy for individual subjects may now be feasible on a routine basis. Code and data are fully integrated into SPM software tool and are made publicly available. In addition, a review on the MRI segmentation using atlas and the MRF over the last 20 years is also provided, with the general mathematical framework clearly derived.
Analysis of the annual cycle of intensity, extent, and width of the Hadley circulation across a 31-yr period (1979–2009) from all existent reanalyses reveals a good agreement among the datasets. All ...datasets show that intensity is at a maximum in the winter hemisphere and at a minimum in the summer hemisphere. Maximum and minimum values of meridional extent are reached in the respective autumn and spring hemispheres. While considering the horizontal momentum balance, where a weakening of the Hadley cell (HC) is expected in association with a widening, it is shown here that there is no direct relationship between intensity and extent on a monthly time scale.
All reanalyses show an expansion in both hemispheres, most pronounced and statistically significant during summer and autumn at an average rate of expansion of 0.55° decade−1in each hemisphere. In contrast, intensity trends are inconsistent among the datasets, although there is a tendency toward intensification, particularly in winter and spring.
Correlations between the HC and tropical and extratropical large-scale modes of variability suggest interactions where the extent of the HC is influenced by El Niño–Southern Oscillation (ENSO) and the annular modes. The cells tend to shrink (expand) during the warm (cold) phase of ENSO and during the low (high) phase of the annular modes. Intensity appears to be influenced only by ENSO and only during spring for the southern cell and during winter for the northern cell.

The authors searched the PubMed, Scielo, Web of Science, Science Direct, EMBASA, EBSCO, and Scopus databases for articles containing the keywords “multisystem inflammatory syndrome in children” or “MIS-C” or “PIMS-TS” or “SIMP” and “COVID-19” or “SARS-CoV-2” published between December 1st, 2019 and July 10th, 2021. Patient characteristics, tissue and organ comorbidities, the incidence of symptoms after COVID-19 infection, treatment, and patient evolution in the articles found were evaluated. The data were abstracted following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines and Newcastle-Ottawa Scale (NOS).
In total, 98 articles (2275 patients) were selected for demographics, clinical treatment, and outcomes of patients diagnosed with MIS-C. The average age of children with MIS-C, 56.8% of whom were male, was of nine years. Fever (100%), gastrointestinal (GI) (82%), and abdominal pain (68%) were the decisive symptoms for the diagnosis of MIS-C. Shock and/or hypotension were common in patients with MIS-C. Cardiac symptoms (66%) predominated over respiratory (39%) and neurological (28%) symptoms. MIS-C treatment followed the common guidelines for treating children with septic shock and Kawasaki disease (KD) and proved to be effective.
This meta-analysis highlights the main clinical symptoms used for the diagnosis of MIS-C, the differences between MIS-C and KD, and the severity of the inflammatory process and urgency for hospital care.
