Introduction Transcranial direct current stimulation (tDCS) has been shown to modulate cortical excitability and to improve motor learning (Nitsche and Paulus, 2000; Reis et al., 2009; Fritsch et ...al., 2010) . To date there is no data available on the safety aspects of tDCS covering a broad range of stimulation intensities, thus limitations for dosages applied to humans are still based empirically on other types of electrical brain stimulation (Agnew and McCreery, 1987). Objectives The aim of this study was to investigate lesion threshold, accompanying neurodegeneration and possible microstructural changes in glial cells in an animal model of tDCS. Methods The tDCS electrode was placed in a screw tube surgically fixed on the skull above the left primary motor cortex of 47 Male Sprague Dawley rats. Anodal tDCS was applied in different intensities (254.8, 127.4, 63.7, 31.8, 15.9 and 8 A/sqm) or as sham stimulation for 20 min under slight isoflurane anesthesia. For each intensity animals were sacrificed either 24 h (acute effects) or 7 days (chronic effects) after intervention. Histological analyses of neurodegeneration (HE-staining, Fluoro-Jade C), astrocyte (anti-GFAP) and microglial (anti-CD11b OX-42) cell counts/density and/or morphological changes were evaluated. Results Macroscopical lesions could be observed at tDCS intensity of 127 A/sqm and above whereas the lesion dose 50 (LD 50) for microscopical neurodegeneration was determined at 60 A/sqm. Threshold for damage in neuronal and glial cells and change in morphology (activation) was identical. These changes could be detected both in the acute and chronic group. Notably, signs of still ongoing neurodegeneration, though to a slighter extent, were seen 7 days after stimulation. Brain swelling, which was seen underneath the stimulation electrode 24 h following the highest intensity used, was not present one week after stimulation. Conclusions Our results demonstrate that the LD 50 intensity of anodal tDCS for unwanted structural effects is 75 times higher than the intensities applied to humans to date. Additional safety derives from the fact that structures between brain and stimulation electrode, which lead to energy absorbtion, are thicker in humans compared to rats. Cell death, as a trigger for glial reaction, continues even 7 days after the intervention. Based on our results we conclude that higher tDCS intensities in human studies can be applied without risking structural damage.
We present time-resolved conductance measurements and charge spectra for the conduction-band states of InAs quantum dots after creating metastable holes by illumination. We demonstrate an electrical ...way of measuring the conduction-band energy offset and inverse tunnel rates of electrons from a two-dimensional electron gas into neutral (X0), single positively (X1+), and double positively (X2+) charged exciton states. The experiment also gives information about the metastable hole storage time and discharge dynamics.
We investigated using poly(3,4-ethylenedioxythiophene) (PEDOT) to lower the impedance of small, gold recording electrodes with initial impedances outside of the effective recording range. Smaller ...electrode sites enable more densely packed arrays, increasing the number of input and output channels to and from the brain. Moreover, smaller electrode sizes promote smaller probe designs; decreasing the dimensions of the implanted probe has been demonstrated to decrease the inherent immune response, a known contributor to the failure of long-term implants. As expected, chronically implanted control electrodes were unable to record well-isolated unit activity, primarily as a result of a dramatically increased noise floor. Conversely, electrodes coated with PEDOT consistently recorded high-quality neural activity, and exhibited a much lower noise floor than controls. These results demonstrate that PEDOT coatings enable electrode designs 15 µm in diameter.
We study the conductance of a quantum wire in the presence of weak electron-electron scattering. In a sufficiently long wire the scattering leads to full equilibration of the electron distribution ...function in the frame moving with the electric current. At nonzero temperature this equilibrium distribution differs from the one supplied by the leads. As a result the contact resistance increases, and the quantized conductance of the wire acquires a quadratic in temperature correction. The magnitude of the correction is found by analysis of the conservation laws of the system and does not depend on the details of the interaction mechanism responsible for equilibration.
We have observed room temperature (RT) electrical spin injection in an InAs quantum dot (QD) light emitting diode (LED) grown on a p-type GaAs substrate from a ferromagnetic Fe/Tb electrode with ...strong out-of-plane anisotropy in remanence, i.e. without applied magnetic field. The QDs in the LED emit at 1275nm (ground state luminescence), which is beyond the range for highly sensitive detectors, and therefore not optimum for various applications, e.g. quantum information studies. We will present two different ways to blue-shift the emission wavelength and discuss the advantages and drawbacks of the experiments.
Zusammenfassung
Die Durchführung von Verfahren der assistierten Reproduktion zur Behandlung subfertiler Paare nimmt weltweit seit einigen Jahrzehnten deutlich zu. Mit dem Heranwachsen der Nachkommen ...gewinnen auch deren allgemeine und gesundheitliche Entwicklung sowie deren Fertilität an Bedeutung. Bisher liegen nur begrenzte Daten zum Vergleich der Pubertätsentwicklung und des reproduktiv-endokrinen Profils bei Jugendlichen vor, die nach einer Therapie mittels intrazytoplasmatischer Spermieninjektion (ICSI) oder nach Spontankonzeption geboren wurden. Diese Daten konnten aktuell durch eine in Deutschland durchgeführte prospektive, kontrollierte Nachbeobachtungsstudie bei Jugendlichen im Alter von 14 bis 18 Jahren erweitert werden (ICSI:

= 274; Spontankonzeption:
n
= 273). Sowohl für die Mädchen als auch für die Jungen zeigte sich dabei eine vergleichbare und altersentsprechende Pubertätsentwicklung anhand der Tanner-Stadien und des Menarchealters. In linearen Regressionsanalysen zeigte sich auch eine unauffällige weibliche reproduktiv-endokrine Situation, jedoch bei den männlichen Jugendlichen eine Tendenz zu niedrigeren Inhibin-B-Serumspiegeln sowie signifikant höhere Östradiolspiegel und ein signifikant niedrigerer Testosteron-Östradiol-Quotient. Diese Daten stehen im Einklang mit anderen aktuell publizierten Studienergebnissen. Auch wenn die Daten eine Einschränkung der endokrinen Hodenfunktion bei den nach ICSI-Therapie konzipierten Jugendlichen andeuten, ist eine Beurteilung der reproduktiven Funktion gegenwärtig noch nicht möglich.
Zusammenfassung
Zur gesundheitlichen Entwicklung von Kindern, die nach einer Kinderwunschbehandlung konzipiert und geboren wurden, gibt es immer mehr Daten. Daten zu Schwangerschaftsverlauf, ...neonatalem Outcome und frühkindlicher Entwicklung werden nun durch Informationen zur Pubertätsentwicklung ergänzt. Ein nächster Meilenstein sind erste Daten zu kardiometabolischen Risikoprofilen. Es ist möglich, dass durch epigenetische Einflüsse kardiometabolische Parameter verändert werden. Diese Einflüsse können durch In-vitro-Techniken forciert werden. Gegenstand dieser Übersichtsarbeit ist die Deutsche ICSI-Follow-up-Studie, die seit Anfang der 2000er-Jahre eine bereits in der Schwangerschaft rekrutierte Kohorte von nach intrazytoplasmatischer Spermieninjektion (ICSI) konzipierten Kindern erfasst. Im mittleren Alter von etwa 16 Jahren wurden 275 Jugendliche nach ICSI und 273 Kontrollen untersucht. Zur Erfassung der kardiometabolischen Entwicklung wurden eine körperliche Untersuchung der Kinder sowie eine Analyse von Laborwerten des Fett- und Glukose-Insulin-Stoffwechsels bei den Kindern und Müttern durchgeführt. Zudem erfolgte eine strukturierte Erfassung von Daten zur psychisch-physischen Entwicklung der Kinder, Mütter und Väter. Unterschiede in den erhobenen Messwerten wurden nach der Adjustierung für elterliche Faktoren in den meisten Fällen relativiert oder verschwanden komplett. Im Gegensatz zu anderen Kohorten wurde ein erhöhter Blutdruck bei nach ICSI geborenen Kindern in dieser Studie nicht nachgewiesen. Insgesamt findet sich kein Hinweis auf relevante kardiometabolische Probleme der nach ICSI geborenen Kinder. Da sich die Beobachtungen in diesem Alter naturgemäß auf Surrogatparameter stützen, sind für eine bessere Bewertung weitere Studien in höherem Lebensalter notwendig.
