Our objective was to establish a diabetes mellitus type 2 (DM2) model in rats using a high-fat diet and streptozotocin (HF-STZ). Male Wistar rats (240-250g) were divided into a control group ...(commercial feed), and HF-STZ group, (66.5%-commercial feed, 13.5%-lard, and 20%-sugar). STZ (40mg/kg i.p.) or vehicle was administered on the 13th day. An oral glucose tolerance test (OGTT) was performed (2.5mg of glucose/kg v.o.) on both groups. After 39 days of treatment, blood and tissue samples were collected for analyses. The weight gain after STZ administration was lower in the HF-STZ group than in the control group with reductions in muscle mass and adipose tissue. The HF-STZ group showed hyperglycemia after STZ administration (glucose on day 39: HF-STZ: 499 ± 60; control: 134 ± 9mg/dL). Serum glucagon was 23% lower, and insulin levels were unaltered. The HOMA index was 4-times higher in the HF-STZ. The HF-STZ group showed increased post-prandial (330%) and fasting (125%) triglycerides, and while glycogen content in the liver and muscles decreased (70-80%). The area under the curve (OGTT) was 282% higher in the HF-STZ group. The combination of high-fat diet with STZ (i.p) generated rats with hyperglycemia associated with hypertriglyceridemia and introduced many other alterations present in human DM2.
PM2.5 severely affects human health. Remotely sensed (RS) data can be used to estimate PM2.5 concentrations and population exposure, and therefore to explain acute respiratory disorders. However, ...available global PM2.5 concentration forecast products derived from models assimilating RS data have not yet been exploited to generate early alerts for respiratory problems in Brazil. We investigated the feasibility of building such an early warning system. For this, PM2.5 concentrations on a 4-day horizon forecast were provided by the Copernicus Atmosphere Monitoring Service (CAMS) and compared with the number of severe acute respiratory disease (SARD) cases. Confounding effects of the meteorological conditions were considered by selecting the best linear regression models in terms of Akaike Information Criterion (AIC), with meteorological features and their two-way interactions as explanatory variables and PM2.5 concentrations and SARD cases, taken separately, as response variables. Pearson and Spearman correlation coefficients were then computed between the residuals of the models for PM2.5 concentration and SARD cases. The results show a clear tendency to positive correlations between PM2.5 and SARD in all regions of Brazil but the South one, with Spearman’s correlation coefficient reaching 0.52 (p < 0.01). Positive significant correlations were also found in the South region by previously correcting the effects of viral infections on the SARD case dynamics. The possibility of using CAMS global PM2.5 concentration forecast products to build an early warning system for pollution-related effects on human health in Brazil was therefore established. Further investigations should be performed to determine alert threshold(s) and possibly build combined risk indicators involving other risk factors for human respiratory diseases. This is of particular interest in Brazil, where the COVID-19 pandemic and biomass burning are occurring concomitantly, to help minimize the effects of PM emissions and implement mitigation actions within populations.

allele in three long-distance migrant species of shorebird and one non-migrant. Significant differences among species were found in the number and morphological complexity of the astrocytes, as well as in the size of the microsatellites of the
gene. We found significant associations between the size of the
microsatellites, the migratory distances, and the degree of morphological complexity of the astrocytes. We suggest that associations between astrocyte number and morphological complexity,
microsatellite size, and migratory behavior may be part of the adaptive response to the migratory process of shorebirds.

, that migrates from northern Canada and Alaska to South America. Before the transatlantic non-stop long-distance component of their flight, the birds make a stopover at the Bay of Fundy in Canada. To test our hypothesis, we estimated total numbers and compared the three-dimensional (3-D) morphological features of adult
astrocytes captured in the Bay of Fundy (
= 249 cells) with those from birds captured in the coastal region of Bragança, Brazil, during the wintering period (
= 250 cells). Optical fractionator was used to estimate the number of astrocytes and for 3-D reconstructions we used hierarchical cluster analysis. Both morphological phenotypes showed reduced morphological complexity after the long-distance non-stop flight, but the reduction in complexity was much greater in Type I than in Type II astrocytes. Coherently, we also found a significant reduction in the total number of astrocytes after the transatlantic flight. Taken together these findings suggest that the long-distance non-stop flight altered significantly the astrocytes population and that morphologically distinct astrocytes may play different physiological roles during migration.

transcripts were significantly increased in samples from APL patients compared with those of healthy donors (all

< 0.05). However, this had no effect on clinical outcomes.
STMN1
expression was associated with proliferation- and metabolism-related gene signatures in APL. Our data confirmed that STMN1 was highly expressed in early hematopoietic progenitors and reduced during cell differentiation, including the ATRA-induced granulocytic differentiation model. STMN1 phosphorylation was predominant in a pool of mitosis-enriched APL cells. In NB4 and NB4-R2 cells, STMN1 knockdown decreased autonomous cell growth (all

Finally, treatment with paclitaxel (as a single agent or combined with ATRA) induced microtubule stabilization, resulting in mitotic catastrophe with repercussions for cell viability, even in ATRA-resistant APL cells. This study provides new insights into the STMN1 functions and microtubule dynamics in APL.

S. frugiperda
damage, followed by BRS 4103. In addition, higher levels of mono, homo-, or sesquiterpenes, together with green leaf volatiles (GLVs) were the most attractive blend for
T. remus;

The aim of this study was to evaluate thermogenesis in the interscapular brown adipose tissue (IBAT) of rats submitted to low-protein, high-carbohydrate (LPHC) diet and the involvement of adrenergic ...stimulation in this process. Male rats (~100 g) were submitted to LPHC (6 %-protein; 74 %-carbohydrate) or control (C; 17 %-protein; 63 %-carbohydrate) isocaloric diets for 15 days. The IBAT temperature was evaluated in the rats before and after the administration of noradrenaline (NA) (20 µg 100 g b w⁻¹ min⁻¹). The expression levels of uncoupling protein 1 (UCP1) and other proteins involved in the regulation of UCP1 expression were determined by Western blot (Student’s t test, P ≤ 0.05). The LPHC diet promoted a 1.1 °C increase in the basal temperature of IBAT when compared with the basal temperature in the IBAT of the C group. NA administration promoted a 0.3 °C increase in basal temperature in the IBAT of the C rats and a 0.5 °C increase in the IBAT of the LPHC group. The level of UCP1 increased 60 % in the IBAT of LPHC-fed rats, and among the proteins involved in its expression, such as β3-AR and α1-AR, there was a 40 % increase in the levels of p38-MAPK and a 30 % decrease in CREB when compared to the C rats. The higher sympathetic flux to IBAT, which is a consequence of the administration of the LPHC diet to rats, activates thermogenesis and increases the expression of UCP1 in the tissue. Our results suggest that the increase in UCP1 content may occur via p38 MAPK and ATF2.
ABSTRACT Articles on the applicability of briquetting to the production of mushrooms are rare. Therefore, this research provides the scientific community and, mainly, producers, unpublished technical ...information regarding the productive yield of oyster mushroom cultivated in sugarcane bagasse briquettes supplemented with bioproducts of cereals. In 30 days of axenic cultivation, 6 tons-force briquettes with 80% humidity resulted in higher productivity (30.4%), and generated the lowest physical volume of exhausted compound (44.8%), thus, overcoming the technical performance of the other briquetting matrices tested, and even of methods traditionally used for the preparation of substrate for P. ostreatus cultivation on a commercial scale. The conclusion is, therefore, that the axenic cultivation of P. ostreatus var. Florida in sugarcane bagasse briquettes, supplemented with bioproducts of cereals, has technically qualified as an original and efficient method for the production of high biological value protein by the recycling of agricultural residues and can therefore guarantee multiple economic benefits to the agribusiness of the oyster mushroom, in addition to allowing reduction of environmental impacts by the restricted generation of exhausted compound in the post-harvest stage.
Protein-peptide interactions play a fundamental role in a wide variety of biological processes, such as cell signaling, regulatory networks, immune responses, and enzyme inhibition. Peptides are ...characterized by low toxicity and small interface areas; therefore, they are good targets for therapeutic strategies, rational drug planning and protein inhibition. Approximately 10% of the ethical pharmaceutical market is protein/peptide-based. Furthermore, it is estimated that 40% of protein interactions are mediated by peptides. Despite the fast increase in the volume of biological data, particularly on sequences and structures, there remains a lack of broad and comprehensive protein-peptide databases and tools that allow the retrieval, characterization and understanding of protein-peptide recognition and consequently support peptide design.
We introduce Propedia, a comprehensive and up-to-date database with a web interface that permits clustering, searching and visualizing of protein-peptide complexes according to varied criteria. Propedia comprises over 19,000 high-resolution structures from the Protein Data Bank including structural and sequence information from protein-peptide complexes. The main advantage of Propedia over other peptide databases is that it allows a more comprehensive analysis of similarity and redundancy. It was constructed based on a hybrid clustering algorithm that compares and groups peptides by sequences, interface structures and binding sites. Propedia is available through a graphical, user-friendly and functional interface where users can retrieve, and analyze complexes and download each search data set. We performed case studies and verified that the utility of Propedia scores to rank promissing interacting peptides. In a study involving predicting peptides to inhibit SARS-CoV-2 main protease, we showed that Propedia scores related to similarity between different peptide complexes with SARS-CoV-2 main protease are in agreement with molecular dynamics free energy calculation.
Propedia is a database and tool to support structure-based rational design of peptides for special purposes. Protein-peptide interactions can be useful to predict, classifying and scoring complexes or for designing new molecules as well.
