•Beef cattle enteric methane is reasonably well predicted using dietary forage content, body weight and dry matter intake.•Separate beef cattle enteric methane models for forage contents ≤ 18% and ≥ ...25% data contribute to improved prediction.•Depending on the predictor variables used, global region-specific beef cattle enteric methane models may perform the best.•Predicting enteric methane from energy conversion factors requires adequate forage content-based and region-specific values.
Enteric methane (CH4) production attributable to beef cattle contributes to global greenhouse gas emissions. Reliably estimating this contribution requires extensive CH4 emission data from beef cattle under different management conditions worldwide. The objectives were to: 1) predict CH4 production (g d−1 animal−1), yield g (kg dry matter intake; DMI)−1 and intensity g (kg average daily gain)−1 using an intercontinental database (data from Europe, North America, Brazil, Australia and South Korea); 2) assess the impact of geographic region, and of higher- and lower-forage diets. Linear models were developed by incrementally adding covariates. A K-fold cross-validation indicated that a CH4 production equation using only DMI that was fitted to all available data had a root mean square prediction error (RMSPE; % of observed mean) of 31.2%. Subsets containing data with ≥25% and ≤18% dietary forage contents had an RMSPE of 30.8 and 34.2%, with the all-data CH4 production equation, whereas these errors decreased to 29.3 and 28.4%, respectively, when using CH4 prediction equations fitted to these subsets. The RMSPE of the ≥25% forage subset further decreased to 24.7% when using multiple regression. Europe- and North America-specific subsets predicted by the best performing ≥25% forage multiple regression equation had RMSPE of 24.5 and 20.4%, whereas these errors were 24.5 and 20.0% with region-specific equations, respectively. The developed equations had less RMSPE than extant equations evaluated for all data (22.5 vs. 23.2%), for higher-forage (21.2 vs. 23.1%), but not for the lower-forage subsets (28.4 vs. 27.9%). Splitting the dataset by forage content did not improve CH4 yield or intensity predictions. Predicting beef cattle CH4 production using energy conversion factors, as applied by the Intergovernmental Panel on Climate Change, indicated that adequate forage content-based and region-specific energy conversion factors improve prediction accuracy and are preferred in national or global inventories.
DNA lesions encountered by replicative polymerases threaten genome stability and cell cycle progression. Here we report the identification of mutations in TRAIP, encoding an E3 RING ubiquitin ligase, ...in patients with microcephalic primordial dwarfism. We establish that TRAIP relocalizes to sites of DNA damage, where it is required for optimal phosphorylation of H2AX and RPA2 during S-phase in response to ultraviolet (UV) irradiation, as well as fork progression through UV-induced DNA lesions. TRAIP is necessary for efficient cell cycle progression and mutations in TRAIP therefore limit cellular proliferation, providing a potential mechanism for microcephaly and dwarfism phenotypes. Human genetics thus identifies TRAIP as a component of the DNA damage response to replication-blocking DNA lesions.

Transformations des rites funéraires, notamment sous l’effet de la ...déchristianisation de l’Occident, diffusion d’un pluralisme religieux, pratiques individuelles en sont les traits les plus marquants. Mais c’est davantage l’acte de mourir, voire la définition même de la mort, qui semblent aujourd’hui changer radicalement. Cette mutation, profonde, n’est pourtant pas un fait nouveau. Elle n’est que le prolongement d’une autre transformation, plus ancienne, qui a touché l’ensemble du monde occidental dès le XVIIIe siècle, où l’on a commencé à déplacer et éloigner les sépultures à la périphérie des villes. La « transition » – si ce n’est la rupture – qui s’est alors produite en seulement quelques générations est venue bouleverser un régime funéraire qui s’était imposé depuis la fin de l’Antiquité, articulant des espaces destinés aux morts aux lieux sacrés, et faisant cohabiter les vivants et les défunts. Cet ouvrage vient analyser la manière dont s’est mis en place cet effacement progressif à travers les siècles, en tentant de dresser une histoire longue des rapports entre les vivants et les morts.
Bloom syndrome, caused by biallelic mutations in BLM, is characterized by prenatal-onset growth deficiency, short stature, an erythematous photosensitive malar rash, and increased cancer ...predisposition. Diagnostically, a hallmark feature is the presence of increased sister chromatid exchanges (SCEs) on cytogenetic testing. Here, we describe biallelic mutations in TOP3A in ten individuals with prenatal-onset growth restriction and microcephaly. TOP3A encodes topoisomerase III alpha (TopIIIα), which binds to BLM as part of the BTRR complex, and promotes dissolution of double Holliday junctions arising during homologous recombination. We also identify a homozygous truncating variant in RMI1, which encodes another component of the BTRR complex, in two individuals with microcephalic dwarfism. The TOP3A mutations substantially reduce cellular levels of TopIIIα, and consequently subjects’ cells demonstrate elevated rates of SCE. Unresolved DNA recombination and/or replication intermediates persist into mitosis, leading to chromosome segregation defects and genome instability that most likely explain the growth restriction seen in these subjects and in Bloom syndrome. Clinical features of mitochondrial dysfunction are evident in several individuals with biallelic TOP3A mutations, consistent with the recently reported additional function of TopIIIα in mitochondrial DNA decatenation. In summary, our findings establish TOP3A mutations as an additional cause of prenatal-onset short stature with increased cytogenetic SCEs and implicate the decatenation activity of the BTRR complex in their pathogenesis.
The European Space Agency’s INTErnational Gamma-Ray Astrophysics Laboratory (ESA/INTEGRAL) was launched aboard a Proton-DM2 rocket on 17 October 2002 at 06:41 CEST, from Baikonur in Kazakhstan. Since ...then, INTEGRAL has been providing long, uninterrupted observations (up to about 47h, or 170ksec, per satellite orbit of 2.7 days) with a large field-of-view (FOV, fully coded: 100 deg2), millisecond time resolution, keV energy resolution, polarization measurements, as well as additional wavelength coverage at optical wavelengths. This is realized by two main instruments in the 15keV to 10MeV energy range, the spectrometer SPI (spectral resolution 3keV at 1.8MeV) and the imager IBIS (angular resolution: 12arcmin FWHM), complemented by X-ray (JEM-X; 3–35keV) and optical (OMC; Johnson V-band) monitor instruments. All instruments are co-aligned to simultaneously observe the target region. A particle radiation monitor (IREM) measures charged particle fluxes near the spacecraft. The Anti-coincidence subsystems of the main instruments, built to reduce the background, are also very efficient all-sky γ-ray detectors, which provide virtually omni-directional monitoring above ∼75keV. Besides the long, scheduled observations, INTEGRAL can rapidly (within a couple of hours) re-point and conduct Target of Opportunity (ToO) observations on a large variety of sources.
INTEGRAL observations and their scientific results have been building an impressive legacy: The discovery of currently more than 600 new high-energy sources; the first-ever direct detection of 56Ni and 56Co radio-active decay lines from a Type Ia supernova; spectroscopy of isotopes from galactic nucleo-synthesis sources; new insights on enigmatic positron annihilation in the Galactic bulge and disk; and pioneering gamma-ray polarization studies. INTEGRAL is also a successful actor in the new multi-messenger astronomy introduced by non-electromagnetic signals from gravitational waves and from neutrinos: INTEGRAL found the first prompt electromagnetic radiation in coincidence with a binary neutron star merger.
Up to now more than 1750 scientific papers based on INTEGRAL data have been published in refereed journals. In this paper, we will give a comprehensive update of the satellite status after more than 18 years of operations in a harsh space environment, and an account of the successful Ground Segment.
Abstract The immune system is constantly exposed to dying cells, most of which arise during central tolerance and from effete circulating immune cells. Under homeostatic conditions, phagocytes ...(predominantly macrophages and dendritic cells) belonging to the innate immune system, rapidly ingest cells and their debris. Apoptotic cell removal requires recognition of altered self on the apoptotic membrane, a process which is facilitated by natural antibodies and serum opsonins. Recognition, may be site and context specific. Uptake and ingestion of apoptotic cells promotes an immunosuppressive environment that avoids inflammatory responses to self-antigens. However, it does not preclude a T cell response and it is likely that constant exposure to self-antigen, particularly by immature dendritic cells, leads to T cell tolerance. Tolerance occurs by several different mechanisms including anergy and deletion (for CD8+T cells) and induction of T regulatory cells (for CD4+T cells). Failed apoptotic cell clearance promotes immune responses to self-antigens, especially when the cellular contents are leaked from the cell (necrosis). Inflammatory responses may be induced by nucleic acid stimulation of Toll like receptors and other immune sensors, specific intracellular proteins and non-protein (uric acid) stimulation of inflammasomes.
