The break‐up of the Soviet Union in 1991 triggered cropland abandonment on a continental scale, which in turn led to carbon accumulation on abandoned land across Eurasia. Previous studies have ...estimated carbon accumulation rates across Russia based on large‐scale modelling. Studies that assess carbon sequestration on abandoned land based on robust field sampling are rare. We investigated soil organic carbon (SOC) stocks using a randomized sampling design along a climatic gradient from forest steppe to Sub‐Taiga in Western Siberia (Tyumen Province). In total, SOC contents were sampled on 470 plots across different soil and land‐use types. The effect of land use on changes in SOC stock was evaluated, and carbon sequestration rates were calculated for different age stages of abandoned cropland. While land‐use type had an effect on carbon accumulation in the topsoil (0–5 cm), no independent land‐use effects were found for deeper SOC stocks. Topsoil carbon stocks of grasslands and forests were significantly higher than those of soils managed for crops and under abandoned cropland. SOC increased significantly with time since abandonment. The average carbon sequestration rate for soils of abandoned cropland was 0.66 Mg C ha−1 yr−1 (1–20 years old, 0–5 cm soil depth), which is at the lower end of published estimates for Russia and Siberia. There was a tendency towards SOC saturation on abandoned land as sequestration rates were much higher for recently abandoned (1–10 years old, 1.04 Mg C ha−1 yr−1) compared to earlier abandoned crop fields (11–20 years old, 0.26 Mg C ha−1 yr−1). Our study confirms the global significance of abandoned cropland in Russia for carbon sequestration. Our findings also suggest that robust regional surveys based on a large number of samples advance model‐based continent‐wide SOC prediction.

We present here a versatile resource for plant biologists comprising a set of cloning vectors and 96 standardized parts to enable Golden Gate construction of multigene constructs for plant transformation. Parts include promoters, untranslated sequences, reporters, antigenic tags, localization signals, selectable markers, and terminators. The comparative performance of parts in the model plant Nicotiana benthamiana is discussed.
The role of tracheostomy in coronavirus disease 2019 (COVID-19) is unclear, with several consensus guidelines advising against this practice. We developed both a dedicated airway team and coordinated ...education programme to facilitate ward management of tracheostomised COVID-19 patients. Here, we report outcomes in the first 100 COVID-19 patients who underwent tracheostomy at our institution.
This was a prospective observational cohort study of patients confirmed to have COVID-19 who required mechanical ventilation at Queen Elizabeth Hospital, Birmingham, UK. The primary outcome measure was 30-day survival, accounting for severe organ dysfunction (Acute Physiology and Chronic Health APACHE-II score>17). Secondary outcomes included duration of ventilation, ICU stay, and healthcare workers directly involved in tracheostomy care acquiring COVID-19.
A total of 164 patients with COVID-19 were admitted to the ICU between March 9, 2020 and April 21, 2020. A total of 100 patients (mean standard deviation age: 55 12 yr; 29% female) underwent tracheostomy; 64 (age: 57 14 yr; 25% female) did not undergo tracheostomy. Despite similar APACHE-II scores, 30-day survival was higher in 85/100 (85%) patients after tracheostomy, compared with 27/64 (42%) non-tracheostomised patients {relative risk: 3.9 (95% confidence intervals CI: 2.3–6.4); P<0.0001}. In patients with APACHE-II scores ≥17, 68/100 (68%) tracheotomised patients survived, compared with 12/64 (19%) non-tracheotomised patients (P<0.001). Tracheostomy within 14 days of intubation was associated with shorter duration of ventilation (mean difference: 6.0 days 95% CI: 3.1–9.0; P<0.0001) and ICU stay (mean difference: 6.7 days 95% CI: 3.7–9.6; P<0.0001). No healthcare workers developed COVID-19.
Independent of the severity of critical illness from COVID-19, 30-day survival was higher and ICU stay shorter in patients receiving tracheostomy. Early tracheostomy appears to be safe in COVID-19.
Pathogenic Xanthomonas bacteria cause disease on more than 400 plant species. These Gram-negative bacteria utilize the type III secretion system to inject type III effector proteins (T3Es) directly ...into the plant cell cytosol where they can manipulate plant pathways to promote virulence. The host range of a given Xanthomonas species is limited, and T3E repertoires are specialized during interactions with specific plant species. Some effectors, however, are retained across most strains, such as Xanthomonas Outer Protein L (XopL). As an 'ancestral' effector, XopL contributes to the virulence of multiple xanthomonads, infecting diverse plant species. XopL homologs harbor a combination of a leucine-rich-repeat (LRR) domain and an XL-box which has E3 ligase activity. Despite similar domain structure there is evidence to suggest that XopL function has diverged, exemplified by the finding that XopLs expressed in plants often display bacterial species-dependent differences in their sub-cellular localization and plant cell death reactions. We found that XopL from X. euvesicatoria (XopLXe) directly associates with plant microtubules (MTs) and causes strong cell death in agroinfection assays in N. benthamiana. Localization of XopLXe homologs from three additional Xanthomonas species, of diverse infection strategy and plant host, revealed that the distantly related X. campestris pv. campestris harbors a XopL (XopLXcc) that fails to localize to MTs and to cause plant cell death. Comparative sequence analyses of MT-binding XopLs and XopLXcc identified a proline-rich-region (PRR)/α-helical region important for MT localization. Functional analyses of XopLXe truncations and amino acid exchanges within the PRR suggest that MT-localized XopL activity is required for plant cell death reactions. This study exemplifies how the study of a T3E within the context of a genus rather than a single species can shed light on how effector localization is linked to biochemical activity.
The relative abundance of specific fatty acids in milk can be important for consumer health and manufacturing properties of dairy products. Understanding of genes controlling milk fat synthesis may ...contribute to the development of dairy products with high quality and nutritional value. This study aims to identify key genes and genetic variants affecting de novo synthesis of the short- and medium-chained fatty acids C4:0 to C14:0. A genome-wide association study using 609,361 SNP markers and 1,811 animals was performed to detect genomic regions affecting fatty acid levels. These regions were further refined using sequencing data to impute millions of additional genetic variants. Results suggest associations of PAEP with the content of C4:0, AACS with the content of fatty acids C4:0-C6:0, NCOA6 or ACSS2 with the longer chain fatty acids C6:0-C14:0, and FASN mainly associated with content of C14:0. None of the top-ranking markers caused amino acid shifts but were mostly situated in putatively regulating regions and suggested a regulatory role of the QTLs. Sequencing mRNA from bovine milk confirmed the expression of all candidate genes which, combined with knowledge of their roles in fat biosynthesis, supports their potential role in de novo synthesis of bovine milk fatty acids.
The increasing interest in early identification of people at risk of developing dementia, has led to the development of numerous models aimed at estimating the likelihood of progression from mild ...cognitive impairment (MCI) to dementia. It is important to study both the need for and possible outcomes related with such prediction models, including the impact of risk predictions on perceived quality of life (QoL).
This study aimed to quantify the impact that receiving a risk prediction on progression from MCI to dementia has on QoL.
A Discrete Choice Experiment (DCE) and Time Trade Off (TTO) study were performed. Participants completed choice tasks related to dementia prognosis while imagining having MCI. We collected DCE data by an online survey, and TTO data via videoconferencing interviews. DCE data were analyzed using a mixed multinomial logit model and were anchored to a health state utility scale using mean observed TTO valuations.
296 people participated in the DCE and 42 in the TTO. Moderate and high predicted dementia risks were associated with decrements in utility (-0.05 and -0.18 respectively), compared to no prognostic information. Low predicted risk was associated with an increase in utility (0.06), as well as the availability of medication or lifestyle interventions (0.05 and 0.13 respectively).
This study shows a significant impact of dementia risk predictions on QoL and highlights the importance of caution when sharing information about expected MCI disease courses.
In the following paper, we present an industry perspective of inertial sensors for navigation purposes driven by applications and customer needs. Microelectromechanical system (MEMS) inertial sensors ...have revolutionized consumer, automotive, and industrial applications and they have started to fulfill the high end tactical grade performance requirements of hybrid navigation systems on a series production scale. The Fiber Optic Gyroscope (FOG) technology, on the other hand, is further pushed into the near navigation grade performance region and beyond. Each technology has its special pros and cons making it more or less suitable for specific applications. In our overview paper, we present latest improvements at NG LITEF in tactical and navigation grade MEMS accelerometers, MEMS gyroscopes, and Fiber Optic Gyroscopes, based on our long-term experience in the field. We demonstrate how accelerometer performance has improved by switching from wet etching to deep reactive ion etching (DRIE) technology. For MEMS gyroscopes, we show that better than 1°/h series production devices are within reach, and for FOGs we present how limitations in noise performance were overcome by signal processing. The paper also intends a comparison of the different technologies, emphasizing suitability for different navigation applications, thus providing guidance to system engineers.
Synthetic aperture radar (SAR) is more sensitive to the dielectric properties and structure of the targets and less affected by weather conditions than optical sensors, making it more capable of ...detecting changes induced by management practices in agricultural fields. In this study, the capability of C-band SAR data for detecting crop seeding and harvest events was explored. The study was conducted for the 2019 growing season in Temiskaming Shores, an agricultural area in Northern Ontario, Canada. Time-series SAR data acquired by Sentinel-1 constellation with the interferometric wide (IW) mode with dual polarizations in VV (vertical transmit and vertical receive) and VH (vertical transmit and horizontal receive) were obtained. interferometric SAR (InSAR) processing was conducted to derive coherence between each pair of SAR images acquired consecutively in time throughout the year. Crop seeding and harvest dates were determined by analyzing the time-series InSAR coherence and SAR backscattering. Variation of SAR backscattering coefficients, particularly the VH polarization, revealed seasonal crop growth patterns. The change in InSAR coherence can be linked to change of surface structure induced by seeding or harvest operations. Using a set of physically based rules, a simple algorithm was developed to determine crop seeding and harvest dates, with an accuracy of 85% (n = 67) for seeding-date identification and 56% (n = 77) for harvest-date identification. The extra challenge in harvest detection could be attributed to the impacts of weather conditions, such as rain and its effects on soil moisture and crop dielectric properties during the harvest season. Other factors such as post-harvest residue removal and field ploughing could also complicate the identification of harvest event. Overall, given its mechanism to acquire images with InSAR capability at 12-day revisiting cycle with a single satellite for most part of the Earth, the Sentinel-1 constellation provides a great data source for detecting crop field management activities through coherent or incoherent change detection techniques. It is anticipated that this method could perform even better at a shorter six-day revisiting cycle with both satellites for Sentinel-1. With the successful launch (2019) of the Canadian RADARSAT Constellation Mission (RCM) with its tri-satellite system and four polarizations, we are likely to see improved system reliability and monitoring efficiency.

Teleosts exhibit extensive diversity of sex determination (SD) systems and mechanisms, providing the opportunity to study the evolution of SD and sex chromosomes. Here we sequenced the genome of the ...common lumpfish (Cyclopterus lumpus Linnaeus), a species of increasing importance to aquaculture, and identified the SD region and master SD locus using a 70 K single nucleotide polymorphism array and tissue‐specific expression data. The chromosome‐level assembly identified 25 diploid chromosomes with a total size of 572.89 Mb, a scaffold N50 of 23.86 Mb and genome annotation‐predicted 21,480 protein‐coding genes. Genome‐wide association analysis located a highly sex‐associated region on chromosome 13, suggesting that anti‐Müllerian hormone (AMH) is the putative SD factor. Linkage disequilibrium and heterozygosity across chromosome 13 support a proto‐XX/XY system, with an absence of widespread chromosome divergence between sexes. We identified three copies of AMH in the lumpfish primary and alternate haplotype assemblies localized in the SD region. Comparison to sequences from other teleosts suggested a monophyletic relationship and conservation within the Cottioidei. One AMH copy showed similarity to AMH/AMHY in a related species and was also the only copy with expression in testis tissue, suggesting this copy may be the functional copy of AMH in lumpfish. The two other copies arranged in tandem inverted duplication were highly similar, suggesting a recent duplication event. This study provides a resource for the study of early sex chromosome evolution and novel genomic resources that benefits lumpfish conservation management and aquaculture.
