A web-based registry collected retrospective data on patients with TNFRSF1A sequence variants and inflammatory symptoms. Participating hospitals included paediatric rheumatology centres and adult centres with a specific interest in autoinflammatory diseases. Cases were independently validated by experts in the disease.
Complete information on 158 validated patients was available. The most common TNFRSF1A variant was R92Q (34% of cases), followed by T50M (10%). Cysteine residues were disrupted in 27% of cases, accounting for 39% of sequence variants. A family history was present in 19% of patients with R92Q and 64% of those with other variants. The median age at which symptoms began was 4.3 years but 9.1% of patients presented after 30 years of age. Attacks were recurrent in 88% and the commonest features associated with the pathogenic variants were fever (88%), limb pain (85%), abdominal pain (74%), rash (63%) and eye manifestations (45%). Disease associated with R92Q presented slightly later at a median of 5.7 years with significantly less rash or eye signs and more headaches. Children were more likely than adults to present with lymphadenopathy, periorbital oedema and abdominal pains. AA amyloidosis has developed in 16 (10%) patients at a median age of 43 years.
In this, the largest reported case series to date, the genetic heterogeneity of TRAPS is accompanied by a variable phenotype at presentation. Patients had a median 70 symptomatic days a year, with fever, limb and abdominal pain and rash the commonest symptoms. Overall, there is little evidence of a significant effect of age or genotype on disease features at presentation.
Methane is an important greenhouse gas and energy resource generated dominantly by methanogens at low temperatures and through the breakdown of organic molecules at high temperatures. However, ...methane-formation temperatures in nature are often poorly constrained. We measured formation temperatures of thermogenic and biogenic methane using a "clumped isotope" technique. Thermogenic gases yield formation temperatures between 157° and 221°C, within the nominal gas window, and biogenic gases yield formation temperatures consistent with their comparatively lower-temperature formational environments (<50°C). In systems where gases have migrated and other proxies for gas-generation temperature yield ambiguous results, methane clumped-isotope temperatures distinguish among and allow for independent tests of possible gas-formation models.
This article provides an overview of the latest research developments on binary and ternary oxide coatings that have the potential to be used as solid lubricants at elevated temperatures. The review ...focuses on understanding the major mechanisms that lead to a reduction in friction and/or wear in high temperature lubricious oxides. Changes in the structural, chemical, and electronic properties of these oxides as a function of temperature will be correlated to their mechanical and tribological performance using a range of experimental tools in addition to modeling based on ab initio calculations and molecular dynamics simulation methods. This review also includes a discussion of the industrial applications of these coatings as well as of potential improvements to the coating design and other anticipated future developments.

Thermo-oxidation of biomass is an important process that occurs through a variety of reaction pathways depending on the chemical nature of the molecules and reaction conditions. These processes can ...be modeled using reactive molecular dynamics to study chemical reactions and the evolution of converted molecules over time. The advantage of this approach is that many molecules can be modeled, but it is challenging to use the large amount of data obtained from such a simulation to determine reaction products and pathways. In this study, we developed a tracking approach to identify the reaction pathways of the dominant reaction products from reactive molecular dynamics simulations. We demonstrated the approach for thermo-oxidation reactions of modified model lignin compounds. For two modified lignin structures, we tracked the evolving chemical species to find the most common reaction products. Subsequently, we monitored specific bonds to determine the individual steps in the reaction process. This combined approach of reactive molecular dynamics and tracking enabled us to identify the most likely thermo-oxidation pathways. Accumulating data support that vitamin D possesses several biological and molecular actions apart from its role in calcium homeostasis. Immune cells express vitamin D receptor and are capable of ...metabolizing vitamin D. Within this context, experimental studies show that vitamin D modulates immune and inflammatory responses. Epidemiologic evidence linking poor vitamin D status to autoimmune diseases, type 2 diabetes, and cardiovascular disease suggests that insufficient vitamin D may be involved in the etiology of such disorders. Given the impact of immune and inflammatory abnormalities in the development of chronic diseases, including autoimmune disorders, it is possible that vitamin D might reduce chronic disease risk by modulating the immune system.
A hybrid approach for the synthesis of planar thinned antenna arrays is presented. The proposed solution exploits and combines the most attractive features of a particle swarm algorithm and those of ...a combinatorial method based on the noncyclic difference sets of Hadamard type. Numerical experiments validate the proposed solution, showing improvements with respect to previous results.

A web-based registry retrospectively collected data on patients with CAPS. Experts in the disease independently validated all cases. Patients carrying NLRP3 variants and germline-mutation-negative patients were included.
136 patients were analysed. The median age at disease onset was 9 months, and the median duration of follow-up was 15 years. Skin rash, musculoskeletal involvement and fever were the most prevalent features. Neurological involvement (including severe complications) was noted in 40% and 12% of the patients, respectively, with ophthalmological involvement in 71%, and neurosensory hearing loss in 42%. 133 patients carried a heterozygous, germline mutation, and 3 patients were mutation-negative (despite complete NLRP3 gene screening). Thirty-one different NLRP3 mutations were recorded; 7 accounted for 78% of the patients, whereas 24 rare variants were found in 27 cases. The latter were significantly associated with early disease onset, neurological complications (including severe complications) and severe musculoskeletal involvement. The T348M variant was associated with early disease onset, chronic course and hearing loss. Neurological involvement was less strongly associated with V198M, E311 K and A439 V alleles. Early onset was predictive of severe neurological complications and hearing loss.
Patients carrying rare NLRP3 variants are at risk of severe CAPS; onset before the age of 6 months is associated with more severe neurological involvement and hearing loss. These findings may have an impact on treatment decisions.

–SCR) and for the low-temperature ...selective oxidation of methane to methanol (MTM). Accessing a quantitative understanding of Cu–speciation in this material is a key step to unveil structure–performance relationships for these high-impact processes. Herein, we select Cu–CHA as a case study to demonstrate the potential of chemometric approaches, such as multivariate curve resolution (MCR) applied in combination with principal component analysis (PCA). We employ these methods to assist the interpretation of X-ray absorption spectroscopy (XAS) experiments in the near-edge (XANES) region, determining the spectroscopic signatures and concentration profiles of the pure Cu–species formed. We pinpoint the composition impact on the material reducibility and highlight Cu–speciation–productivity relationships for the MTM process. Furthermore, we report novel insights on the formation of O
2
-derived species in Cu–CHA, obtained from MCR analysis of high energy resolution fluorescence detected (HERFD) XANES data collected during thermal treatment of Cu–CHA in both He and O
2
gas flow. Multivariate analysis, in combination with the superior energy resolution adopted, allows us to identify an additional Cu(II) species. This component, different from the previously characterized ZCu(II)OH moiety, is only formed at significant concentrations in O
2
and it is envisaged to play an important role in the MTM conversion.
To develop evidence based points to consider the use of imaging in the diagnosis and management of juvenile idiopathic arthritis (JIA) in clinical practice. The task force comprised a group of ...paediatric rheumatologists, rheumatologists experienced in imaging, radiologists, methodologists and patients from nine countries. Eleven questions on imaging in JIA were generated using a process of discussion and consensus. Research evidence was searched systematically for each question using MEDLINE, EMBASE and Cochrane CENTRAL. Imaging modalities included were conventional radiography, ultrasound, MRI, CT, scintigraphy and positron emission tomography. The experts used the evidence obtained from the relevant studies to develop a set of points to consider. The level of agreement with each point to consider was assessed using a numerical rating scale. A total of 13 277 references were identified from the search process, from which 204 studies were included in the systematic review. Nine points to consider were produced, taking into account the heterogeneity of JIA, the lack of normative data and consequent difficulty identifying pathology. These encompassed the role of imaging in making a diagnosis of JIA, detecting and monitoring inflammation and damage, predicting outcome and response to treatment, use of guided therapies, progression and remission. Level of agreement for each proposition varied according to the research evidence and expert opinion. Nine points to consider and a related research agenda for the role of imaging in the management of JIA were developed using published evidence and expert opinion.
