This effect was attributed to nuclear translocation of the transcription factor NFE2L2 resulting in upregulation of several antioxidative enzymes. In vivo, defective VSMC autophagy led to upregulation of MMP9, TGFB and CXCL12 and promoted postinjury neointima formation and diet-induced atherogenesis. Lesions of VSMC-specific atg7 knockout mice were characterized by increased total collagen deposition, nuclear hypertrophy, CDKN2A upregulation, RB hypophosphorylation, and GLB1 activity, all features typical of cellular senescence. To conclude, autophagy is crucial for VSMC function, phenotype, and survival. Defective autophagy in VSMCs accelerates senescence and promotes ligation-induced neointima formation and diet-induced atherogenesis, implying that autophagy inhibition as therapeutic strategy in the treatment of neointimal stenosis and atherosclerosis would be unfavorable. Conversely, stimulation of autophagy could be a valuable new strategy in the treatment of arterial disease.
O Brasil apresenta uma população de 213 milhões de habitantes, com uma composição étnica oriunda de indígenas, africanos e europeus. A investigação da distribuição dos alelos dos antígenos de ...neutrófilos humanos (HNA) em regiões representativas torna-se uma importante ferramenta para se avaliar os riscos de aloimunização em uma população tão miscigenada.
Investigar a distribuição dos alelos HNA em doadores de sangue das regiões norte, nordeste e sudeste do Brasil, que apresentam uma ancestralidade indígena, negroide e caucasiana, respectivamente.
Trata-se de um estudo transversal, envolvendo amostras de doadores de sangue do norte do país (Pará, 50 amostras); nordeste (Alagoas, 50 amostras); e sudeste (São Paulo, 300 amostras). A genotipagem dos sistemas HNA-1 e HNA-4; HNA-3 e HNA-5 foi realizada através de PCR-SSP e PCR-RFLP respectivamente.
Entre os sistemas analisados os que apresentaram uma diferença estatística significativa entre as regiões estudadas foram HNA-1 e HNA-5. Os alelos FCGR3B*01 e *02 apresentaram uma distribuição inversa na região norte (0,554 e 0,376) quando comparado com as regiões sudeste e nordeste do país (0,315 e 0,637; e 0,336 e 0,572, respectivamente. p < 0,0001 em ambas as comparações). A presença do alelo FCGR3B*03, mais frequente em negroides, foi marcante na região nordeste do país (0,090 vs. 0,048 (p = 0,0002) e 0,069 (p = 0,0459) nas regiões sudeste e norte, respectivamente. O alelo FCGR3B*null foi identificado apenas na região norte do país (0,029). Observou-se uma grande diferença na frequência alélica do sistema HNA-5 nas regiões sudeste e nordeste: ITGAL*01 (0,711 e 0,397, respectivamente. p < 0,0001) e ITGAL*02 (0,289 e 0,602, respectivamente. p < 0,0001). Os sistemas HNA - 3 e 4 não apresentaram diferença significativa entre as regiões brasileiras.
A proporção dos alelos FCGR3B*01 e *02 na região norte se assemelha àquela descrita em populações indígenas (Brasil e Argentina) e países asiáticos, evidenciando a influência indígena na região. A maior frequência do alelo FCGR3B*03 na região nordeste é comparável à descrita em africanos. A presença do alelo FCGR3B*null apenas na região norte assemelha-se à descrita em tailandeses. Os resultados da região sudeste se aproximam aos observados em caucasianos. A distribuição dos alelos do sistema HNA-5 estão invertidas em comparação às populações estudadas. Pode se assemelhar à população africana, entretanto não há dados disponíveis na literatura.
Low density sugarcane plantation (LDSP) has been implemented by some sugarcane producers in Brazil, aiming to save seeds and operational costs. The study was carried out in the municipality of Areia, ...Paraíba, Brazil. Five planting densities were used, varying from 5 to 25 m-2 of buds arranged in randomized blocks, with four replications. Data were measured annually over three cultivation cycles (2017 to 2020), during which the field was fertilized with NPK and the harvests were carried out manually without prior burning. The lower planting density presents higher productivity only in the cane plant (101.03 t ha-1) due to the higher plant height (2.37 m) and the higher number of stalks (11 stalks m-2), suggesting that these variables are due to the greater availability of light, water and photosynthate. However, there is a drastic reduction in sugarcane yield for this lower population in the 2nd ratoon by up to 65.62%, which is correlated with number of stalks per meter. We demonstrate the agronomic viability of LDSP in the population of 10 buds m-2 in relation to conventional planting of sugarcane until the 2nd ratoon. Data are important for future studies to present additional considerations for other production factors, such as the effects of mechanized harvesting and the management of nutrients and water, assessing the sustainability of this large-scale planting system.
Descrito pela primeira vez em 1950, o sistema de grupo sanguíneo Duffy é caracterizado por uma glicoproteína (ACKR1) expressa na membrana celular dos eritrócitos, bem como em outros tecidos. ...Indivíduos que possuem o polimorfismo rs2814778 no gene ACKR1 apresentam um fenótipo Fya e Fyb negativo (Duffy-null), frequentemente relacionado a neutropenia étnica benigna, embora este mecanismo ainda não esteja completamente esclarecido. Por esse motivo, a avaliação deste fenótipo deve fazer parte do roteiro de investigação dos pacientes com neutropenia, sobretudo naqueles em que há suspeita de neutropenia étnica benigna.
Correlacionar a fenotipagem por técnica imuno-hematológica dos antígenos Fya e Fyb, a genotipagem para o sistema Duffy e a presença do polimorfismo rs2814778 em pacientes neutropênicos, de modo a definir se apenas a fenotipagem é suficiente para a determinação do fenótipo Duffy-null, em pacientes portadores de neutropenia.
Trata-se de um estudo transversal incluindo 41 pacientes, 28 adultos e 13 pediátricos atendidos no período de dois anos no Ambulatório de Neutropenias da Disciplina de Hematologia e Hemoterapia da Universidade Federal de São Paulo (UNIFESP). As amostras foram submetidas a fenotipagem dos antígenos Fya e Fyb, genotipagem do gene ACRK1 e a pesquisa do polimorfismo rs2814778.
Do total de pacientes analisados, 31,70% apresentavam idade < 18 anos (pediátricos) e 68,29% ≥ 18 anos (adultos). Dentre o grupo de pacientes pediátricos, a prevalência do fenótipo Duffy-null foi de 61,53%, enquanto no grupo de pacientes adultos foi de 82,14%. Considerando o total de pacientes, a prevalência do fenótipo foi de 75,60%. Quando comparados os testes imuno-hematológicos e moleculares, 97,46% apresentaram resultados concordantes e apenas um paciente (2,54%) apresentou resultado discrepante, demonstrando alta correlação entre as técnicas (coeficiente κ = 0,935). Considerando estes dados, pode-se observar que a fenotipagem para o sistema Duffy apresentou sensibilidade de 100%, especificidade de 90% e acurácia de 97%.
A pesquisa do fenótipo Duffy-null é um passo essencial na investigação de pacientes portadores de neutropenia. Embora a fenotipagem do sistema Duffy por métodos imuno-hematológicos seja amplamente disponível na maior parte dos serviços, a genotipagem apresenta custo mais elevado e pode não ser acessível. A alta concordância entre as técnicas analisadas demonstrou segurança na realização apenas dos testes imuno-hematológico para determinação do fenótipo Fya e Fyb, sem que seja necessária a realização de análise genética, rotineiramente, na população neutropênica.
A concordância obtida entre os testes realizados foi alta, demonstrando que a fenotipagem do sistema Duffy aparenta ser um teste sensível e acurado para a determinação do fenótipo Duffy-null nos pacientes portadores de neutropenia.

Numerous studies have investigated dietary approaches to prevent chronic lifestyle-related diseases, including hypertension. Spent brewer’s yeast is the second largest byproduct originated by the brewing industry and it deserves considerable attention because of its high nutritional value, ca. 40% of its dry mass is rich in protein which can be hydrolyzed into biologically active peptides. To upgrade this byproduct, the aim of this study was initially in vitro assessment of biological properties, e.g. ACE inhibition and antioxidant activity, and then, the in vivo effect in short-term oral antihypertensive effect of hydrolyzed yeast fractions on a well characterized model to study hypertension - Spontaneously Hypertensive Rats (SHR). Here, it was demonstrated that the fraction with molecular weight below 3 kDa containing tri and tetra- peptides with hydrophobic amino acid residues – SPQW, PWW and RYW, causes the most noticeable decrease in systolic, diastolic and mean blood pressure of SHR and shows highest antioxidant effect. These properties highlight the potential use of yeast extract as nutraceutical or functional food ingredient for the management and treatment of hypertension with antioxidant effect.
Introdução: O Transplante de medula óssea é um dos pilares no tratamento de algumas doenças onco-hematológicas. Embora haja muitos estudos sobre essa temática, faltam dados epidemiológicos e ...segurança em perfis diversos. O objetivo desse estudo é quantificar e relatar o perfil e eficácia de pacientes submetidos a (TMOs) ressaltando a patologia prevalescente e o índice de suporte hemoterápico por um serviço de hematologia e hemoterapia na cidade do Rio de Janeiro. No presente trabalho foi avaliado o perfil dos pacientes das diversas instituições no qual realizamos o suporte de atendimento, quantidade de (TMOs) realizados, se autólogos e/ou alogênicos, perfil epidemiológico, necessidade de suporte transfusional, taxa de altas e óbitos e doença de maior prevalência no período de 12 meses. Materiais e métodos: Estudo retrospectivo em pacientes portadores de doenças onco- hematológicas a transplante autólogo ou alogênico de medula óssea envolvendo as unidades que realizam este procedimento, no período de 01/06/2022 a 30/06/2023. Para entender melhor o perfil, os dados foram obtidos em planilhas próprias e em software de gerenciamento das unidades envolvidas. Resultados: Foram avaliados 61 pacientes com mediana de idade de 54 anos (variando entre 5–75 anos), submetidos a coleta de células progenitoras hematopoiéticas periféricas através de coleta por aférese, sendo 100% dos casos em maquina Spectra Optia. O sexo dominante foi o masculino 37 (60,70%), o maior público foram os portadores do tipo sanguíneo O positivo 30 (49,18%), A positivo 15 (24,59%), B positivo 7 (11,48%), A Negativo 4 (6,56%), O negativo 3 (4,92%), B negativo 1 (1,64%) e AB positivo 1 (1,64%). A patologia prevalente foi Mieloma Múltiplo 34 (55,74%), Linfomas 16 (26,23%), Leucemias 7 (11,48%) e outras doenças onco-hematológicas como Neuroblastoma e Amiloidose 4 (6,56%). A análise demonstrou significância estatística para o grau de resposta ao tratamento aplicado previamente ao transplante autólogo de medula óssea 52 (85,25%). Os TMOs alogênicos sendo 9 (14,75%) em sua totalidade, 89% foram alogênicos aparentados e somente 11%, realizados através de doador não aparentado. Em todos os pacientes, a mobilização foi realizada com uso de granulokine e em 24 destes houve a necessidade também da utilização do Mozobil (39,34%) para obtenção de uma resposta mais eficiente do produto. Foram observados que a média transfusional foi de 7,8 hemocomponentes por paciente, onde o hemocomponente predominante foi concentrado de plaqueta por aférese 266 (60,71%), seguido de concentrado de hemácia 127 (28,95%) e concentrado de plaqueta randomizada 45 (10,74%). Observou-se que no TMO (alogênico) demandou um consumo de 40,18% enquanto no TMO (autólogo) foi consumido 59,82% dos hemocomponentes. A sobrevida percebida em um ano de análise foi de Alta 59 (96,72%) e somente 2(3,38%) foram a óbito. Destes pacientes que evoluíram ao óbito foram submetidos a TMOs por métodos distintos sendo um autólogo (sem comorbidades) e um alogênico, onde o paciente (possuía comorbidades DM e HAS) e realizou TMO haploidentico (50% compatível). Conclusão: De acordo com os resultados obtidos, foi possível observar que o transplante de medula óssea é uma estratégia terapêutica eficaz no alcance de um bom prognóstico junto as doenças onco-hematológicas supracitadas, fato este evidenciado devido ao alto índice de alta hospitalar percebida no período analisado.
Ultrasound standing waves can be used to separate emulsions. So far, they have been applied to oil-in-water emulsions with low continuous phase viscosity. This technique has the potential to be used ...for novel applications such as separating lipids from algal biomass; however, this requires the methodology to be optimized to process viscous emulsions. We have addressed this issue by studying the effects of bulk phase viscosity (1–23 mPa·s), emulsion droplet size (4.5–20 μm), power (10–54 W/L), and frequency (1 and 2 MHz) of ultrasound on the separation efficiency of model mineral oil-in-water–glycerol-mixture emulsions. For the small droplet size (4.5 μm) emulsion in water, the maximum separation achieved increased from 36 to 79% when ultrasound power increased from 10 to 54 W/L. However, for the large droplet size (11 μm) emulsion, the maximum separation was greater than 95% and was independent of ultrasound power. The maximum separation efficiency for small droplet size (4.5–6 μm) emulsions decreased from 80 to 14% when the viscosity increased from 1 to 23 mPa·s. However, for the large droplet size (11–20 μm) emulsion, the maximum separation efficiency decreased from 98 to 62% when the viscosity of the bulk phase was increased from 1 to 23 mPa·s. The experimental results were then interpreted using analytical and numerical simulations by calculating the time required for the emulsion droplets to migrate to the nearest pressure antinodal plane under the influence of ultrasound standing waves. Further experiments showed that increasing the ultrasound frequency from 1 to 2 MHz increased the maximum separation from 36 to 86% for fine emulsions and water as the continuous phase.

The present study aimed to evaluate the effect of continuous butyrate administration in dairy calves' liquid diet considering diarrhea, metabolic profile, gastrointestinal ...development, and corporal growth. Immediately after birth, calves were randomly allocated into 2 groups of 62 calves (50 females and 12 males), with access to water and a solid feed ad libitum. The Butyrate Group (BG) received 4 g/day of sodium butyrate (Admix® Easy - Adisseo) diluted in the whole milk, and the Control Group (CG) received whole milk with no supplementation. Sodium butyrate was administered from day one of life until the weaning at 90 d. Feces consistency was assessed daily for the first 30 d of life and characterized by scores from 0 to 4 (0 and 1 for normal, and 2, 3, and 4 for abnormal feces). Diarrhea was diagnosed when the animals had abnormal feces and fever. Morbidity, recurrence, mortality, and lethality data were recorded and compared between the groups. Average daily weight gain (ADG) and corporal growth (body weight, thoracic perimeter, height at the withers, and croup width) were evaluated weekly, from the first day to the 30th, and later at the 45th, 60th, and 90th d of life. Blood samples were taken weekly for up to 30 d to determine the circulating levels of total calcium, phosphorus, chloride, bicarbonate, glucose, β-hydroxybutyrate, and nonesterified fatty acids (NEFA). The males were euthanized at 15 (n = 6 per group) and 30 d (n = 6 per group) for morphometric, histological, and gene expression analysis of the gastrointestinal tract. The results showed that the Butyrate Group had a lower rate of morbidity (BG = 30% vs. CG = 50%) and recurrence (BG = 26.7% vs. CG = 60%) of diarrhea than the Control Group. In addition, the BG had abnormal feces for a shorter period (BG = 4.64 ± 0.47 d vs. CG = 8.6 ± 0.65 d). The ADG tended to be higher in BG than CG up to 30 and 60 d. Metabolic evaluations showed the lowest levels of glucose and highest levels of NEFA in BG. On d 30 of life, rumen papillae length, papilla area, duodenum villus length, and crypt depth were higher in BG than in CG. The duodenal gene expression at 30 d showed that animals with diarrhea episodes that did not receive butyrate had the highest levels of transcripts for the LCT and GLP2 genes. In addition, in different ways, both butyrate and neonatal diarrhea affected the gene expression of IGF1, SLC5A1 and AQP3. These results allow us to conclude that continuous supplementation with sodium butyrate improves gastrointestinal development, reduces the occurrence of diarrhea, and makes clinical conditions milder with faster recovery, favoring a higher ADG in the first 30 and 60 d of life. Based on these results, we conclude that sodium butyrate can be indicated for liquid diet supplementation to accelerate gastrointestinal tract development and prevent severe cases of neonatal diarrhea, tending to improve average daily gain until the weaning.
The observational evidence for the acceleration of the Universe demonstrates that canonical theories of cosmology and particle physics are incomplete, if not incorrect, and that new physics is out ...there, waiting to be discovered. Forthcoming high-resolution ultrastable spectrographs will play a crucial role in this quest for new physics, by enabling a new generation of precision consistency tests. Here we focus on astrophysical tests of the stability of nature's fundamental couplings, and by using principal component analysis techniques further calibrated by existing VLT data we discuss how the improvements that can be expected with ESPRESSO and ELT-HIRES will impact on fundamental cosmology. In particular we show that a 20 to 30 night program on ELT-HIRES will allow it to play a leading role in fundamental cosmology.
We report magnetic and spectroscopic observations and modelling of the Of?p star CPD -28... 2561. Using more than 75 new spectra, we have measured the equivalent width variations and examined the ...dynamic spectra of photospheric and wind-sensitive spectral lines. A period search results in an unambiguous 73.41 d variability period. High-resolution spectropolarimetric data analysed using least-squares deconvolution yield a Zeeman signature detected in the mean Stokes V profile corresponding to phase 0.5 of the spectral ephemeris. Interpreting the 73.41 d period as the stellar rotational period, we have phased the equivalent widths and inferred longitudinal field measurements. The phased magnetic data exhibit a weak sinusoidal variation, with maximum of about 565 G at phase 0.5, and a minimum of about -335 G at phase 0.0, with extrema approximately in phase with the (double-wave) H... equivalent width variation. Modelling of the H... equivalent width variation assuming a quasi-3D magnetospheric model produces a unique solution for the ambiguous couplet of inclination and magnetic obliquity angles: (i, ...) or (..., i) = (35..., 90...). Adopting either geometry, the longitudinal field variation yields a dipole polar intensity Bd = 2.6 ± 0.9 kG, consistent with that obtained from direct modelling of the Stokes V profiles. We derive a wind magnetic confinement parameter ..., leading to an Alfven radius ..., and a Kepler radius ... This supports a physical scenario in which the H... emission and other line variability have their origin in an oblique, corotating 'dynamical magnetosphere' structure resulting from a magnetically channelled wind. Nevertheless, the details of the formation of spectral lines and their variability within this framework remain generally poorly understood. (ProQuest: ... denotes formulae/symbols omitted.)
