The peroxisome proliferator activated receptor-gamma (PPARgamma) protein is a nuclear transcriptional activator with importance in diabetes management as the molecular target for the ...thiazolidinedione (TZD) family of drugs. Substantial evidence indicates that the TZD family of PPARgamma agonists may retard the development of atherosclerosis. However, recent clinical data have suggested that at least one TZD may increase the risk of myocardial infarction and death from cardiovascular disease. In this study, we used a genetic approach to disrupt PPARgamma signaling to probe the protein's role in smooth muscle cell (SMC) responses that are important for atherosclerosis.
SMC isolated from transgenic mice harboring the dominate-negative P465L mutation in PPARgamma (PPARgamma(L/+)) exhibited greater proliferation and migration then did wild-type cells. Upregulation of ETS-1, but not ERK activation, correlated with enhanced proliferative and migratory responses PPARgamma(L/+) SMCs. After arterial injury, PPARgamma(L/+) mice had a approximately 4.3-fold increase in the development of intimal hyperplasia.
These findings are consistent with a normal role for PPARgamma in inhibiting SMC migration and proliferation in the context of restenosis or atherosclerosis.

The influence of seed suspension of Trigonella foenum graceum (commonly known as fenugreek) was investigated for wound healing properties in excision, incision, and dead space wound models in rats. ...The seed suspension of T. foenum graceum 1ml/kg was given either topically or orally once a day. The granulation tissues formed were removed after 4, 8, 12, and 16 days of post excision wounding; and biochemical parameters such as DNA, total protein, total collagen, and hexosamine were estimated. The suspension increased cellular proliferation and collagen synthesis at the wound site, as evidenced by increases in DNA, protein, and collagen contents of the granulation tissues. Quicker and better maturation and cross-linking of collagen was ob-served in the suspension-treated rats, as indicated by the high stability of acid soluble collagen and increase in aldehyde content and tensile strength. The orally treated wound was found to epithelize faster, and the rate of wound contraction was higher, as compared with the control wounds. The results show that T. foenum graceum produced different actions on the various phases of wound repair.
A fluorous linker-assisted synthetic protocol has been developed for preparation of sclerotigenin-type benzodiazepine-quinazolinone library containing 144 analogues. Amide coupling of fluorous ...trimethoxybenzyl (TMB)-protected amino esters with anthranilic acids followed by base-promoted cyclizations afforded 4-benzodiazepine-2,5-diones. Further derivatization of benzodiazepinediones by reacting with azidobenzoyl chlorides, cyclization, and fluorous linker cleavage afforded the desired compound library. The reaction intermediates were purified by fluorous solid-phase extraction (F-SPE) and final products were further purified by prep-HPLC.
In multi input multi output orthogonal frequency division multiplexing systems, a group of low complexity subspace based time domain channel estimation methods are studied. These methods are based on ...parametric channel model, where the response of the channel is considered as a collection of sparse propagation paths. Considering the channel correlation matrix, translate estimation of channel parameters into an unconstrained minimized problem. To solve this problem, subspace tracking based Kalman filter method is proposed, which employs the constant subspace to construct state equation and measurement equation. The Least Mean Square and Recursive Least Square algorithms are applied and evaluated. These methods represent a group of low complexity subspace schemes. The approach can be extended to multi carrier code-division multiple-access systems. The simulation results prove that the Kalman filter method in time domain channel estimation can track faster fading channel, and is more accurate with low complexity.
The effects of fabric orientation on the mechanical behavior of a Nicalon/SiC composite were investigated by performing flexure tests at room temperature in air, and at 1000°C in an argon ...environment. The specimen configurations were designated as edge-on and transverse, depending on whether the load was applied parallel or normal to the fabric plies, respectively. The monotonic and fatigue behavior of the material was not noticeably affected by fabric orientation, at both room and elevated temperatures. The interlaminar pores in the material appeared to have acted as stress concentration sites. The cracks initiated from these sites linked up the interlaminar pores, resulting in specimen severance under monotonic and fatigue loadings, in both edge-on and transverse orientations.

A tetrapeptide derivative PEP1261 {Boc-Lys-(Boc)-Arg-Asp-Ser-(tBu)-OtBu}, corresponding to residues 39-42 of human lactoferrin, was tested for its antiinflammatory action in adjuvant induced ...arthritis in rats. Administration of heat killed Mycobacterium tuberculosis (500 μg/0.1 ml of paraffin oil) intradermally into the foot pad of right hind paw resulted in an increased paw volume and an increase in the levels of reactive oxygen species and β-glucuronidase as well as a decrease in the antioxidants levels. PEP1261, at an effective dose of 10 mg/kg body wt., exhibited a significant antiarthritic activity as evidenced by lowering of paw volume and inhibited the free radicals toxicity by increasing the antioxidants levels. This peptide derivative was also shown to have a membrane stabilizing action by significantly decreasing the total and free activity of β-glucuronidase and inhibiting the rate of release of the enzyme from lysosomal rich fraction. Histopathological studies confirmed the above results by showing a decrease in mononuclear cell infiltration, hypertrophy, hyperplasia and pannus formation after PEP1261 treatment in arthritic rats.PUBLICATION ABSTRACT
