Fructans, which are β-(2,1) and/or β-(2,6) linked polymers of fructose, are important storage carbohydrates in many plants. They are mobilized via fructan exohydrolases (FEHs). The cloning, mapping, ...and functional analysis of the first 1-FEH (EC 3.2.1.153) from Lolium perenne L. var. Bravo is described here. By screening a perennial ryegrass cDNA library, a 1-FEH cDNA named Lp1-FEHa was cloned. The Lp1-FEHa deduced protein has a low iso-electric point (5.22) and it groups together with plant FEHs and cell-wall type invertases. The deduced amino acid sequence shows 75% identity to wheat 1-FEH w2. The Lp1-FEHa gene was mapped at a distal position on the linkage group 3 (LG3). Functional characterization of the recombinant protein in Pichia pastoris demonstrated that it had high FEH activity towards 1-kestotriose, 1,1-kestotetraose, and inulin, but low activity against 6-kestotriose and levan. Like other fructan-plant FEHs, no hydrolase activity could be detected towards sucrose, convincingly demonstrating that the enzyme is not a classic invertase. The expression pattern analysis of Lp1-FEHa revealed transcript accumulation in leaf tissues accumulating fructans while transcript level was low in the photosynthetic tissues. The high expression level of this 1-FEH in conditions of active fructan synthesis, together with its low expression level when fructan contents are low, suggest that it might play a role as a β-(2,1) trimming enzyme acting during fructan synthesis in concert with fructan synthesis enzymes.
Timothy (Phleum pratense L.) is an important forage grass used for pasture, hay and silage in regions with cool and humid growth seasons. Harvesting conditions could reduce its nutritive value, ...particularly with extended wilting periods. To understand how daytime or night‐time wilting influences the nutritive value of timothy, this study investigated the metabolism of non‐structural carbohydrates, including fructan and starch, together with total soluble protein and amino acid patterns in timothy plants harvested at two maturity stages (heading and anthesis) and wilted under controlled conditions for 24 h at two temperatures (15°C, 20°C) and two light regimes (darkness, light) by simulating different wilting management practices. Correlation analysis with the whole dataset showed that soluble protein, glucose and starch contents declined in plant tissues concomitantly with water loss, while amino acid, sucrose and fructose contents increased. Transient increase in amino acid content suggests that the decrease in protein content was due to proteolysis during wilting. Sucrose and fructose contents generally increased in plant tissues harvested at anthesis and wilted in light whereas they were unaffected in plants wilted in darkness. Fructan content remained stable. Fructan exohydrolase (FEH) and soluble acid invertase (INV) activities were well preserved during the first 12 h of wilting and might facilitate the fermentation process at the beginning of ensiling by supplying fructose from fructans and hexoses from sucrose to the fermentive bacteria.
Sainfoin (Onobrychis viciifolia) is a temperate legume that contains condensed tannins (CT), i.e., polyphenols that are able to bind proteins and thus reduce protein degradation in the rumen. Few ...studies have investigated the relation between plant phenology, CT characteristics and nutritive value of sainfoin. In this study we investigated differences among three sainfoin varieties (Esparcette, Ambra and Villahoz) that were of different geographical origin, and which were cultivated at the same site over the course of two growth cycles and compared with a tannin‐free legume, lucerne (Aubigny). Plants were harvested on nine dates in the first and four dates in the second growth cycle. Phenological stages were analysed for leaf‐to‐whole‐plant ratio, nitrogen (N), organic matter digestibility (OMd) by the pepsin‐cellulase method, cell‐wall concentration, N solubility (solN) plus CT concentration, biological activity and structures. Leaf‐to‐whole‐plant, cell‐wall concentration, N of sainfoin and lucerne were closely related to the phenological stage. Although sainfoin developed earlier than lucerne, its pepsin‐cellulase digestibility was higher than or close to lucerne. Esparcette had the lowest OMd, N and solN and the highest CT concentration. CT concentration, proportion of prodelphinidins (PD) and mean degree of polymerization (mDP) increased with the phenological stage (or plant maturity), and these parameters were associated with lower biological activity of CT. Sainfoin varieties differed in their relationships between phenological stages, nutritional parameters and CT characteristics. The results demonstrated that sainfoin can be a valuable alternative forage legume to lucerne.
This work focuses on the study of the physical aging of a selenium glass by differential scanning calorimetry (DSC) and fast scanning calorimetry (FSC). Aged samples of 30 and 40 years old at room ...temperature were analysed by classical DSC, and their enthalpies of recovery were calculated, showing that the glass has reached its thermodynamic equilibrium and is stable over time. The study of accelerated physical aging by using FSC on a rejuvenated sample allows reproducing the way by which this glass has reached its thermodynamic equilibrium at the laboratory time scale. We evidence that the glass reaches its equilibrium state in one or two steps, depending on the gap between the aging temperature and the glass transition temperature, resulting from the difference in the mechanisms governing the relaxation process. Furthermore, a second phenomenon is evidenced, consisting in the crystallization of the glass once the equilibrium has been reached for aging temperatures close to the glass transition temperature.

In recent years, the number of cases of listeriosis has increased worldwide. Ninety-five isolates of Listeria monocytogenes recovered from Portuguese human cases of listeriosis have been ...characterized by biotyping (cadmium and arsenic sensitivity), polymerase chain reaction (PCR) grouping, and by pulsed-field gel electrophoresis (PFGE) applying the enzymes AscI and ApaI. Isolates were classified into one of three PCR groups; IVb (71.6%), IIb (17.9%), and IIa (10.5%). Four biotypes were differentiated: sensitive to arsenic/cadmium (48.4%), arsenic-sensitive and cadmium-resistant (25.3%), resistant to arsenic and sensitive to cadmium (18.9%), and resistant to both heavy metals (7.4%). Combined analyses of AscI and ApaI patterns yielded a total of 58 PFGE types with five sets (G, Jb, KKa, Me, and U) of Portuguese strains, each of which were indistinguishable by PFGE typing. In the present study, it was demonstrated that there are recurrent pulsotypes and that some were the same pulsotypes linked to outbreaks in France. In addition, there are some pulsotypes spread throughout the country, while others only appear in a restricted region. This study allowed the assembly of a first large pulsotype database of Portuguese clinical strains.
When animals are fed a grass and legume mixture, digestive interactions can occur in the rumen between the substrates contained in the different plants, and the response of the animal to the ...combination of forages can differ from the balanced median values of their components considered individually. Our objective was to assess the associative effects between temperate forages in 8 grass-legume binary combinations on in vitro rumen fermentation characteristics to highlight synergies or antagonisms in terms of nutritional and environmental impacts. Two grasses (cocksfoot, CF; ryegrass, RG) and 4 legume species (alfalfa, A; white clover, WC; red clover, RC; sainfoin, SAN) were incubated alone and in grass-legume mixture (1:1, wt/wt) in batches containing buffered rumen fluid during 3.5 and 24 h. For each substrate and each incubation time, parameters describing the degradation of the energetic and nitrogenous compounds and their partition into fermentation end products were measured. Data were subjected to ANOVA using a mixed procedure to test quadratic contrasts. At 3.5 h of incubation, many quadratic effect were observed. The presence of A, WC, or RC in mixtures quadratically increased the NH^sub 3^-N production (up to +28% when compared with the calculated value, P < 0.001), whereas the presence of condensed tannin (CT) activity in SAN considerably decreased it (up to -67%, quadratic effect, P < 0.001) and the N disappearance. To a lesser extent, the presence of SAN reduced the proportion of methane in the gas produced (up to 7%, quadratic effect, P = 0.018). Generally, the degradation of OM and NDF was not stimulated by mixing grass and legume, except for SAN. However, the presence of SAN severely impaired the NDF digestion at the early phase of fermentation. At 24 h of incubation, few associative effects were observed in comparison with those observed at 3.5 h of incubation, but the effect of CT of SAN on N metabolism was still clearly present. This study shows that, among the grass-legume mixtures tested in vitro, only the presence of SAN can interact with the grasses to reduce the degradation of proteins and the production of methane but with a transitory negative effect on fiber digestion. The main storage compounds in Lolium perenne are fructans with prevailing β(2–6) linkages. A cDNA library of L. perenne was screened using Poa secunda sucrose:fructan 6-fructosyltransferase (6-SFT) ...as a probe. A full-length Lp6-SFT clone was isolated as shown by heterologous expression in Pichia pastoris. High levels of Lp6-SFT transcription were found in the growth zone of elongating leaves and in mature leaf sheaths where fructans are synthesized. Upon fructan synthesis induction, Lp6-SFT transcription was high in mature leaf blades but with no concomitant accumulation of fructans. In vitro studies with the recombinant Lp6-SFT protein showed that both 1-kestotriose and 6G-kestotriose acted as fructosyl acceptors, producing 1- and 6-kestotetraose (bifurcose) and 6G,6-kestotetraose, respectively. Interestingly, bifurcose formation ceased and 6G,6-kestotetraose was formed instead, when recombinant fructan:fructan 6G-fructosyltransferase (6G-FFT) of L. perenne was introduced in the enzyme assay with sucrose and 1-kestotriose as substrates. The remarkable absence of bifurcose in L. perenne tissues might be explained by a higher affinity of 6G-FFT, as compared with 6-SFT, for 1-kestotriose, which is the first fructan formed. Surprisingly, recombinant 6-SFT from Hordeum vulgare, a plant devoid of fructans with internal glucosyl residues, also produced 6G,6-kestotetraose from sucrose and 6G-kestotriose. In the presence of recombinant L. perenne 6G-FFT, it produced 6G,6-kestotetraose from 1-kestotriose and sucrose, like L. perenne 6-SFT. Thus, we demonstrate that the two 6-SFTs have close catalytic properties and that the distinct fructans formed in L. perenne and H. vulgare can be explained by the presence of 6G-FFT activity in L. perenne and its absence in H. vulgare.
Schools, as social systems, may knowingly or unintentionally perpetuate inequities through unchallenged oppressive systems. This paper focuses on mathematics as a subject area in school practices in ...which inequities seem to be considered normal. Issues of racism and racialization in the discipline of mathematics are predominantly lived through the practice of streaming where students are enrolled in courses of different levels of difficulty. Such practice denies marginalized groups of students the full benefit of rich learning experiences. These issues should be of concern for activists, advocates, and allies as well as individuals and groups who are systematically and directly affected. The purpose of this paper is to make visible issues of racism and racialization in school mathematics to a range of stakeholders that include: school administrators, teachers, students, parents, education advocates, academics, educational researchers, and politicians. The ultimate goal is that the knowledge gained through this call to action will contribute toward eliminating social injustice in all school systems, particularly as it relates to skin colour, country of origin, culture, language, customs, and religion.
The goal of this work is to provide an answer to the important scientific question: will a glass-forming liquid reach its equilibrium state or not before to crystallize during physical aging process, ...i.e. when it is maintained below its glass transition temperature? For this goal the physical aging process in a 62.5GeS2-12.5Sb2S3-25CsCl glass has been investigated for a long period (5 months) at 30°C below the glass transition temperature Tg. A crystallization was observed during this process, and was characterized through X-Ray Diffraction (XRD), Transmission Electron Microscopy (TEM) and Temperature Modulated Differential Scanning Calorimetry (MT-DSC) analyses. By evaluating the enthalpy loss related to the physical aging and by comparing this experimental quantity to the theoretical one, we have proved that this glass has reached its thermodynamic equilibrium state before crystallizing. Thus, this work evidences experimentally the ability for a glass former to crystallize under Tg after reaching its equilibrium state.
Twenty-five different Staphylococcus aureus strains that are widespread in France were screened by various methods for heterogeneous and low-level resistance to vancomycin. Population analysis on ...brain–heart infusion agar containing 4 mg/L of the drug detected resistant cells at frequencies of 10–7 to 10–6 in five multiply resistant strains. There was no antagonism between vancomycin and β-lactam antibiotics. One of the five strains, isolated in 1993, was considered a putative progenitor of a French nosocomial S. aureus strain isolated in 1998 and for which the vancomycin MIC was 8 mg/L.
