The current investigation of marine water from 30 sites adjacent to stormwater outlets across the entire Sydney estuary is the first such research in Australia. The number of analytes detected were: 8/59 pharmaceutical compounds (codeine, paracetamol, tramadol, venlafaxine, propranolol, fluoxetine, iopromide and carbamazepine), 7/38 of the pesticides (2,4-dichlorophenoxyacetic acid (2,4-D), 3,4-dichloroaniline, carbaryl, diuron, 2-methyl-4-chlorophenoxyacetic acid (MCPA), mecoprop and simazine) and 0/3 of the personal care products (PCPs) analysed. An artificial sweetener (acesulfame) was detected, however none of the nine antibiotics analysed were identified. Sewage water is not discharged to this estuary, except infrequently as overflow during high-precipitation events. The presence of acesulfame (a recognised marker of domestic wastewater) and pharmaceuticals in water from all parts of the estuary after a dry period, suggests sewage water is leaking into the stormwater system in this catchment. The pesticides are applied to the environment and were discharged via stormwater to the estuary.
ABSTRACT
The use of acoustic Doppler current profilers (ADCP) for discharge measurements and three‐dimensional flow mapping has increased rapidly in recent years and has been primarily driven by ...advances in acoustic technology and signal processing. Recent research has developed a variety of methods for processing data obtained from a range of ADCP deployments and this paper builds on this progress by describing new software for processing and visualizing ADCP data collected along transects in rivers or other bodies of water. The new utility, the Velocity Mapping Toolbox (VMT), allows rapid processing (vector rotation, projection, averaging and smoothing), visualization (planform and cross‐section vector and contouring), and analysis of a range of ADCP‐derived datasets. The paper documents the data processing routines in the toolbox and presents a set of diverse examples that demonstrate its capabilities. The toolbox is applicable to the analysis of ADCP data collected in a wide range of aquatic environments and is made available as open‐source code along with this publication. Published 2012. The accuracy of state‐of‐the‐art global barotropic tide models is assessed using bottom pressure data, coastal tide gauges, satellite altimetry, various geodetic data on Antarctic ice shelves, and ...independent tracked satellite orbit perturbations. Tide models under review include empirical, purely hydrodynamic (“forward”), and assimilative dynamical, i.e., constrained by observations. Ten dominant tidal constituents in the diurnal, semidiurnal, and quarter‐diurnal bands are considered. Since the last major model comparison project in 1997, models have improved markedly, especially in shallow‐water regions and also in the deep ocean. The root‐sum‐square differences between tide observations and the best models for eight major constituents are approximately 0.9, 5.0, and 6.5 cm for pelagic, shelf, and coastal conditions, respectively. Large intermodel discrepancies occur in high latitudes, but testing in those regions is impeded by the paucity of high‐quality in situ tide records. Long‐wavelength components of models tested by analyzing satellite laser ranging measurements suggest that several models are comparably accurate for use in precise orbit determination, but analyses of GRACE intersatellite ranging data show that all models are still imperfect on basin and subbasin scales, especially near Antarctica. For the M2 constituent, errors in purely hydrodynamic models are now almost comparable to the 1980‐era Schwiderski empirical solution, indicating marked advancement in dynamical modeling. Assessing model accuracy using tidal currents remains problematic owing to uncertainties in in situ current meter estimates and the inability to isolate the barotropic mode. Velocity tests against both acoustic tomography and current meters do confirm that assimilative models perform better than purely hydrodynamic models.

Mutations in the p53 tumor-suppressor gene are prevalent in human cancers. The majority of p53 mutations are missense, which can be classified into contact mutations (that directly disrupts the ...DNA-binding activity of p53) and structural mutations (that disrupts the conformation of p53). Both of the mutations can disable the normal wild-type (WT) p53 activities. Nevertheless, it has been amply documented that small molecules can rescue activity from mutant p53 by restoring WT tumor-suppressive functions. These compounds hold promise for cancer therapy and have now entered clinical trials. In this study, we show that cruciferous-vegetable-derived phenethyl isothiocyanate (PEITC) can reactivate p53 mutant under in vitro and in vivo conditions, revealing a new mechanism of action for a dietary-related compound. PEITC exhibits growth-inhibitory activity in cells expressing p53 mutants with preferential activity toward p53(R175), one of the most frequent 'hotspot' mutations within the p53 sequence. Mechanistic studies revealed that PEITC induces apoptosis in a p53(R175) mutant-dependent manner by restoring p53 WT conformation and transactivation functions. Accordingly, in PEITC-treated cells the reactivated p53(R175) mutant induces apoptosis by activating canonical WT p53 targets, inducing a delay in S and G2/M phase, and by phosphorylating ATM/CHK2. Interestingly, the growth-inhibitory effects of PEITC depend on the redox state of the cell. Further, PEITC treatments render the p53(R175) mutant sensitive to degradation by the proteasome and autophagy in a concentration-dependent manner. PEITC-induced reactivation of p53(R175) and its subsequent sensitivity to the degradation pathways likely contribute to its anticancer activities. We further show that dietary supplementation of PEITC is able to reactivate WT activity in vivo as well, inhibiting tumor growth in xenograft mouse model. These findings provide the first example of mutant p53 reactivation by a dietary compound and have important implications for cancer prevention and therapy.
Questions: Functional redundancy in assemblages may insure ecosystem processes after perturbation, potentially causing temporary or permanent local species extinctions. Yet, functional redundancy has ...only been inferred by indirect evidence or measured by methods that may not be the most appropriate. Here, we apply an existing method to measure functional redundancy, which is the fraction of species diversity not expressed by functional diversity, to assess whether functional redundancy affects community resilience after disturbance. Location: Subtropical grassland, south Brazil (30°05′46″S, 51°40′37″W). Method: Species traits and community composition were assessed in quadrats before grazing and after community recovery. Grazing intensity (G) was measured in each quadrat. We used traits linked to grazing intensity to define functional redundancy (FR) as the difference of Gini–Simpson index of species diversity (D) and Rao's quadratic entropy (Q). Also, with the same traits, we defined community functional stability (S) as the similarity between trait-based community composition before grazing and 47 and 180 d after grazing ending. Using path analysis we assessed different postulated causal models linking functional diversity (Q), functional redundancy (FR), grazing intensity (G) and community-weighted mean traits to community stability (S) under grazing. Results: Path analysis revealed the most valid causal model FR → S ← G, with a significant positive path coefficient for FR → S and a marginally significant negative one for S ← G. Since FR and G were independent in their covariation and in their effects on S, the model discriminated community resistance to grazing (the effect of G on S) from community resilience after grazing caused by functional redundancy (indicated by the effect of FR on S). Conclusion: We show that expressing functional redundancy mathematically is a useful tool for testing causal models linking diversity to community stability. The results support the conclusion that functional redundancy enhanced community resilience, therefore corroborating the insurance hypothesis.
The COHERENT experiment is well poised to test sub-GeV dark matter models using detectors sensitive to coherent elastic neutrino-nucleus scattering (CEvNS) in the π+ decay-at-rest (π-DAR) neutrino ...beam produced by the Spallation Neutron Source. We show a planned 750-kg single-phase liquid argon scintillation detector would place leading limits on scalar light dark matter models for dark matter particles produced through vector and leptophobic portals in the absence of other effects beyond the standard model. The characteristic timing profile of a π-DAR beam allows a unique opportunity for constraining systematic uncertainties on the standard model background using a time window where dark matter signal is not expected, enhancing expected sensitivity. Additionally, we discuss future prospects which show that an on-axis CEvNS detector would probe the thermal abundance for a scalar dark matter candidate for all couplings α′ ≤ 1 for 15 MeV dark matter with just 1.0 tonne-yr of exposure with increased exposure testing a wider range of dark matter masses and spins.
Sustainability concerns as well as recent increases in fertilizer prices exacerbates the need to optimise the use of biowastes as fertilizers. For this reason, we investigated how different ...pretreatments affect the P dynamics when biofertilizers are placed in the soil.
Sewage sludge (SS), sewage sludge ash (SS-ash), meat and bone meal (MBM), and the solid fraction of biogas digestate (BGF) were pretreated with H2SO4, NaOH, and Ca(OH)2 and incubated for 2 and 12 days, respectively, in a one-dimensional reaction system for detailed studies of the interactions in the biomaterial-soil interface and the soil adjacent to the placement zone.
Our results showed that acidification and treatment with NaOH increased the P solubility of the biomaterials. The P loss from the biomaterial layer to the soil was correlated with water-extractable P in the biomaterials (0.659) and water-extractable P in the soil (0.809). Acidification significantly increased the total amount of P depleted from the biomaterial to the soil whereas NaOH pre-treatment did not. However, for NaOH-treated SS and SS-ash, the apparent recoveries were significantly higher compared to the acidification due to a decrease in soil P sorption capacity as the soil pH increased due to residual alkalinity in the biomaterials.
Acidification showed promising results by increasing the P solubility of all the biomaterials, and the alkalinization of SS and SS-ash with NaOH by increasing the apparent recovery in the soil. However, further studies are needed to assess the effects of these treatments on plant growth and P uptake.
