Resveratrol offers pleiotropic health benefits including a reported ability to inhibit lipopolysaccharide (LPS)-induced cytokine production. The aim of this work was to prepare, characterize, and ...evaluate a resveratrol nanoparticulate formulation based on zein. For this purpose, the oral bioavailability of the encapsulated polyphenol as well as its anti-inflammatory effects in a mouse model of endotoxic shock was studied. The resveratrol-loaded nanoparticles displayed a mean size of 307 ± 3 nm, with a negative zeta potential (−51.1 ± 1.55 mV), and a polyphenol loading of 80.2 ± 3.26 μg/mg. In vitro, the release of resveratrol from the nanoparticles was found to be pH independent and adjusted well to the Peppas–Sahlin kinetic model, suggesting a mechanism based on the combination of diffusion and erosion of the nanoparticle matrix. Pharmacokinetic studies demonstrated that zein-based nanoparticles provided high and prolonged plasma levels of the polyphenol for at least 48 h. The oral bioavailability of resveratrol when administered in these nanoparticles increased up to 50% (19.2-fold higher than for the control solution of the polyphenol). Furthermore, nanoparticles administered daily for 7 days at 15 mg/kg were able to diminish the endotoxic symptoms induced in mice by the intraperitoneal administration of LPS (i.e., hypothermia, piloerection, and stillness). In addition, serum tumor necrosis factor-alpha (TNF-α) levels were slightly lower (approximately 15%) than those observed in the control.
Resveratrol is a naturally occurring polyphenol that provides several health benefits including cardioprotection and cancer prevention. However, its biological activity is limited by a poor ...bioavailability when taken orally. The aim of this work was to evaluate the capability of casein nanoparticles as oral carriers for resveratrol. Nanoparticles were prepared by a coacervation process, purified and dried by spray-drying. The mean size of nanoparticles was around 200 nm with a resveratrol payload close to 30 μg/mg nanoparticle. In vitro studies demonstrated that the resveratrol release from casein nanoparticles was not affected by the pH conditions and followed a zero-order kinetic. When nanoparticles were administered orally to rats, they remained within the gut, displaying an important capability to reach the intestinal epithelium. No evidence of nanoparticle "translocation" were observed. The resveratrol plasma levels were high and sustained for at least 8 h with a similar profile to that observed for the presence of the major metabolite in plasma. The oral bioavailability of resveratrol when loaded in casein nanoparticles was calculated to be 26.5%, 10 times higher than when the polyphenol was administered as oral solution. Finally, a good correlation between in vitro and in vivo data was observed.
Blue LUminous Events (BLUEs) are transient corona discharges occurring in thunderclouds and characterized by strong 337.0 nm light flashes with absent (or weak) 777.4 nm component. We present the ...first nighttime climatology of BLUEs as detected by the Modular Multispectral Imaging Array of the Atmosphere‐Space Interaction Monitor showing their worldwide geographical and seasonal distribution. A total (land and ocean) of ∼11 BLUEs occur around the globe every second at local midnight and the average BLUE land/sea ratio is ∼7:4. The northwest region of Colombia shows an annual nighttime peak. Globally, BLUEs are maximized during the boreal summer‐autumn, contrary to lightning which is maximed in the boreal summer. The geographical distribution of nighttime BLUEs shows three main regions in, by order of importance, the Americas, Europe/Africa and Asia/Australia.

ABSTRACT Our Galaxy is known to contain a central boxy/peanut-shaped bulge, yet the importance of a classical, pressure-supported component within the central part of the Milky Way is still being ...debated. It should be most visible at low metallicity, a regime that has not yet been studied in detail. Using metallicity-sensitive narrow-band photometry, the Pristine Inner Galaxy Survey (PIGS) has collected a large sample of metal-poor ($\rm {Fe/H}\, \lt -1.0$) stars in the inner Galaxy to address this open question. We use PIGS to trace the metal-poor inner Galaxy kinematics as function of metallicity for the first time. We find that the rotational signal decreases with decreasing Fe/H , until it becomes negligible for the most metal-poor stars. Additionally, the velocity dispersion increases with decreasing metallicity for $-3.0 \lt \rm {Fe/H}\, \lt -0.5$, with a gradient of −44 ± 4 km s−1 dex−1. These observations may signal a transition between Galactic components of different metallicities and kinematics, a different mapping on to the boxy/peanut-shaped bulge for former disc stars of different metallicities and/or the secular dynamical and gravitational influence of the bar on the pressure-supported component. Our results provide strong constraints on models that attempt to explain the properties of the inner Galaxy.
ABSTRACT
The Pristine survey uses narrow-band photometry to derive precise metallicities down to the extremely metal-poor regime ($ \rm Fe/H \lt -3$), and currently consists of over 4 million ...FGK-type stars over a sky area of $\sim 2500\, \mathrm{deg}^2$. We focus our analysis on a subsample of ∼80 000 main-sequence turn-off stars with heliocentric distances between 6 and 20 kpc, which we take to be a representative sample of the inner halo. The resulting metallicity distribution function (MDF) has a peak at $ \rm Fe/H =-1.6$, and a slope of Δ(LogN)/$\Delta \rm Fe/H = 1.0 \pm 0.1$ in the metallicity range of $-3.4\; \lt\; \rm Fe/H\; \lt -2.5$. This agrees well with a simple closed-box chemical enrichment model in this range, but is shallower than previous spectroscopic MDFs presented in the literature, suggesting that there may be a larger proportion of metal-poor stars in the inner halo than previously reported. We identify the Monoceros/TriAnd/ACS/EBS/A13 structure in metallicity space in a low-latitude field in the anticentre direction, and also discuss the possibility that the inner halo is dominated by a single, large merger event, but cannot strongly support or refute this idea with the current data. Finally, based on the MDF of field stars, we estimate the number of expected metal-poor globular clusters in the Milky Way halo to be 5.4 for $ \rm Fe/H\; \lt\; -2.5$ and 1.5 for $ \rm Fe/H\; \lt\; -3$, suggesting that the lack of low-metallicity globular clusters in the Milky Way is not due simply to statistical undersampling.
Abstract
This study summarizes the revision performed on the surface layer formulation of the Weather Research and Forecasting (WRF) model. A first set of modifications are introduced to provide more ...suitable similarity functions to simulate the surface layer evolution under strong stable/unstable conditions. A second set of changes are incorporated to reduce or suppress the limits that are imposed on certain variables in order to avoid undesired effects (e.g., a lower limit in u*). The changes introduced lead to a more consistent surface layer formulation that covers the full range of atmospheric stabilities. The turbulent fluxes are more (less) efficient during the day (night) in the revised scheme and produce a sharper afternoon transition that shows the largest impacts in the planetary boundary layer meteorological variables. The most important impacts in the near-surface diagnostic variables are analyzed and compared with observations from a mesoscale network.
This is the first of two papers that document the creation of the New European Wind Atlas (NEWA). It describes the sensitivity analysis and evaluation procedures that formed the basis for choosing ...the final setup of the mesoscale model simulations of the wind atlas. The suitable combination of model setup and parameterizations, bound by practical constraints, was found for simulating the climatology of the wind field at turbine-relevant heights with the Weather Research and Forecasting (WRF) model. Initial WRF model sensitivity experiments compared the wind climate generated by using two commonly used planetary boundary layer schemes and were carried out over several regions in Europe. They confirmed that the most significant differences in annual mean wind speed at 100 m a.g.l. (above ground level) mostly coincide with areas of high surface roughness length and not with the location of the domains or maximum wind speed. Then an ensemble of more than 50 simulations with different setups for a single year was carried out for one domain covering northern Europe for which tall mast observations were available. We varied many different parameters across the simulations, e.g. model version, forcing data, various physical parameterizations, and the size of the model domain. These simulations showed that although virtually every parameter change affects the results in some way, significant changes in the wind climate in the boundary layer are mostly due to using different physical parameterizations, especially the planetary boundary layer scheme, the representation of the land surface, and the prescribed surface roughness length. Also, the setup of the simulations, such as the integration length and the domain size, can considerably influence the results. We assessed the degree of similarity between winds simulated by the WRF ensemble members and the observations using a suite of metrics, including the Earth Mover's Distance (EMD), a statistic that measures the distance between two probability distributions. The EMD was used to diagnose the performance of each ensemble member using the full wind speed and direction distribution, which is essential for wind resource assessment. We identified the most realistic ensemble members to determine the most suitable configuration to be used in the final production run, which is fully described and evaluated in the second part of this study (Dörenkämper et al., 2020).

The present work describes the encapsulation of probiotics using a by-product as wall material and a process feasible to be scaled-up: coacervation of soybean protein concentrate (SPC) by using calcium salts and spray-drying. SPC was extracted from soybean flour, produced during the processing of soybean milk, by alkaline extraction following isoelectric precipitation. Two probiotic strains were selected for encapsulation (Lactobacillus plantarum CECT 220 and Lactobacillus casei CECT 475) in order to evaluate the ability of SPC to encapsulate and protect bacteria from stress conditions. The viability of these encapsulated strains under in vitro gastrointestinal conditions and shelf-life during storage were compared with the most common forms commercialized nowadays. Results show that SPC is a feasible material for the development of probiotic microparticles with adequate physicochemical properties and enhanced significantly both probiotic viability and tolerance against simulated gastrointestinal fluids when compared to current available commercial forms.
Abstract Zein nanoparticles were evaluated as nanocarriers to promote the oral bioavailability of quercetin and, thus, improve its anti-inflammatory effect on a mouse model of induced endotoxemia. ...For this purpose, the flavonoid and 2-hydroxypropyl-β-cyclodextrin were encapsulated in zein nanoparticles. The resulting nanoparticles displayed a mean size of about 300 nm and the payload was calculated to be close to 70 μg/mg nanoparticle. The release of quercetin from zein nanoparticles followed a zero-order kinetic. After oral administration, nanoparticles provided high and sustained levels of quercetin in plasma and the relative oral bioavailability was calculated to be approx. 60%. Animals treated with quercetin-loaded nanoparticles (1 dose every two days; 1 week) presented endotoxic symptoms less severe than those observed in animals treated with the oral solution of the flavonoid (1 dose every day; 1 week). This was further corroborated by the significantly low circulating TNF-alpha in the quercetin-loaded nanoparticles treated mice.
This is the second of two papers that document the creation of the New European Wind Atlas (NEWA). In Part 1, we described the sensitivity experiments and accompanying evaluation done to arrive at ...the final mesoscale model setup used to produce the mesoscale wind atlas. In this paper, Part 2, we document how we made the final wind atlas product, covering both the production of the mesoscale climatology generated with the Weather Research and Forecasting (WRF) model and the microscale climatology generated with the Wind Atlas Analysis and Applications Program (WAsP). The paper includes a detailed description of the technical and practical aspects that went into running the mesoscale simulations and the downscaling using WAsP. We show the main results from the final wind atlas and present a comprehensive evaluation of each component of the NEWA model chain using observations from a large set of tall masts located all over Europe. The added value of the WRF and WAsP downscaling of wind climatologies is evaluated relative to the performance of the driving ERA5 reanalysis and shows that the WRF downscaling reduces the mean wind speed bias and spread relative to that of ERA5 from -1.50±1.30 to 0.02±0.78 m s−1. The WAsP downscaling has an added positive impact relative to that of the WRF model in simple terrain. In complex terrain, where the assumptions of the linearized flow model break down, both the mean bias and spread in wind speed are worse than those from the raw mesoscale results.
