Litopenaeus vannamei
(LvAK) was purified and its activity was assayed for phosphorylation of TDP using ATP as phosphate donor. Moreover, by using high-pressure liquid chromatography (HPLC) the phosphate transfer reaction was followed. Also, LvAK tryptophan fluorescence emission changes were detected by dTDP titration, suggesting that the hydrophobic environment of Trp 221, which is located in the top of the active site, is perturbed upon dTDP binding. The kinetic constants for both substrates Arg and dTDP were calculated by isothermal titration calorimetry (ITC). Besides, docking calculations suggested that dTDP could bind LvAK in the same cavity where ATP bind, and LvAK basic residues (Arg124, 126 and 309) stabilize the dTDP phosphate groups and the pyrimidine base interact with His284 and Ser122. These results suggest that LvAK bind and phosphorylate dTDP being ATP the phosphate donor, thus describing a novel alternate nucleoside diphosphate kinase-like activity for this enzyme.

A cohort prospective 5 years follow-up study has been designed including 25000 patients with hypertension, diabetes and/or dyslipidemia attended in usual clinical practice. All information is registered in a unique electronic health recording system (ABUCASIS) that is the usual way to register clinical practice in the Valencian Health System (primary and secondary care). The system covers about 95% of population (near 5 million people). The system is linked with database of mortality register, hospital withdrawals, prescriptions and assurance databases in which each individual have a unique identification number. Diagnoses in clinical practice are always registered based on IDC-9. Occurrence of CV disease was the main outcomes of interest. Risk survival analysis methods will be applied to estimate the cumulative incidence of developing CV events over time.

We aimed to estimate the attributable risk associated with chronic ...kidney disease Epidemiology Collaboration-estimated glomerular filtration rate (eGFR), for all-cause mortality, and cardiovascular hospitalization.
Prospective study of study participants with cardiovascular risk factors in 2008-2012. We included 52 007 cardiovascular disease-free men and women aged 30 years or older with hypertension, diabetes, or dyslipidemia, who underwent routine health examinations in primary care.
A total of 6639 (12.8%) patients had eGFR below 60 ml/min per 1.73 m and among them 1782 (3.4%) had 45 ml/min per 1.73 m or lower. During an average follow-up time of 3.2 years, 54.12 deaths, 99.98 coronary heart disease (CHD) hospitalizations, and 90.64 stroke hospitalizations/10 000 person-years were recorded. The population attributable risks associated with having a GFR lower than 60 ml/min per 1.73 m were 6.9% (95% confidence interval = 2.07, 10.65) for all-cause mortality, 6.8% (4.3, 9.4) for CHD hospitalization, and 4.1% (1.02, 7.00) for stroke hospitalization. Participants with increasing number of cardiovascular risk factors displayed increasing population attributable risks associated to a GFR less than 60 ml/min per 1.73 m for all-cause mortality and CHD (P heterogeneity 0.002 and 0.05, respectively).
In a large general practice cohort of patients with cardiovascular disease risk factors, decreasing eGFR levels were associated with additional attributed risk of mortality and cardiovascular disease. Our findings underscore that intensified efforts are needed to reduce the cardiovascular disease burden associated to chronic kidney disease.
This paper introduces a novel and systematic approach to formulate a simple and dimensionally homogeneous Jacobian matrix for the planar 3-RRR parallel manipulator. Based on such a matrix, the ...equation of the velocity ellipsoid is derived, and a velocity optimization procedure is proposed. As a main result, optimal directions for obtaining the best velocity performance of the end effector of the manipulator are mathematically formulated and geometrically located. Detailed numerical examples are given to illustrate the applicability of the proposed approach.
This paper presents a systematic approach to formulate a closed-form solution of the forward displacement analysis for a Schönflies parallel manipulator. The effectiveness of the method relies upon ...the solution of only one polynomial equation in terms of just one of the involved variables. Once the roots of this polynomial are found, the remaining variables are readily determined from backward substitution. In addition, the proposed method works equally well for general geometries and for special geometries of the mobile platform and fixed platform of the manipulator. Finally, a fully detailed case study is reported.
Tumor‐associated stroma biomarkers are emerging as key signaling molecules of metastasis in colorectal cancer (CRC). In this sense, periostin (POSTN), a protein of the extracellular matrix from the ...stromal compartment, is envisioned as a potential stromal prognostic biomarker, which facilitates the application of pertinent treatments. In this work, we report an easy‐to‐handle amperometric sandwich‐based immunosensing strategy for the determination of POSTN involving commercial magnetic microparticles (MBs), disposable carbon electrodes, and the horseradish peroxidase (HRP)/H2O2/hydroquinone (HQ) electrochemical system. The method allowed a dynamic linear range between 0.47 and 25 ng mL−1 and a limit of detection, LOD, of 0.14 ng mL−1 compatible with clinical demands. The method was applied to the analysis of POSTN in a variety of cancer‐related real bio‐scenarios including cell extracts and secretomes from CRC cells, and plasma from CRC patients at different stages. The potentials of this firstly developed MBs‐assisted immunoplatform are justified by distinguishing between metastatic abilities of cultured CRC cells through the analysis of their extracts and secretomes, and by differentiating between healthy controls and CRC patients in just 60 min. Therefore, the developed immunoplatform can be envisaged as a novel and profitable tool for exploring the most uncharacterized but prospective tumor areas.

The use of tissue adhesives has been proposed as an anastomosis reinforcement; however, their efficacy has not been evaluated in a contaminated environment. The objective of this study was to ...determine if the use of sutures reinforced with ethyl-2-cyanoacrylate for colonic anastomoses in the presence of fecal peritonitis, in a murine animal model, decreases the frequency of dehiscence.
Wistar rats were used. Fecal peritonitis was established until reaching 18 hours of evolution. Then, resection and anastomosis of the colon were performed with only polydioxanone (PDS) sutures in the control group and reinforcement with ethyl-2-cyanoacrylate in the experimental group. The dehiscence frequency and burst pressure were evaluated six days after the anastomosis was performed.
We included 30 Wistar rats, all males, with a median age of five months and an average weight of 350.43 g. Anastomotic dehiscence was observed in 53.33% of the control group, in contrast with 13.33% of the experimental group (p = 0.020). There was no significant difference in burst pressure between the two groups.
The use of ethyl-2-cyanoacrylate, in an experimental murine animal model, as reinforcement in colonic anastomoses in the presence of fecal peritonitis decreases the frequency of anastomotic dehiscence, although it does not increase resistance to burst pressure.
