The enantiomers of 1-methyl-3-(10H-phenothiazine-10-ylmethyl)-1-azoniabicyclo2 ,2,2octane iodide (1) were prepared by chiral chromatographic resolution of the precursor mequitazine (2). The ...(+)-(S)-enantiomer 1b is 10-fold more potent than (-)-(R)-enantiomer 1a as a histamine antagonist, while the two enantiomers show the same antimuscarinic activity in vitro. The absolute configuration of the more active dextrorotatory isomer has been determined by X-ray analysis. Conformational analysis and molecular modeling suggest that the (+)-(S)-enantiomer can adopt a conformation similar to that attributed to the receptor binding conformers of classical antihistamines.
A combined histologic, immunohistologic, enzyme histochemical, and immunologic study has been carried out in a 7‐year‐old girl with recurring extramediastinal monocentric giant lymph node hyperplasia ...of hyaline‐vascular type. A large panel of monoclonal and polyclonal antibodies to lymphoid and nonlymphoid cell markers were tested on frozen and paraffin‐embedded lymph node tissue as well as on cell suspension and peripheral blood. Tissue enzyme histochemical study, including a conventional hematologic panel, was performed on frozen and plastic‐embedded sections. The pattern was dominated by nodular aggregates of round BA‐1+ Leu‐14+ HLA‐DR+ ATPase+ lymphocytes with polyclonal sIgD and sIgM positivity and lacking clg and BA‐2 staining. Leu‐l+/Leu‐4+, OKT6+, OKT10+, Leu‐7+, and CALLA+ cells were few or absent in the nodules, whereas DRC‐1+ BA‐2+ HLA‐DR+ 5′‐Nuc+ cells formed a dendritic network in the outer portion of the nodules. No immunoreactivity for lymphoid and nonlymphoid cell markers, including cytokeratin and keratin, was detected in centrinodular histiocytic‐like cells. Particularly, the Hassall's‐like structures contained a target‐like positivity for laminin, and consisted of flattened acid phosphatase (AP), alpha‐naphthyl acetate esterase (ANAE), 5′‐nucIeotidase (5′‐Nuc), and adenosine tri‐phosphatase (ATPase) positive cells, whose enzyme profile overlapped with that of the histiocytic‐like cells. The extranodular areas were mainly composed of Leu‐1+/Leu‐4+ lymphocytes with Leu‐3a+/ OKT4+ phenotype and, to a lesser extent, of OKT6+ OKT10+ lymphoid cells and scattered cells with markers of histiocytic lineage. The abundant vascular component was generally identified by laminin positivity and, in smaller proportion, it was positive for Factor VIII‐related antigen. Most of the medium‐sized vessels with high endothelium had marked AP, ANAE, and ATPase activities. The process observed resulted from vascularized nodular aggregates of nontransformed B‐cells with the phenotype of primary follicle lymphocytes, associated to centrinodular histiocytic‐like cells with a distinct enzyme profile.
Nifedipine is a calcium-channel antagonist with effective antihypertensive activity and has been suggested for the treatment of high blood pressure as an alternative to vasodilators. The aim of this ...study was to define the acute effect of nifedipine and in particular the dose-effect relationship, effects on veins, influence on adrenergic reflexes, and effectiveness on hypertension according to severity and etiologic type. The effects of nifedipine on blood pressure, heart rate, forearm blood flow, peripheral vascular resistance, orthostatic and cold reflexes, and venous tone were examined in 45 patients with hypertension of different etiologies (essential, renovascular, and renal parenchymal) and different World Health Organization grades. The antihypertensive effect was dose dependent, but a dose of 20 mg has nearly maximal activity with acceptable side effects. The drug acts by lowering peripheral vascular resistance, and this lowering is directly related to baseline values; therefore, the antihypertensive effect increases with severity of the hypertension. Nifedipine had the same effect in all three etiologic groups of hypertension studied. The drug seems to increase venous tone, since it caused venoconstriction when locally injected in hand veins. Nifedipine did not alter adrenergic reflexes induced by both cold application and standing and was well tolerated. In conclusion, the calcium antagonist nifedipine for its characteristics of action, at least in acute administration, seems to be a useful alternative in the treatment of various forms of hypertension.
A combined morphological, immunohistological, and enzyme histochemical analysis was performed on frozen and fixed lymph node tissue in a case of histiocytic necrotizing lymphadenitis (HNL) using ...conventional histology, a panel of monoclonal and polyclonal antibodies, and a series of common haematological enzyme reactions. Histology showed multiple paracortical necrotizing foci which, in a prominently necrobiotic background devoid of granulocytes, contained large numbers of foamy histiocytes and macrophages intermingled with cells resembling degenerating plasmacytoid T-cells. Most of the histiocytes were alpha1-antichymotrypsin positive and foamy cells were also distinctly Leu-M1 positive. Strong granular acid phosphatase (AP) positivity was present in the cytoplasm of the macrophages and histiocytes. The cells with plasmacytoid features showed weaker and homogeneously diffuse AP staining. Alpha-naphthyl acetate esterase (ANAE) activity was much less striking than AP in the necrotizing foci and most of the ANAE negative cells corresponded to those with plasmacytoid features. No cells with B-cell lineage markers were present within the necrotizing foci; most of the occasional T-cells (Leu-1+, Leu-4+) present in the foci were Leu-2a+ (OKT8+) whereas OKT10+ lymphoid cells were abundant and appeared to correspond with the cells with plasmacytoid features. Our combined data confirm that the special type of necrosis found in HNL develops within foci of plasmacytoid T-cells undergoing regressive changes and apparently exhibiting distinct immunohistological and enzyme histochemical features.

Les tensions générées par ce type d'intérêts contradictoires sont particulièrement sensibles en Corse en raison de l'existence de grandes zones préservées et strictement protégées. Cette recherche se propose d'éclairer deux niveaux de régulation de ces intérêts : d'une part la régulation légale par les cours de justice qui arbitrent en général des conflits d'urbanisme dont l'échelle spatiale se situe au niveau de la commune, d'autres part la régulation politique au niveau régional au travers de procédures de concertation visant à réformer les règles de protection des zones côtières.

The tensions generated by these contradictory interests are particularly sensitive in Corsica, because a large part of coastal spaces still remain preserved and strictly protected. The present research is interested on two levels of regulation of these interests: judicial regulation by the administrative courts, relating to conflicts on a district level (building projects), and on the regional level, political governance through a public consultation procedure which focuses on planning lax reforms. The inquiry shows how these two levels seems to be connected: the legal strategies of local representatives can be explained by characteristics of judicial regulation.

Solutions containing Zn(II) and Cu(II) complexes with 15aneN(3)O(2) rapidly adsorb atmospheric CO(2) to give {ZnL(3)( (3)-CO(3))}.(ClO(4))(4) (2) and {CuL(3)( (3)-CO(3))}.(ClO(4))(4) (4) complexes. ...The crystal structures of both complexes have been solved (for 2, space group R3c, a, b = 22.300(5) Å, c = 17.980(8) Å, V = 7743(4) Å(3), Z = 6, R = 0.0666, R(w)(2) = 0.1719; for 4, space group R3c, a, b = 22.292(7) Å, c = 10.096(8) Å, V = 7788(5) Å(3), Z = 6, R = 0.0598, R(w)(2) = 0.1611), and the spectromagnetic behavior of 4 has been studied. In both compounds a carbonate anion triply bridges three metal cations. Each metal is coordinated by one oxygen of the carbonate, three nitrogens, and an oxygen of the macrocycle; the latter donor weakly interacts with the metals. Although the two compounds are isomorphous, they are not isostructural, because the coordination geometries of Zn(II) in 2 and Cu(II) in 4 are different. The mixed complex {CuZn(2)L(3)( (3)-CO(3))}.(ClO(4))(4) has been synthesized. X-ray analysis (space group R3c, a, b = 22.323(7) Å, c = 17.989(9) Å, V = 7763(5) Å(3), Z = 6, R = 0.0477, R(w)(2) = 0.1371) and EPR measurements are in accord with a (3)-carbonate bridging one Cu(II) and two Zn(II) ions in {CuZn(2)L(3)( (3)-CO(3))}(4+). Both the Zn(II) and Cu(II) cations exhibit the same coordination sphere, almost equal to that found in the trinuclear Zn(II) complex 2. The systems Zn(II)/L and Cu(II)/Lhave been studied by means of potentiometric measurements in 0.15 mol dm(-)(1) NaCl and in 0.1 mol dm(-)(3) NaClO(4) aqueous solutions; the species present in solution and their stability constants have been determined. In both systems ML(2+) species and hydroxo complexes M(II)LOH(+) (M = Zn, Cu) are present in solution. In the case of Cu(II), a CuL(OH)(2) complex is also found. The process of CO(2) fixation is due to the presence of such hydroxo-species, which can act as nucleophiles toward CO(2). In order to test the nucleophilic ability of the Zn(II) complexes, the kinetics of the promoted hydrolysis of p-nitrophenyl acetate has been studied. The ZnLOH(+) complex promotes such a reaction, where the Zn(II)-bound OH(-) acts as a nucleophile to the carbonyl carbon. The equilibrium constants for the addition of HCO(3)(-) and CO(3)(2)(-) to the ZnL(2+) complex have been potentiometrically determined. Only ML(HCO(3))(+) and ML(CO(3)) species are found in aqueous solution. A mechanism for the formation of {ML(3)( (3)-CO(3))}.(ClO(4))(4) is suggested.
