Background. Enteroaggregative Escherichia coli (EAEC) is a cause of epidemic and sporadic diarrhea, yet its role as an enteric pathogen is not fully understood. Methods. We characterized 121 EAEC ...strains isolated in 2008 as part of a case-control study of moderate to severe acute diarrhea among children 0-59 months of age in Bamako, Mali. We applied multiplex polymerase chain reaction and comparative genome hybridization to identify potential virulence factors among the EAEC strains, coupled with classification and regression tree modeling to reveal combinations of factors most strongly associated with illness. Results. The gene encoding the autotransporter protease SepA, originally described in Shigella species, was most strongly associated with diarrhea among the EAEC strains tested (odds ratio, 5.6 95% confidence interval, 1.92-16.17; P = .0006). In addition, we identified 3 gene combinations correlated with diarrhea: (1) a clonal group positive for sepA and a putative hemolysin; (2) a group harboring the EAST-1 enterotoxin and the flagellar type H33 but no other previously identified EAEC virulence factor; and (3) a group carrying several of the typical EAEC virulence genes. Conclusion. Our data suggest that only a subset of EAEC strains are pathogenic in Mali and suggest that sepA may serve as a valuable marker for the most virulent isolates.
Better understanding of root function is central for the development of plants with more efficient nutrient uptake and translocation. We here present a method for multielement bioimaging at the ...cellular level in roots of the genetic model system Arabidopsis (Arabidopsis thaliana). Using conventional protocols for microscopy, we observed that diffusible ions such as potassium and sodium were lost during sample dehydration. Thus, we developed a protocol that preserves ions in their native, cellular environment. Briefly, fresh roots are encapsulated in paraffin, cryo-sectioned, and freeze dried. Samples are finally analyzed by laser ablation-inductively coupled plasma-mass spectrometry, utilizing a specially designed internal standard procedure. The method can be further developed to maintain the native composition of proteins, enzymes, RNA, and DNA, making it attractive in combination with other omics techniques. To demonstrate the potential of the method, we analyzed a mutant of Arabidopsis unable to synthesize the metal chelator nicotianamine. The mutant accumulated substantially more zinc and manganese than the wild type in the tissues surrounding the vascular cylinder. For iron, the images looked completely different, with iron bound mainly in the epidermis of the wild-type plants but confined to the cortical cell walls of the mutant. The method offers the power of inductively coupled plasma-mass spectrometry to be fully employed, thereby providing a basis for detailed studies of ion transport in roots. Being applicable to Arabidopsis, the molecular and genetic approaches available in this system can now be fully exploited in order to gain a better mechanistic understanding of these processes.

Urban growth and sprawl have long put pressure on rural areas, leading to a growing zone of 'rurban areas' where a multitude of ...functions, urban or rural, compete for land. The development is acknowledged in European spatial planning documents, which also recommend common solutions. E.g. the spatial policy discussed and recommended in the European Spatial Development Perspective (ESDP) emphasises the compact city concept, rural-urban linkages and conservation of nature and cultural heritage. This paper compares planning practice for rurban areas in three cases: Roskilde municipality (Denmark), Staffanstorp municipality (Sweden) and Werv-area (the Netherlands). All three areas belong to the rurban zone and are selected to exemplify pro-active planning. The analysis focuses on how the concept of compact city is perceived and implemented, how rurban areas are managed in order to avoidfurther urban encroachment, and how resilient green landscapes are ensured. The results reveal significant differences in approaches, reflecting variations in the public involvement in rurban areas development, the role of different administrative levels and the use of zonation. Variation in the use of zonation encapsulates important differences between the cases: pre-zonation based on local discretion (Sweden), zonation based on national legislation and principles with little room of manoeuvre for the municipality (Denmark) and post-zonation with deliberate introduction of urban elements in rural areas and negotiations between stakeholders (the Netherlands).
