Abstract Background Exosomes are nanometer-sized vesicles that are released by normal and neoplastic cells. Previous studies have focused on the interaction between tumour-derived exosomes and the ...immune system, as a consequence of immune suppression or enhancement. However, the effects of tumour-derived exosomes on tumour cells themselves have not been well studied. Aims To investigate the effects of gastric cancer exosomes on tumour cell proliferation and the possible mechanisms. Methods By serial centrifugation and sucrose gradient ultracentrifugation, we isolated and purified the exosomes from gastric cancer SGC7901 cells, then viewed them by electron microscopy. Cell proliferation was measured by 3-(4,5-dimethylthiazol)-2,5-diphenyltetrazolium bromide assay. Protein expression was assayed by Western blotting. Results SGC7901-cell-derived exosomes promoted the proliferation of SGC7901 and BGC823 cells. The increase in proliferation induced by exosomes was accompanied by activation of Akt and extracellular-regulated protein kinase, and phosphoinositide 3-kinase or extracellular-regulated protein kinase inhibitor partially reversed the proliferative effect of exosomes. Moreover, the exosome-induced increase in activity of Akt and extracellular-regulated protein kinase coincided with decreased expression of the Casitas B-lineage lymphoma family of ubiquitin ligases. Conclusion Gastric cancer exosomes promoted tumour cell proliferation, at least in part, by activation of PI3K/Akt and mitogen-activated protein kinase/extracellular-regulated protein kinase pathways. The decreased expression of Casitas B-lineage lymphoma proteins might have contributed to the activation of Akt and extracellular-regulated protein kinase.
Four extreme haze episodes occurred in October 2014 in the North China Plain (NCP). To clarify the formation mechanism of hazes in autumn, strengthened observations were conducted in Beijing from 5 ...October to 2 November. The meteorological parameters, satellite data, chemical compositions and optical properties of aerosols were obtained. The hazes originated from the NCP, developing in the southwest and northeast directions, with the highest concentration of PM2.5 of 469 mu g m-3 in Beijing. The NCP was dominated by a weak high pressure system during the haze episode, which resulted in low surface wind speed and relatively stagnant weather. Moreover, the wind slowed down around Beijing city. The secondary aerosols NO3- was always higher than that of SO42-, which indicated the motor vehicles played a more important part in the hazes in October 2014, even though the oxidation rate from SO2 to SO42- was faster than that of NOx to NO3-. Sudden increases of the concentrations of organic matter, Cl- and BC (black carbon) before each haze episode implied that regional transport of pollutants by biomass burning was important for haze formation during autumn. A satellite map of fire points and the backward trajectories of the air masses also indicated this pollution source. The distinct decrease in the PBL (planetary boundary layer) height during four haze episodes restrained the vertical dispersion of the air pollutants. Water vapor also played a vital role in the formation of hazes by accelerating the chemical transformation of secondary pollutants, leading to hygroscopic growth of aerosols and altering the thermal balance of the atmosphere.

A double-peak X-ray burst from the Galactic magnetar SGR J1935+2154 was discovered to be associated with the two radio pulses of FRB 200428 separated by 28.97 ± 0.02 ms. Precise measurements ...of the timing and spectral properties of the X-ray bursts are helpful for understanding the physical origin of fast radio bursts. In this paper, we have reconstructed some information about the hard X-ray events, which were lost because the High Energy X-ray Telescope (HE) on board the Insight-HXMT mission was saturated by this extremely bright burst, and used the information to improve the temporal and spectral analyses of the X-ray burst. The arrival times of the two X-ray peaks by fitting the new Insight-HXMT/HE lightcurve with multi-Gaussian profiles are 2.77 ± 0.45 ms and 34.30 ± 0.56 ms after the first peak of FRB 200428, respectively, while these two parameters are 2.57 ± 0.52 ms and 32.5 ± 1.4 ms if the fitting profile is a fast-rise and exponential decay function. The spectrum of the two X-ray peaks could be described by a cutoff power law with cutoff energy ∼60 keV and photon index ∼1.4; the latter is softer than the result obtained in Li et al. when the two X-ray peaks appeared.

, the cyclotron absorption line energy anticorrelates with luminosity. Below the critical luminosity, a positive correlation is observed. Therefore, 1A 0535+262 becomes the second source after V0332+53, which clearly shows an anticorrelation above and transition between correlation and anticorrelation around the critical luminosity. The evolution of both the observed CRSF line energy and broadband X-ray continuum spectrum throughout the outburst exhibits significant differences during the rising and fading phases; that is, for a similar luminosity, the spectral parameters take different values, which results in hysteresis patterns for several spectral parameters including the cyclotron line energy. We argue that, similar to V0332+53, these changes might be related to the different geometry of the emission region in rising and declining parts of the outburst, probably due to changes in the accretion disk structure and its interaction with the magnetosphere of the neutron star.
High-β_{θe} (a ratio of the electron thermal pressure to the poloidal magnetic pressure) steady-state long-pulse plasmas with steep central electron temperature gradient are achieved in the ...Experimental Advanced Superconducting Tokamak. An intrinsic current is observed to be modulated by turbulence driven by the electron temperature gradient. This turbulent current is generated in the countercurrent direction and can reach a maximum ratio of 25% of the bootstrap current. Gyrokinetic simulations and experimental observations indicate that the turbulence is the electron temperature gradient mode (ETG). The dominant mechanism for the turbulent current generation is due to the divergence of ETG-driven residual flux of current. Good agreement has been found between experiments and theory for the critical value of the electron temperature gradient triggering ETG and for the level of the turbulent current. The maximum values of turbulent current and electron temperature gradient lead to the destabilization of an m/n=1/1 kink mode, which by counteraction reduces the turbulence level (m and n are the poloidal and toroidal mode number, respectively). These observations suggest that the self-regulation system including turbulence, turbulent current, and kink mode is a contributing mechanism for sustaining the steady-state long-pulse high-β_{θe} regime.
Abstract We report the phase-resolved spectral results of the first Galactic pulsating ultraluminous X-ray source (PULX) Swift J0243.6+6124, modeling its 2017–2018 outburst peak using data collected ...by the Hard X-ray Modulation Telescope (Insight-HXMT). The broad energy coverage of Insight-HXMT allows us to obtain a more accurate spectral continuum to reduce the coupling of broad iron line profiles with other components. We use three different continuum spectrum models but obtain similar iron line results. For the first time, we detect the pulse characteristics of the broad iron line in a PULX. The variation in the width and intensity of this iron line with σ ∼ 1.2–1.5 keV has a phase offset of about 0.25 from the pulse phase. We suggest that the uneven irradiation of the thick inner disk by the accretion column produces the modulated variation of the broad iron line. In addition, the nonpulsed narrow line is suggested to come from the outer disk region.

Transition metal oxides show great potential as electrocatalysts, owing to the low cost and rich chemical states. However, the limited surface areas, low intrinsic activity and poor ...hydrogen evolution reaction (HER) activity greatly restrict the application for overall water splitting. Herein, we have constructed S doped NiCo2O4 nanosheet arrays by Ar plasma (Ar-NiCo2O4|S) to enhance active sites and boost catalytic kinetics. Consequently, the Ar-NiCo2O4|S shows the improved performances for HER and oxygen evolution reaction (OER). Further, as bifunctional electrocatalysts, Ar-NiCo2O4|S exhibit a voltage of 1.63 V at 10 mA cm−2, as well as good stability.

Cyclotron line scattering features are detected in a few tens of X-ray pulsars (XRPs) and used as direct indicators of a strong magnetic field at the surface of accreting neutron stars ...(NSs). In a few cases, cyclotron lines are known to be variable with the accretion luminosity of XRPs. It is accepted that the observed variations of cyclotron line scattering features are related to variations of geometry and dynamics of accretion flow above the magnetic poles of an NS. A positive correlation between the line centroid energy and luminosity is typical for subcritical XRPs, where the accretion results in hot spots at the magnetic poles. The negative correlation was proposed to be a specific feature of bright supercritical XRPs, where radiation pressure supports accretion columns above the stellar surface. The cyclotron line in the spectra of the Be-transient XRP GRO J1008-57 is detected at energies from ∼75 to 90 keV, the highest observed energy of a cyclotron line feature in XRPs. We report the peculiar relation of cyclotron line centroid energies with luminosity in GRO J1008-57 during the Type II outburst in 2017 August observed by Insight-HXMT. The cyclotron line energy was detected to be negatively correlated with the luminosity at 3.2 × 10

. We speculate that the observed peculiar behavior of a cyclotron line would be due to variations of accretion channel geometry.
Cygnus X-1, as the first discovered black hole binary, is a key source for understanding the mechanisms of state transitions, and the scenarios of accretion in extreme gravity fields. We present a ...spectral-timing analysis of observations taken with the Insight-HXMT mission, focusing on the spectral-state dependent timing properties in the broad energy range of 1–150 keV, thus extending previous RXTE-based studies to both lower and higher energies. Our main results are the following: a) We successfully use a simple empirical model to fit all spectra, confirming that the reflection component is stronger in the soft state than in the hard state; b) The evolution of the total fractional root mean square (rms) depends on the selected energy band and the spectral shape, which is a direct result of the evolution of the power spectral densities (PSDs); c) In the hard/intermediate state, we see clear short-term variability features and a positive correlation between central frequencies of the variability components and the soft photon index Γ(sub 1), also at energies above 15 keV. The power spectrum is dominated by red noise in the soft state instead. These behaviors can be traced to at least 90 keV; d) The coherence and the phase-lag spectra show different behaviors dependent on different spectral shapes.
