Abstract Objectives To review the history, theory and current applications of Weibull analyses sufficient to make informed decisions regarding practical use of the analysis in dental material ...strength testing. Data References are made to examples in the engineering and dental literature, but this paper also includes illustrative analyses of Weibull plots, fractographic interpretations, and Weibull distribution parameters obtained for a dense alumina, two feldspathic porcelains, and a zirconia. Sources Informational sources include Weibull's original articles, later articles specific to applications and theoretical foundations of Weibull analysis, texts on statistics and fracture mechanics and the international standards literature. Study selection The chosen Weibull analyses are used to illustrate technique, the importance of flaw size distributions, physical meaning of Weibull parameters and concepts of “equivalent volumes” to compare measured strengths obtained from different test configurations. Conclusions Weibull analysis has a strong theoretical basis and can be of particular value in dental applications, primarily because of test specimen size limitations and the use of different test configurations. Also endemic to dental materials, however, is increased difficulty in satisfying application requirements, such as confirming fracture origin type and diligence in obtaining quality strength data.
Abstract Objectives Determination of material and fractographic properties of a dental indirect resin composite material. Methods A resin composite (Paradigm, 3M-ESPE, MN) was characterized by ...strength, static elastic modulus, Knoop hardness, fracture toughness and edge toughness. Fractographic analyses of the broken bar surfaces was accomplished with a combination of optical and SEM techniques, and included determination of the type and size of the failure origins, and fracture mirror and branching constants. Results The flexure test mean strength ± standard deviation was 145 ± 17 MPa, and edge toughness, Te , was 172 ± 12 N/mm. Knoop hardness was load dependent, with a plateau at 0.99 ± 0.02 GPa. Mirrors in the bar specimens were measured with difficulty, resulting in a mirror constant of approximately 2.6 MPa m1/2 . Fracture in the bar specimens initiated at equiaxed material flaws that had different filler concentrations that sometimes were accompanied by partial microcracks. Using the measured flaw sizes, which ranged from 35 to 100 μm in size, and using estimates of the stress intensity shape factors, fracture toughness was estimated to be 1.1 ± 0.2 MPa m1/2. Significance Coupling the flexure tests with fractographic examination enabled identification of the intrinsic strength limiting flaws. The same techniques could be useful in determining if clinical restorations of similar materials fail from the same causes. The existence of a strong load-dependence of the Knoop hardness of the resin composite is not generally mentioned in the literature, and is important for material comparisons and wear evaluation studies. Finally, the edge toughness test was found promising as a quantitative measure of resistance to edge chipping, an important failure mode in this class of materials.

. Despite the conservation of SAPK pathways within these pathogenic fungi, evidence is emerging that their role and regulation has significantly diverged. However, consistent with stress adaptation being a common virulence trait, SAPK pathways are important pathogenicity determinants in all these major human pathogens. Thus, the development of drugs which target fungal SAPKs has the exciting potential to generate broad-acting antifungal treatments.
Abstract Objectives Presentation of a methodological approach using stereo and scanning electron microscope examination for the failure analysis of an alumina all-ceramic premolar crown (Procera® ...AllCeram). Methods The recovered part of a fractured Procera alumina crown was examined utilizing first a stereomicroscope and second a scanning electron microscope (SEM). The stereomicroscope analysis was performed at low magnifications with oblique lighting in order to enhance spatial relationships and gross detection of crack features. A preliminary fracture surface map of the stereo observations was drawn and used as a guide for the SEM analysis that followed. Specific sites of interest identified under the stereo microscope were analyzed using the SEM at high magnifications searching for small fracture features such as wake hackle and twist hackle within the veneering ceramic in order to confirm the direction of crack propagation. Results At low magnifications and oblique illumination, the stereomicroscope analysis provided an excellent overview of the fractured topography, showing sites of major interest such as a primary edge chip at a margin, a compression curl indicating the end of the fracture event as well as larger hackle lines distributed over the cracked surface. The greater magnifications with the SEM analysis of the sites of interest showed the presence of wake and twist hackle, indicators of the crack propagation direction. A general map of the fracture events could be reconstructed starting with a primary veneer edge chip at the mesial margin. Hackle and wake hackle of the crack front emanating from this margin arose from hoop stresses and propagated through the full crown thickness towards the distal end of the restoration where the compression curl was located. Additional occlusal surface damage in the form of veneer chipping containing arrest lines and twist hackle running in the opposite direction as the main crack path were observed, but occurred as a secondary event without penetrating the alumina core material. Significance Stereo and scanning electron microscopy are complementary analysis techniques useful for the mapping and interpretation of the fracture surface. This case examination is intended to guide the clinical researcher in using qualitative (descriptive) fractography as a tool for understanding the failure process in brittle restorative materials, as well as for assessing possible design inadequacies.
Candida albicans is a major fungal pathogen of humans, causing approximately 400,000 life-threatening systemic infections world-wide each year in severely immunocompromised patients. An important ...fungicidal mechanism employed by innate immune cells involves the generation of toxic reactive oxygen species (ROS), such as superoxide and hydrogen peroxide. Consequently, there is much interest in the strategies employed by C. albicans to evade the oxidative killing by macrophages and neutrophils. Our understanding of how C. albicans senses and responds to ROS has significantly increased in recent years. Key findings include the observations that hydrogen peroxide triggers the filamentation of this polymorphic fungus and that a superoxide dismutase enzyme with a novel mode of action is expressed at the cell surface of C. albicans. Furthermore, recent studies have indicated that combinations of the chemical stresses generated by phagocytes can actively prevent C. albicans oxidative stress responses through a mechanism termed the stress pathway interference. In this review, we present an up-date of our current understanding of the role and regulation of oxidative stress responses in this important human fungal pathogen.
Abstract Objectives To identify the strength limiting flaws in in vitro test specimens of a fine-grained feldspathic dental porcelain. Methods Four-point flexural strengths were measured for 26 test ...specimens. The fracture origin site of every test specimen was studied using stereoptical and scanning electron microscopy. A fractographically labeled Weibull strength distribution graph was prepared. Results The complex microstructure of the feldspathic dental porcelain included a variety of feldspars, tridymite, and a feldspathoid as well as pores/bubbles and residual glass. The relatively high flexural strength is due in part to the fine grain size. Fractography revealed five flaw types that controlled strength: baseline microstructural flaws, pores/bubbles, side wall grinding damage, corner machining damage, and inclusions. The baseline microstructural flaws probably were clusters of particular crystalline phases. Significance Each flaw type probably has a different severity and size distribution, and hence has a different strength distribution. The Weibull strength distribution graph blended the strength distributions of the five flaw types and the apparent good fit of the combined data to a unimodal strength distribution was misleading. Polishing failed to eliminate deeper transverse grinding cracks and corner damage from earlier preparation steps in many of the test pieces. Bend bars should be prepared carefully with longitudinal surface grinding whenever possible and edge chamfers should be carefully applied. If the grinding and preparation flaws were eliminated, the Weibull modulus for this feldspathic porcelain would be greater than 30. Pores/bubbles sometimes controlled strength, but only if they touched each other or an exposed surface. Isolated interior bubble/pores were harmless.
Fungal pathogenicity has arisen in polyphyletic manner during evolution, yielding fungal pathogens with diverse infection strategies and with differing degrees of evolutionary adaptation to their ...human host. Not surprisingly, these fungal pathogens display differing degrees of resistance to the reactive oxygen and nitrogen species used by human cells to counteract infection. Furthermore, whilst evolutionarily conserved regulators, such as Hog1, are central to such stress responses in many fungal pathogens, species-specific differences in their roles and regulation abound. In contrast, there is a high degree of commonality in the cellular responses to reactive oxygen and nitrogen species evoked in evolutionarily divergent fungal pathogens.
Interactions between bacterial and fungal cells shape many polymicrobial communities. Bacteria elaborate diverse strategies to interact and compete with other organisms, including the deployment of ...protein secretion systems. The type VI secretion system (T6SS) delivers toxic effector proteins into host eukaryotic cells and competitor bacterial cells, but, surprisingly, T6SS-delivered effectors targeting fungal cells have not been reported. Here we show that the 'antibacterial' T6SS of Serratia marcescens can act against fungal cells, including pathogenic Candida species, and identify the previously undescribed effector proteins responsible. These antifungal effectors, Tfe1 and Tfe2, have distinct impacts on the target cell, but both can ultimately cause fungal cell death. 'In competition' proteomics analysis revealed that T6SS-mediated delivery of Tfe2 disrupts nutrient uptake and amino acid metabolism in fungal cells, and leads to the induction of autophagy. Intoxication by Tfe1, in contrast, causes a loss of plasma membrane potential. Our findings extend the repertoire of the T6SS and suggest that antifungal T6SSs represent widespread and important determinants of the outcome of bacterial-fungal interactions.

Abstract Objectives To demonstrate the effectiveness of in vivo replicas of fractured ceramic surfaces for descriptive fractography as applied to the analysis of clinical failures. Methods The ...fracture surface topography of partially failed veneering ceramic of a Procera Alumina molar and an In-Ceram Zirconia premolar were examined utilizing gold-coated epoxy poured replicas viewed using scanning electron microscopy. The replicas were inspected for fractographic features such as hackle, wake hackle, twist hackle, compression curl and arrest lines for determination of the direction of crack propagation and location of the origin. Results For both veneering ceramics, replicas provided an excellent reproduction of the fractured surfaces. Fine details including all characteristic fracture features produced by the interaction of the advancing crack with the material's microstructure could be recognized. The observed features are indicators of the local direction of crack propagation and were used to trace the crack's progression back to its initial starting zone (the origin). Drawbacks of replicas such as artifacts (air bubbles) or imperfections resulting from inadequate epoxy pouring were noted but not critical for the overall analysis of the fractured surfaces. Significance The replica technique proved to be easy to use and allowed an excellent reproduction of failed ceramic surfaces. It should be applied before attempting to remove any failed part remaining in situ as the fracture surface may be damaged during this procedure. These two case studies are intended as an introduction for the clinical researcher in using qualitative (descriptive) fractography as a tool for understanding fracture processes in brittle restorative materials and, secondarily, to draw conclusions as to possible design inadequacies in failed restorations.
