Enzyme activity plays an important role in the functioning of aquatic ecosystems. It is sensitive to changes in environmental conditions such as pH, temperature, and nutrient concentration. The ...objective of this work was to determine the acid phosphatase activity (AcPA) in the Almendares and San Juan rivers (western Cuba) and its relationship with physicochemical and microbiological indicators. For this purpose, AcPA, temperature, pH, total dissolved solids, electrical conductivity, dissolved oxygen, concentration of nitrates, nitrites, ammonium, phosphates, total heterotrophs, enterococci,
Escherichia coli
, thermotolerant coliforms, chlorophyll
a
, and chemical oxygen demand (COD) were determined at three sampling stations on the Almendares River and at three sampling stations on the San Juan River. In addition, the nutrient pollution index (
NPI
) and the
N:P
ratio were calculated. In both ecosystems, spatio-temporal variability was observed in the enzymatic activity. In the Almendares River (polluted ecosystem), AcPA was positively correlated with nitrate concentration and COD, while in the San Juan River (slightly contaminated ecosystem), the AcPA correlated negatively with the pH and
NPI
and positively with the concentrations of total heterotrophs,
Escherichia coli
, chlorophyll
a
, and the
N:P
ratio. These results show the impact of anthropogenic pollution on AcPA in freshwater ecosystems with a tropical climate.
In recent years, the concern derived from the presence of emerging contaminants in the environment and the possible effects on the One Health trilogy has increased. This study determined the ...concentration of pharmaceutical contaminants of emerging concern and their relationship with the extracellular enzymatic activity of microbial communities from two rivers in western Cuba. Two sampling stations were analyzed; one in the Almendares River (urban) and the other in the San Juan River (rural), taking into account the pollution sources that arrive at these stations and previous physicochemical characterizations. Extracellular protease, acid phosphatase, alkaline phosphatase, lipase, and catalase activities in water and sediments were determined and correlated with contaminants of emerging concern determined by liquid chromatography with mass spectrometry. This study evidenced the presence of different pharmaceutical contaminants found in the categories of antihypertensives, stimulants, anti-inflammatories, and antibiotics in both rivers. Concentrations of contaminants of emerging concern were greater in the Almendares River compared to the San Juan River. In addition, through the canonical redundancy analysis, the influence of these contaminants on the extracellular enzymatic activities of microbial communities was documented, where in most cases they inhibit protease, phosphatase, and lipase activities and enhance catalase activity in response to oxidative stress. The present investigation constitutes the first report in Cuba of the presence of pharmaceutical contaminants of emerging concern and one of the few works that exist in the Latin American region.

Genotypic and phenotypic characterization of
Bacillus
spp. from polluted freshwater has been poorly addressed. The objective of this research was to determine the diversity and enzymatic ...potentialities of
Bacillus
spp. strains isolated from the Almendares River.
Bacilli
strains from a polluted river were characterized by considering the production of extracellular enzymes using API ZYM. 14 strains were selected and identified using 16S rRNA,
gyrB
and
aroE
genes. Genotypic diversity of the
Bacillus
spp. strains was evaluated using pulsed field gel electrophoresis. Furthermore, the presence of genetic determinants of potential virulence toxins of the
Bacillus cereus
group and proteinaceous crystal inclusions of
Bacillus thuringiensis
was determined. 10 strains were identified as
B. thuringiensis
, two as
Bacillus megaterium
, one as
Bacillus pumilus
and one as
Bacillus subtilis
. Most strains produced proteases, amylases, phosphatases, esterases, aminopeptidases and glucanases, which reflect the abundance of biopolymeric matter in Almendares River. Comparison of the typing results revealed a spatio-temporal distribution among
B. thuringiensis
strains along the river. The results of the present study highlight the genotypic and phenotypic diversity of
Bacillus
spp. strains from a polluted river, which contributes to the knowledge of genetic diversity of
Bacilli
from tropical polluted freshwater ecosystems.
