This bibliographic review provides an overview of techniques used to detect marine litter using remote sensing. The review classified studies in terms of platform (satellite, aircrafts, drones), ...sensors (passive or active), spectral (visible, infrared, microwaves), spatial resolution (<1 to >30 m), type and size (macroplastics, microplastics), or classification methodology (sighting, photointerpretation, supervised). Most studies applied satellite information to address marine litter using multi- and hyper- spectral optical sensors. The correspondence analysis on analyzed variables exhibited that aircrafts with high spatial resolution (<3 m) with optical sensors (λ = 400 to 2500 nm) seem to be the most optimum combination to target marine litter, while satellites carrying Synthetic Aperture Radar (SAR) sensors (λ = 3.1 to 5.6 cm) may detect sea-slicks associated to surfactants that might contain high concentration of microplastics. Gaps indicate that future goals in marine litter detection should be addressed with platforms including optical and SAR sensors.

-acetylation of chitin by chitinases and deacetylases generates a series of derivatives including chitosans and chitooligosaccharides (COS), which are involved in molecular ...recognition events such as modulation of cell signaling and morphogenesis, immune responses, and host-pathogen interactions. Chitosans and COS are also attractive scaffolds for the development of bionanomaterials for drug/gene delivery and tissue engineering applications. Most of the biological activities associated with COS seem to be largely dependent not only on the degree of polymerization but also on the acetylation pattern, which defines the charge density and distribution of GlcNAc and GlcNH₂ moieties in chitosans and COS. -acetylases (CDAs) catalyze the hydrolysis of the acetamido group in GlcNAc residues of chitin, chitosan, and COS. The deacetylation patterns are diverse, some CDAs being specific for single positions, others showing multiple attack, processivity or random actions. This review summarizes the current knowledge on substrate specificity of bacterial and fungal CDAs, focusing on the structural and molecular aspects of their modes of action. Understanding the structural determinants of specificity will not only contribute to unravelling structure-function relationships, but also to use and engineer CDAs as biocatalysts for the production of tailor-made chitosans and COS for a growing number of applications.
Surface water availability is a fundamental environmental variable to implement effective climate adaptation and mitigation plans, as expressed by scientific, financial and political stakeholders. ...Recently published requirements urge the need for homogenised access to long historical records at a global scale, together with the standardised characterisation of the accuracy of observations. While satellite altimeters offer world coverage measurements, existing initiatives and online platforms provide derived water level data. However, these are sparse, particularly in complex topographies. This study introduces a new methodology in two steps (1) teroVIR, a virtual station extractor for a more comprehensive global and automatic monitoring of water bodies, and (2) teroWAT, a multi-mission, interoperable water level processor, for handling all terrain types. L2 and L1 altimetry products are used, with state-of-the-art retracker algorithms in the methodology. The work presents a benchmark between teroVIR and current platforms in West Africa, Kazakhastan and the Arctic: teroVIR shows an unprecedented increase from 55% to 99% in spatial coverage. A large-scale validation of teroWAT results in an average of unbiased root mean square error ubRMSE of 0.638 m on average for 36 locations in West Africa. Traditional metrics (ubRMSE, median, absolute deviation, Pearson coefficient) disclose significantly better values for teroWAT when compared with existing platforms, of the order of 8 cm and 5% improved respectively in error and correlation. teroWAT shows unprecedented excellent results in the Arctic, using an L1 products-based algorithm instead of L2, reducing the error by almost 4 m on average. To further compare teroWAT with existing methods, a new scoring option, teroSCO, is presented, measuring the quality of the validation of time series transversally and objectively across different strategies. Finally, teroVIR and teroWAT are implemented as platform-agnostic modules and used by flood forecasting and river discharge methods as relevant examples. A review of various applications for miscellaneous end-users is given, tackling the educational challenge raised by the community.
Bacillus subtilis PdaC (BsPdaC) is a membrane-bound, multidomain peptidoglycan N-deacetylase acting on N-acetylmuramic acid (MurNAc) residues and conferring lysozyme resistance to modified cell wall ...peptidoglycans. BsPdaC contains a C-terminal family 4 carbohydrate esterase (CE4) catalytic domain, but unlike other MurNAc deacetylases, BsPdaC also has GlcNAc deacetylase activity on chitooligosaccharides (COSs), characteristic of chitin deacetylases. To uncover the molecular basis of this dual activity, here we determined the X-ray structure of the BsPdaC CE4 domain at 1.54 Å resolution and analyzed its mode of action on COS substrates. We found that the minimal substrate is GlcNAc3 and that activity increases with the degree of glycan polymerization. COS deacetylation kinetics revealed that BsPdaC operates by a multiple-chain mechanism starting at the internal GlcNAc units and leading to deacetylation of all but the reducing-end GlcNAc residues. Interestingly, BsPdaC shares higher sequence similarity with the peptidoglycan GlcNAc deacetylase SpPgdaA than with other MurNAc deacetylases. Therefore, we used ligand-docking simulations to analyze the dual GlcNAc- and MurNAc-binding specificities of BsPdaC and compared them with those of SpPgdA and BsPdaA, representing peptidoglycan deacetylases highly specific for GlcNAc or MurNAc residues, respectively. BsPdaC retains the conserved Asp-His-His metal-binding triad characteristic of CE4 enzymes acting on GlcNAc residues, differing from MurNAc deacetylases that lack the metal-coordinating Asp residue. BsPdaC contains short loops similar to those in SpPgdA, resulting in an open binding cleft that can accommodate polymeric substrates. We propose that PdaC is the first member of a new subclass of peptidoglycan MurNAc deacetylases.
Chitin deacetylases (CDAs) act on chitin polymers and low molecular weight oligomers producing chitosans and chitosan oligosaccharides. Structurally-defined, partially deacetylated ...chitooligosaccharides produced by enzymatic methods are of current interest as bioactive molecules for a variety of applications. Among Pochonia chlamydosporia (Pc) annotated CDAs, gene pc_2566 was predicted to encode for an extracellular CE4 deacetylase with two CBM18 chitin binding modules. Chitosan formation during nematode egg infection by this nematophagous fungus suggests a role for their CDAs in pathogenicity. The P. chlamydosporia CDA catalytic domain (PcCDA) was expressed in E. coli BL21, recovered from inclusion bodies, and purified by affinity chromatography. It displays deacetylase activity on chitooligosaccharides with a degree of polymerization (DP) larger than 3, generating mono- and di-deacetylated products with a pattern different from those of closely related fungal CDAs. This is the first report of a CDA from a nematophagous fungus. On a DP5 substrate, PcCDA gave a single mono-deacetylated product in the penultimate position from the non-reducing end (ADAAA) which was then transformed into a di-deacetylated product (ADDAA). This novel deacetylation pattern expands our toolbox of specific CDAs for biotechnological applications, and will provide further insights into the determinants of substrate specificity in this family of enzymes.
Remote sensing, and particularly Sinthetic Aperture Radar (SAR) is a potential tool to monitor the presence of plastic on the ocean surface, and AI algorithms can learn to find patterns in the ...satellite signals that lead to authomatic detection of plastic patches. The main challenge to train AI algorithms for this purpose is the lack of insitu ground truthdata that allow to reliably label the images. This paper describes a first pilot of binary pixel classification (plastic vs. non-plastic) in SAR imagery using Random Forest, GaussianNaïve Bayes and Support Vector Machines classifiers. It also describes the strategy used to generate ground truth data using Sentinel-1 and Sentinel-2 in the Balearic Islands.
Inhibiting MYC has long been considered unfeasible, although its key role in human cancers makes it a desirable target for therapeutic intervention. One reason for its perceived undruggability was ...the fear of catastrophic side effects in normal tissues. However, we previously designed a dominant-negative form of MYC called Omomyc and used its conditional transgenic expression to inhibit MYC function both in vitro and in vivo. MYC inhibition by Omomyc exerted a potent therapeutic impact in various mouse models of cancer, causing only mild, well-tolerated, and reversible side effects. Nevertheless, Omomyc has been so far considered only a proof of principle. In contrast with that preconceived notion, here, we show that the purified Omomyc mini-protein itself spontaneously penetrates into cancer cells and effectively interferes with MYC transcriptional activity therein. Efficacy of the Omomyc mini-protein in various experimental models of non-small cell lung cancer harboring different oncogenic mutation profiles establishes its therapeutic potential after both direct tissue delivery and systemic administration, providing evidence that the Omomyc mini-protein is an effective MYC inhibitor worthy of clinical development.
Recently, intermittent fasting has gained relevance as a strategy to lose weight and improve health as an alternative to continuous caloric restriction. However, the metabolic impact and the ...sex-related differences are not fully understood. The study aimed to compare the response to a continuous or intermittent caloric restriction in male and female rats following a previous induction of obesity through a cafeteria diet by assessing changes in body weight, energy intake, metabolic parameters, and gene expression in liver hepatic and adipose tissue. The continuous restriction reduced the energy available by 30% and the intermittent restriction consisted of a 75% energy reduction on two non-consecutive days per week. The interventions reduced body weight and body fat in both sexes, but the loss of WAT in females was more marked in both models of caloric restriction, continuous and intermittent. Both caloric restrictions improved insulin sensitivity, but more markedly in females, which showed a more pronounced decrease in HOMA-IR score and an upregulation of hepatic IRS2 and Sirt1 gene expression that was not observed in males. These findings suggest the fact that females are more sensitive than males to reduced caloric content in the diet.
Objective:
To assess the outcome of adolescents with anorexia nervosa (AN) about 20 years after first treatment.
Methods:
Sixty-two women diagnosed with AN during adolescence were invited to ...participate. Of these 62 patients, 38 agreed to participate and were assessed with a battery of questionnaires and interviews. A control group of 30 women of similar age was also assessed.
Results:
Of the patients who completed the full assessment, 13 (34%) presented some degree of eating disorder (ED) at follow-up (10 (26%) met full Diagnostic and Statistical Manual of Mental Disorders (5th ed.; DSM-5) criteria for an ED and 3 (8%) showed partial remission of an ED). The remaining 25 (66%) patients had fully recovered from AN. The duration of untreated illness before admission was significantly associated with an increased risk of a current ED (odds ratio (OR) = 3.334 (1.3–8.7); p = .014). Of the patients who had recovered totally from their ED, 24% showed another psychiatric disorder. This percentage rose to 70% in patients with a current ED.
Conclusion:
Sixty-six percent of adolescents who completed the assessment achieved remission of their AN. Comorbidity was more common in the current ED group. The variable that best predicted complete remission was the number of years without treatment, showing the importance of detection and early intervention.
