Aims: To examine the effects of five inhibitors of methanogenesis, 2‐bromoethanesulphonate (BES), 3‐bromopropanesulphonate (BPS), lumazine, propynoic acid and ethyl 2‐butynoate, on CH4 production of ...the ruminal methanogens Methanobrevibacter ruminantium, Methanosarcina mazei and Methanomicrobium mobile.
Methods and Results: Methanogens were grown in MS medium including 25% (v/v) clarified ruminal fluid. Methane production was measured after 4 and 6 days of incubation. Methanobrevibacter ruminantium was the most sensitive species to BES, propynoic acid and ethyl 2‐butynoate. Methanosarcina mazei was the least sensitive species to those chemical additives, and Mm. mobile was intermediate. BPS failed to inhibit any of the methanogens. All three species were almost completely inhibited by 50‐ and 100%‐lumazine saturated media, but the inhibition was somewhat lower with a 25%‐lumazine saturated media.
Conclusions: There were important differences among species of methanogens regarding their sensitivity to the different inhibitors. In general, Ms. mazei was the most resistant to inhibitors, Mb. ruminantium the least resistant, and Mm. mobile was intermediate.
Significance and Impact of the Study: Differences among methanogens regarding their resistance to chemical inhibitors should be considered when designing strategies of inhibition of ruminal methanogenesis, as selection of resistant species may result.
In full and partial vertical profiles of the electric field in thunderstorms during the Severe Thunderstorm Electrification and Precipitation Study in 2000, we find evidence for inverted‐polarity ...electrical structures in the convective region of thunderstorms. The evidence consists of polarities opposite from normal (1) in the peaks in the profile of the vertical component of the electric field, Ez, and (2) in the vertical sequence of inferred charges. In storms that possibly are inverted in polarity, the lowest peak in Ez inside the cloud is negative, followed by a positive peak farther up. Near storm top, the uppermost peak is negative, and the uppermost inferred charge is positive. The sounding data do not conclusively prove that inverted‐polarity thunderstorms exist, but they support it. If additional data corroborate that such storms do exist, it will require modification of at least some aspects of our conceptual models of storm electrification.

Background Preterm delivery remains the leading cause of perinatal mortality. Risk factors and biomarkers have traditionally failed to identify the majority of preterm deliveries. Objective To ...develop and validate a mass spectrometry–based serum test to predict spontaneous preterm delivery in asymptomatic pregnant women. Study Design A total of 5501 pregnant women were enrolled between 170/7 and 286/7 weeks gestational age in the prospective Proteomic Assessment of Preterm Risk study at 11 sites in the United States between 2011 and 2013. Maternal blood was collected at enrollment and outcomes collected following delivery. Maternal serum was processed by a proteomic workflow, and proteins were quantified by multiple reaction monitoring mass spectrometry. The discovery and verification process identified 2 serum proteins, insulin-like growth factor–binding protein 4 (IBP4) and sex hormone–binding globulin (SHBG), as predictors of spontaneous preterm delivery. We evaluated a predictor using the log ratio of the measures of IBP4 and SHBG (IBP4/SHBG) in a clinical validation study to classify spontaneous preterm delivery cases (<370/7 weeks gestational age) in a nested case-control cohort different from subjects used in discovery and verification. Strict blinding and independent statistical analyses were employed. Results The predictor had an area under the receiver operating characteristic curve value of 0.75 and sensitivity and specificity of 0.75 and 0.74, respectively. The IBP4/SHBG predictor at this sensitivity and specificity had an odds ratio of 5.04 for spontaneous preterm delivery. Accuracy of the IBP4/SHBG predictor increased using earlier case-vs-control gestational age cutoffs (eg, <350/7 vs ≥350/7 weeks gestational age). Importantly, higher-risk subjects defined by the IBP4/SHBG predictor score generally gave birth earlier than lower-risk subjects. Conclusion A serum-based molecular predictor identifies asymptomatic pregnant women at risk of spontaneous preterm delivery, which may provide utility in identifying women at risk at an early stage of pregnancy to allow for clinical intervention. This early detection would guide enhanced levels of care and accelerate development of clinical strategies to prevent preterm delivery.
The field program of the Thunderstorm Electrification and Lightning Experiment (TELEX) took place in central Oklahoma, May–June 2003 and 2004. It aimed to improve understanding of the ...interrelationships among microphysics, kinematics, electrification, and lightning in a broad spectrum of storms, particularly squall lines and storms whose electrical structure is inverted from the usual vertical polarity. The field program was built around two permanent facilities: the KOUN polarimetric radar and the Oklahoma Lightning Mapping Array. In addition, balloon-borne electric-field meters and radiosondes were launched together from a mobile laboratory to measure electric fields, winds, and standard thermodynamic parameters inside storms. In 2004, two mobile C-band Doppler radars provided high-resolution coordinated volume scans, and another mobile facility provided the environmental soundings required for modeling studies. Data were obtained from 22 storm episodes, including several small isolated thunderstorms, mesoscale convective systems, and supercell storms. Examples are presented from three storms. A heavy-precipitation supercell storm on 29 May 2004 produced greater than three flashes per second for 1.5 h. Holes in the lightning density formed and dissipated sequentially in the very strong updraft and bounded weak echo region of the mesocyclone. In a small squall line on 19 June 2004, most lightning flashes in the stratiform region were initiated in or near strong updrafts in the convective line and involved positive charge in the upper part of the radar bright band. In a small thunderstorm on 29 June 2004, lightning activity began as polarimetric signatures of graupel first appeared near lightning initiation regions.

and Remote Ionospheric Sensing (IRIS) suite, complementary to NRL sensors, and comprising three highly miniaturised payloads provided to Dstl by University College London (UCL), University of Bath, and University of Surrey/Surrey Satellite Technology Ltd (SSTL). One IRIS suite will be flown on each satellite, and incorporates an ion/neutral mass spectrometer, a tri-band global positioning system (GPS) receiver for ionospheric remote sensing, and a radiation environment monitor. From the US, NRL have provided two 1U Triple Tiny Ionospheric Photometers (Tri-TIPs) on each satellite (Nicholas et al., 2019), observing the ultraviolet 135.6 nm emission of atomic oxygen at night-time to characterize the two-dimensional distribution of electrons.
Abstract
Balloon soundings were made through two supercell storms during the Severe Thunderstorm Electrification and Precipitation Study (STEPS) in summer 2000. Instruments measured the vector ...electric field, temperature, pressure, relative humidity, and balloon location. For the first time, soundings penetrated both the strong updraft and the rainy downdraft region of the same supercell storm. In both storms, the strong updraft had fewer vertically separated charge regions than found near the rainy downdraft, and the updraft’s lowest charge was elevated higher, its bottom being near the 40-dBZ boundary of the weak-echo vault. The simpler, elevated charge structure is consistent with the noninductive graupel–ice mechanism dominating charge generation in updrafts. In the weak-echo vault, the amount of frozen precipitation and the time for particle interactions are too small for significant charging. Inductive charging mechanisms and lightning may contribute to the additional charge regions found at lower altitudes outside the updraft. Lightning mapping showed that the in-cloud channels of a positive ground flash could be in any one of the three vertically separated positive charge regions found outside the updraft, but were in the middle region, at 6–8 km MSL, for most positive ground flashes. The observations are consistent with the electrical structure of these storms having been inverted in polarity from that of most storms elsewhere. It is hypothesized that the observed inverted-polarity cloud flashes and positive ground flashes were caused by inverted-polarity storm structure, possibly due to a larger than usual rime accretion rate for graupel in a strong updraft.
On 28-29 June 2004 a multicellular thunderstorm west of Oklahoma City, Oklahoma, was probed as part of the Thunderstorm Electrification and Lightning Experiment field program. This study makes use of ...radar observations from the Norman, Oklahoma, polarimetrie Weather Surveillance Radar-1988 Doppler, three-dimensional lightning mapping data from the Oklahoma Lightning Mapping Array (LMA), and balloon-borne vector electric field meter (EFM) measurements. The storm had a low flash rate (30 flashes in 40 min). Four charge regions were inferred from a combination of LMA and EFM data. Lower positive charge near 4 km and midlevel negative charge from 4.5 to 6 km MSL (from 0' to - 6.5'C) were generated in and adjacent to a vigorous updraft pulse. Further midlevel negative charge from 4.5 to 6 km MSL and upper positive charge from 6 to 8 km (from -6.5' to -19'C) were generated later in quantity sufficient to initiate lightning as the updraft decayed. A negative screening layer was present near the storm top (8.5 km MSL, - 25'C). Initial lightning flashes were between lower positive and midlevel negative charge and started occurring shortly after a cell began lofting hydrometeors into the mixed phase region, where graupel was formed. A leader from the storm's first flash avoided a region where polarimetrie radar suggested wet growth and the resultant absence of noninductive charging of those hydrometeors. Initiation locations of later flashes that propagated into the upper positive charge tracked the descending location of a polarimetrie signature of graupel. As the storm decayed, electric fields greater than 160 kV msup -1 exceeded the minimum threshold for lightning initiation suggested by the hypothesized runaway breakdown process at 5.5 km MSL, but lightning did not occur. The small spatial extent (asymptotically =100 m) of the large electric field may not have been sufficient to allow runaway breakdown to fully develop and initiate lightning. PUBLICATION ABSTRACT
Several lines of evidence suggest that norepinephrine (NE) can modulate seizure activity. However, the experimental methods used in the past cannot exclude the possible role of other ...neurotransmitters coreleased with NE from noradrenergic terminals. We have assessed the seizure susceptibility of genetically engineered mice that lack NE. Seizure susceptibility was determined in the dopamine beta-hydroxylase null mutant (Dbh -/-) mouse using four different convulsant stimuli: 2,2,2-trifluroethyl ether (flurothyl), pentylenetetrazol (PTZ), kainic acid, and high-decibel sound. Dbh -/- mice demonstrated enhanced susceptibility (i.e., lower threshold) compared with littermate heterozygous (Dbh +/-) controls to flurothyl, PTZ, kainic acid, and audiogenic seizures and enhanced sensitivity (i.e., seizure severity and mortality) to flurothyl, PTZ, and kainic acid. c-Fos mRNA expression in the cortex, hippocampus (CA1 and CA3), and amygdala was increased in Dbh -/- mice in association with flurothyl-induced seizures. Enhanced seizure susceptibility to flurothyl and increased seizure-induced c-fos mRNA expression were reversed by pretreatment with L-threo-3, 4-dihydroxyphenylserine, which partially restores the NE content in Dbh -/- mice. These genetically engineered mice confirm unambiguously the potent effects of the noradrenergic system in modulating epileptogenicity and illustrate the unique opportunity offered by Dbh -/- mice for elucidating the pathways through which NE can regulate seizure activity.

Development of the body plan is controlled by large networks of regulatory genes. A gene regulatory network that controls the specification of endoderm and mesoderm in the sea urchin embryo is ...summarized here. The network was derived from large-scale perturbation analyses, in combination with computational methodologies, genomic data, cis-regulatory analysis, and molecular embryology. The network contains over 40 genes at present, and each node can be directly verified at the DNA sequence level by cis-regulatory analysis. Its architecture reveals specific and general aspects of development, such as how given cells generate their ordained fates in the embryo and why the process moves inexorably forward in developmental time.
