Patches of riparian woody vegetation potentially help mitigate environmental impacts of agriculture and safeguard biodiversity. We investigated the effects of riparian forest on invertebrate ...diversity in coupled stream-riparian networks using a case study in the Zwalm river basin (Flanders, Belgium). Agriculture is one of the main pressures in the basin and riparian forest is limited to a number of isolated patches. Our 32 study sites comprised nine unshaded “unbuffered” sites which were paired with nine shaded “buffered” sites on the same stream reach, along with five ‘least-disturbed’ sites and nine downstream sites. We sampled water chemistry, habitat characteristics and stream and riparian invertebrates (carabid beetles and spiders) at each site. Three methods were used to quantify riparian attributes at different spatial scales: a visually-assessed qualitative index, quantitative estimates of habitat categories in six rectangular plots (10 × 5 m) and geographic information system (GIS)-derived land cover data. We investigated relationships between invertebrates and riparian attributes at different scales with linear regression and redundancy analyses. Spiders and carabids were most associated with local riparian attributes. In contrast, aquatic macroinvertebrates were strongly influenced by the extent of riparian vegetation in a riparian band upstream (100–300 m). These findings demonstrate the value of quantifying GIS-based metrics of riparian cover over larger spatial scales into assessments of the efficacy of riparian management as a complement to more detailed local scale riparian assessments in situ. Our findings highlight the value of even small patches of riparian vegetation in an otherwise extensively disturbed landscape in supporting biodiversity of both terrestrial and freshwater invertebrates and emphasize the need to consider multiple spatial scales in riparian management strategies which aim to mitigate human impacts on biodiversity in stream-riparian networks.
Developing a general, predictive understanding of ecological systems requires knowing how much structural and functional relationships can cross scales and contexts. Here, we introduce the CROSSLINK ...project that investigates the role of forested riparian buffers in modified European landscapes by measuring a wide range of ecosystem attributes in stream-riparian networks. CROSSLINK involves replicated field measurements in four case-study basins with varying levels of human development: Norway (Oslo Fjord), Sweden (Lake Mälaren), Belgium (Zwalm River), and Romania (Argeş River). Nested within these case-study basins include multiple, independent stream-site pairs with a forested riparian buffer and unbuffered section located upstream, as well as headwater and downstream sites to show cumulative land-use impacts. CROSSLINK applies existing and bespoke methods to describe habitat conditions, biodiversity, and ecosystem functioning in aquatic and terrestrial habitats. Here, we summarize the approaches used, detail protocols in supplementary materials, and explain how data is applied in an optimization framework to better manage tradeoffs in multifunctional landscapes. We then present results demonstrating the range of riparian conditions present in our case-study basins and how these environmental states influence stream ecological integrity with the commonly used macroinvertebrate Average Score Per Taxon (ASPT) index. We demonstrate that a qualitative index of riparian integrity can be positively associated with stream ecological status. This introduction to the CROSSLINK project shows the potential for our replicated study with its panoply of ecosystem attributes to help guide management decisions regarding the use of forested riparian buffers in human-impacted landscapes. This knowledge is highly relevant in a time of rapid environmental change where freshwater biodiversity is increasingly under pressure from a range of human impacts that include habitat loss, pollution, and climate change.
Water quality assessments provide essential information for protecting aquatic habitats and stakeholders downstream of mining sites. Moreover, mining companies must comply with environmental quality ...standards and include public participation in water quality monitoring (WQM) practices. However, overarching challenges beyond corporate environmental responsibility are the scientific soundness, political relevance and harmonization of WQM practices. In this study, a mountainous watershed supporting large-scale gold mining in the headwaters, besides urban and agricultural landuses at lower altitudes, is assessed in the dry season. Conventional physicochemical and biological (Biological Monitoring Water Party-Colombia index) freshwater quality parameters were evaluated, including hydromorphological and land-use characteristics. According to the indicators used, water quality deterioration by mining was absent, in contrast to the effects of urban economic activities, hydromorphological alterations and (less important) agricultural pollutants. We argue that mining impacts are hardly captured due to the limited ecological knowledge of high-mountain freshwaters, including uncharacterized mining-specific bioindicators, environmental baselines and groundwater processes, as well as ecotoxicological and microbial freshwater quality components. Lessons for overcoming scientific and operational challenges are drawn from joint efforts among governments, academia and green economy competitiveness. Facing a rapid development of extractive industries, interinstitutional and multidisciplinary collaborations are urgently needed to implement more integrated freshwater quality indicators of complex mining impacts.
Although pesticides are frequently used for agriculture in the Galapagos Islands (Ecuador), there are, to date, no investigations of pesticide occurrences in its coastal waters. We examined the ...presence of pesticide residues in the coastal waters of urban areas in two islands of the Galapagos archipelago using a repeated sampling design. Quantification was performed by solid-phase extraction, followed by chemical analysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS) and gas chromatography-electron capture detector (GC-ECD). The diversity and concentration of pesticide residues in Santa Cruz island were higher compared to Isabela island. In total, sixteen pesticides were detected, including three persistent organic pollutants. Carbendazim (23.93 μg·L−1), cadusafos (4.74 μg·L−1), DDT (2.99 μg·L−1), diuron (1.61 μg·L−1) and aldrin (1.55 μg·L−1) were detected with the highest concentrations between samples. Repetitions in locations show that concentrations of pesticide residues varied considerably in space and time. Comparison with local products indicated agricultural activities on the islands as a possible source. Furthermore, evaluation through ecological risk quotients showed that the observed concentration levels of seven pesticides pose a relatively high risk for three biotic groups (i.e. algae, invertebrates and fishes). Taken together, this study provides insights into the need to regulate, monitor and assess the presence of pesticides in the islands. At a global scale, this study is moreover valuable for the many islands that are facing the same challenges.

Oceanic islands harbor unique yet fragile marine ecosystems that require evidence-based environmental management. Among these islands, the Galapagos archipelago is well known for its fish diversity, ...but the factors that structure communities within and between its islands remain poorly understood. In this study, water quality, physical habitats and geographical distance were assessed as potential predictors for the diversity and structure of fish assemblages. Differences in the structure of fish assemblages of the two studied islands (Santa Cruz and Floreana) were most likely driven by temperature and nutrient concentrations. In the relatively highly populated island Santa Cruz, the structure of fish assemblages was more affected by water conditions than physical habitats while the contrary was true for the more pristine area of Floreana. A wide variety of species with different geographical origins were distributed over the different islands, which indicates that most fish species are able to reach the islands of the archipelago. However, temperature gradients and elevated nutrient levels cause large differences in the structure of local fish assemblages. In addition, in Santa Cruz nutrient concentrations were negatively correlated with α diversity. Since pollution is a clear pressure on the fish assemblages of oceanic islands, environmental management of the coastal areas is of paramount importance.

Polyunsaturated fatty acids (PUFAs) have key biological roles in fish cells. We recently showed that the phospholipid composition of rainbow trout liver cells (RTL-W1 cell line) modulates their tolerance to an acute cadmium (Cd) challenge. Here, we investigated (i) the extent to which PUFAs and Cd impact fatty acid homeostasis and metabolism in these cells and (ii) possible mechanisms by which specific PUFAs may confer cytoprotection against Cd. First, RTL-W1 cells were cultivated for one week in growth media spiked with 50 μmol L−1 of either alpha-linolenic acid (ALA, 18:3n-3), eicosapentaenoic acid (EPA, 20:5n-3), linoleic acid (LA, 18:2n-6) or arachidonic acid (AA, 20:4n-6) in order to modulate their fatty acid profile. Then, the cells were challenged with Cd (0, 50 or 100 μmol L−1) for 24 h prior to assaying viability, fatty acid profile, intracellular Cd content, proteomic landscape and expression levels of genes involved in fatty acid metabolism, synthesis of PUFA-derived signalling molecules and stress response. We observed that the fatty acid supply and, to a lesser extent, the exposure to Cd influenced cellular fatty acid homeostasis and metabolism. The cellular fatty acid composition of fish liver cells modulated their tolerance to an acute Cd challenge. Enrichments in ALA, EPA, and, to a lesser extent, AA conferred cytoprotection while enrichment in LA had no impact on cell viability. The present study ruled out the possibility that cytoprotection reflects a decreased Cd burden. Our results rather suggest that the PUFA-derived cytoprotection against Cd occurs through a reduction of the oxidative stress induced by Cd and a differential induction of the eicosanoid cascade, with a possible role of peroxiredoxin and glutaredoxin (antioxidant enzymes) as well as cytosolic phospholipase A2 (enzyme initiating the eicosanoid cascade).

The present study aimed at investigating interactive effects between dietary lipids and both short- and long-term exposures to a low, environmentally realistic, cadmium (Cd) concentration. Juvenile rainbow trout were fed four isolipidic diets (31.7 g/kg) enriched in either linoleic acid (LA, 18:2n-6), alpha-linolenic acid (ALA, 18:3n-3), eicosapentaenoic acid (EPA, 20:5n-3) or docosahexaenoic acid (DHA, 22:6n-3). From the 4th week of this 10-week experiment, the lipid level of the diet was increased (120.0 g/kg) and half of the fish fed each diet were aqueously exposed to Cd (0.3 μg/L) while the other half were not exposed to Cd (control). Fish were sampled and their liver was harvested for fatty acid profile, hepatic Cd and calcium concentrations, total glutathione level and gene expression assessment, either (i) after 4 weeks of feeding and 24 h of Cd contamination (day 29) (short-term Cd exposure) or (ii) after 10 weeks of feeding and 6 weeks of Cd contamination (day 70) (long-term Cd exposure). We found that both dietary lipids and Cd exposure influenced fatty acid homeostasis and metabolism. The hepatic fatty acid profile mostly reflected that of the diet (e.g. n-3/n-6 ratio) with some differences, including selective retention of specific long chain polyunsaturated fatty acids (LC-PUFAs) like DHA and active biotransformation of dietary LA and ALA into LC-PUFAs. Cd effects on hepatic fatty acid profiles were influenced by the duration of the exposure and the nutritional status of the fish. The effects of diet and Cd exposure on the fatty acid profiles were only sparsely explained by variation of the expression pattern of genes involved in fatty acid metabolism. The biological responses to Cd were also influenced by dietary lipids. Fish fed the ALA-enriched diet seemed to be the least affected by the Cd exposure, as they showed a higher detoxifying ability against Cd with an early upregulation of protective metallothionein a (MTa) and apoptosis regulator BCL2-Like1 (BCLx) genes, an increased long-term phospholipid synthesis and turnover and fatty acid bioconversion efficiency, as well as a lower long-term accumulation of Cd in their liver. In contrast, fish fed the EPA-enriched diet seemed to be the most sensitive to a long-term Cd exposure, with an impaired growth performance and a decreased antioxidant capacity (lower glutathione level). Our results highlight that low, environmentally realistic aqueous concentrations of Cd can affect biological response in fish and that these effects are influenced by the dietary fatty acid composition.
Riparian forest buffers have multiple benefits for biodiversity and ecosystem services in both freshwater and terrestrial habitats but are rarely implemented in water ecosystem management, partly ...reflecting the lack of information on the effectiveness of this measure. In this context, social learning is valuable to inform stakeholders of the efficacy of riparian vegetation in mitigating stream degradation. We aim to develop a Bayesian belief network (BBN) model for application as a learning tool to simulate and assess the reach- and segment-scale effects of riparian vegetation properties and land use on instream invertebrates. We surveyed reach-scale riparian conditions, extracted segment-scale riparian and subcatchment land use information from geographic information system data, and collected macroinvertebrate samples from four catchments in Europe (Belgium, Norway, Romania, and Sweden). We modelled the ecological condition based on the Average Score Per Taxon (ASPT) index, a macroinvertebrate-based index widely used in European bioassessment, as a function of different riparian variables using the BBN modelling approach. The results of the model simulations provided insights into the usefulness of riparian vegetation attributes in enhancing the ecological condition, with reach-scale riparian vegetation quality associated with the strongest improvements in ecological status. Specifically, reach-scale buffer vegetation of score 3 (i.e. moderate quality) generally results in the highest probability of a good ASPT score (99–100%). In contrast, a site with a narrow width of riparian trees and a small area of trees with reach-scale buffer vegetation of score 1 (i.e. low quality) predicts a high probability of a bad ASPT score (74%). The strengths of the BBN model are the ease of interpretation, fast simulation, ability to explicitly indicate uncertainty in model outcomes, and interactivity. These merits point to the potential use of the BBN model in workshop activities to stimulate key learning processes that help inform the management of riparian zones.

Long chain polyunsaturated fatty acids (LC-PUFAs) such as eicosapentaenoic acid (EPA, 20:5n-3) affect zooplankton fitness and ability to cope with environmental stressors. However, the impact of ...LC-PUFAs on zooplankton sensitivity to chemical stressors is unknown. Here, we aimed to document the interaction between EPA and cadmium (Cd), as model chemical stressor, in Daphnia magna. A life-history experiment was performed in which daphnid neonates were raised into adulthood on three diets of different lipid composition: (i) algae mix; (ii) algae mix supplemented with control liposomes; (iii) algae mix supplemented with liposomes containing EPA. Juveniles (3rd, 4th and 5th brood) released by daphnids during this life-history experiment were sampled, challenged with Cd during 48 h and their immobility was assessed. At the end of this life-history experiment, another immobilisation test was performed with adults from each treatment. Daphnids absorbed, incorporated and transferred ingested EPA to their offspring. Liposome feeding increased adult tolerance to Cd. The presence of EPA in liposomes did not increase adult tolerance to Cd. Offspring's tolerance to Cd was influenced by the brood number and the maternal diet. It was positively correlated with the PUFA level in body neutral lipids, especially alpha-linolenic acid (ALA, 18:3n-3) and negatively correlated with the saturated fatty acid level in body neutral lipids, especially stearic acid (18:0). Overall, these results emphasize the importance of dietary lipids and maternal transfer of body lipids in D. magna sensitivity to Cd and highlight the need to take into account these parameters in ecotoxicological studies and risk assessment.

Few studies have been done to evaluate the transfer of catabolic plasmids from an introduced donor strain to indigenous microbial populations as a means to remediate contaminated soils. In this work ...we determined the effect of the conjugative transfer of two 2,4-D degradative plasmids to indigenous soil bacterial populations on the rate of 2,4-D degradation in soil. We also assessed the influence of the presence of 2,4-D on the number of transconjugants formed. The two plasmids used, pEMT1k and pEMT3k, encode 2,4-D degradative genes (tfd) that differ in DNA sequence as well as gene organisation, and confer different growth rates to Ralstonia eutropha JMP228 when grown with 2,4-D as a sole carbon source. In an agricultural soil (Ardoyen) treated with 2,4-D (100 ppm) there were ca. 10(7) CFU of transconjugants per gram bearing pEMT1k as well as a high number of pEMT3k bearing transconjugants (ca 10(6) CFU/g). In this soil the formation of a high number of 2,4-D degrading transconjugants resulted in faster degradation of 2,4-D as compared to the uninoculated control soil. In contrast, only transconjugants with pEMT1k were detected (at a level of ca. 10(3) CFU/g soil) in the untreated Ardoyen soil. High numbers of transconjugants that carried pEMT1k were also found in a second experiment done using forest soil (Lembeke) treated with 100 ppm 2,4-D. However, unlike in the Ardoyen soil, no transconjugants with pEMT3k were detected and the transfer of plasmid pEMT1k to indigenous bacteria did not result in a higher rate of decrease of 2,4-D. This may be because 2,4-D was readily metabolised by indigenous bacteria in this soil. The results indicate that bioaugmentation with catabolic plasmids may be a viable means to enhance the bioremediation of soils which lack an adequate intrinsic ability to degrade a given xenobiotic.
