Abstract
Variability is a powerful tool to investigate properties of X-ray binaries (XRB), in particular for Ultraluminous X-ray sources (ULXs) that are mainly detected in the X-ray band. For most ...ULXs the nature of the accretor is unknown, although a few ULXs have been confirmed to be accreting at super-Eddington rates on to a neutron star (NS). Monitoring these sources is particularly useful both to detect transients and to derive periodicities, linked to orbital and super-orbital modulations. Here, we present the results of our monitoring campaign of the galaxy NGC 925, performed with the Neil Gehrels Swift Observatory. We also include archival and literature data obtained with Chandra, XMM–Newton, and NuSTAR. We have studied spectra, light curves, and variability properties on days to months time-scales. All the three ULXs detected in this galaxy show flux variability. ULX-1 is one of the most luminous ULXs known, since only 10 per cent of the ULXs exceed a luminosity of ∼5 × 1040 erg s−1, but despite its high flux variability we found only weak spectral variability. We classify it as in a hard ultraluminous regime of super-Eddington accretion. ULX-2 and ULX-3 are less luminous but also variable in flux and possibly also in spectral shape. We classify them as in between the hard and the soft ultraluminous regimes. ULX-3 is a transient source: by applying a Lomb–Scargle algorithm, we derive a periodicity of ∼126 d, which could be associated with an orbital or superorbital origin.

The absence of spin pulsations when the broad complex is detected suggests that the mechanism responsible for the aperiodic modulation also dampens the spin signal's pulsed fraction. If true, this represents an additional obstacle in the detection of new PULXs, suggesting an even larger occurrence of PULXs among ULXs.

Ultraluminous X-ray sources (ULXs) are extragalactic X-ray emitters located off-center of their host galaxy and with a luminosity in excess of a few \({10^{39}\text{ erg s}^{-1}}\), if emitted ...isotropically. The discovery of periodic modulation revealed that in some ULXs the accreting compact object is a neutron star, indicating luminosities substantially above their Eddington limit. The most extreme object in this respect is \({NGC 5907~ULX-1}\) (ULX1), with a peak luminosity that is 500 times its Eddington limit. During a Chandra observation to probe a low state of ULX1, we detected diffuse X-ray emission at the position of ULX1. Its diameter is \(2.7 \pm 1.0\) arcsec and contains 25 photons, none below 0.8 keV. We interpret this extended structure as an expanding nebula powered by the wind of ULX1. Its diameter of about \({200\text{ pc}}\), characteristic energy of \({\sim 1.9\text{ keV}}\), and luminosity of \({\sim 2\times10^{38}\text{ erg s}^{-1}}\) imply a mechanical power of \({1.3\times10^{41}\text{ erg s}^{-1}}\) and an age \({\sim 7 \times 10^{4}\text{ yr}}\). This interpretation suggests that a genuinely super-Eddington regime can be sustained for time scales much longer than the spin-up time of the neutron star powering the system. As the mechanical power from a single ULX nebula can rival the injection rate of cosmic rays of an entire galaxy, ULX nebulae could be important cosmic ray accelerators.
In recent years, the Mediterranean Sea has become an accumulation zone for waste generated by the 22 countries bordering its shores. Although the effects of plastic litter on the marine environment ...and on organisms have recently been studied in other areas, further information is needed for the Mediterranean Sea and, in particular, about plastics additives inputs and interactions with the biota and the trophic network, such as phthalates and bisphenol A. Plastic material production, use and disposal contribute also to the release of heavy metals into the environment, such as mercury (Hg), often used during the production of chlorine, the primary ingredient in PVC, lead (Pb) and cadmium (Cd), which are used as stabilizers in PVC and leach out of products during use and disposal. Our research aims to evaluate phthalates, bisphenol A and heavy metals contamination in
(Pisces, Trichiuridae), which could be considered as a potential sentinel species. For the evaluation of toxicological effects, we evaluated the expression of vitellogenin and metallothioneins 1. In all samples analyzed, we have not found microplastics in the gastrointestinal tract but chemical analysis revealed the presence of high content of phthalates, and in particular high quantities of DIDP, DEHP, bis-benzylester phthalate, bis-butyl ester phthalate and mono-N-butyl ester phthalate in different organs. Instead, trace elements detected in tissue revealed a trend of concentrations generally higher in liver and intestine than gill and muscle tissues. Immunohistochemical analysis for anti-metallothionein 1 antibody showed a strong positivity of liver cells, both in females and males. Analysis for the anti-vitellogenin antibody showed in females a strong positivity both in the liver cells and in the gonads, in male specimens was found to be always negative except for a specimen, in which it was highlighted a positivity in some areas of the liver and of the gonad.
Osteosarcoma (OS) is the most common primary bone tumor in children and adolescent. Surgery and multidrug chemotherapy are the standard of treatment achieving 60-70% of event-free survival for ...localized disease at diagnosis. However, for metastatic disease, the prognosis is dismal. Exploiting immune system activation in the setting of such unfavorable mesenchymal tumors represents a new therapeutic challenge.
In immune competent OS mouse models bearing two contralateral lesions, we tested the efficacy of intralesional administration of a TLR9 agonist against the treated and not treated contralateral lesion evaluating abscopal effect. Multiparametric flow cytometry was used to evaluate changes of the tumor immune microenviroment. Experiments in immune-deficient mice allowed the investigation of the role of adaptive T cells in TLR9 agonist effects, while T cell receptor sequencing was used to assess the expansion of specific T cell clones.
TLR9 agonist strongly impaired the growth of locally-treated tumors and its therapeutic effect also extended to the contralateral, untreated lesion. Multiparametric flow cytometry showed conspicuous changes in the immune landscape of the OS immune microenvironment upon TLR9 engagement, involving a reduction in M2-like macrophages, paralleled by increased infiltration of dendritic cells and activated CD8 T cells in both lesions. Remarkably, CD8 T cells were needed for the induction of the abscopal effect, whereas they were not strictly necessary for halting the growth of the treated lesion. T cell receptor (TCR) sequencing of tumor infiltrating CD8 T cells showed the expansion of specific TCR clones in the treated tumors and, remarkably, their selected representation in the contralateral untreated lesions, providing the first evidence of the rewiring of tumor-associated T cell clonal architectures.
Overall these data indicate that the TLR9 agonist acts as an in situ anti-tumor vaccine, activating an innate immune response sufficient to suppress local tumor growth while inducing a systemic adaptive immunity with selective expansion of CD8 T cell clones, which are needed for the abscopal effect.
Iron toxicity is associated with organ injury and has been reported in various clinical conditions, such as hemochromatosis, thalassemia major, and myelodysplastic syndromes. Therefore, iron ...chelation therapy represents a pivotal therapy for these patients during their lifetime. The aim of the present study was to assess the iron chelating properties of α-lipoic acid (ALA) and how such an effect impacts on iron overload mediated toxicity. Human mesenchymal stem cells (HS-5) and animals (zebrafish,
= 10 for each group) were treated for 24 h with ferric ammonium citrate (FAC, 120 µg/mL) in the presence or absence of ALA (20 µg/mL). Oxidative stress was evaluated by reduced glutathione content, reactive oxygen species formation, mitochondrial dysfunction, and gene expression of heme oxygenase-1b and mitochondrial superoxide dismutase; organ injury, iron accumulation, and autophagy were measured by microscopical, cytofluorimetric analyses, and inductively coupled plasma‒optical mission Spectrometer (ICP-OES). Our results showed that FAC results in a significant increase of tissue iron accumulation, oxidative stress, and autophagy and such detrimental effects were reversed by ALA treatment. In conclusion, ALA possesses excellent iron chelating properties that may be exploited in a clinical setting for organ preservation, as well as exhibiting a good safety profile and low cost for the national health system.
This study investigated the phenolic compounds and antioxidant capacity of fruit extracts of
(Ker Gawl.) Haw.(OD) and
(L.) Mill.(OFI), yellow (F1) and red (F2) varieties. In order to evaluate the ...antioxidant activity of these extracts on human sperm quality after thawing, the semen parameters (vitality, motility, acrosome reaction, oxidative stress, and DNA fragmentation) were analysed after 1 h of exposure. The results showed that OD has higher phenolic content and antioxidant power than OFI, and that they are higher in F2 than F1. Furthermore, regarding the activity of extracts on thawed sperm, the results showed a significant increase in motility in samples treated with OFI F1 and OD extracts, while an improvement in vitality and acrosome reaction and a reduction of DNA fragmentation were observed in all exposed samples compared to the control. Finally, a reduction of oxidative stress was observed in samples exposed to OFI F2 and OD than control.
