Obesity and insulin resistance are the major predisposing factors to comorbidities, such as Type 2 diabetes, nonalcoholic fatty liver disease, cardiovascular and neurodegenerative diseases, and ...several types of cancer. The prevalence of obesity is still increasing worldwide and now affects a large number of individuals. Here, we review the role of the gut microbiota in the pathophysiology of insulin resistance/obesity. The human intestine is colonized by ∼100 trillion bacteria, which constitute the gut microbiota. Studies have shown that lean and overweight rodents and humans may present differences in the composition of their intestinal flora. Over the past 10 years, data from different sources have established a causal link between the intestinal microbiota and obesity/insulin resistance. It is important to emphasize that diet-induced obesity promotes insulin resistance by mechanisms independent and dependent on gut microbiota. In this review, we present several mechanisms that contribute to explaining the link between intestinal flora and insulin resistance/obesity. The LPS from intestinal flora bacteria can induce a chronic subclinical inflammatory process and obesity, leading to insulin resistance through activation of TLR4. The reduction in circulating SCFA may also have an essential role in the installation of reduced insulin sensitivity and obesity. Other mechanisms include effects of bile acids, branched-chain amino acids (BCAA), and some other lesser-known factors. In the near future, this area should open new therapeutic avenues for obesity/insulin resistance and its comorbidities.
Under salt stress conditions, plant growth is reduced due to osmotic, nutritional and oxidative imbalance. However, salicylic acid acts in the mitigation of this abiotic stress by promoting an ...increase in growth, photosynthesis, nitrogen metabolism, synthesis of osmoregulators and antioxidant enzymes. In this context, the objective was to evaluate the effect of salicylic acid doses on the growth and physiological changes of eggplant seedlings under salt stress. The experiment was conducted in a greenhouse, where the treatments were distributed in randomized blocks using a central composite matrix Box with five levels of electrical conductivity of irrigation water (CEw) (0.50; 1.08; 2.50; 3.92 and 4.50 dS m-1), associated with five doses of salicylic acid (SA) (0.00; 0.22; 0.75; 1.28 and 1.50 mM), with four repetitions and each plot composed of three plants. At 40 days after sowing, plant height, stem diameter, number of leaves, leaf area, electrolyte leakage, relative water content, and total dry mass were determined. ECw and SA application influenced the growth and physiological changes of eggplant seedlings. Increasing the ECw reduced growth in the absence of SA. Membrane damage with the use of SA remained stable up to 3.9 dS m-1 of ECw. The relative water content independent of the CEw increased with 1.0 mM of SA. The use of SA at the concentration of 1.0 mM mitigated the deleterious effect of salinity on seedling growth up to 2.50 dS m-1 of ECw.
Low density sugarcane plantation (LDSP) has been implemented by some sugarcane producers in Brazil, aiming to save seeds and operational costs. The study was carried out in the municipality of Areia, ...Paraíba, Brazil. Five planting densities were used, varying from 5 to 25 m-2 of buds arranged in randomized blocks, with four replications. Data were measured annually over three cultivation cycles (2017 to 2020), during which the field was fertilized with NPK and the harvests were carried out manually without prior burning. The lower planting density presents higher productivity only in the cane plant (101.03 t ha-1) due to the higher plant height (2.37 m) and the higher number of stalks (11 stalks m-2), suggesting that these variables are due to the greater availability of light, water and photosynthate. However, there is a drastic reduction in sugarcane yield for this lower population in the 2nd ratoon by up to 65.62%, which is correlated with number of stalks per meter. We demonstrate the agronomic viability of LDSP in the population of 10 buds m-2 in relation to conventional planting of sugarcane until the 2nd ratoon. Data are important for future studies to present additional considerations for other production factors, such as the effects of mechanized harvesting and the management of nutrients and water, assessing the sustainability of this large-scale planting system.
Polystyrene and poly(vinyl alcohol) nanofibres containing cellulose nanocrystals (CNCs) were successfully produced by electrospinning. Knowledge of the local orientation of CNCs in electrospun fibres ...is critical to understand and exploit their mechanical properties. The orientation of CNCs in these electrospun fibres was investigated using transmission electron microscopy (TEM) and Raman spectroscopy. A Raman band located at ~1095 cm⁻¹, associated with the C–O ring stretching of the cellulose backbone, was used to quantify the orientation of the CNCs within the fibres. Raman spectra were fitted using a theoretical model to characterize the extent of orientation. From these data, it is observed that the CNCs have little orientation along the direction parallel to the axis of the fibres. Evidences for both oriented and non-oriented regions of CNCs in the fibres are presented from TEM images of nanofibres. These results contradict previously published work in this area and micromechanical modelling calculations suggest a uniform orientation of CNCs in electrospun polymer fibres. It is demonstrated that this explains why the mechanical properties of electrospun fibre mats containing CNCs are not always the same as that would be expected for a fully oriented system.
This work focuses on active galactic nuclei (AGNs) and on the relation between the sizes of the hot dust continuum and the broad-line region (BLR). We find that the continuum size measured using ...optical/near-infrared interferometry (OI) is roughly twice that measured by reverberation mapping (RM). Both OI and RM continuum sizes show a tight relation with the H
β
BLR size, with only an intrinsic scatter of 0.25 dex. The masses of supermassive black holes (BHs) can hence simply be derived from a dust size in combination with a broad line width and virial factor. Since the primary uncertainty of these BH masses comes from the virial factor, the accuracy of the continuum-based BH masses is close to those based on the RM measurement of the broad emission line. Moreover, the necessary continuum measurements can be obtained on a much shorter timescale than those required monitoring for RM, and they are also more time efficient than those needed to resolve the BLR with OI. The primary goal of this work is to demonstrate a measuring of the BH mass based on the dust-continuum size with our first calibration of the
R

relation. The current limitation and caveats are discussed in detail. Future GRAVITY observations are expected to improve the continuum-based method and have the potential of measuring BH masses for a large sample of AGNs in the low-redshift Universe.
The present study sought to evaluate the antimicrobial and anti-adherent potential of Eucalyptus radiata essential oil against food-borne strains of Escherichia coli. The study was performed using ...the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). In addition, the disk diffusion technique was used to evaluate the association of Eucalyptus radiata essential oil with synthetic antimicrobials. The Minimum Inhibitory Adherence Concentration (MIC) was also performed. The results revealed that E. radiata showed antimicrobial activity against the E. coli strains tested, with MIC values ranging from 500 μg/mL to 1000 μg/mL and MBC values ranging from 500 μg/mL to 1,024 μg/mL. As for the associations, it was observed that E. radiata oil exhibited a synergistic effect for some antibiotics, especially Ceftriaxone, with greater interference from the essential oil. Furthermore, it was effective in inhibiting the adherence of bacterial strains of E. coli, showing a more significant antibiofilm effect than the antibacterial agent 0.12% chlorhexidine digluconate. In summary, the essential oil of E. radiata showed antimicrobial potential against strains of E. coli of food origin, and can therefore, through in-depth studies, be used alone or in association with synthetic antimicrobials to combat infections caused by this pathogen.
Abstract Macrophages are frequently identified in solid tumors, playing important roles in cancer progression. Their remarkable plasticity makes them very sensitive to environmental factors, ...including the extracellular matrix (ECM). In the present work, we investigated the impact of human tumor colorectal matrices on macrophage polarization and on macrophage-mediated cancer cell invasion. Accordingly, we developed an innovative 3D-organotypic model, based on the decellularization of normal and tumor tissues derived from colorectal cancer patients' surgical resections. Extensive characterization of these scaffolds revealed that DNA and other cell constituents were efficiently removed, while native tissue characteristics, namely major ECM components, architecture and mechanical properties, were preserved. Notably, normal and tumor decellularized matrices distinctly promoted macrophage polarization, with macrophages in tumor matrices differentiating towards an anti-inflammatory M2-like phenotype (higher IL-10, TGF-β and CCL18 and lower CCR7 and TNF expression). Matrigel invasion assays revealed that tumor ECM-educated macrophages efficiently stimulated cancer cell invasion through a mechanism involving CCL18. Notably, the high expression of this chemokine at the invasive front of human colorectal tumors correlated with advanced tumor staging. Our approach evidence that normal and tumor decellularized matrices constitute excellent scaffolds when trying to recreate complex microenvironments to understand basic mechanisms of disease or therapeutic resistance.
In this study, videothermometry's application in detecting mammary tumors in dogs is explored in-depth. The research hypothesizes that this technique can effectively identify cancerous tissues during ...surgery by analyzing thermal patterns. The methodology involved comparing thermal imaging results from dogs with palpable mammary nodules against a control group, focusing on capturing real-time thermal patterns. Results were significant, showing distinct thermal patterns in carcinomas. This indicates videothermometry's capability in accurately identifying micro metastases and differentiating between neoplastic and non-neoplastic changes. The study concludes that videothermometry has considerable potential in enhancing surgical precision, especially in tumor resection and safety margin definition, but emphasizes the need for further research to thoroughly understand the thermal signatures of various mammary tumors in dogs.
The yellow fever virus (YFV) epidemic in Brazil is the largest in decades. The recent discovery of YFV in Brazilian
species mosquitos highlights a need to monitor the risk of reestablishment of urban ...YFV transmission in the Americas. We use a suite of epidemiological, spatial, and genomic approaches to characterize YFV transmission. We show that the age and sex distribution of human cases is characteristic of sylvatic transmission. Analysis of YFV cases combined with genomes generated locally reveals an early phase of sylvatic YFV transmission and spatial expansion toward previously YFV-free areas, followed by a rise in viral spillover to humans in late 2016. Our results establish a framework for monitoring YFV transmission in real time that will contribute to a global strategy to eliminate future YFV epidemics.
By using the GRAVITY instrument with the near-infrared (NIR) Very Large Telescope Interferometer (VLTI), the structure of the broad (emission-)line region (BLR) in active galactic nuclei (AGNs) can ...be spatially resolved, allowing the central black hole (BH) mass to be determined. This work reports new NIR VLTI/GRAVITY interferometric spectra for four type 1 AGNs (Mrk 509, PDS 456, Mrk 1239, and IC 4329A) with resolved broad-line emission. Dynamical modelling of interferometric data constrains the BLR radius and central BH mass measurements for our targets and reveals outflow-dominated BLRs for Mrk 509 and PDS 456. We present an updated radius-luminosity (R-L) relation independent of that derived with reverberation mapping (RM) measurements using all the GRAVITY-observed AGNs. We find our R-L relation to be largely consistent with that derived from RM measurements except at high luminosity, where BLR radii seem to be smaller than predicted. This is consistent with RM-based claims that high Eddington ratio AGNs show consistently smaller BLR sizes. The BH masses of our targets are also consistent with the standard M BH - σ * relation. Model-independent photocentre fitting shows spatial offsets between the hot dust continuum and the BLR photocentres (ranging from ∼17 μas to 140 μas) that are generally perpendicular to the alignment of the red- and blueshifted BLR photocentres. These offsets are found to be related to the AGN luminosity and could be caused by asymmetric K -band emission of the hot dust, shifting the dust photocentre. We discuss various possible scenarios that can explain this phenomenon.
