Education and training of surgeons has traditionally focused on the development of individual knowledge, technical skills and decision making. Knowledge about endoscopic instruments is one of the ...core elements of urological training. We assessed the precourse knowledge of newly appointed urology trainees and the impact of boot camp in improving their knowledge.
Newly appointed specialty trainees in urology took part in a pilot 5-day urology simulation boot camp (USBC). The aim of the USBC was to improve their confidence, procedural performance and non-technical skills, with one of the modules looking at the trainees' knowledge about common endoscopic instruments in urology. Delegates were first asked to identify and assemble the instruments, followed by one-to-one teaching about the instruments. An Objective Structured Assessment Tool was used to assess their knowledge in the identification and assembly of the cystoscope, resectoscope and optical urethrotome, before and at the end of the course.
Data of two successive boot camps were collected to assess knowledge of instruments of newly appointed urology trainees. Majority of the trainees had good precourse knowledge of the cystoscopy kit, with 84% able to correctly identify the parts. Seventy-six per cent of candidates were able to identify the resectoscope equipment, but only approximately a third of trainees were able to correctly identify the urethrotome kit. The assembly of cystoscope, resectoscope and urethrotome was performed correctly in 74%, 42% and 32% at baseline and 94%, 90% and 77% postcourse, respectively. Overall performance improved significantly in the postcourse assessment (<0.001).
This urology boot camp has addressed gaps in trainees' core equipment knowledge and guided them to improve their knowledge with respect to identification and assembly of cystoscope, resectoscope and urethrotome.
Francisella tularensis is the causative agent of a fatal human disease, tularemia. F. tularensis was used in bioweapon programs in the past and is now classified as a category A select agent owing to ...its possible use in bioterror attacks. Despite over a century since its discovery, an effective vaccine is yet to be developed. In this study four transposon insertion mutants of F. tularensis live vaccine strain (LVS) in Na/H antiporter (FTL_0304), aromatic amino acid transporter (FTL_0291), outer membrane protein A (OmpA)-like family protein (FTL_0325) and a conserved hypothetical membrane protein gene (FTL_0057) were evaluated for their attenuation and protective efficacy against F. tularensis SchuS4 strain. All four mutants were 100-1000 fold attenuated for virulence in mice than parental F. tularensis. Except for the FTL_0304, single intranasal immunization with the other three mutants provided 100% protection in BALB/c mice against intranasal challenge with virulent F. tularensis SchuS4. Differences in the protective ability of the FTL_0325 and FTL_0304 mutant which failed to provide protection against SchuS4 were investigated further. The results indicated that an early pro-inflammatory response and persistence in host tissues established a protective immunity against F. tularensis SchuS4 in the FTL_0325 immunized mice. No differences were observed in the levels of serum IgG antibodies amongst the two vaccinated groups. Recall response studies demonstrated that splenocytes from the FTL_0325 mutant immunized mice induced significantly higher levels of IFN-γ and IL-17 cytokines than the FTL_0304 immunized counterparts indicating development of an effective memory response. Collectively, this study demonstrates that persistence of the vaccine strain together with its ability to induce an early pro-inflammatory innate immune response and strong memory responses can discriminate between successful and failed vaccinations against tularemia. This study describes a live attenuated vaccine which may prove to be an ideal vaccine candidate for prevention of respiratory tularemia.
In this study, micron-sized flake shaped carbonyl iron (CI) water-based MR fluids were prepared with adding laponite and oleic acid as an additive and surfactant, respectively. The MR suspensions are ...comprised of the fixed CI particles and water weight %, while weight % of laponite and oleic acid changes from 1 to 3 wt% and 0.5 to 1.5 wt%, respectively. The remarkable enhancement in magnetorheological properties was obtained with improved sedimentation stability for CI/water MR suspensions with the addition of laponite and oleic acid. It was found that at the lowest magnetic field strength, the higher laponite concentration is effective, while at the highest magnetic field strength, the smaller concentration was effective. It was because of the combined effect of the field-induced CI chains and the laponite clay gel network. Its storage moduli showed a stable plateau area for whole angular frequencies, suggesting distinguished solid-like behavior of the MR fluid. Finally, a novel correlation was obtained between the initial settling rate of the CI particles and magnetorheological behavior of CI/laponite/OA MR suspensions with 1 wt% laponite and 0.5 wt% oleic acid, which has less zero-field, high on-state shear stress with enhanced sedimentation stability. The prepared MR fluids are a reliable industrial application vibration-isolation, clutch, and brake.

Creep and recovery experiments showed that fluid acts as a linear viscoelastic material at lower stress levels. As the stress levels increase, the contribution of retardation strain and viscous strain decreases, and it acts like nonlinear viscoelastic material. In summary, this work is expected to obtain MR fluid results for application in MR devices under applied strain, frequencies, and constant stress levels.

Francisella tularensis, the causative agent of tularemia, is one of the deadliest agents of biological warfare and bioterrorism. Extremely high virulence of this bacterium is associated with its ability to dampen or subvert host innate immune response. The objectives of this study were to identify factors and understand the mechanisms of host innate immune evasion by F. tularensis. We identified and explored the pathogenic role of a mutant interrupted at gene locus FTL_0325, which encodes an OmpA-like protein. Our results establish a pathogenic role of FTL_0325 and its ortholog FTT0831c in the virulent F. tularensis SchuS4 strain in intramacrophage survival and suppression of proinflammatory cytokine responses. This study provides mechanistic evidence that the suppressive effects on innate immune responses are due specifically to these proteins and that FTL_0325 and FTT0831c mediate immune subversion by interfering with NF-κB signaling. Furthermore, FTT0831c inhibits NF-κB activity primarily by preventing the nuclear translocation of p65 subunit. Collectively, this study reports a novel F. tularensis factor that is required for innate immune subversion caused by this deadly bacterium.
Pseudomonas is an efficient plant growth-promoting rhizobacteria (PGPR); however, intolerance to drought and high temperature limit its application in agriculture as a bioinoculant. Transposon 5 ...(Tn5) mutagenesis was used to generate a stress tolerant mutant from a PGPR Pseudomonas putida NBRI1108 isolated from chickpea rhizosphere. A mutant NBRI1108T, selected after screening of nearly 10,000 transconjugants, exhibited significant tolerance towards high temperature and drought. Southern hybridization analysis of EcoRI and XhoI restricted genomic DNA of NBRI1108T confirmed that it had a single Tn5 insertion. The metabolic changes in the polar and non-polar extracts of NBRI1108 and NBRI1108T were examined using 1H, 31P nuclear magnetic resonance (NMR) spectroscopy and gas chromatography-mass spectrometry (GC-MS). Thirty six chemically diverse metabolites consisting of amino acids, fatty acids and phospholipids were identified and quantified. Insertion of Tn5 influenced amino acid and phospholipid metabolism and resulted in significantly higher concentration of aspartic acid, glutamic acid, glycinebetaine, glycerophosphatidylcholine (GPC) and putrescine in NBRI1108T as compared to that in NBRI1108. The concentration of glutamic acid, glycinebetaine and GPC increased by 34%, 95% and 100%, respectively in the NBRI1108T as compared to that in NBRI1108. High concentration of glycerophosphatidylethanolamine (GPE) and undetected GPC in NBRI1108 indicates that biosynthesis of GPE may have taken place via the methylation pathway of phospholipid biosynthesis. However, high GPC and low GPE concentration in NBRI1108T suggest that methylation pathway and phosphatidylcholine synthase (PCS) pathway of phospholipid biosynthesis are being followed in the NBRI1108T. Application of multivariate principal component analysis (PCA) on the quantified metabolites revealed clear variations in NBRI1108 and NBRI1108T in polar and non-polar metabolites. Identification of abiotic stress tolerant metabolites from the NBRI1108T suggest that Tn5 mutagenesis enhanced tolerance towards high temperature and drought. Tolerance to drought was further confirmed in greenhouse experiments with maize as host plant, where NBRI1108T showed relatively high biomass under drought conditions.
The influenza virus is classified into four types A, B, C, and D, but type A and B are responsible for major illnesses in people with influenza A being the only virus responsible for flu pandemics ...due to the presence of two surface proteins called hemagglutinin (H) and neuraminidase (N) on the virus. The two subtypes of influenza A virus, H1N1 and H3N2, have been known to cause many flu pandemics. Both subtypes change genetically and antigenically to produce variants (clades and subclades, also know as groups and subgroups). H3N2 tends to change rapidly, both genetically and antigenically whereas that of H1N1 generally tends to have smaller changes. Influenza A (H3N2) viruses have evolved to form many separate, genetically different clades that continue to co-circulate. Influenza A(H3N2) viruses have caused significant deaths as per WHO report. The review describes methods for detection of influenza A(H3N2) viruses by conventional serological methods as well as the advanced methods of molecular biology and biosensors. All these methods are based on different parameters and have different targets but the goal is to improve specificity and increase sensitivity. Amongst the molecular methods, real-time polymerase chain reaction (RT-PCR) is considered a gold standard test due to its many advantages whereas a number of other molecular methods are time-consuming, complex to perform or lack specificity. The review also considers bio-sensing methods for simple, rapid, highly sensitive, and specific detection of H3N2. The classification and principle of various H3N2 biosensors are also discussed.
This study deals with the performance modeling and reliability analysis of a redundant machining system composed of several functional machines. To analyze the more realistic scenarios, the concepts ...of switching failure and geometric reneging are included. The time-to-breakdown and repair time of operating and standby machines are assumed to follow the exponential distribution. For the quantitative assessment of the machine interference problem, various performance measures such as mean-time-to-failure, reliability, reneging rate, etc. have been formulated. To show the practicability of the developed model, a numerical illustration has been presented. For the practical justification and validity of the results established, the sensitivity analysis of reliability indices has been presented by varying different system descriptors.
Chromium (VI) is one of the hazardous heavy metal, heavily discharged into the soil and severely hampers the plants yield. The TiO2 NPs was selected due to its potential to alleviate the heavy metals ...toxicity. This manuscript unravels the mechanisms for Cr(VI) induced toxicity and how foliar application of TiO2 NPS potentially ameliorate the toxicity by regulating the photosynthetic attributes, DNA damage, antioxidants defense machinery, and phytochelatins synthesis in Helianthus annuus L. Plants were exposed to Cr(VI) concentrations 0, 15, 30, and 60 mg Cr(VI) kg−1 of soil, and TiO2 NPS (15 mg L−1, 25 nm size) were foliar sprayed thrice to the plants at three days interval. The maximum accumulation of total chromium was recorded in root (12.53 µg g−1 DW) followed by shoot (5.67 µg g−1 DW) at 60 mg Cr(VI) treatment. The presence and localization of TiO2 NPs inside the plant leaf cells were confirmed by TEM-EDS analysis. The results revealed that Cr(VI) exposure had a dose-dependent inhibitory effects on photosynthetic attributes, structure of guard and epidermal cells, photosynthetic pigments; inducing impacts on H2O2 and MDA productions, DNA damage, AsA-GSH cycle, and most importantly on PC2, and PC3 synthesis which is rarely reported. However, TiO2 NPs exposure minimized Cr(VI) induced toxicity through reduction of total chromium accumulation, H2O2 and MDA productions, thereby reducing DNA damage reported first time under combined treatment of Cr(VI)+ TiO2 NPs as evidenced through comet assay. It also positively regulate the photosynthetic pigments, AsA-GSH cycle, and modulates PC2 and PC3 synthesis which have crucial impacts on ROS quenching and Cr(VI) detoxification, respectively, and in turn, minimizes Cr(VI) toxicity in H. annuus L. Besides, this study strengthens the less acknowledged report that Cr(VI) is an inducer of PCs synthesis and also confirms that TiO2 NPs potentially counteract Cr(VI) toxicity.
