Six dogs had perivascular extravasation of antineoplastic agents during IV administration. Treatment with 300 units of hyaluronidase injected locally in the affected site was initiated following the ...extravasation event. Injections were repeated weekly until signs of toxic effects within the tissues resolved satisfactorily. All dogs recovered within 6 weeks, and residual fibrosis at the extravasation sites most was considered minimal. Many chemotherapeutic agents cause severe cytotoxic reactions when extravasated during cycles of chemotherapy, resulting in tissue necrosis with ulceration and sloughing of skin during the following weeks. Surgical treatments and skin grafting are often necessary to achieve healing. The sequelae of extravasation may result in discontinuation of chemotherapy or euthanasia of the animal. Hyaluronidase appears to be a safe treatment for the adverse effects of extravasation of various chemotherapeutic agents and may be used effectively to reduce the severity of cutaneous toxicosis.
A thirteen-year-old female spayed with a history of hydronephrosis was presented for a routine abdominal ultrasonographic exam. The imaging exam showed a mass involving the pancreas and a large mass ...affecting the spleen. Exploratory laparotomy evidenced a mass in the pancreas and another involving one-third of the spleen. The patient had partial pancreatectomy and splenectomy. The histopathology report came back with a diagnosis of pancreatic carcinoma. Adjuvant chemotherapy was declined. The cat is still free of gross tumor recurrence and metastatic disease after twenty-six months. Early diagnosed and aggressively treated feline pancreatic carcinoma might yield a favorable prognosis.

A 10-year-old Great Pyrenees was presented for anorexia and weight loss. On physical examination, the dog was emaciated and showed a large ulcerated lesion on the right lower lip in addition to an ...enlarged right testicle. Fine-needle aspiration biopsy of the testicle and surgical biopsy of the lip lesion were performed; the histopathological report was consistent with metastatic seminoma. The diagnostic and therapeutic approach in this unusual metastatic seminoma is presented and compared to the previous literature. A multimodality therapy consisting of surgery and chemotherapy is proposed for the clinical management of metastatic seminoma in dogs.

A preliminary phase I study on dogs with stage III splenic hemangiosarcoma was designed in order to evaluate the efficacy and toxicity of the protease inhibitor Indinavir to delay the progression of this advanced neoplasm. The results suggest that Indinavir is potentially beneficial in dogs affected by microscopic residual disease.

Primary melanoma of the gastrointestinal tract is a rare, highly malignant neoplasm that is associated with an unfavorable long-term prognosis. Animal studies are needed to further characterize this ...tumor and to develop new and more effective protocols. A spontaneous canine intestinal melanoma is described which, because of its local aggressive behavior and advanced stage, was not treatable with conventional strategies, thus being a potential candidate for investigational trials.
HtrA1 is a serine protease homologue to the bacterial serine-protease HtrA, also known as DegP, a heat shock-induced envelope-associated serine protease. It has been shown that over-expression of ...HtrA1 in human cancer cells inhibits cell growth and proliferation in vitro and in vivo, thus, suggesting a possible role as a tumor suppressor. The expression of HtrA1 was investigated in depth by means of immunohistochemistry in a large group of human lung cancer specimens and corresponding lymph node metastases. Univariate analysis showed, that the only statistically significant correlation was found between the HtrA1 expression level detected in the primary tumors and in the lymph node metastases. This result was also confirmed when the analysis was restricted only to the cases where both the primary tumor and the autologous lymph node metastasis were available. Our data suggest that HtrA1 may be involved in lung cancer progression by targeting several molecular pathways.
Canine apocrine gland carcinoma is a locally aggressive neoplasm that can occasionally lead to metastatic spread, thus mimicking the behavior of their human counterpart. In this paper we describe the ...successful treatment of a cervical metastatic spread of this neoplasia by using mitoxantrone selectively driven within the tumor cells by trains of biphasic pulses. The dog experienced tumor reduction from the first cycle of electrochemotherapy (ECT) and complete remission by the time of its fourth session. Neither systemic or local toxicities were detected during the whole course of therapy. The dog is in complete remission after six months from his last treatment. Electrochemotherapy is a safe and efficacious therapy for metastatic carcinoma and warrants further investigation.
