Nitrogen (N), phosphorus (P) and potassium (K) budgets were calculated for nine organic farms in the UK. The farms were situated on sandy loams, silty clay loams and silty loams over chalk with ...stockless farming systems and cattle, pig and poultry enterprises with a significant proportion of arable cropping. A soil surface nutrient budget was calculated for the target rotation on each farm using information about field management and measurements of the soil, crops and manure. Losses of N through leaching and volatilization were calculated independently using the NITCAT and MANNER models. Nutrient budgets for seven of the farm rotations showed an N surplus, six a P surplus and three a K surplus. The ratio of N inputs supplied in the form of biological fixation : manure : atmospheric deposition was approximately 2 : 2 : 1 for stocked systems and 2 : 0 : 1 for stockless systems. Phosphorus surpluses resulted from supplementary P fertilizer (rock phosphate) and additional feed for non-ruminant livestock. The stockless system without P fertilizer resulted in a large P deficit and stocked systems, which relied on recycling manure alone, a small P deficit. Only rotations with large returns of manure or imported feed showed a K surplus or a balanced K budget.
We collated contact tracing data from COVID-19 clusters in Singapore and Tianjin, China and estimated the extent of pre-symptomatic transmission by estimating incubation periods and serial intervals. ...The mean incubation periods accounting for intermediate cases were 4.91 days (95%CI 4.35, 5.69) and 7.54 (95%CI 6.76, 8.56) days for Singapore and Tianjin, respectively. The mean serial interval was 4.17 (95%CI 2.44, 5.89) and 4.31 (95%CI 2.91, 5.72) days (Singapore, Tianjin). The serial intervals are shorter than incubation periods, suggesting that pre-symptomatic transmission may occur in a large proportion of transmission events (0.4-0.5 in Singapore and 0.6-0.8 in Tianjin, in our analysis with intermediate cases, and more without intermediates). Given the evidence for pre-symptomatic transmission, it is vital that even individuals who appear healthy abide by public health measures to control COVID-19.

°C. We aimed to clarify the mechanisms of short-term (2-weeks) nitrogen (N) cycling processes and microbial community composition in relation to dissolved organic carbon (DOC) and N (DON) availability and selected soil properties. N cycling was measured by
15N pool dilution and microbial community composition by denaturing gradient gel electrophoresis (DGGE), phospholipid fatty acid (PLFA) and community level physiological profiles (CLPP). Soil DOC increased in the order of arable<grassland<forest soil while DON and gross N fluxes increased in the order of forest<arable<grassland soil; land use had no affect on respiration rate. Soil DOC was lower, while respiration, DON and gross N fluxes were higher at 25 than 5
°C. Gross N fluxes, respiration and bacterial biomass were all positively correlated with each other. Gross N fluxes were positively correlated with pH and DON, and negatively correlated with organic matter, fungal biomass, DOC and DOC/DON ratio. Respiration rate was positively correlated with bacterial biomass, DON and DOC/DON ratio. Multiple linear modelling indicated that soil pH, organic matter, bacterial biomass, DON and DOC/DON ratio were important in predicting gross N mineralization. Incubation temperature, pH and total-C were important in predicting gross nitrification, while gross N mineralization, gross nitrification and pH were important in predicting gross N immobilization. Permutation multivariate analysis of variance indicated that DGGE, CLPP and PLFA profiles were all significantly

affected by land use and incubation temperature. Multivariate regressions indicated that incubation temperature, pH and organic matter content were important in predicting DGGE, CLPP and PLFA profiles. PLFA and CLPP were also related to DON, DOC, ammonium and nitrate contents. Canonical correlation analysis showed that PLFA and CLPP were related to differences in the rates of gross N mineralization, gross nitrification and soil respiration. Our study indicates that vegetation type and/or management practices which control soil pH and mediate dissolved organic matter availability were important predictors of gross N fluxes and microbial composition in this short-term experiment.
ERA-Interim/Land is a global land surface reanalysis data set covering the period 1979-2010. It describes the evolution of soil moisture, soil temperature and snowpack. ERA-Interim/Land is the result ...of a single 32-year simulation with the latest ECMWF (European Centre for Medium-Range Weather Forecasts) land surface model driven by meteorological forcing from the ERA-Interim atmospheric reanalysis and precipitation adjustments based on monthly GPCP v2.1 (Global Precipitation Climatology Project). The horizontal resolution is about 80 km and the time frequency is 3-hourly. ERA-Interim/Land includes a number of parameterization improvements in the land surface scheme with respect to the original ERA-Interim data set, which makes it more suitable for climate studies involving land water resources. The quality of ERA-Interim/Land is assessed by comparing with ground-based and remote sensing observations. In particular, estimates of soil moisture, snow depth, surface albedo, turbulent latent and sensible fluxes, and river discharges are verified against a large number of site measurements. ERA-Interim/Land provides a global integrated and coherent estimate of soil moisture and snow water equivalent, which can also be used for the initialization of numerical weather prediction and climate models.
To improve cancer therapy, it is critical to target metastasizing cells. Circulating tumor cells (CTCs) are rare cells found in the blood of patients with solid tumors and may play a key role in ...cancer dissemination. Uncovering CTC phenotypes offers a potential avenue to inform treatment. However, CTC transcriptional profiling is limited by leukocyte contamination; an approach to surmount this problem is single cell analysis. Here we demonstrate feasibility of performing high dimensional single CTC profiling, providing early insight into CTC heterogeneity and allowing comparisons to breast cancer cell lines widely used for drug discovery.
We purified CTCs using the MagSweeper, an immunomagnetic enrichment device that isolates live tumor cells from unfractionated blood. CTCs that met stringent criteria for further analysis were obtained from 70% (14/20) of primary and 70% (21/30) of metastatic breast cancer patients; none were captured from patients with non-epithelial cancer (n = 20) or healthy subjects (n = 25). Microfluidic-based single cell transcriptional profiling of 87 cancer-associated and reference genes showed heterogeneity among individual CTCs, separating them into two major subgroups, based on 31 highly expressed genes. In contrast, single cells from seven breast cancer cell lines were tightly clustered together by sample ID and ER status. CTC profiles were distinct from those of cancer cell lines, questioning the suitability of such lines for drug discovery efforts for late stage cancer therapy.
For the first time, we directly measured high dimensional gene expression in individual CTCs without the common practice of pooling such cells. Elevated transcript levels of genes associated with metastasis NPTN, S100A4, S100A9, and with epithelial mesenchymal transition: VIM, TGFß1, ZEB2, FOXC1, CXCR4, were striking compared to cell lines. Our findings demonstrate that profiling CTCs on a cell-by-cell basis is possible and may facilitate the application of 'liquid biopsies' to better model drug discovery.
The top priority of fire management agencies in Canada is to protect human life and property. Here we investigate if decades of aggressive fire suppression in the boreal biome of Canada has reduced ...the proportion of recently burned forests (RBF; <30 years) near human communities, and thereby inadvertently increased the risk of wildfire. We measured the percentage of RBF, which are usually less flammable than older forests, up to a 25-km radius around communities compared to that in the surrounding regional fire regime zone. Our analysis of 160 communities across boreal Canada shows that 54.4% exhibited a deficit or lack of RBF, whereas only 15.0% showed a surplus. Overall, a majority (74.4%) of communities are surrounded by a low (≤10%) proportion of RBF, indicating a higher vulnerability of those communities to wildfire. These findings suggest that suppression policies are increasing flammability in the wildland-urban interface of boreal Canada.
A diverse range of materials and properties are exhibited by layered perovskites. We report on the synthesis, characterization, and computational investigation of a new ferroelectricCsBi2Ti2NbO10an ...n = 3 member of the Dion–Jacobson (DJ) family. Structural studies using variable-temperature neutron powder diffraction indicate that a combination of octahedral rotations and polar displacements result in the polar structure. Density functional theory calculations reveal that the wider perovskite blocks in CsBi2Ti2NbO0 stabilize proper ferroelectricity, in contrast to the hybrid-improper ferroelectricity reported for all other DJ phases. Our results raise the possibility of a new class of proper ferroelectric materials analogous to the well-known Aurivillius phases.
This meta‐analytic review of 232 effect sizes showed that, across five categories, attitudes were more negative toward older than younger adults. Perceived age differences were largest for age ...stereotypes and smallest for evaluations. As predicted by social role theory (Eagly, 1987), effect sizes were reduced when detailed information was provided about the person being rated. The double standard of aging emerged for evaluations and behavior/behavioral intentions, but was reversed for the competence category. Perceptions depended on respondent age also. Results demonstrated both the multi‐dimensionality and the complexity of attitudes toward older adults (Hummert, 1999; Kite & Wagner, 2002).
COVID-19 remains a major public health concern, with large resurgences even where there has been widespread uptake of vaccines. Waning immunity and the emergence of new variants will shape the ...long-term burden and dynamics of COVID-19. We explore the transition to the endemic state, and the endemic incidence in British Columbia (BC), Canada and South Africa (SA), to compare low and high vaccination coverage settings with differing public health policies, using a combination of modelling approaches. We compare reopening (relaxation of public health measures) gradually and rapidly as well as at different vaccination levels. We examine how the eventual endemic state depends on the duration of immunity, the rate of importations, the efficacy of vaccines and the transmissibility. These depend on the evolution of the virus, which continues to undergo selection. Slower reopening leads to a lower peak level of incidence and fewer overall infections in the wave following reopening: as much as a 60% lower peak and a 10% lower total in some illustrative simulations; under realistic parameters, reopening when 70% of the population is vaccinated leads to a large resurgence in cases. The long-term endemic behaviour may stabilize as late as January 2023, with further waves of high incidence occurring depending on the transmissibility of the prevalent variant, duration of immunity, and antigenic drift. We find that long term endemic levels are not necessarily lower than current pandemic levels: in a population of 100,000 with representative parameter settings (Reproduction number 5, 1-year duration of immunity, vaccine efficacy at 80% and importations at 3 cases per 100K per day) there are over 100 daily incident cases in the model. Predicted prevalence at endemicity has increased more than twofold after the emergence and spread of Omicron. The consequent burden on health care systems depends on the severity of infection in immunized or previously infected individuals.
