In no antibiotics ever (NAE) broilers, enteric diseases pose a threat to intestinal health and generally welfare, which can be exacerbated because of stocking density. Through knowledge of litter ...condition and management, disease can be minimized, and broiler welfare can be improved. To evaluate how stocking density influences NAE broilers raised in conventional housing, we evaluated production traits for broilers raised at two stocking densities within a single commercial house. Over the course of 4 flocks, 78,960 Cobb 500 broilers were raised in an industry-style tunnel ventilated house. The house was divided into four equally sized pens, each representing one of two stocking densities. An industry standard stocking density (SSD; 0.23 m2 per bird) and low stocking density (LSD; 0.27 m2 per bird) were each assigned to two pens per flock and were alternated for each subsequent flock raised. Litter moisture content, body weight, mortality, and feed conversion (FCR) were evaluated and averaged over all four flocks for both stocking densities. Data were analyzed in JMP with an ANOVA, and means were separated by Tukey's honestly significant difference. As expected, the litter moisture content was significantly larger in the more densely packed SSD pens at weeks 2, 3, 4, and 6, totaling 0.242 moisture content on average at week 6 vs. 0.217 in LSD pens at the same flock age (P = 0.035). Weekly body weight and final flock FCR were not significantly impacted by stocking density. No association was observed in mortality between the broilers raised in SSD and LSD. The results from this study indicate that the two densities examined were comparable in their growth and efficiency. Additional management pressure would exist to handle the increase in litter moisture in flocks placed at SSD in a production setting; although, raised in the same barn at the same time, the impact of SSD vs. LSD was minimal in this study.
Increased demand in consumer choice has resulted in a wide variety of egg selection available in the retail market. Specialty and designer chicken eggs represent a portion of the table egg market ...that is increasing in size. Egg quality is known to be of great importance in all eggs as it relates to food safety, consumer preferences, and product value. In this study, egg quality characteristics were analyzed using a one-way ANOVA to evaluate 2 commercially available conventional egg brands (A and B) and 4 commercially available designer egg brands (C–F). Three hundred nine eggs were evaluated for shell and content weight, dimensional measurements, and breakage force. Calculations were completed to determine %yolk and albumen, yolk index, and Haugh units (HU), followed by an accelerated lipid oxidation study. No significant variation exists in breakage force. Brands A–E meet AA grade standard at a score of 72 HU or above, while brand F, a pasture-raised brand, meets the A grade standard, falling between 60 and 71 HU. Brand F has the highest yolk fan color value (10.41 ± 0.193, P < 0.001) and the lowest yolk index (0.523 ± 0.013, P < 0.05). In addition, brand F has the lowest albumen height (P < 0.001). As albumen height is an indication of freshness, and as all eggs were of equivalent age, it is possible that brand F exhibits overall lower quality than other brands. The conventionally raised white eggs of A experienced the greatest increase in % free fatty acids, which would likely result in off-flavors from hydrolytic rancidity. The organic cage-free D eggs have a significantly greater peroxide value (17.3 ± 2.9, P < 0.001), relative to all other brands, and is over the 10 mEg/kg threshold, which would be considered an unsuitable product for consumption. Ultimately, the measures of egg quality used in this study are essential for evaluating the delivery of the specialty market to the consumer and may indicate that improved measures of quality are needed to truly differentiate between the different egg types and their quality.
The major histocompatibility complex (MHC) is a highly polymorphic region on chromosome 16, which contains numerous immune response genes, and is known to influence disease susceptibility and ...resistance in chickens. Variability of MHC-B haplotypes in various well-known and commercially utilized breeds has previously been identified. This study aims to understand MHC-B diversity in the Silkie breed using a high-density SNP panel that encompasses the chicken MHC-B region. DNA was obtained from 74 females and 27 males from a commercial Silkie breeder colony that is maintained through minimal genetic selection practices. A previously described panel of 90 SNPs, all located within the MHC-B region, was used to evaluate MHC-B variability in the commercial Silkie breeder colony. MHC-B haplotypes identified from the individual SNP information in the Silkie colony were compared to published haplotypes from the same region. Of the 27 haplotypes identified in the Silkie population, 8 have been previously described. Nineteen haplotypes are unique to the Silkie population and include one novel recombinant and 2 additional possible novel recombinants. Six haplotypes were found at a frequency greater than 5% of the population, of which 4 are novel. Finally, Hardy Weinberg Equilibrium (HWE) was calculated for the observed haplotypes, which were found to be in HWE. This study shows considerable MHC-B diversity in the Silkie breed and adds further information on variability of the MHC-B region in the chicken.
Inhaled mucoactive agents are used in respiratory disease to improve mucus properties and enhance secretion clearance. The effect of mannitol, recombinant human deoxyribonuclease/dornase alfa ...(rhDNase) and hypertonic saline (HS) or normal saline (NS) are not well described in chronic lung conditions other than cystic fibrosis (CF). The aim of this review was to determine the benefit and safety of inhaled mucoactive agents outside of CF. We searched Medline, Embase, CINAHL and CENTRAL for randomized controlled trials investigating the effects of mucoactive agents on lung function, adverse events (AEs), health-related quality of life (HRQOL), hospitalization, length of stay, exacerbations, sputum clearance and inflammation. There were detrimental effects of rhDNase in bronchiectasis, with average declines of 1.9-4.3% in forced expiratory volume in 1 s (FEV
) and 3.7-5.4% in forced vital capacity (FVC) (n = 410, two studies), and increased exacerbation risk (relative risk = 1.35, 95% CI = 1.01-1.79 n = 349, one study). Some participants exhibited a reduction in FEV
(≥10-15%) with mucoactive agents on screening (mannitol = 158 of 1051 participants, rhDNase = 2 of 30, HS = 3 of 80). Most AEs were mild and transient, including bronchospasm, cough and breathlessness. NS eased symptomatic burden in COPD, while NS and HS improved spirometry, HRQOL and sputum burden in non-CF bronchiectasis. Mannitol improved mucociliary clearance in asthma and bronchiectasis, while the effects of N-acetylcysteine were unclear. In chronic lung diseases outside CF, there are small benefits of mannitol, NS and HS. Adverse effects of rhDNase suggest this should not be administered in non-CF bronchiectasis.
Clostridioides difficile spores were previously demonstrated to survive industrial laundering. Understanding interactions between heat, disinfectants and soiling (e.g. bodily fluids) affecting C. ...difficile spore survival could inform the optimization of healthcare laundry processes. Reducing spore attachment to linen could also enhance laundering efficacy. This study aimed to compare the sensitivity of C. difficile spores to heat and detergent, with and without soiling and to investigate adherence to cotton. Survival of C. difficile spores exposed to industrial laundering temperatures (71–90°C), reference detergent and industrial detergent was quantified with and without soiling. The adherence to cotton after 0 and 24 h air drying was determined with the exosporium of C. difficile spores partially or fully removed. Clostridioides difficile spores were stable at 71°C for 20 min (≤0·37 log10 reduction) while 90°C was sporicidal (3 log10 reduction); soiling exerted a protective effect. Industrial detergent was more effective at 71°C compared to 25°C (2·81 vs 0·84 log10 reductions), however, specifications for sporicidal activity (>3 log10 reduction) were not met. Clostridioides difficile spores increasingly adhered to cotton over time, with 49% adherence after 24 h. Removal of the exosporium increased adherence by 19–23% compared to untreated spores. Further understanding of the role of the exosporium in attachment to cotton could enhance spore removal and aid decontamination of linen.
Significance and Impact of the Study: Clostridioides difficile spores survive on healthcare linens during industrial laundering. This study demonstrates that C. difficile spores are resistant to thermal and chemo‐thermal disinfection parameters recommended for healthcare laundry in the UK. The interactions between soiling, heat and disinfectants on C. difficile spore inactivation were established, which may aid in optimization of healthcare laundry processes. Clostridioides difficile spores attached to cotton over time, with the exosporium playing a role in adherence. These findings may inform novel strategies to prevent attachment to textiles, improving spore removal from linen by detergents during laundering.
: An increasing global burden of geriatric hip fractures is anticipated. The appropriate treatment for fractures is of ongoing interest and becoming more relevant with an aging population and finite ...health resources. Trochanteric fractures constitute approximately half of all hip fractures with the medial calcar critical to fracture stability. In the management of unstable trochanteric fractures, it is assumed that intramedullary nails and longer implants will lead to less failure. However, the lack of power, inclusion of older generation femoral nails, and a variable definition of stability complicate interpretation of the literature.
: Between January 2012 and December 2017, a retrospective analysis of operatively treated geriatric trochanteric hip fracture patients were examined at a Level 1 Trauma Centre. The treatment was with a long and short version of one type of trochanteric nail. Unstable trochanteric fractures with medial calcar comminution were examined (AO31A2.3, 2.3 & 3.3). The length of the medial calcar loss, nail length, demographics, fracture morphology, and relevant technical factors were examined in univariate and multivariate analysis using competing risk regression analysis. The primary outcome was failure of fixation with post-operative death the competing event and powered to previously reported failure rates.
: Unstable patterns with medial calcar comminution loss constituted 617 (56%) of operatively treated trochanteric fractures. Failure occurred in 16 (2.6%) at a median post-operative time of 111 days (40-413). In univariate and multivariate analysis, only younger age was a significant predictor of failure (years; SHR: 0.91, CI 95%: 0.86-0.96,
< 0.001). Nail length, medial calcar loss, varus reduction, and other technical factors did not influence nail failure.
: In a cohort of unstable geriatric trochanteric hip fractures with medial calcar insufficiency, only younger patient age was predictive of nail failure. Neither the length of the medial calcar fragment or nail was predictive of failure.
Ascites syndrome is the most severe manifestation of pulmonary hypertension in fast-growing broilers. The disease can be attributed to increased body weights of birds, where the higher metabolic load ...is not matched by sufficient oxygen supply to the cells and tissues. Although there are environmental components, the disease exhibits moderate to high heritability. The current study uses high throughput whole genome resequencing (WGR) to identify genes and chromosomal regions associated with ascites.
The WGR data identified the CPQ gene on chromosome 2. The association was confirmed by genotyping a large collection of DNAs from phenotyped birds from three distinct broiler lines using SNPs in intron 6 and exon 8 of the CPQ gene. By combining the genotype data for these two SNP loci, we identified three different alleles segregating in the three broiler lines. Particular genotypes could be associated with resistance to ascites. We further determined that particular genotypes most associated with resistance overexpress CPQ mRNA in three tissues which might explain the role of these alleles in contributing to resistance.
Our findings indicate CPQ is an important determinant of pulmonary hypertension syndrome leading to ascites in broilers. We identified particular SNPs that can be used for marker-assisted selection of broilers for resistance to the disease. Our findings validate WGR as a highly efficient approach to map determinants contributing to complex phenotypic or disease-related traits. The CPQ gene has been associated with pulmonary hypertension in genome-wide association studies in humans. Therefore, ascites investigations in broilers are likely to provide insights into some forms of hypertension in humans.
We present the Migdal In Galactic Dark mAtter expLoration (MIGDAL) experiment aiming at the unambiguous observation and study of the so-called Migdal effect induced by fast-neutron scattering. It is ...hoped that this elusive atomic process can be exploited to enhance the reach of direct dark matter search experiments to lower masses, but it is still lacking experimental confirmation. Our goal is to detect the predicted atomic electron emission which is thought to accompany nuclear scattering with low, but calculable, probability, by deploying an Optical Time Projection Chamber filled with a low-pressure gas based on CF4. Initially, pure CF4 will be used, and then in mixtures containing other elements employed by leading dark matter search technologies — including noble species, plus Si and Ge. High resolution track images generated by a Gas Electron Multiplier stack, together with timing information from scintillation and ionisation readout, will be used for 3D reconstruction of the characteristic event topology expected for this process — an arrangement of two tracks sharing a common vertex, with one belonging to a Migdal electron and the other to a nuclear recoil. Different energy-loss rate distributions along both tracks will be used as a powerful discrimination tool against background events. In this article we present the design of the experiment, informed by extensive particle and track simulations and detailed estimations of signal and background rates. In pure CF4 we expect to observe 8.9 (29.3) Migdal events per calendar day of exposure to an intense D–D (D–T) neutron generator beam at the NILE facility located at the Rutherford Appleton Laboratory (UK). With our nominal assumptions, 5σ median discovery significance can be achieved in under one day with either generator.
