Bacteria within family S24-7 (phylum Bacteroidetes) are dominant in the mouse gut microbiota and detected in the intestine of other animals. Because they had not been cultured until recently and the ...family classification is still ambiguous, interaction with their host was difficult to study and confusion still exists regarding sequence data annotation.
We investigated family S24-7 by combining data from large-scale 16S rRNA gene analysis and from functional and taxonomic studies of metagenomic and cultured species.
A total of 685 species was inferred by full-length 16S rRNA gene sequence clustering. While many species could not be assigned ecological habitats (93,045 samples analyzed), the mouse was the most commonly identified host (average of 20% relative abundance and nine co-occurring species). Shotgun metagenomics allowed reconstruction of 59 molecular species, of which 34 were representative of the 16S rRNA gene-derived species clusters. In addition, cultivation efforts allowed isolating five strains representing three species, including two novel taxa. Genome analysis revealed that S24-7 spp. are functionally distinct from neighboring families and versatile with respect to complex carbohydrate degradation.
We provide novel data on the diversity, ecology, and description of bacterial family S24-7, for which the name Muribaculaceae is proposed.
The dynamics of energetic particles in strong electromagnetic fields can be heavily influenced by the energy loss arising from the emission of radiation during acceleration, known as radiation ...reaction. When interacting with a high-energy electron beam, today’s lasers are sufficiently intense to explore the transition between the classical and quantum radiation reaction regimes. We present evidence of radiation reaction in the collision of an ultrarelativistic electron beam generated by laser-wakefield acceleration (ϵ>500MeV) with an intense laser pulse (a0>10). We measure an energy loss in the postcollision electron spectrum that is correlated with the detected signal of hard photons (γrays), consistent with a quantum description of radiation reaction. The generatedγrays have the highest energies yet reported from an all-optical inverse Compton scattering scheme, with critical energyϵcrit>30MeV.
Alzheimer’s disease (AD) is the leading cause of dementia and the most common neurodegenerative disease in the elderly. Furthermore, AD has provided the most positive indication to support the fact ...that inflammation contributes to neurodegenerative disease. The exact etiology of AD is unknown, but environmental and genetic factors are thought to contribute, such as advancing age, family history, presence of chronic diseases such as cardiovascular disease (CVD) and diabetes, and poor diet and lifestyle. It is hypothesised that early prevention or management of inflammation could delay the onset or reduce the symptoms of AD. Normal physiological changes to the brain with ageing include depletion of long chain omega-3 fatty acids and brains of AD patients have lower docosahexaenoic acid (DHA) levels. DHA supplementation can reduce markers of inflammation. This review specifically focusses on the evidence in humans from epidemiological, dietary intervention, and supplementation studies, which supports the role of long chain omega-3 fatty acids in the prevention or delay of cognitive decline in AD in its early stages. Longer term trials with long chain omega-3 supplementation in early stage AD are warranted. We also highlight the importance of overall quality and composition of the diet to protect against AD and dementia.
Ambient concentrations of ice-forming particles measured during ship expeditions are collected and summarised with the aim of determining the spatial distribution and variability in ice nuclei in ...oceanic regions.
The presented data from literature and previously unpublished data from over 23 months of ship-based measurements stretch from the Arctic to the Southern Ocean and include a circumnavigation of Antarctica. In comparison to continental observations, ship-based measurements of ambient ice nuclei show 1 to 2 orders of magnitude lower mean concentrations. To quantify the geographical variability in oceanic areas, the concentration range of potential ice nuclei in different climate zones is analysed by meridionally dividing the expedition tracks into tropical, temperate and polar climate zones. We find that concentrations of ice nuclei in these meridional zones follow temperature spectra with similar slopes but vary in absolute concentration. Typically, the frequency with which specific concentrations of ice nuclei are observed at a certain temperature follows a log-normal distribution. A consequence of the log-normal distribution is that the mean concentration is higher than the most frequently measured concentration. Finally, the potential contribution of ship exhaust to the measured ice nuclei concentration on board research vessels is analysed as function of temperature. We find a sharp onset of the influence at approximately −36 ∘C but none at warmer temperatures that could bias ship-based measurements.
As the Arctic warms at twice the global rate, radiative feedbacks from clouds will lead to compounding impacts on the surface energy budget that affect both regional and global weather, and climate. ...In a future warmer world, the Arctic is projected to become cloudier. However, the formation and evolution of Arctic clouds remain highly uncertain in part due to a limited understanding of current and future sources of ice nucleating particles (INPs). In particular, the sources and abundance of biologically-derived INPs are poorly characterized, yet they may be pivotal for cloud ice formation, especially at temperatures in which Arctic mixed-phase clouds (AMPCs) persist (i.e. >−15 °C). Here, we show for the first time that permafrost is a remarkably rich source of biologically-derived INPs, both heat labile (probably proteinaceous) and other organic INPs of biomolecular origin (41%-100% and 99%-100% of the total INPs, respectively). INP concentrations in 1000 to 30 000 year old permafrost were comparable to the most active of other Arctic and midlatitude soil sources (up to 1010 INPs per gram of soil). Thawing of permafrost-which promotes metabolic activity in microbes-and subsequent mobilization of those soils directly into the atmosphere or into lakes, rivers, and the ocean, suggests the intriguing possibility that increasing emissions of INPs from this hitherto overlooked reservoir could be widespread, and, in time, greatly impact Arctic cloud cloud glaciation and radiative properties. This discovery is timely given the rapidly-thawing permafrost in Alaska and across Earth's high latitudes. Since permafrost covers 15% of Northern Hemisphere land, this novel and prevalent INP source may become central to predictions of aerosol-cloud-precipitation interactions in AMPCs.
Semiconductor quantum dots in cavities are promising single-photon sources. Here, we present a path to deterministic operation, by harnessing the intrinsic linear dipole in a neutral quantum dot via ...phonon-assisted excitation. This enables emission of fully polarized single photons, with a measured degree of linear polarization up to 0.994±0.007, and high population inversion-85% as high as resonant excitation. We demonstrate a single-photon source with a polarized first lens brightness of 0.50±0.01, a single-photon purity of 0.954±0.001, and single-photon indistinguishability of 0.909±0.004.
Ear tattooing is a routine procedure performed on laboratory, commercial and companion rabbits for the purpose of identification. Although this procedure is potentially painful, it is usually ...performed without the provision of analgesia, so compromising animal welfare. Furthermore, current means to assess pain in rabbits are poor and more reliable methods are required. The objectives of this study were to assess the physiological and behavioural effects of ear tattooing on rabbits, evaluate the analgesic efficacy of topical local anaesthetic cream application prior to this procedure, and to develop a scale to assess pain in rabbits based on changes in facial expression.
In a crossover study, eight New Zealand White rabbits each underwent four different treatments of actual or sham ear tattooing, with and without prior application of a topical local anaesthetic (lidocaine/prilocaine). Changes in immediate behaviour, heart rate, arterial blood pressure, serum corticosterone concentrations, facial expression and home pen behaviours were assessed. Changes in facial expression were examined to develop the Rabbit Grimace Scale in order to assess acute pain. Tattooing without EMLA cream resulted in significantly greater struggling behaviour and vocalisation, greater facial expression scores of pain, higher peak heart rate, as well as higher systolic and mean arterial blood pressure compared to all other treatments. Physiological and behavioural changes following tattooing with EMLA cream were similar to those in animals receiving sham tattoos with or without EMLA cream. Behavioural changes 1 hour post-treatment were minimal with no pain behaviours identifiable in any group. Serum corticosterone responses did not differ between sham and tattoo treatments.
Ear tattooing causes transient and potentially severe pain in rabbits, which is almost completely prevented by prior application of local anaesthetic cream. The Rabbit Grimace Scale developed appears to be a reliable and accurate way to assess acute pain in rabbits.
Abstract
The Arctic is warming faster than anywhere else on Earth, prompting glacial melt, permafrost thaw, and sea ice decline. These severe consequences induce feedbacks that contribute to ...amplified warming, affecting weather and climate globally. Aerosols and clouds play a critical role in regulating radiation reaching the Arctic surface. However, the magnitude of their effects is not adequately quantified, especially in the central Arctic where they impact the energy balance over the sea ice. Specifically, aerosols called ice nucleating particles (INPs) remain understudied yet are necessary for cloud ice production and subsequent changes in cloud lifetime, radiative effects, and precipitation. Here, we report observations of INPs in the central Arctic over a full year, spanning the entire sea ice growth and decline cycle. Further, these observations are size-resolved, affording valuable information on INP sources. Our results reveal a strong seasonality of INPs, with lower concentrations in the winter and spring controlled by transport from lower latitudes, to enhanced concentrations of INPs during the summer melt, likely from marine biological production in local open waters. This comprehensive characterization of INPs will ultimately help inform cloud parameterizations in models of all scales.
Each successive generation of X-ray machines has opened up new frontiers in science, such as the first radiographs and the determination of the structure of DNA. State-of-the-art X-ray sources can ...now produce coherent high-brightness X-rays of greater than kiloelectronvolt energy and promise a new revolution in imaging complex systems on nanometre and femtosecond scales. Despite the demand, only a few dedicated synchrotron facilities exist worldwide, in part because of the size and cost of conventional (accelerator) technology. Here we demonstrate the use of a new generation of laser-driven plasma accelerators, which accelerate high-charge electron beams to high energy in short distances, to produce directional, spatially coherent, intrinsically ultrafast beams of hard X-rays. This reduces the size of the synchrotron source from the tens of metres to the centimetre scale, simultaneously accelerating and wiggling the electron beam. The resulting X-ray source is 1,000 times brighter than previously reported plasma wigglers and thus has the potential to facilitate a myriad of uses across the whole spectrum of light-source applications.
Additive manufacture (AM) is receiving significant attention globally, reflected in the volume of research being carried out to support the commercialisation of the technology for industrial ...applications and the interest shown by government and policy makers in the technology. The lack of distinction between 3D printing and AM, as well as the portrayal of some highly publicised applications, may imply that the technology is now firmly established. However, this is not the case. The aim of this study is to identify the current barriers to the progression of AM for end-use products from an industrial perspective and to understand the nature of those barriers. Case study research has been conducted with organisations in the UK aerospace, automotive, defence, heavy machinery and medical device industries. Eighteen barriers are identified: education, cost, design, software, materials, traceability, machine constraints, in-process monitoring, mechanical properties, repeatability, scalability, validation, standards, quality, inspection, tolerances, finishing and sterilisation. Explanation building and logic models are used to generalise the findings. The results are discussed in the context of current academic research on AM. The outcomes of this study help to inform the frontiers of research in AM and how AM research agendas can be aligned with the requirements for industrial applications.
