The atmospheric pollen loads of Hobart, Tasmania, Australia, were monitored between September 2007 and July 2009. To examine the match of the airborne pollen composition with the flowering duration ...of their contributing plants, the phenology of native and non-native plants in various habitats near the pollen-trapping site was undertaken between August 2008 and July 2009. The pollen load was found to have a strong seasonal component associated with the start of spring in September. This is incongruent with the peak flowering season of the total taxa in October. In most taxa, atmospheric pollen signatures appeared before flowering was observed in the field. The presence of most pollen types in the atmosphere also exceeded the observed flowering duration of potential pollen-source taxa. Reasons for this may be related to the sampling effort of phenological monitoring, pollen blown in from earlier flowering populations outside of the sampling area, the ability of pollen to be reworked, and the large pollen production of some wind-pollinated taxa. In 2007-2008, 15 pollen types dominated the atmosphere, accounting for 90% of the airborne pollen load. The top six pollen types belonged to Betula, Cupressaceae, Myrtaceae, Salix, Poaceae and Ulmus. Comparatively, the annual pollen load of Hobart is lower than in most other Australian cities; however, the pollen signal of Betula is inordinately high. Native plants play a minor role as pollen contributors, despite the proximity of native habitats to the pollen-sampling location. The implications of the aerobiological observations are discussed in relation to public health.
Previous publications on stapled peptide inhibitors against Mdm2/Mdm4-p53 interactions have established that this new class of drugs have the potential to be easily optimised to attain high binding ...affinity and specificity, but the mechanisms controlling their cellular uptake and target engagement remain elusive and controversial. To aid in understanding the rules of peptide and staple design, and to enable rapid optimisation, we employed the newly-developed cellular thermal shift assay (CETSA). CETSA was able to validate stapled peptide binding to Mdm2 and Mdm4, and the method was also used to determine the extent of cellular uptake, cellular availability, and intracellular binding of the endogenous target proteins in its native environment. Our data suggest that while the stapled peptides engage their targets intracellularly, more work is needed to improve their cellular entry and target engagement efficiency in vivo. CETSA now provides a valuable tool to optimize such in vivo properties of stapled peptides.
Babies born at lower gestational ages or smaller birthweights have a greater risk of poorer health in later life. Both the causes of these sub-optimal birth outcomes and the mechanism by which the ...effects are transmitted over decades are the subject of extensive study. We investigated whether a transcriptomic signature of either birthweight or gestational age could be detected in umbilical cord RNA.
The gene expression patterns of 32 umbilical cords from Singaporean babies of Chinese ethnicity across a range of birthweights (1698-4151 g) and gestational ages (35-41 weeks) were determined. We confirmed the differential expression pattern by gestational age for 12 genes in a series of 127 umbilical cords of Chinese, Malay and Indian ethnicity.
We found that the transcriptome is substantially influenced by gestational age; but less so by birthweight. We show that some of the expression changes dependent on gestational age are enriched in signal transduction pathways, such as Hedgehog and in genes with roles in cytokine signalling and angiogenesis. We show that some of the gene expression changes we report are reflected in the epigenome.
We studied the umbilical cord which is peripheral to disease susceptible tissues. The results suggest that soma-wide transcriptome changes, preserved at the epigenetic level, may be a mechanism whereby birth outcomes are linked to the risk of adult metabolic and arthritic disease and suggest that greater attention be given to the association between premature birth and later disease risk.
