This study investigated the interactions between organic chemicals and components of the soil matrix and their effects on subsequent microbial mineralization kinetics. Five 14C‐labeled chemicals ...(anionic, cationic, and nonionic surfactants) were aseptically sorbed to montmorillonite, kaolinite, illite, sand, humic acids, and fulvic acids. Small amounts of these sorbed chemicals were dosed to an acidic, sludge‐amended agricultural soil (Rossmoyne) to a final added chemical concentration of 50 ng g−1. Controls received the same final added concentration of the chemicals in water. The ratio of sorbed chemical to soil was kept low to minimize changes to the soil mineralogy, chemistry, and microbiology. Microbial mineralization of the chemicals to 14CO2 was measured over a period of 60 to 70 d, and the data were fitted to first‐order and 3/2‐order mineralization models. Association with the soil constituents inhibited the mineralization of the chemicals in the following rank (from least to greatest effect): controls ≈ sand < kaolinite < illite < montmorillonite ≈ humic acids < fulvic acids. These experiments demonstrated that interactions with some soil constituents (kaolinite, illite, and sand) had little effect on the microbial metabolism of these chemicals, while montmorillonite, humic acids, and especially fulvic acids significantly decreased the bioavailability of the chemicals to the microbial community. The first group of soil constituents had little influence on the mineralization kinetic parameters, whereas the latter significantly reduced at least one of the parameter estimates. The parent soil, possibly via interactions with its mineral surfaces, also had effects on the degradation of the chemicals, since soil microbial biomass and physiological activity were not correlated with any of the mineralization kinetic parameter estimates. These experiments demonstrate that the environmental form of a chemical has a significant influence on its eventual microbial metabolism and is an important parameter to consider when investigating the fate of chemicals in soil environments.
The 2-amino-3-benzoylthiophene PD 81,723 has been shown to exhibit allosteric enhancement of adenosine A1 receptor binding and function. The aim of this study was to clarify the mechanism of this ...effect using membranes purified from rat brain and Chinese hamster ovary (CHO)-A1 cells that stably express the rat adenosine A1 receptor as well as intact CHO-A1 and nontransfected CHO cells. In membranes containing 100 microM magnesium, (2-amino-4, 5-dimethyl-3-thienyl)-3-(trifluoromethyl)phenylmethanone (PD 81, 723) significantly increased the affinity of the adenosine A1 receptor agonist, cyclopentyladenosine, for the low-affinity receptor without affecting high-affinity binding or Bmax. In intact cells, PD 81,723 inhibited basal adenylyl cyclase (AC) activity as well as forskolin-, cholera toxin-, and pertussis toxin-stimulated AC activity in CHO-A1 and CHO cells. Basal AC activity was inhibited 49% in CHO and 82% in CHO-A1 cells by 30 microM PD 81,723. In CHO-A1 cells, half-maximal inhibition of forskolin-stimulated AC occurred at 5 microM PD 81,723 compared to 10 microM in CHO cells. Cholera toxin-stimulated AC was reduced 90% in both CHO and CHO-A1 cells by 30 microM PD 81,723. At the same concentration of PD 81,723, pertussis toxin-stimulated AC activity was reduced 86% (CHO-A1) and 77% (CHO). 3Hforskolin was displaced from purified rat liver AC by PD 81,723 with an IC50 of 96 microM. These results demonstrate that two mechanisms appear to contribute to the observed effects of PD 81, 723. One mechanism is allosteric enhancement of adenosine A1 receptor function. Results from transfected and nontransfected cells suggest that PD 81,723 also inhibits AC directly by binding to the catalytic unit at or near the forskolin-binding site.

The identification of a functional T-cadherin isoform and the coexpression of T-cadherins and N-cadherin by individual cells suggest that specific adhesive interactions of embryonic cells may involve a complex interplay between multiple cadherins.
Matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry is now an essential tool in biopolymer analysis. Sensitivity and mass range are unsurpassed, but mass ...measurement accuracy and resolution have been limited. With delayed extraction and a reflecting analyzer, mass measurements using MALDI-TOF can be made with an accuracy of a few parts per million (ppm). It is possible to distinguish Lys from Gln in peptides, and to determine the elemental composition of smaller molecules (mass 100-500). In database searching strategies, a smaller mass window, resulting from an increase in mass accuracy, greatly decreases the number of possible candidates. Mass measurement accuracy with errors less than 5 ppm is demonstrated on a mixture of 12 peptides ranging in mass from ca. 900 to 3700 Da. Mass measurements on 13 peaks in an unseparated tryptic digest of myoglobin gave results with an overall average error less than 3.5 ppm, with a maximum error of 7 ppm.
We have investigated liver LDL receptor mRNA expression in nontransgenic, human cholesteryl ester transfer protein (CETP) transgenic, and human apolipoprotein (Apo) B/CETP double transgenic mice fed ...a normal chow diet and a high fat, high cholesterol diet (HFHC). Three weeks of HFHC feeding increased total serum cholesterol 1.5-fold in the nontransgenic, 3.1-fold in the CETP transgenic, and 3.4-fold in the ApoB/CETP double transgenic mice. To examine the liver LDL receptor mRNA expression among the different groups of mice fed the normal diet or fed the HFHC diet, we developed a quantitative reverse-transcribed polymerase chain reaction assay in which the LDL receptor mRNA level was normalized with the beta-actin mRNA. The results show that on the normal chow diet, the LDL receptor mRNA expression levels were lower in the ApoB/CETP mice than in the nontransgenic mice and the human CETP transgenic mice. Liver LDL receptor gene expression was lower in all groups of mice fed the HFHC diet, with the lowest level of expression in the ApoB/CETP mice. Similar results were obtained by Northern blot analysis. In addition, we have previously shown that the cytokine oncostatin M (OM) increases LDL receptor gene expression in HepG2 cells. In this study, we used the ApoB/CETP mice as the model system to examine the in vivo activity of OM on liver LDL receptor gene expression. Our data show that OM increased the level of liver LDL receptor mRNA up to 80% to 90% when the animals were fed the HFHC diet. The results from these studies demonstrate that the expression of the liver LDL receptor in the ApoB/CETP mice is suppressed compared with nontransgenic mice and that the expression of the hepatic LDL receptor gene in these mice is subjected to the normal cholesterol feedback regulation. In addition, LDL receptor gene expression in these mice is also inducible by a positive regulator.
This study reports new electrocardiographic (ECG) predictors of radiofrequency catheter ablation failure and recurrence in idiopathic right ventricular outflow tract (RVOT) ventricular tachycardia ...(VT) or ectopy based on 91 consecutive patients. Procedural success and failure rates were 85% (77/91) and 15% (14/91), respectively. Twenty three percent (18/77) had recurrence during the follow-up period of 1 to 120 months (mean 56 ± 31 months). Baseline RVOT VT/ectopy on 12-lead ECG taken prior to ablation from 91 patients were retrospectively analyzed. Ablation performed with RVOT ectopy (isolated ectopies, bigeminy, trigeminy, or couplets) as template arrhythmia was more likely to fail (30% vs. 8%,
P = .02) as opposed to RVOT VT (sustained or nonsustained). VT/ectopy-QRS morphology variation was more observed in failed ablations (36% vs. 7%,
P = .001). Significantly wider mean VT/ectopy QRS in leads I, II, AVR, V2, V3, V5, and V6 were noted in failed ablation group. Mean R wave amplitude reached statistical significance only in lead II (22.0 ± 5.1 mV for failed vs. 17.8 ± 5.2 mV for successful outcomes;
P = .009). QRS morphologic variation (47% vs. 16%;
P = .009) was the only statistically significant ECG to be more common in patients with arrhythmia recurrence. In conclusion, ablation with ectopy over VT as template arrhythmia, presence of QRS morphologic variation, wider mean QRS width, and taller mean R-wave amplitude in lead II were identified ECG predictors of failed RVOT VT/Ectopy ablation. The only ECG predictor of recurrence was the presence of RVOT VT or ectopy QRS morphologic variation.
This study examined effects of aging on the cardiac response in vitro to daunorubicin, a cancer chemotherapeutic agent that causes cardiotoxicity. Left ventricular trabeculae carneae from adult (aged ...6–9 months) and old (aged 24–28 months) Fischer 344 rats were placed in oxygenated, physiological buffer. Preparations were treated with daunorubicin (175 μM) or saline (controls) over a 210-minute study period. Daunorubicin-induced decline in contractility (DS and dS/dt) was greater in old compared to adult myocardium (p <.02). Similarly, cardiac relaxation (90% relaxation time) was more impaired by daunorubicin in older preparations (p < 01). Although daunorubicin concentrations were unaffected by age, daunorubicinol concentrations in ventricular strips increased with time to a greater extent in the older group (p < .05). This study suggests that senescence increases the acute in vitro cardiotoxicity of daunorubicin and that the metabolite, daunorubicinol, may contribute to this toxicity.
Manned exploration of Mars may result in the contamination of that planet with terrestrial microbes, a situation requiring assessment of the survival potential of possible contaminating organisms. In ...this study, the survival of Bacillius subtilis, Azotobacter chroococcum, and the enteric bacteriophage MS2 was examined in clays representing terrestrial (Wyoming type montmorillonite) or Martian (Fe(3+)-montmorillonite) soils exposed to terrestrial and Martian environmental conditions of temperature and atmospheric pressure and composition, but not to UV flux or oxidizing conditions. Survival of bacteria was determined by standard plate counts and biochemical and physiological measurements over 112 days. Extractable lipid phosphate was used to measure microbial biomass, and the rate of 14C-acetate incorporation into microbial lipids was used to determine physiological activity. MS2 survival was assayed by plaque counts. Both bacterial types survived terrestrial or Martian conditions in Wyoming montmorillonite better than Martian conditions in Fe(3+)-montmorillonite. Decreased survival may have been caused by the lower pH of the Fe(3+)-montmorillonite compared to Wyoming montmorillonite. MS2 survived simulated Mars conditions better than the terrestrial environment, likely due to stabilization of the virus caused by the cold and dry conditions of the simulated Martian environment. The survival of MS2 in the simulated Martian environment is the first published indication that viruses may be able to survive in Martian type soils. This work may have implications for planetary protection for future Mars missions.
Hot subdwarf stars are mostly stripped red giants that can exhibit photometric variations due to stellar pulsations, eclipses, the reflection effect, ellipsoidal modulation, and Doppler beaming. ...Detailed studies of their light curves help constrain stellar parameters through asteroseismological analyses or binary light curve modeling and generally improve our capacity to draw a statistically meaningful picture of this enigmatic stage of stellar evolution. From an analysis of Gaia DR2 flux errors, we have identified around 1200 candidate hot subdwarfs with inflated flux errors for their magnitudes - a strong indicator of photometric variability. As a pilot study, we obtained 2-min cadence TESS Cycle 2 observations of 187 candidate hot subdwarfs with anomalous Gaia flux errors. More than 90% of our targets show significant photometric variations in their TESS light curves. Many of the new systems found are cataclysmic variables, but we report the discovery of several new variable hot subdwarfs, including HW Vir binaries, reflection effect systems, pulsating sdBVs stars, and ellipsoidally modulated systems. We determine atmospheric parameters for select systems using follow-up spectroscopy from the 3-m Shane telescope. Finally, we present a Fourier diagnostic plot for classifying binary light curves using the relative amplitudes and phases of their fundamental and harmonic signals in their periodograms. This plot makes it possible to identify certain types of variables efficiently, without directly investigating their light curves, and may assist in the rapid classification of systems observed in large photometric surveys.
A series of 17 cysteine, N-acetyl cysteine, glutathione, and N-trifluoroacetyl glutathione conjugates have been prepared, and their thermospray (TSP) spectra have been recorded in the positive and ...negative ion modes. The compounds undergo extensive fragmentation, which primarily occurs at the carbon-sulfur bonds. For most of the compounds, positive ion TSP is more sensitive than negative ion thermospray. Probably due to the thermal lability of these adducts, the quality of the spectra obtained are dependent on source conditions, requiring fine control of the vaporization/desolvation process.
