Large quantities of organic carbon are stored in frozen soils (permafrost) within Arctic and sub-Arctic regions. A warming climate can induce environmental changes that accelerate the microbial ...breakdown of organic carbon and the release of the greenhouse gases carbon dioxide and methane. This feedback can accelerate climate change, but the magnitude and timing of greenhouse gas emission from these regions and their impact on climate change remain uncertain. Here we find that current evidence suggests a gradual and prolonged release of greenhouse gas emissions in a warming climate and present a research strategy with which to target poorly understood aspects of permafrost carbon dynamics.
As Arctic regions warm and frozen soils thaw, the large organic carbon pool stored in permafrost becomes increasingly vulnerable to decomposition or transport. The transfer of newly mobilized carbon ...to the atmosphere and its potential influence upon climate change will largely depend on the degradability of carbon delivered to aquatic ecosystems. Dissolved organic carbon (DOC) is a key regulator of aquatic metabolism, yet knowledge of the mechanistic controls on DOC biodegradability is currently poor due to a scarcity of long-term data sets, limited spatial coverage of available data, and methodological diversity. Here, we performed parallel biodegradable DOC (BDOC) experiments at six Arctic sites (16 experiments) using a standardized incubation protocol to examine the effect of methodological differences commonly used in the literature. We also synthesized results from 14 aquatic and soil leachate BDOC studies from across the circum-arctic permafrost region to examine pan-arctic trends in BDOC. An increasing extent of permafrost across the landscape resulted in higher DOC losses in both soil and aquatic systems. We hypothesize that the unique composition of (yedoma) permafrost-derived DOC combined with limited prior microbial processing due to low soil temperature and relatively short flow path lengths and transport times, contributed to a higher overall terrestrial and freshwater DOC loss. Additionally, we found that the fraction of BDOC decreased moving down the fluvial network in continuous permafrost regions, i.e. from streams to large rivers, suggesting that highly biodegradable DOC is lost in headwater streams. We also observed a seasonal (January-December) decrease in BDOC in large streams and rivers, but saw no apparent change in smaller streams or soil leachates. We attribute this seasonal change to a combination of factors including shifts in carbon source, changing DOC residence time related to increasing thaw-depth, increasing water temperatures later in the summer, as well as decreasing hydrologic connectivity between soils and surface water as the thaw season progresses. Our results suggest that future climate warming-induced shifts of continuous permafrost into discontinuous permafrost regions could affect the degradation potential of thaw-released DOC, the amount of BDOC, as well as its variability throughout the Arctic summer. We lastly recommend a standardized BDOC protocol to facilitate the comparison of future work and improve our knowledge of processing and transport of DOC in a changing Arctic.
Research has traditionally focused on atmospheric release of carbon from thawing permafrost, yet overlooked waterborne release pathways likely contribute significantly, especially in a warming ...Arctic. To address this knowledge gap and better constrain the fate of carbon in the North, we recommend inter-disciplinary efforts bridging physical, chemical and computational research.
The Arctic is a water-rich region, with freshwater systems covering about 16 % of the northern permafrost landscape. Permafrost thaw creates new freshwater ecosystems, while at the same time ...modifying the existing lakes, streams, and rivers that are impacted by thaw. Here, we describe the current state of knowledge regarding how permafrost thaw affects lentic (still) and lotic (moving) systems, exploring the effects of both thermokarst (thawing and collapse of ice-rich permafrost) and deepening of the active layer (the surface soil layer that thaws and refreezes each year). Within thermokarst, we further differentiate between the effects of thermokarst in lowland areas vs. that on hillslopes. For almost all of the processes that we explore, the effects of thaw vary regionally, and between lake and stream systems. Much of this regional variation is caused by differences in ground ice content, topography, soil type, and permafrost coverage. Together, these modifying factors determine (i) the degree to which permafrost thaw manifests as thermokarst, (ii) whether thermokarst leads to slumping or the formation of thermokarst lakes, and (iii) the manner in which constituent delivery to freshwater systems is altered by thaw. Differences in thaw-enabled constituent delivery can be considerable, with these modifying factors determining, for example, the balance between delivery of particulate vs. dissolved constituents, and inorganic vs. organic materials. Changes in the composition of thaw-impacted waters, coupled with changes in lake morphology, can strongly affect the physical and optical properties of thermokarst lakes. The ecology of thaw-impacted lakes and streams is also likely to change; these systems have unique microbiological communities, and show differences in respiration, primary production, and food web structure that are largely driven by differences in sediment, dissolved organic matter, and nutrient delivery. The degree to which thaw enables the delivery of dissolved vs. particulate organic matter, coupled with the composition of that organic matter and the morphology and stratification characteristics of recipient systems will play an important role in determining the balance between the release of organic matter as greenhouse gases (CO2 and CH4), its burial in sediments, and its loss downstream. The magnitude of thaw impacts on northern aquatic ecosystems is increasing, as is the prevalence of thaw-impacted lakes and streams. There is therefore an urgent need to quantify how permafrost thaw is affecting aquatic ecosystems across diverse Arctic landscapes, and the implications of this change for further climate warming.
The future trajectory of greenhouse gas concentrations depends on interactions between climate and the biogeosphere. Thawing of Arctic permafrost could release significant amounts of carbon into the ...atmosphere in this century. Ancient Ice Complex deposits outcropping along the ~7,000-kilometre-long coastline of the East Siberian Arctic Shelf (ESAS), and associated shallow subsea permafrost, are two large pools of permafrost carbon, yet their vulnerabilities towards thawing and decomposition are largely unknown. Recent Arctic warming is stronger than has been predicted by several degrees, and is particularly pronounced over the coastal ESAS region. There is thus a pressing need to improve our understanding of the links between permafrost carbon and climate in this relatively inaccessible region. Here we show that extensive release of carbon from these Ice Complex deposits dominates (57 ± 2 per cent) the sedimentary carbon budget of the ESAS, the world’s largest continental shelf, overwhelming the marine and topsoil terrestrial components. Inverse modelling of the dual-carbon isotope composition of organic carbon accumulating in ESAS surface sediments, using Monte Carlo simulations to account for uncertainties, suggests that 44 ± 10 teragrams of old carbon is activated annually from Ice Complex permafrost, an order of magnitude more than has been suggested by previous studies. We estimate that about two-thirds (66 ± 16 per cent) of this old carbon escapes to the atmosphere as carbon dioxide, with the remainder being re-buried in shelf sediments. Thermal collapse and erosion of these carbon-rich Pleistocene coastline and seafloor deposits may accelerate with Arctic amplification of climate warming.
While over 99% of coastal arctic rivers drain small catchments, future projections of land-ocean fluxes are based on data from large rivers. We encourage inclusion of and increased focus on smaller ...catchments to support representative assessments of arctic ecosystem change.
Climate change is increasing the frequency and intensity of thermokarst, and accelerating the delivery of terrestrial organic material from previously sequestered sources to aquatic systems, where it ...is subject to further biochemical alteration. Rapid climate change in the glacially conditioned ice-rich and ice-marginal terrain of the Peel Plateau, western Canada, is accelerating thaw-driven mass wasting in the form of retrogressive thaw slumps, which are rapidly increasing in area, volume and thickness of permafrost thawed. Despite major perturbation of downstream sedimentary and geochemical fluxes, few studies have examined changes in flux and composition of particulate organic carbon (POC) in streams and rivers as a result of permafrost thaw. Here we show that the orders of magnitude increase in total organic carbon, nitrogen, and phosphorus mobilized to streams from thaw slumps on the Peel Plateau is almost entirely due to POC and associated particulate nitrogen and phosphorus release. Slump-mobilized POC is compositionally distinct from its dissolved counterpart and appears to contain relatively greater amounts of degraded organic matter, as inferred from base-extracted fluorescence of particulate organic matter. Thus, slump-mobilized POC is potentially more recalcitrant than POC present in non-slump affected stream networks. Furthermore a substantial portion of POC mobilized from thaw slumps will be constrained within primary sediment stores in valley bottoms, where net accumulation is currently exceeding net erosion, resulting in century to millennial scale sequestration of thermokarst-mobilized POC. This study highlights the pressing need for better knowledge of sedimentary cascades, mobilization, and storage reservoirs in slump-affected streams, and baseline assessments of the biodegradability of POC and cycling of particulate nutrients within a sedimentary cascade framework. Explicit incorporation of POC dynamics into our understanding of land-water carbon mobilization in the face of permafrost thaw is critical for understanding implications of thermokarst for regional carbon cycling and fluvial ecosystems.

Preclinical cell-based assays that recapitulate human disease play an important role in drug repurposing. We previously developed a functional forskolin induced swelling (FIS) assay using patient-derived intestinal organoids (PDIOs), allowing functional characterization of CFTR, the gene mutated in people with cystic fibrosis (pwCF). CFTR function-increasing pharmacotherapies have revolutionized treatment for approximately 85% of people with CF who carry the most prevalent F508del-CFTR mutation, but a large unmet need remains to identify new treatments for all pwCF.
We used 76 PDIOs not homozygous for F508del-CFTR to test the efficacy of 1400 FDA-approved drugs on improving CFTR function, as measured in FIS assays. The most promising hits were verified in a secondary FIS screen. Based on the results of this secondary screen, we further investigated CFTR elevating function of PDE4 inhibitors and currently existing CFTR modulators.
In the primary screen, 30 hits were characterized that elevated CFTR function. In the secondary validation screen, 19 hits were confirmed and categorized in three main drug families: CFTR modulators, PDE4 inhibitors and tyrosine kinase inhibitors. We show that PDE4 inhibitors are potent CFTR function inducers in PDIOs where residual CFTR function is either present, or created by additional compound exposure. Additionally, upon CFTR modulator treatment we show rescue of CF genotypes that are currently not eligible for this therapy.
This study exemplifies the feasibility of high-throughput compound screening using PDIOs. We show the potential of repurposing drugs for pwCF carrying non-F508del genotypes that are currently not eligible for therapies.

On‐going shrinkage of Greenland's icecap, permafrost thaw, and changes in precipitation are exposing its landscapes to erosion and remobilization of ancient organic carbon (OC) held in soils and ...sedimentary rocks. The fate of this OC and potential feedbacks to climate are still unclear. Here, we show that the glacial Zackenberg river (Northeastern Greenland) exports aged particulate OC (POC, uncalibrated radiocarbon ages of ∼4,000 years). Many of the smaller periglacial streams affected by abrupt permafrost thaw transport substantially older POC (up to 32,000 years), especially with enhanced discharge following intense precipitation. Mineralogical analysis, and density and size fractionation of soils and both glacial and nonglacial river sediments reveal that OC is largely associated with phyllosilicate minerals, suggesting stabilization against microbial processing. Enhanced export of ancient, mineral‐associated OC as a consequence of summer rainfall may accelerate translocation of OC from terrestrial to marine environments, but could have limited consequences for climate.

On‐going Arctic warming leads to shrinking ice sheets and permafrost thaw, which promotes erosion of ancient carbon stocks. The decomposition of this remobilized carbon to CO2 or CH4 would in turn fuel further climate warming. Here we show that the glacial Zackenberg river in Northeastern Greenland likely exports a mixture of aged soil organic matter, recent bacteria and ice algae biomass residues, and deposited organic aerosols. In contrast, many of the smaller, nonglacial streams in the study area carried significantly older carbon from deeper soil layers or sedimentary rocks. More intense rainfall will likely increase these carbon fluxes. However, close interactions of organic matter with mineral surfaces may hamper greenhouse gas production and instead promote burial in marine sediments. Therefore, the impact on atmospheric CO2 and climate may be limited.

Whole-grain wheat (WGW) is described as nutritionally superior to refined wheat (RW) and thus advocated as the healthy choice, although evidence from intervention studies is often inconsistent. The ...liver, as the central organ in energy metabolism, might be an important target organ for WGW interventions.
The aim of this study was to investigate the potential benefits of WGW consumption compared with RW consumption on liver health and associated parameters.
We performed a double-blind, parallel trial in which 50 overweight 45- to 70-y-old men and postmenopausal women were randomly allocated to a 12-wk intervention with either WGW (98 g/d) or RW (98 g/d) products. Before and after the intervention we assessed intrahepatic triglycerides (IHTGs) and fat distribution by proton magnetic resonance spectroscopy/magnetic resonance imaging, fecal microbiota composition, adipose tissue gene expression, and several fasting plasma parameters, as well as postprandial plasma lipids after a mixed meal.
Fasting plasma cholesterol, triglycerides, nonesterified fatty acids, and insulin were not affected by RW or WGW intervention. We observed a substantial increase of 49.1% in IHTGs in the RW when compared with the WGW group (P = 0.033). Baseline microbiota composition could not predict the increase in IHTGs after RW, but gut microbiota diversity decreased in the RW group when compared with the WGW group (P = 0.010). In the WGW group, we observed increased postprandial triglyceride levels compared with the RW group (P = 0.020). In addition, the WGW intervention resulted in a trend towards lower fasting levels of the liver acute-phase proteins serum amyloid A (P = 0.057) and C-reactive protein (P = 0.064) when compared to the RW intervention.
A 12-wk RW intervention increases liver fat and might contribute to the development of nonalcoholic fatty liver disease, whereas a 12-wk 98-g/d WGW intervention prevents a substantial increase in liver fat. Our results show that incorporating feasible doses of WGW in the diet at the expense of RW maintains liver health. The study was registered at clinicaltrials.gov as NCT02385149.
