The aim of this study was to assess validation evidence for a sedation scale for dogs. We hypothesized that the chosen sedation scale would be unreliable when used by different raters and show poor ...discrimination between sedation protocols. A sedation scale (range 0-21) was used to score 62 dogs scheduled to receive sedation at two veterinary clinics in a prospective trial. Scores recorded by a single observer were used to assess internal consistency and construct validity of the scores. To assess inter-rater reliability, video-recordings of sedation assessment were randomized and blinded for viewing by 5 raters untrained in the scale. Videos were also edited to allow assessment of inter-rater reliability of an abbreviated scale (range 0-12) by 5 different raters.
Both sedation scales exhibited excellent internal consistency and very good inter-rater reliability (full scale, intraclass correlation coefficient ICC
= 0.95; abbreviated scale, ICC
= 0.94). The full scale discriminated between the most common protocols: dexmedetomidine-hydromorphone (median range of sedation score, 11 1-18, n = 20) and acepromazine-hydromorphone (5 0-15, n = 36, p = 0.02).
The hypothesis was rejected. Full and abbreviated scales showed excellent internal consistency and very good reliability between multiple untrained raters. The full scale differentiated between levels of sedation.
Three variants of schlieren techniques are employed to investigate the development of second-mode instability waves in the hypersonic boundary layer of a slender cone in a reflected shock tunnel. ...First, a previously proposed technique using high frame rate (i.e., at least as high as the dominant instability frequency) schlieren visualization with a continuous light source is shown to provide repeatable measurements of the instability propagation speed and frequency. A modified version of the technique is then introduced whereby a pulsed light source allows the use of a higher-resolution camera with a lower frame rate: this provides significant benefits in terms of spatial resolution and total recording time. A detailed picture of the surface-normal intensity distribution for individual wave packets is obtained, and the images provide comprehensive insight into the unsteady flow structures within the boundary layer. Finally, two-point schlieren deflectometry is implemented and shown to be capable of providing second-mode growth information in the challenging shock tunnel environment.
Vertically aligned GaN nanorod arrays with nonpolar InGaN/GaN multi quantum wells (MQW) were grown by MOVPE on c-plane GaN-on-sapphire templates. The chemical and structural properties of single ...nanorods are optically investigated with a spatial resolution beyond the diffraction limit using tip-enhanced Raman spectroscopy (TERS). This enables the local mapping of variations in the chemical composition, charge distribution, and strain in the MQW region of the nanorods. Nanoscale fluctuations of the In content in the InGaN layer of a few percent can be identified and visualized with a lateral resolution below 35 nm. We obtain evidence for the presence of indium clustering and the formation of cubic inclusions in the wurtzite matrix near the QW layers. These results are directly confirmed by high-resolution TEM images, revealing the presence of stacking faults and different polymorphs close to the surface near the MQW region. The combination of TERS and HRTEM demonstrates the potential of this nanoscale near-field imaging technique, establishing TERS as a very potent, comprehensive, and nondestructive tool for the characterization and optimization of technologically relevant semiconductor nanostructures.
We developed a new nitrogen oxide (NOx) and carbon monoxide (CO) emission inventory for the Los Angeles‐South Coast Air Basin (SoCAB) expanding the Fuel‐based Inventory for motor‐Vehicle Emissions ...and applied it in regional chemical transport modeling focused on the California Nexus of Air Quality and Climate Change (CalNex) 2010 field campaign. The weekday NOx emission over the SoCAB in 2010 is 620 t d−1, while the weekend emission is 410 t d−1. The NOx emission decrease on weekends is caused by reduced diesel truck activities. Weekday and weekend CO emissions over this region are similar: 2340 and 2180 t d−1, respectively. Previous studies reported large discrepancies between the airborne observations of NOx and CO mixing ratios and the model simulations for CalNex based on the available bottom‐up emission inventories. Utilizing the newly developed emission inventory in this study, the simulated NOx and CO mixing ratios agree with the observations from the airborne and the ground‐based in situ and remote sensing instruments during the field study. The simulations also reproduce the weekly cycles of these chemical species. Both the observations and the model simulations indicate that decreased NOx on weekends leads to enhanced photochemistry and increase of O3 and Ox (=O3 + NO2) in the basin. The emission inventory developed in this study can be extended to different years and other urban regions in the U.S. to study the long‐term trends in O3 and its precursors with regional chemical transport models.

The high-frequency peaked BL Lac PKS 2155-304 at redshift z = 0.116 is a well-known VHE (>100 GeV) gamma -ray emitter. Since 2002 its VHE flux has been monitored using the H.E.S.S. stereoscopic array ...of imaging atmospheric Cerenkov telescopes in Namibia. During the 2006 July dark period, the average VHE flux was measured to be more than 10 times typical values observed from the object. This article focuses solely on an extreme gamma -ray outburst detected in the early hours of 2006 July 28 (MJD 53,944). The average flux observed during this outburst is I(>200 GeV) = (1.72 plus or minus 0.05 unk plus or minus 0.34 unk) x 10 super(-9) cm-z s-i, corresponding to similar to 7 times the flux, I(>200 GeV), observed from the Crab Nebula. Peak fluxes are measured with 1 minute timescale resolution at more than twice this average value. Variability is seen up to similar to 600 s in the Fourier power spectrum, and well-resolved bursts varying on timescales of similar to 200 s are observed. There are no strong indications for spectral variability within the data. Assuming the emission region has a size comparable to the Schwarzschild radius of a similar to 10 super(9) M unk black hole, Doppler factors greater than 100 are required to accommodate the observed variability timescales.
Summary
Chronic lymphocytic leukaemia (CLL) cells encounter T‐cells and proliferate in response to T‐cell signals in the lymph node microenvironment. In this report we determined interleukin 21 ...(IL21) function in CLL and showed that IL21 and interleukin 4 (IL4) act co‐operatively to promote leukaemic cell proliferation without apoptosis or differentiation We further show that IL21 increased side population (SP) cells, which are associated with resistance to chemotherapy and increased self‐renewal capacity in CLL. IL21 and IL4 are the major cytokines produced by the recently described CD4+ T follicular helper (Tfh) cell subset. Determination of Tfh cells in peripheral blood showed that patients had significantly increased numbers as compared to normal subjects although no association was found between Tfh numbers and IGHV gene mutational status or clinical stage. Our data suggests that the Tfh cytokines, IL4 and IL21, contribute to driving leukaemic cell proliferation in the lymph node microenvironment, and may contribute to the specific production of cells resistant to conventional chemotherapy. We suggest that increased circulating Tfh cells is a component of T‐cell dysregulation in CLL. Our findings have implications for the therapeutic use of IL21.
A significant fraction of the energy density of the interstellar medium is in the form of high-energy charged particles (cosmic rays). The origin of these particles remains uncertain. Although it is ...generally accepted that the only sources capable of supplying the energy required to accelerate the bulk of Galactic cosmic rays are supernova explosions, and even though the mechanism of particle acceleration in expanding supernova remnant (SNR) shocks is thought to be well understood theoretically, unequivocal evidence for the production of high-energy particles in supernova shells has proven remarkably hard to find. Here we report on observations of the SNR RX J1713.7 - 3946 (G347.3 - 0.5), which was discovered by ROSAT in the X-ray spectrum and later claimed as a source of high-energy γ-rays of TeV energies (1 TeV = 1012 eV). We present a TeV γ-ray image of the SNR: the spatially resolved remnant has a shell morphology similar to that seen in X-rays, which demonstrates that very-high-energy particles are accelerated there. The energy spectrum indicates efficient acceleration of charged particles to energies beyond 100 TeV, consistent with current ideas of particle acceleration in young SNR shocks.
Centaurus A (Cen A) is the nearest radio galaxy discovered as a very-high-energy (VHE; 100 GeV–100 TeV) γ-ray source by the High Energy Stereoscopic System (H.E.S.S.). It is a faint VHE γ-ray ...emitter, though its VHE flux exceeds both the extrapolation from early Fermi-LAT observations as well as expectations from a (misaligned) single-zone synchrotron-self Compton (SSC) description. The latter satisfactorily reproduces the emission from Cen A at lower energies up to a few GeV. New observations with H.E.S.S., comparable in exposure time to those previously reported, were performed and eight years of Fermi-LAT data were accumulated to clarify the spectral characteristics of the γ-ray emission from the core of Cen A. The results allow us for the first time to achieve the goal of constructing a representative, contemporaneous γ-ray core spectrum of Cen A over almost five orders of magnitude in energy. Advanced analysis methods, including the template fitting method, allow detection in the VHE range of the core with a statistical significance of 12σ on the basis of 213 hours of total exposure time. The spectrum in the energy range of 250 GeV–6 TeV is compatible with a power-law function with a photon index Γ = 2.52 ± 0.13stat ± 0.20sys. An updated Fermi-LAT analysis provides evidence for spectral hardening by ΔΓ ≃ 0.4 ± 0.1 at γ-ray energies above 2.8+1.0−0.6 2.8−0.6+1.0 $2.8\substack{+1.0 \\-0.6}$ GeV at a level of 4.0σ. The fact that the spectrum hardens at GeV energies and extends into the VHE regime disfavour a single-zone SSC interpretation for the overall spectral energy distribution (SED) of the core and is suggestive of a new γ-ray emitting component connecting the high-energy emission above the break energy to the one observed at VHE energies. The absence of significant variability at both GeV and TeV energies does not yet allow disentanglement of the physical nature of this component, though a jet-related origin is possible and a simple two-zone SED model fit is provided to this end.
Cannabidiol (CBD) is a non-intoxicating phytocannabinoid whose purported therapeutic benefits and impression of a high safety profile has promoted its increasing popularity. CBD’s popularity is also ...increasing among children and adolescents who are being administered CBD, off label, for the treatment of numerous symptoms associated with autism spectrum disorder, attention deficit hyperactivity disorder, anxiety, and depression. The relative recency of its use in the adolescent population has precluded investigation of its impact on the developing brain and the potential consequences that may present in adulthood. Therefore, there’s an urgency to identify whether prolonged adolescent CBD exposure has substantive impacts on the developing brain that impact behavioral and cognitive processes in adulthood. Here, we tested the effect of twice-daily intraperitoneal administrations of CBD (20 mg/kg) in male and female C57BL/6J mice during the adolescent period of 25–45 days on weight gain, and assays for locomotor behavior, anxiety, and spatial memory. Prolonged adolescent CBD exposure had no detrimental effects on locomotor activity in the open field, anxiety behavior on the elevated plus maze, or spatial memory in the Barnes Maze compared to vehicle-treated mice. Interestingly, CBD-treated mice had a faster rate of learning in the Barnes Maze. However, CBD-treated females had reduced weight gain during the exposure period. We conclude that prolonged adolescent CBD exposure in mice does not have substantive negative impacts on a range of behaviors in adulthood, may improve the rate of learning under certain conditions, and impacts weight gain in a sex-specific manner.
