Abstract Targeting of specific receptors on antigen-presenting cells is an appealing prospect in the production of novel nanoparticulate vaccines. In particular, the targeting of vaccines to ...dendritic cell (DC) subsets has been shown in models to significantly improve the induction of immune responses. This paper describes the evaluation of natural ligands, mannan and chitosan, and monoclonal antibodies as targeting motifs to enhance uptake of PLGA nanoparticle carriers by bovine DCs. To assess enhancement of uptake after the addition of natural ligands a bovine monocyte derived DC (MoDC) model was used. For the assessment of monoclonal antibody targeting, the model was expanded to include afferent lymph DCs (ALDCs) in a competitive uptake assay. Mannan, proved unsuccessful at enhancing uptake or targeting by MoDCs. Chitosan coated particle uptake could be impeded by the addition of mannan suggesting uptake may be mediated through sugar receptors. Inclusion of monoclonal antibodies specific for the DEC-205 (CD205) receptor increased the number of receptor expressing DCs associated with particles as well as the number of particles taken up by individual cells. These results support the further evaluation of active targeting of nanovaccines to DCs to enhance their immunogenicity in cattle and other large mammalian species including humans.

Vaccination is considered the most successful health intervention; yet incomplete immunisation coverage continues to risk outbreaks of vaccine preventable diseases worldwide. Vaccination ...coverage improvement through a single‐dose prime‐boost technology would revolutionise modern vaccinology, impacting on disease prevalence, significantly benefiting health care and lowering economic burden of disease.
Key findings
Over the past 30 years, there have been efforts to develop a single‐dose delayed release vaccine technology that could replace the repeated prime‐boost immunisations required for many current vaccines. Biocompatible polymers have been employed to encapsulate model vaccines for delayed delivery in vivo, using either continuous or pulsed release. Biomaterial considerations, safety aspects, particle characteristics and immunological aspects of this approach are discussed in detail.
Summary
Despite many studies showing the feasibility of vaccine encapsulation for single‐dose prime‐boost administration, none have been translated into convincing utility in animal models or human trials. Further development of the encapsulation technology, through optimising the particle composition, formulation, antigen loading efficacy and stability, could lead to the application of this important approach in vaccine deployment. If successful, this would provide a solution to better global vaccination coverage through a reduction in the number of immunisations needed to achieve protection against infectious diseases. This review provides an overview of single‐dose vaccination in the context of today's vaccine needs and is derived from a body of literature that has not been reviewed for over a decade.
A vaccine for Chlamydia trachomatis is of urgent medical need. We explored bioinformatic approaches to generate an immunogen against C. trachomatis that would induce cross-serovar T-cell responses as ...(i) CD4(+) T cells have been shown in animal models and human studies to be important in chlamydial protection and (ii) antibody responses may be restrictive and serovar specific.
A consensus antigen based on over 1,500 major outer membrane protein (MOMP) sequences provided high epitope coverage against the most prevalent C. trachomatis strains in silico. Having designed the T-cell immunogen, we assessed it for immunogenicity in prime-boost regimens. This consensus MOMP transgene was delivered using plasmid DNA, Human Adenovirus 5 (HuAd5) or modified vaccinia Ankara (MVA) vectors with or without MF59(®) adjuvanted recombinant MOMP protein.
Different regimens induced distinct immune profiles. The DNA-HuAd5-MVA-Protein vaccine regimen induced a cellular response with a Th1-biased serum antibody response, alongside high serum and vaginal MOMP-specific antibodies. This regimen significantly enhanced clearance against intravaginal C. trachomatis serovar D infection in both BALB/c and B6C3F1 mouse strains. This enhanced clearance was shown to be CD4(+) T-cell dependent. Future studies will need to confirm the specificity and precise mechanisms of protection.
A C. trachomatis vaccine needs to induce a robust cellular response with broad cross-serovar coverage and a heterologous prime-boost regimen may be an approach to achieve this.
Poly(lactic-co-glycolic acid) (PLGA) is the most commonly described biocompatible copolymer used in biomedical applications. In this work, a green synthetic approach based on the biocompatible zinc ...proline complex, as an initiator for PLGA synthesis, is reported for the first time for the synthesis of methoxy-poly(ethylene glycol)-block-poly(l-lactic-co-glycolic acid) (mPEG-PLGA). mPEG-PLGA with controlled molecular weight and narrow polydispersity was synthesised. Its potential for delivery of irinotecan (Ir), a poorly water-soluble chemotherapeutic drug used for the treatment of colon and pancreatic cancer, was studied. Nanoparticles of controlled size (140-160 nm), surface charge (∼-10 mV), release properties and cytotoxicity against CT-26 (colon) and BxPC-3 (pancreatic) cancer cells, were prepared. Tumor accumulation was confirmed by optical imaging of fluorescently labelled nanoparticles. Unlike Tween® 80 coated NP-Ir, the Pluronic® F-127 coated NP-Ir exhibits significant tumor growth delay compared to untreated and blank formulation treated groups in the CT-26 subcutaneous tumor model, after 4 treatments of 30 mg irinotecan per kg dose. Overall, this proof-of-concept study demonstrates that the newly synthesized copolymer, via a green route, is proven to be nontoxic, requires fewer purification steps and has potential applications in drug delivery.
Point of use drinking water treatment with the BioSand filter (BSF) allows people to treat their water in the home. The purpose of this research was to document the ability of the Hydraid ...plastic-housing BSF to reduce diarrheal disease in households who received a BSF in a randomized controlled trial. The trial of the Hydraid plastic-housing BSF was carried out in rural, mountainous communities in Copan, Honduras during April of 2008 to February of 2009. A logistic regression adjusting for clustering showed that the incidence of diarrheal disease in children under 5 years was reduced by approximately 45% (odds ratio = 0.55, 95% confidence interval = 0.28, 1.10) in households that had a BSF compared with those households without a BSF, but this finding fluctuated depending on season and was not statistically significant. Households with a BSF had significantly better drinking water quality regardless of water source or season.
A variety of high-quality spectroscopic studies have contributed to knowledge of the formyl cation, HCO super(+), and its rare isotopoiogues, but technical limitations have previously limited precise ...determinations of the far-infrared, or terahertz spectrum. This study extends the microwave, millimeter, and submillimeter spectroscopy of HCO super(+) into the terahertz range. The resulting measurements and predictions are of sufficient coverage to adequately address astrophysical questions about this species using the Herschel Space Observatory or the Atacama Large Millimeter Array.
Abstract
Metal nanoparticles have been exploited as vaccine delivery platforms for various diseases. In particular, Ag and Au NPs of different size and shape have been shown to promote delivery of ...antigens and act as efficient adjuvants. Yet, there is limited description of cuboidal Ag nanocubes (AgNCs) and Au nanocages (AuNCs) as antigen carriers for vaccination. Here, we first investigated the intrinsic protein binding ability of AgNCs and AuNCs, using ovalbumin (OVA) as a model. Next, the effect of AgNCs and AuNCs on bone marrow-derived dendritic cells (BMDCs) in terms of cytotoxicity and maturation was assessed. Finally, we investigated humoral and cellular immune responses of AgNCs-OVA and AuNCs-OVA following intramuscular immunization and their prophylactic effects in B16F10-OVA mice tumor model. In terms of OVA loading efficiency, AgNCs were more potent than AuNCs, despite their non-porous form. Both cubic nanomaterials were found not to induce BMDCs maturation in vitro or DCs maturation in spleen. Following the administration of nanovaccine doses, serum IgG responses were comparable between groups. However, there were significant alterations in relative frequencies of lymphocyte sub-populations. Overall, AgNCs-OVA and AuNCs-OVA immunization via intramuscular route did not alter tumour growth. This study has demonstrated the intrinsic immunomodulatory properties of AgNCs and AuNCs, preceding their potential use as nanocarriers.

Purpose. To report the outcomes and adverse events of people diagnosed with lumbar disc herniation with associated radiculopathy (LDHR) who were treated with a physiotherapy functional restoration ...programme.
Method. Data on functional outcome (Oswestry score), work status, global rating of change, and adverse events were extracted from the files of all people with LDHR treated by three physiotherapists using functional restoration principles from 2001 to 2009.
Results. Ninety-five participants were included. The Oswestry score reduced by a mean of 15.9 (95%% CI, 11.8-20.1) points between baseline and discharge following a mean (SD) of 8.7 (9.4) months of treatment. The proportion of participants working full-time increased from 37%% to 67%% between baseline and discharge (p < 0.001). Eighty per cent of participants reported overall improvement between baseline and discharge on the global rating of change scale (p < 0.001). Minor adverse events were reported by eight (8%%) participants, while one (1%%) participant developed adhesive capsulitis.
Conclusion. People with LDHR who undertook a physiotherapy functional restoration programme achieved significant improvements in Oswestry disability scores, work status, and global rating of change, with few adverse events reported. A randomised controlled trial is warranted to determine the efficacy of functional restoration for people with this condition.
Context. Over the past five decades, radio astronomy has shown that molecular complexity is a natural outcome of interstellar chemistry, in particular in star forming regions. However, the pathways ...that lead to the formation of complex molecules are not completely understood and the depth of chemical complexity has not been entirely revealed. In addition, the sulfur chemistry in the dense interstellar medium is not well understood. Aims. We want to know the relative abundances of alkanethiols and alkanols in the Galactic center source Sagittarius B2(N2), the northern hot molecular core in Sgr B2(N), whose relatively small line widths are favorable for studying the molecular complexity in space. Methods. We investigated spectroscopic parameter sets that were able to reproduce published laboratory rotational spectra of ethanethiol and studied effects that modify intensities in the predicted rotational spectrum of ethanol. We used the Atacama Large Millimeter Array (ALMA) in its Cycles 0 and 1 for a spectral line survey of Sagittarius B2(N) between 84 and 114.4 GHz. These data were analyzed by assuming local thermodynamic equilibrium (LTE) for each molecule. Our observations are supplemented by astrochemical modeling; a new network is used that includes reaction pathways for alkanethiols for the first time. Results. We detected methanol and ethanol in their parent 12C species and their isotopologs with one 12C atom substituted by 13C; the latter were detected for the first time unambiguously in the case of ethanol. The 12C/13C ratio is ~25 for both molecules. In addition, we identified CH318 OH with a 16O/18O ratio of ~180 and a 13CH3OH/CH318 OH ratio of ~7.3. Upper limits were derived for the next larger alkanols normal- and iso-propanol. We observed methanethiol, CH3SH, also known as methyl mercaptan, including torsionally excited transitions for the first time. We also identified transitions of ethanethiol (or ethyl mercaptan), though not enough to claim a secure detection in this source. The ratios CH3SH to C2H5SH and C2H5OH to C2H5SH are ≳21 and ≳125, respectively. In the process of our study, we noted severe discrepancies in the intensities of observed and predicted ethanol transitions and propose a change in the relative signs of the dipole moment components. In addition, we determined alternative sets of spectroscopic parameters for ethanethiol. The astrochemical models indicate that substantial quantities of both CH3SH and C2H5SH may be produced on the surfaces of dust grains, to be later released into the gas phase. The modeled ratio CH3SH/C2H5SH = 3.1 is lower than the observed value of ≳21; the model value appears to be affected most by the underprediction of CH3SH relative to CH3OH and C2H5OH, as judged by a very high CH3OH/CH3SH ratio. Conclusions. The column density ratios involving methanol, ethanol, and methanethiol in Sgr B2(N2) are similar to values reported for Orion KL, but those involving ethanethiol are significantly different and suggest that the detection of ethanethiol reported toward Orion KL is uncertain. Our chemical model presently does not permit the prediction of sufficiently accurate column densities of alkanethiols or their ratios among alkanethiols and alkanols. Therefore, additional observational results are required to establish the level of C2H5SH in the dense and warm interstellar medium with certainty.
