We present a 30 h integration Very Large Telescope X-shooter spectrum of the Lyman series forest towards the z = 7.084 quasar ULAS J1120+0641. The only detected transmission at S/N > 5 is confined to ...seven narrow spikes in the Lyα forest, over the redshift range 5.858 <z < 6.122, just longward of the wavelength of the onset of the Lyβ forest. There is also a possible detection of one further unresolved spike in the Lyβ forest at z = 6.854, with S/N = 4.5. We also present revised Hubble Space Telescope F814W photometry of the source. The summed flux from the transmission spikes is in agreement with the F814W photometry, so all the transmission in the Lyman series forest may have been detected. There is a Gunn-Peterson (GP) trough in the Lyα forest from z = 6.122 all the way to the quasar near zone at z = 7.04. The trough, of comoving length 240 h-1 Mpc, is over twice as long as the next longest known GP trough. We combine the spectroscopic and photometric results to constrain the evolution of the Lyα effective optical depth (τGPeff) with redshift, extending a similar analysis by Simpson et al. We find τGPeff ∝ (1 + z)ξ where ξ = 11.2+ 0.4-0.6, for z > 5.5. The data nevertheless provide only a weak limit on the volume-weighted intergalactic medium (IGM) hydrogen neutral fraction at z ~ 6.5, xH i > 10-4, similar to limits at redshift z ~ 6 from less distant quasars. The new observations cannot extend measurements of the neutral fraction of the IGM to higher values because absorption in the Lyα forest is already saturated near z ~ 6. For higher neutral fractions, other methods such as measuring the red damping wing of the IGM will be required.
Tumor progression is a complex, multistep process involving accumulation of genetic aberrations and alterations in gene expression patterns leading to uncontrolled cell division, invasion into ...surrounding tissue and finally dissemination and metastasis. We have previously shown that the Arg/Abl2 non-receptor tyrosine kinase acts downstream of the EGF receptor and Src tyrosine kinases to promote invadopodium function in breast cancer cells, thereby promoting their invasiveness. However, whether and how Arg contributes to tumor development and dissemination in vivo has never been investigated. Using a mouse xenograft model, we show that knocking down Arg in breast cancer cells leads to increased tumor cell proliferation and significantly enlarged tumor size. Despite having larger tumors, the Arg-knockdown (Arg KD) tumor-bearing mice exhibit significant reductions in tumor cell invasion, intravasation into blood vessels and spontaneous metastasis to lungs. Interestingly, we found that proliferation-associated genes in the Ras-MAPK (mitogen-activated protein kinase) pathway are upregulated in Arg KD breast cancer cells, as is Ras-MAPK signaling, while invasion-associated genes are significantly downregulated. These data suggest that Arg promotes tumor cell invasion and dissemination, while simultaneously inhibiting tumor growth. We propose that Arg acts as a switch in metastatic cancer cells that governs the decision to 'grow or go' (divide or invade).
An anterior pathway, concerned with extracting meaning from sound, has been identified in nonhuman primates. An analogous pathway has been suggested in humans, but controversy exists concerning the ...degree of lateralization and the precise location where responses to intelligible speech emerge. We have demonstrated that the left anterior superior temporal sulcus (STS) responds preferentially to intelligible speech (Scott SK, Blank CC, Rosen S, Wise RJS. 2000. Identification of a pathway for intelligible speech in the left temporal lobe. Brain. 123:2400-2406.). A functional magnetic resonance imaging study in Cerebral Cortex used equivalent stimuli and univariate and multivariate analyses to argue for the greater importance of bilateral posterior when compared with the left anterior STS in responding to intelligible speech (Okada K, Rong F, Venezia J, Matchin W, Hsieh IH, Saberi K, Serences JT,Hickok G. 2010. Hierarchical organization of human auditory cortex: evidence from acoustic invariance in the response to intelligible speech. 20: 2486-2495.). Here, we also replicate our original study, demonstrating that the left anterior STS exhibits the strongest univariate response and, in decoding using the bilateral temporal cortex, contains the most informative voxels showing an increased response to intelligible speech. In contrast, in classifications using local "searchlights" and a whole brain analysis, we find greater classification accuracy in posterior rather than anterior temporal regions. Thus, we show that the precise nature of the multivariate analysis used will emphasize different response profiles associated with complex sound to speech processing.
The intestinal microbiota plays a critical role in maintaining human health; however, the mechanisms governing the normal homeostatic number and composition of these microbes are largely unknown. ...Previously it was shown that intestinal alkaline phosphatase (IAP), a small intestinal brush border enzyme, functions as a gut mucosal defence factor limiting the translocation of gut bacteria to mesenteric lymph nodes. In this study the role of IAP in the preservation of the normal homeostasis of the gut microbiota was investigated.
Bacterial culture was performed in aerobic and anaerobic conditions to quantify the number of bacteria in the stools of wild-type (WT) and IAP knockout (IAP-KO) C57BL/6 mice. Terminal restriction fragment length polymorphism, phylogenetic analyses and quantitative real-time PCR of subphylum-specific bacterial 16S rRNA genes were used to determine the compositional profiles of microbiotas. Oral supplementation of calf IAP (cIAP) was used to determine its effects on the recovery of commensal gut microbiota after antibiotic treatment and also on the colonisation of pathogenic bacteria.
IAP-KO mice had dramatically fewer and also different types of aerobic and anaerobic microbes in their stools compared with WT mice. Oral supplementation of IAP favoured the growth of commensal bacteria, enhanced restoration of gut microbiota lost due to antibiotic treatment and inhibited the growth of a pathogenic bacterium (Salmonella typhimurium).
IAP is involved in the maintenance of normal gut microbial homeostasis and may have therapeutic potential against dysbiosis and pathogenic infections.

Absorption of radiation by ice is extremely weak at visible and near-ultraviolet wavelengths, so small amounts of light-absorbing impurities in snow can dominate the absorption of solar radiation at ...these wavelengths, reducing the albedo relative to that of pure snow, contributing to the surface energy budget and leading to earlier snowmelt. In this study Arctic snow is surveyed for its content of light-absorbing impurities, expanding and updating the 1983–1984 survey of Clarke and Noone. Samples were collected in Alaska, Canada, Greenland, Svalbard, Norway, Russia, and the Arctic Ocean during 1998 and 2005–2009, on tundra, glaciers, ice caps, sea ice, frozen lakes, and in boreal forests. Snow was collected mostly in spring, when the entire winter snowpack is accessible for sampling. Sampling was carried out in summer on the Greenland Ice Sheet and on the Arctic Ocean, of melting glacier snow and sea ice as well as cold snow. About 1200 snow samples have been analyzed for this study. The snow is melted and filtered; the filters are analyzed in a specially designed spectrophotometer system to infer the concentration of black carbon (BC), the fraction of absorption due to non-BC light-absorbing constituents and the absorption Ångstrom exponent of all particles. This is done using BC calibration standards having a mass absorption efficiency of 6.0 m2 g−1 at 550 nm and by making an assumption that the absorption Angstrom exponent for BC is 1.0 and for non-BC light-absorbing aerosol is 5.0. The reduction of snow albedo is primarily due to BC, but other impurities, principally brown (organic) carbon, are typically responsible for ~40% of the visible and ultraviolet absorption. The meltwater from selected snow samples was saved for chemical analysis to identify sources of the impurities. Median BC amounts in surface snow are as follows (nanograms of carbon per gram of snow): Greenland 3, Arctic Ocean snow 7, melting sea ice 8, Arctic Canada 8, subarctic Canada 14, Svalbard 13, Northern Norway 21, western Arctic Russia 27, northeastern Siberia 34. Concentrations are more variable in the European Arctic than in Arctic Canada or the Arctic Ocean, probably because of the proximity to BC sources. Individual samples of falling snow were collected on Svalbard, documenting the springtime decline of BC from March through May. Absorption Ångstrom exponents are 1.5–1.7 in Norway, Svalbard, and western Russia, 2.1–2.3 elsewhere in the Arctic, and 2.5 in Greenland. Correspondingly, the estimated contribution to absorption by non-BC constituents in these regions is ~25%, 40%, and 50% respectively. It has been hypothesized that when the snow surface layer melts some of the BC is left at the top of the snowpack rather than being carried away in meltwater. This process was observed in a few locations and would cause a positive feedback on snowmelt. The BC content of the Arctic atmosphere has declined markedly since 1989, according to the continuous measurements of near-surface air at Alert (Canada), Barrow (Alaska), and Ny-Ålesund (Svalbard). Correspondingly, the new BC concentrations for Arctic snow are somewhat lower than those reported by Clarke and Noone for 1983–1984, but because of methodological differences it is not clear that the differences are significant. Nevertheless, the BC content of Arctic snow appears to be no higher now than in 1984, so it is doubtful that BC in Arctic snow has contributed to the rapid decline of Arctic sea ice in recent years.
We present new Spitzer Infrared Array Camera (IRAC) photometry of 12 very late-type T dwarfs: nine have 3.6, 4.5, 5.8, and 8.0 photometry and three have 3.6 and 4.5 photometry only. Combining this ...with previously published photometry, we investigate trends with type and color that are useful for both the planning and interpretation of infrared surveys designed to discover the coldest T or Y dwarfs. The online appendix provides a collation of MKO-system YJHKL'M' and IRAC photometry for a sample of M, L, and T dwarfs. Brown dwarfs with effective temperature (T{sub eff}) below 700 K emit more than half their flux at wavelengths longer than 3 {mu}m, and the ratio of the mid-infrared flux to the near-infrared flux becomes very sensitive to T{sub eff} at these low temperatures. We confirm that the color H (1.6 {mu}m) - 4.5 is a good indicator of T{sub eff} with a relatively weak dependence on metallicity and gravity. Conversely, the colors H - K (2.2 {mu}m) and 4.5 - 5.8 are sensitive to metallicity and gravity. Thus, near- and mid-infrared photometry provide useful indicators of the fundamental properties of brown dwarfs, and if temperature and gravity are known, then mass and age can be reliably determined from evolutionary models. There are 12 dwarfs currently known with H- 4.5 >3.0, and 500 K {approx}< T{sub eff} {approx}<800 K, which we examine in detail. The ages of the dwarfs in the sample range from very young (0.1-1.0 Gyr) to relatively old (3-12 Gyr). The mass range is possibly as low as 5 Jupiter masses to up to 70 Jupiter masses, i.e., near the hydrogen burning limit. The metallicities also span a large range, from m/H = -0.3 to m/H = +0.3. The small number of T8-T9 dwarfs found in the UK Infrared Telescope Infrared Deep Sky Survey to date appear to be predominantly young low-mass dwarfs. Accurate mid-infrared photometry of cold brown dwarfs is essentially impossible from the ground, and extensions to the mid-infrared space missions, warm-Spitzer and Wide-Field Infrared Survey Explorer, are desirable in order to obtain the vital mid-infrared data for cold brown dwarfs, and to discover more of these rare objects.
We identify galaxy groups and clusters in volume-limited samples of the Sloan Digital Sky Survey (SDSS) redshift survey, using a redshift-space friends-of-friends algorithm. We optimize the ...friends-of-friends linking lengths to recover galaxy systems that occupy the same dark matter halos, using a set of mock catalogs created by populating halos of N-body simulations with galaxies. Extensive tests with these mock catalogs show that no combination of perpendicular and line-of-sight linking lengths is able to yield groups and clusters that simultaneously recover the true halo multiplicity function, projected size distribution, and velocity dispersion. We adopt a linking length combination that yields, for galaxy groups with 10 or more members: a group multiplicity function that is unbiased with respect to the true halo multiplicity function; an unbiased median relation between the multiplicities of groups and their associated halos; a spurious group fraction of less than 61%; a halo completeness of more than 697%; the correct projected size distribution as a function of multiplicity; and a velocity dispersion distribution that is 620% too low at all multiplicities. These results hold over a range of mock catalogs that use different input recipes of populating halos with galaxies. We apply our group-finding algorithm to the SDSS data and obtain three group and cluster catalogs for three volume-limited samples that cover 3495.1 deg super(2) on the sky, go out to redshifts of 0.1, 0.068, and 0.045, and contain 57,138, 37,820, and 18,895 galaxies, respectively. We correct for incompleteness caused by fiber collisions and survey edges and obtain measurements of the group multiplicity function, with errors calculated from realistic mock catalogs. These multiplicity function measurements provide a key constraint on the relation between galaxy populations and dark matter halos.

Here we ...design a Pt(II) pincer-type material with selective absorptive and emissive responses to methanol and water. The yellow anhydrous form converts reversibly on a subsecond timescale to a red hydrate in the presence of parts-per-thousand levels of atmospheric water vapour. Exposure to methanol induces a similarly-rapid and reversible colour change to a blue methanol solvate. Stable smart coatings on glass demonstrate robust switching over 10
cycles, and flexible microporous polymer membranes incorporating microcrystals of the complex show identical vapochromic behaviour. The rapid vapochromic response can be rationalised from the crystal structure, and in combination with quantum-chemical modelling, we provide a complete microscopic picture of the switching mechanism. We discuss how this multiscale design approach can be used to obtain new compounds with tailored VOC selectivity and spectral responses.
Next-generation genetic sequencing (NGS) technologies facilitate the screening of multiple genes linked to neurodegenerative dementia, but there are few reports about their use in clinical practice. ...Which patients would most profit from testing, and information on the likelihood of discovery of a causal variant in a clinical syndrome, are conspicuously absent from the literature, mostly for a lack of large-scale studies. We applied a validated NGS dementia panel to 3241 patients with dementia and healthy aged controls; 13,152 variants were classified by likelihood of pathogenicity. We identified 354 deleterious variants (DV, 12.6% of patients); 39 were novel DVs. Age at clinical onset, clinical syndrome and family history each strongly predict the likelihood of finding a DV, but healthcare setting and gender did not. DVs were frequently found in genes not usually associated with the clinical syndrome. Patients recruited from primary referral centres were compared with those seen at higher-level research centres and a national clinical neurogenetic laboratory; rates of discovery were comparable, making selection bias unlikely and the results generalisable to clinical practice. We estimated penetrance of DVs using large-scale online genomic population databases and found 71 with evidence of reduced penetrance. Two DVs in the same patient were found more frequently than expected. These data should provide a basis for more informed counselling and clinical decision making.
The quasar ULAS J1120+0641 at redshiftz= 7.085 has a highly ionized near zone which is smaller than those around quasars of similar luminosity atzsime 6. The spectrum also exhibits evidence for a ...damping wing extending redward of the systemic Ly alpha redshift. We use radiative transfer simulations in a cosmological context to investigate the implications for the ionization state of the inhomogeneous intergalactic medium (IGM) surrounding this quasar. Our simulations show that the transmission profile is consistent with an IGM in the vicinity of the quasar with a volume averaged Hi fraction of and that ULAS J1120+0641 has been bright for 106-107yr. The observed spectrum is also consistent with smaller IGM neutral fractions, , if a damped Ly alpha system in an otherwise highly ionized IGM lies within 5 proper Mpc of the quasar. This is, however, predicted to occur in only similar to 5 per cent of our simulated sightlines for a bright phase of 106-107yr. Unless ULAS J1120+0641 grows during a previous optically obscured phase, the low age inferred for the quasar adds to the theoretical challenge of forming a 2 109Modot black hole at this high redshift.
