Around 80% of domestic heat demand in Great Britain (GB) is supplied by natural gas, but continuing to heat dwellings in this way is unlikely to be compatible with national emission reduction ...targets. Electrical heating using heat pumps is expected to play a significant role in future space heating and hot water provision. The assessment of future heating technologies requires knowledge of the current demand for heat at short time intervals in order to evaluate peak demands and possible storage requirements. Existing half-hourly national heat demand estimates are built on data from small samples of dwellings. This paper provides estimates of GB domestic heat demand under mild, normal and cold weather conditions based on data from over 6000 dwellings collected between May 2009 and July 2010 that participated in the GB smart meter trial. The calculated peak domestic heat demand of 170 GW is around 40% lower than previously calculated suggesting that the difficulties surrounding the electrification of heat are far less profound than previously assumed. These results can be used in the development of future energy pathways and scenarios.

RNA viruses within infected individuals exist as a population of evolutionary-related variants. Owing to evolutionary change affecting the constitution of this population, the frequency and/or ...occurrence of individual viral variants can show marked or subtle fluctuations. Since the development of massively parallel sequencing platforms, such viral populations can now be investigated to unprecedented resolution. A critical problem with such analyses is the presence of sequencing-related errors that obscure the identification of true biological variants present at low frequency. Here, we report the development and assessment of the Quality Assessment of Short Read (QUASR) Pipeline (http://sourceforge.net/projects/quasr) specific for virus genome short read analysis that minimizes sequencing errors from multiple deep-sequencing platforms, and enables post-mapping analysis of the minority variants within the viral population. QUASR significantly reduces the error-related noise in deep-sequencing datasets, resulting in increased mapping accuracy and reduction of erroneous mutations. Using QUASR, we have determined influenza virus genome dynamics in sequential samples from an in vitro evolution of 2009 pandemic H1N1 (A/H1N1/09) influenza from samples sequenced on both the Roche 454 GSFLX and Illumina GAIIx platforms. Importantly, concordance between the 454 and Illumina sequencing allowed unambiguous minority-variant detection and accurate determination of virus population turnover in vitro.

The Galactic Archaeology with HERMES (GALAH) survey is a large high-resolution spectroscopic survey using the newly commissioned High Efficiency and Resolution Multi-Element Spectrograph (HERMES) on ...the Anglo-Australian Telescope. The HERMES spectrograph provides high-resolution (R ∼ 28 000) spectra in four passbands for 392 stars simultaneously over a 2 deg field of view. The goal of the survey is to unravel the formation and evolutionary history of the Milky Way, using fossil remnants of ancient star formation events which have been disrupted and are now dispersed throughout the Galaxy. Chemical tagging seeks to identify such dispersed remnants solely from their common and unique chemical signatures; these groups are unidentifiable from their spatial, photometric or kinematic properties. To carry out chemical tagging, the GALAH survey will acquire spectra for a million stars down to V ∼ 14. The HERMES spectra of FGK stars contain absorption lines from 29 elements including light proton-capture elements, α-elements, odd-Z elements, iron-peak elements and n-capture elements from the light and heavy s-process and the r-process. This paper describes the motivation and planned execution of the GALAH survey, and presents some results on the first-light performance of HERMES.
In this paper, we present Q-U-I JOint Tenerife Experiment (QUIJOTE) 10–20 GHz observations (194 h in total over ≈250 deg2) in intensity and polarisation of G159.6-18.5, one of the most widely studied ...regions harbouring anomalous microwave emission (AME). By combining with other publicly available intensity data, we achieve the most precise spectrum of the AME measured to date in an individual region, with 13 independent data points between 10 and 50 GHz being dominated by this emission. The four QUIJOTE data points provide the first independent confirmation of the downturn of the AME spectrum at low frequencies, initially unveiled by the COSMOlogical Structures On Medium Angular Scales experiment in this region. Our polarisation maps, which have an angular resolution of ≈1° and a sensitivity of ≈ 25 μK beam−1, are consistent with zero polarisation. We obtain upper limits on the polarisation fraction of Π < 6.3 and <2.8 per cent (95 per cent C.L.), respectively, at 12 and 18 GHz (ΠAME < 10.1 and <3.4 per cent with respect to the residual AME intensity), a frequency range where no AME polarisation observations have been reported to date. The combination of these constraints with those from other experiments confirm that all the magnetic dust models based on single-domain grains, and most of those considering randomly oriented magnetic inclusions, predict higher polarisation levels than is observed towards regions with AME. Also, neither of the two considered models of electric dipole emission seems to be compatible with all the observations together. More stringent constraints of the AME polarisation at 10–40 GHz are necessary to disentangle between different models, to which future QUIJOTE data will contribute.
The GLIMPSE (Galactic Legacy Mid-Plane Survey Extraordinaire) Point Source Catalog of 630 million mid-infrared sources toward the inner Galaxy, 10 , "l" 65 , 65 and "b" , 1, was used to determine the ...distribution of stars in Galactic longitude, l, latitude, b, and apparent magnitude, m. The counts versus longitude can be approximated by the modified Bessel function N = N sub(0)(l/l sub(0)) K sub(1)(l/l sub(0)), where l sub(0) is insensitive to limiting magnitude, band choice, and side of Galactic center: l sub(0) = 17-30 with a best-fit value in the 4.5 km band of l sub(0) = 24 c 4. Modeling the source distribution as an exponential disk yields a radial scale length of H sub(*) = 3.9 c 0.6 kpc. There is a pronounced north-south asymmetry in source counts for "l" 30, with 625% more stars in the north. For l = 10-30, there is a strong enhancement of stars of m = 11.5-13.5 mag. A linear bar passing through the Galactic center with half-length R sub(bar) = 4.4 c 0.5 kpc, tilted by h = 44 c 10 to the Sun-Galactic center line, provides the simplest interpretation of these data. We examine the possibility that enhanced source counts at l = 26-28, 31.5-34, and 306-309 are related to Galactic spiral structure. Total source counts are depressed in regions where the counts of red objects (m sub(K)-m sub(8.0) > 3) peak. In these areas, the counts are reduced by extinction due to molecular gas, high diffuse backgrounds associated with star formation, or both.
CRISPR-Cas systems provide bacteria and archaea with adaptive immunity against genetic invaders, such as bacteriophages. The systems integrate short sequences from the phage genome into the bacterial ...CRISPR array. These 'spacers' provide sequence-specific immunity but drive natural selection of evolved phage mutants that escape the CRISPR-Cas defence. Spacer acquisition occurs by either naive or primed adaptation. Naive adaptation typically results in the incorporation of a single spacer. By contrast, priming is a positive feedback loop that often results in acquisition of multiple spacers, which occurs when a pre-existing spacer matches the invading phage. We predicted that single and multiple spacers, representative of naive and primed adaptation, respectively, would cause differing outcomes after phage infection. We investigated the response of two phages, ϕTE and ϕM1, to the Pectobacterium atrosepticum type I-F CRISPR-Cas system and observed that escape from single spacers typically occurred via point mutations. Alternatively, phages escaped multiple spacers through deletions, which can occur in genes encoding structural proteins. Cryo-EM analysis of the ϕTE structure revealed shortened tails in escape mutants with tape measure protein deletions. We conclude that CRISPR-Cas systems can drive phage genetic diversity, altering morphology and fitness, through selective pressures arising from naive and primed acquisition events. We use a new, well‐calibrated 500 year paleorecord off southern California to determine collapse frequency, cross correlation, persistence, and return times of exploited forage fish populations. The ...paleorecord shows that “collapse” (defined as <10% of the mean peak biomass) is a normal state repeatedly experienced by northern anchovy, Pacific hake, and Pacific sardine which were collapsed 29–40% of the time, prior to commercial fishing exploitation. Mean (± SD) persistence of “fishable biomass” (defined as one third mean peak biomass from the paleorecord) was 19 ± 18, 15 ± 17, and 12 ± 7 years for anchovy, hake, and sardine. Mean return times to the same biomass was 8 years for anchovy but 22 years for sardine and hake. Further, we find that sardine and anchovy are positively correlated over 400 years, consistent with coherent declines of both species off California. Persistence and return times combined with positive sardine‐anchovy correlation indicate that on average 1–2 decades of fishable biomass will be followed by 1–2 decades of low forage. Forage populations are resilient on the 500 year time scale, but their collapse and recovery cycle (based on the paleorecord) are suited to alternating periods of high fishing mortality and periods of little or no fishing.

A visual examination of the images from the Galactic Legacy Infrared Mid-Plane Survey Extraordinaire (GLIMPSE) has revealed 322 partial and closed rings that we propose represent partially or fully ...enclosed three-dimensional bubbles. We argue that the bubbles are primarily formed by hot young stars in massive star formation regions. We have found an average of about 1.5 bubbles per square degree. About 25% of the bubbles coincide with known radio H II regions, and about 13% enclose known star clusters. It appears that B4-B9 stars (too cool to produce detectable radio H II regions) probably produce about three-quarters of the bubbles in our sample, and the remainder are produced by young O-B3 stars that produce detectable radio H II regions. Some of the bubbles may be the outer edges of H II regions where PAH spectral features are excited and may not be dynamically formed by stellar winds. Only three of the bubbles are identified as known SNRs. No bubbles coincide with known planetary nebulae or W-R stars in the GLIMPSE survey area. The bubbles are small. The distribution of angular diameters peaks between 1' and 3' with over 98% having angular diameters less than 10' and 88% less than 4'. Almost 90% have shell thicknesses between 0.2 and 0.4 of their outer radii. Bubble shell thickness increases approximately linearly with shell radius. The eccentricities are rather large, peaking between 0.6 and 0.7; about 65% have eccentricities between 0.55 and 0.85.

Achalasia is a relatively rare primary motor esophageal disorder, characterized by absence of relaxations of the lower esophageal sphincter and of peristalsis along the esophageal body. As a result, ...patients typically present with dysphagia, regurgitation and occasionally chest pain, pulmonary complication and malnutrition. New diagnostic methodologies and therapeutic techniques have been recently added to the armamentarium for treating achalasia. With the aim to offer clinicians and patients an up-to-date framework for making informed decisions on the management of this disease, the International Society for Diseases of the Esophagus Guidelines proposed and endorsed the Esophageal Achalasia Guidelines (I-GOAL). The guidelines were prepared according the Appraisal of Guidelines for Research and Evaluation (AGREE-REX) tool, accredited for guideline production by NICE UK. A systematic literature search was performed and the quality of evidence and the strength of recommendations were graded according to the Grading of Recommendations Assessment, Development and Evaluation (GRADE). Given the relative rarity of this disease and the paucity of high-level evidence in the literature, this process was integrated with a three-step process of anonymous voting on each statement (DELPHI). Only statements with an approval rate >80% were accepted in the guidelines. Fifty-one experts from 11 countries and 3 representatives from patient support associations participated to the preparations of the guidelines. These guidelines deal specifically with the following achalasia issues: Diagnostic workup, Definition of the disease, Severity of presentation, Medical treatment, Botulinum Toxin injection, Pneumatic dilatation, POEM, Other endoscopic treatments, Laparoscopic myotomy, Definition of recurrence, Follow up and risk of cancer, Management of end stage achalasia, Treatment options for failure, Achalasia in children, Achalasia secondary to Chagas' disease.
Summary
Background
The epidermal barrier is important for water conservation, failure of which is evident in dry‐skin conditions. Barrier function is fulfilled by the stratum corneum, tight junctions ...(TJs, which control extracellular water) and keratinocyte mechanisms, such as organic osmolyte transport, which regulate intracellular water homeostasis. Organic osmolyte transport by keratinocytes is largely unexplored and nothing is known regarding how cellular and extracellular mechanisms of water conservation may interact.
Objectives
We aimed to characterize osmolyte transporters in skin and keratinocytes, and, using transporter inhibitors, to investigate whether osmolytes can modify TJs. Such modification would suggest a possible link between intracellular and extracellular mechanisms of water regulation in skin.
Methods
Immunostaining and quantitative polymerase chain reaction of organic osmolyte‐treated organ‐cultured skin were used to identify changes to organic osmolyte transporters, and TJ protein and gene expression. TJ functional assays were performed on organic osmolyte‐treated primary human keratinocytes in culture.
Results
Immunostaining demonstrated the expression of transporters for betaine, taurine and myo‐inositol in transporter‐specific patterns. Treatment of human skin with either betaine or taurine increased the expression of claudin‐1, claudin‐4 and occludin. Osmolyte transporter inhibition abolished this response. Betaine and taurine increased TJ function in primary human keratinocytes in vitro.
Conclusions
Treatment of skin with organic osmolytes modulates TJ structure and function, which could contribute to the epidermal barrier. This emphasizes a role for organic osmolytes beyond the maintenance of intracellular osmolarity. This could be harnessed to enhance topical therapies for diseases characterized by skin barrier dysfunction.

Limited studies have shown that organic osmolytes protect keratinocytes from stressors such as ultraviolet, but their transporters have never been shown in human skin.
