Recent advances in fluorescence imaging permit large-scale recording of neural activity and dynamics of neurochemical release with unprecedented resolution in behaving animals. Calcium imaging with ...highly optimized genetically encoded indicators provides a mesoscopic view of neural activity from genetically defined populations at cellular and subcellular resolutions. Rigorously improved voltage sensors and microscopy allow for robust spike imaging of populational neurons in various brain regions. In addition, recent protein engineering efforts in the past few years have led to the development of sensors for neurotransmitters and neuromodulators. Here, we discuss the development and applications of these genetically encoded fluorescent indicators in reporting neural activity in response to various behaviors in different biological systems as well as in drug discovery. We also report a simple model to guide sensor selection and optimization.
Abstract Using data from Wave 4 of the National Longitudinal Study of Adolescent to Adult Health, this study examined the independent relationship of childhood maltreatment type (emotional, sexual, ...physical) on IPV victimization and perpetration; then mutually exclusive categories of IPV involvement (victimization, perpetration, and victimization/perpetration) were investigated. IPV victimization and perpetration were assessed using items from the revised Conflict Tactics Scales. A series of binary regression models and multinomial regression models were estimated. Models were stratified across gender. Results uncovered significant relationships between child physical abuse and IPV victimization as well as IPV perpetration for males and females, but this effect was reduced when emotional maltreatment was added to the model. When IPV victimization/perpetration was considered, maltreatment effects changed. For males, physical maltreatment remained significantly related to victimization only and physical, sexual, and emotional maltreatment were related to victimization/perpetration. For females, physical maltreatment remained significantly related to IPV victimization only and emotional maltreatment was related to perpetration only and to victimization/perpetration. Screening and intervention for maltreatment, including emotional maltreatment, among children as well as adults with IPV histories may be important to preventing first IPV experiences and stemming current involvement.

Australia has one of the highest incidence rates of inflammatory bowel disease (IBD) in the world. Early diagnosis and treatment for IBD is critical. For Crohn disease, in particular, this may change ...the natural history of disease and reduce disability. Faecal calprotectin is a sensitive test that can be used by primary care physicians to assist in determining which patients with gastrointestinal symptoms may have IBD. This allows for prompt identification of patients who may benefit from endoscopy. Regular re-evaluation of disease status with strategies that can safely, readily and reliably detect the presence of inflammation with faecal biomarkers and imaging is important. To avoid the risks of cumulative radiation exposure, magnetic resonance imaging and/or intestinal ultrasound, rather than computed tomography scanning, should be performed when possible. Drug treatments for IBD now include five biological drugs listed by the Pharmaceutical Benefits Scheme: adalimumab, infliximab, golimumab, vedolizumab and ustekinumab. Such developments offer the possibility for improved disease control in selected patients.
Background
The gastrointestinal microbiota is the key antigenic drive in the inflammatory bowel diseases. Randomized controlled trials (RCTs) in ulcerative colitis have established fecal microbiota ...transplantation (FMT) as an effective therapy. We have conducted a systematic review to evaluate the efficacy of FMT in Crohn's disease.
Methods
A systematic literature search was performed through to August 2020 (MEDLINE; Embase). Studies were included if they reported FMT administration in patients with Crohn's disease, and reported on clinical outcomes.
Results
Fifteen studies published between 2014 and 2020, comprising 13 cohort studies and two RCTs, were included in the analysis. The majority of trials evaluated FMT for induction of remission, with follow‐up duration varying from 4 to 52 weeks. One RCT in 21 patients, of single‐dose FMT versus placebo, following steroid‐induced remission, showed a higher rate of steroid‐free clinical remission in the FMT group compared to the control group: 87.5% vs 44.4% at week 10 (P = 0.23). Another RCT, two‐dose FMT in 31 patients, showed an overall clinical remission rate of 36% at week 8, however, with no difference in clinical or endoscopic endpoints between FMT administered by gastroscopy and colonoscopy. Considering all studies, the clinical response rates in early follow up were higher following multiple FMT than with single FMT. FMT dose did not appear to influence clinical outcomes, nor did whether FMT was fresh or frozen. FMT delivered via upper gastrointestinal route demonstrated higher early efficacy rates of 75 to 100% compared with lower delivery route rates of 30% to 58%, but on follow up beyond 8 weeks, this difference was not maintained. Whether pre‐FMT antibiotic administration was beneficial was not able to be determined due to the limited number of patients receiving antibiotics and varying antibiotic regimens. No serious adverse events were reported.
Conclusions
Preliminary studies suggest that FMT may be an effective therapy in Crohn's disease. However large controlled trials are needed. No serious safety concerns have been identified.
Research has demonstrated that exposure to violence can result in many negative consequences for youth, but the degree to which neighborhood conditions may foster resiliency among victims is not well ...understood. This study tests the hypothesis that neighborhood collective efficacy attenuates the relationship between adolescent exposure to violence, substance use, and violence. Data were collected from 1,661 to 1,718 adolescents participating in the Project on Human Development in Chicago Neighborhoods, who were diverse in terms of sex (51 % male, 49 % female), race/ethnicity (48 % Hispanic, 34 % African American, 14 % Caucasian, and 4 % other race/ethnicity), and age (mean age 12 years; range 8–16). Information on neighborhood collective efficacy was obtained from adult residents, and data from the 1990 US. Census were used to control for neighborhood disadvantage. Based on hierarchical modeling techniques to adjust for the clustered data, Bernoulli models indicated that more exposure to violence was associated with a greater likelihood of tobacco, alcohol, and marijuana use and perpetration of violence. Poisson models suggested that victimization was also related to a greater variety of substance use and violent behaviors. A moderating effect of collective efficacy was found in models assessing the variety of substance use; the relationship between victimization and substance use was weaker for youth in neighborhoods with higher versus lower levels of collective efficacy. These findings are consistent with literature indicating that social support can ameliorate the negative impact of victimization. This investigation extends this research to show that neighborhood social support can also help to promote resiliency among adolescents.
A growing body of scholarship faults existing risk/needs assessment models for neglecting the risk factors most relevant to women offenders. In response, a series of gender-responsive assessment ...models were tested for their contributions to widely used gender-neutral risk needs assessments. In six of eight samples studied, subsets of the gender-responsive scales achieved statistically significant contributions to gender-neutral models. Promising results were found for the following: (a) parental stress, family support, self-efficacy, educational assets, housing safety, anger/hostility, and current mental health factors in probation samples; (b) child abuse, anger/hostility, relationship dysfunction, family support, and current mental health factors among prisoners; and (c) adult victimization, anger/hostility, educational assets, and family support among released inmates. The predictive validity of gender-neutral assessments was strong in seven of eight samples studied. However, findings for both gender-neutral and gender-responsive domains suggested different treatment priorities for women from those currently put forward in correctional theory and policy.
The effect of abuse victimization in correctional samples has been researched previously, particularly with an eye toward these experiences on justice-involved youth and prison samples’ offending and ...recidivism behavior. The role of this type of victimization, including physical abuse, sexual abuse, and polyvictimization, is less studied in jail populations. The effect of abuse victimization is also less researched among other outcomes, including behavioral health disorders (BHDs) and substance use disorder (SUD). While the effect of abuse, generally, has been examined, less is known about how abuse perpetrator type and timing of abuse impact justice-involved individuals’ outcomes. Using logistic regressions, we examined the influence of abuse perpetrator type (non-stranger or stranger) and timing (before childhood, after childhood, or before and after childhood) in a population of jailed adults from one state (n = 4,713). Outcomes studied included internalizing BHDs, externalizing BHDs, and severe SUD. Results indicated that abuse perpetrated by a non-stranger yielded a greater impact on mental illness compared to abuse perpetrated by a stranger. In contrast to abuse experienced as an adult, childhood abuse was more consistently associated with internalizing and externalizing disorders but was not related to severe SUD, with an exception of physical abuse. Further, BHDs and SUD were strongly associated with each other. Overall, polyvictimization had the strongest effect on the outcomes compared to either physical abuse or sexual abuse alone. Our findings suggest that screening for abuse experiences as a potential destabilizing factor in justice-involved populations could improve case management and interventions for people incarcerated in jails. Results also highlight the importance of distinguishing between the perpetrator type of abuse and timing of abuse.
This paper examines how social capital aids in post-disaster community recovery and redevelopment. While previous studies on social capital and post-disaster recovery have tended to focus on social ...networks as a source of necessary assistance, the primary focus of this study is on how social capital in the form of collective narratives affects post-disaster recovery. We argue that collective narratives can shape the recovery strategies that individuals adopt. To illustrate this we examine the post-Katrina recovery efforts in St. Bernard Parish, an area devastated by flooding and significant environmental damage. In particular, we focus on the shared narrative that dominated qualitative interview data collected in St. Bernard, namely, its shared identity as a close-knit, family-oriented community comprised of hard workers. This narrative led community members to adopt a strategy that emphasized self-reliance.
This study investigated the differences in microbial community abundance, composition, and diversity throughout the depth profiles in soils collected from corn and soybean fields in Iowa (United ...States) using 16S rRNA amplicon sequencing. The results revealed decreased richness and diversity in microbial communities at increasing soil depth. Soil microbial community composition differed due to crop type only in the top 60 cm and due to location only in the top 90 cm. While the relative abundance of most phyla decreased in deep soils, the relative abundance of the phylum Proteobacteria increased and dominated agricultural soils below the depth of 90 cm. Although soil depth was the most important factor shaping microbial communities, edaphic factors, including soil organic matter, soil bulk density, and the length of time that deep soils were saturated with water, were all significant factors explaining the variation in soil microbial community composition. Soil organic matter showed the highest correlation with the exponential decrease in bacterial abundance with depth. A greater understanding of how soil depth influences the diversity and composition of soil microbial communities is vital for guiding sampling approaches in agricultural soils where plant roots extend beyond the upper soil profile. In the long term, a greater knowledge of the influence of depth on microbial communities should contribute to new strategies that enhance the sustainability of soil, which is a precious resource for food security. IMPORTANCE Determining how microbial properties change across different soils and within the soil depth profile will be potentially beneficial to understanding the long-term processes that are involved in the health of agricultural ecosystems. Most literature on soil microbes has been restricted to the easily accessible surface soils. However, deep soils are important in soil formation, carbon sequestration, and providing nutrients and water for plants. In the most productive agricultural systems in the United States where soybean and corn are grown, crop plant roots extend into the deeper regions of soils (>100 cm), but little is known about the taxonomic diversity or the factors that shape deep-soil microbial communities. The findings reported here highlight the importance of soil depth in shaping microbial communities, provide new information about edaphic factors that influence the deep-soil communities, and reveal more detailed information on taxa that exist in deep agricultural soils.
