Here, we report a simple yet effective surface-modification approach to imparting hydrophobic surfaces with superhydrophilicity using ultralow fouling/functionalizable carboxybetaine (CB) copolymers ...via a dip-coating technique. A new series of CB random copolymers with varying amphiphilicities were synthesized and coated on hydrophobic polypropylene (PP) and polystyrene (PS) surfaces. The nonfouling capability of each coating was screened by an enzyme-linked immunosorbent assay (ELISA) and further comprehensively assessed against 100% human serum by a Micro BCA protein assay kit. The random copolymer containing ∼30 mol % CB units showed superhydrophilicity with the highest air contact angle of more than 165° in DI water and the best nonfouling capability against 100% human blood serum. Surfaces of a 96-well plate coated with the optimal CB random copolymer had a significantly better nonfouling capability than those of a commercial 96-well plate with an ultralow attachment surface. The adhesion of mouse embryonic fibroblast cells (NIH3T3) was completely inhibited on surfaces coated with CB random copolymers. Furthermore, the optimal nonfouling CB copolymer surface was functionalized with an antigen via covalent bonding where its specific interactions with its antibody were verified. Thus, this CB random copolymer is capable of imparting both ultralow fouling and functionalizable capabilities to hydrophobic surfaces for blood-contacting devices.
Atmospheric water vapor is one of the important parameters for weather and climate studies. Generally, atmospheric water vapor can be monitored by some techniques, such as the Global Navigation ...Satellite System (GNSS), radiosonde (RS), remote sensing and numerical weather forecast (NWF). However, the comprehensive retrieval and application of precipitable water vapor (PWV) using multi techniques has been hardly performed before, which becomes the focus of this study. A comprehensive PWV retrieval and application platform (CPRAP) is first established by combing the ground-based (GNSS), space-based (Fengyun-3A, Sentinel-3A) and reanalysis-based (the fifth-generation reanalysis dataset of the European Centre for Medium-Range Weather Forecasting, ERA5) techniques. Additionally, its applications are then extended to drought and rainfall monitoring using the CPRAP-derived PWV. The statistical result shows that PWV derived from ground-based GNSS has high accuracy in China, with the root mean square (RMS), Bias and mean absolute error (MAE) of 2.15, 0.05 and 1.65 mm, respectively, when the RS-derived PWV is regarded as the reference. In addition, the accuracy of PWV derived from the space-based (FY-3A and Sentinel-3A) techniques technique is also validated and the RMS, Bias and MAE of a Medium Resolution Spectral Imager (MERSI) onboard Fengyun-3A (FY-3A) and an Ocean and Land Color Instrument (OLCI) onboard Sentinel-3A are 4.46/0.56/3.61 mm and 2.95/0.01/1.37 mm, respectively. Then, the performance of ERA5-derived PWV is evaluated based on GNSS-derived and RS-derived PWV. The result also shows good accuracy of ERA5-provided PWV with the averaged RMS, Bias and MAE of 1.86/0.11/1.48 mm and 0.90/−0.05/1.51 mm, respectively. Finally, the PWV data derived from the established CPRAP are further used for drought and rainfall monitoring. The applied results reveal that the calculated the standardized precipitation evapotranspiration index (SPEI) using the CPRAP-derived PWV can monitor the drought and the correlation coefficient ranges from 0.83 to 0.9 when compared with the SPEI. Furthermore, in this paper correlation analysis between PWV derived from the CPRAP and rainfall, and its potential for rainfall monitoring was also validated. Such results verify the significance of the established CPRAP for weather and climate studies.
Elementary tellurium is currently of great interest as an element with potential promise in nano-technology applications because of the recent discovery regarding its three two-dimensional phases and ...the existence of Weyl nodes around its Femi level. Here, we report on the unique nano-photonic properties of elemental tellurium particles Te(0), as harvest from a culture of a tellurium-oxyanion respiring bacteria. The bacterially-formed nano-crystals prove effective in the photonic applications tested compared to the chemically-formed nano-materials, suggesting a unique and environmentally friendly route of synthesis. Nonlinear optical measurements of this material reveal the strong saturable absorption and nonlinear optical extinctions induced by Mie scattering over broad temporal and wavelength ranges. In both cases, Te-nanoparticles exhibit superior optical nonlinearity compared to graphene. We demonstrate that biological tellurium can be used for a variety of photonic applications which include their proof-of-concept for employment as ultrafast mode-lockers and all-optical switches.
For biotherapeutics that require multiple administrations to fully cure diseases, the induction of undesirable immune response is one common cause for the failure of their treatment. Covalent binding ...of hydrophilic polymers to proteins is commonly employed to mitigate potential immune responses. However, while this technique is proved to partially reduce the antibodies (Abs) reactive to proteins, it may induce Abs toward their associated polymers and thus result in the loss of efficacy. Zwitterionic poly(carboxybetaine) (PCB) is recently shown to improve the immunologic properties of proteins without inducing any antipolymer Abs against itself. However, it is unclear if the improved immunologic profiles can translate to better clinical outcomes since improved immunogenicity cannot directly reflect amelioration in efficacy. Here, a PCB nanocage (PCB NC) is developed, which can physically encase proteins while keeping their structure intact. PCB NC encapsulation of uricase, a highly immunogenic enzyme, is demonstrated to eradicate all the immune responses. To bridge the gap between immunogenicity and efficacy studies, the therapeutic performance of PCB NC uricase is evaluated and compared with its PEGylated counterpart in a clinical‐mimicking gouty rat model to determine any loss of efficacy evoked after five administrations.
A zwitterionic poly(carboxybetaine) (PCB) nanocage (PCB NC) is developed to physically encase proteins while keeping their structure intact. PCB NC encapsulation of uricase is manifested to eliminate all the possible immune responses. A high therapeutic performance of PCB‐NC‐shrouded uricase is demonstrated in a gouty rat model without evoking efficacy loss even after five repetitive administrations, greatly outperforming the industry‐standard PEGylated counterpart.
Repairing point cloud holes has become an important problem in the research of 3D laser point cloud data, which ensures the integrity and improves the precision of point cloud data. However, for the ...point cloud data with non-characteristic holes, the boundary data of point cloud holes cannot be used for repairing. Therefore, this paper introduces photogrammetry technology and analyzes the density of the image point cloud data with the highest precision. The 3D laser point cloud data are first formed into hole data with sharp features. The image data are calculated into six density image point cloud data. Next, the barycenterization Bursa model is used to fine-register the two types of data and to delete the overlapping regions. Then, the cross-section is used to evaluate the precision of the combined point cloud data to get the optimal density. A three-dimensional model is constructed for this data and the original point cloud data, respectively and the surface area method and the deviation method are used to compare them. The experimental results show that the ratio of the areas is less than 0.5%, and the maximum standard deviation is 0.0036 m and the minimum is 0.0015 m.
Blood contact with high surface area medical devices, such as dialysis and extracorporeal life support (ECLS), induces rapid surface coagulation. Systemic anticoagulation, such as heparin, is thus ...necessary to slow clot formation, but some patients suffer from bleeding complications. Both problems might be reduced by 1) replacing heparin anticoagulation with artificial surface inhibition of the protein adsorption that initiates coagulation and 2) selective inhibition of the intrinsic branch of the coagulation cascade. This approach was evaluated by comparing clot formation and bleeding times during short-term ECLS using zwitterionic polycarboxybetaine (PCB) surface coatings combined with either a potent, selective, bicyclic peptide inhibitor of activated Factor XII (FXII900) or standard heparin anticoagulation. Rabbits underwent venovenous ECLS with small sham oxygenators for 60 min using three means of anticoagulation (n = 4 ea): (1) PCB coating + FXII900 infusion, (2) PCB coating + heparin infusion with an activated clotting time of 220–300s, and (3) heparin infusion alone. Sham oxygenator blood clot weights in the PCB + FXII900 and PCB + heparin groups were 4% and 25% of that in the heparin group (p < 10−6 and p < 10−5), respectively. At the same time, the bleeding time remained normal in the PCB + FXII900 group (2.4 ± 0.2 min) but increased to 4.8 ± 0.5 and 5.1 ± 0.7 min in the PCB + heparin and heparin alone groups (p < 10−4 and 0.01). Sham oxygenator blood flow resistance was significantly lower in the PCB + FXII900 and PCB + heparin groups than in the heparin only group (p < 10−6 and 10−5). These results were confirmed by gross and scanning electron microscopy (SEM) images and fibrinopeptide A (FPA) concentrations. Thus, the combined use of PCB coating and FXII900 markedly reduced sham oxygenator coagulation and tissue bleeding times versus the clinical standard of heparin anticoagulation and is a promising anticoagulation method for clinical ECLS.
For the accurate and high-precision measurement of the deformation field in mining areas using different data sources, the probability integral model was used to process deformation data obtained ...from an Unmanned Aerial Vehicle (UAV), Differential InSAR (DInSAR), and Small Baseline Subset InSAR (SBAS-InSAR) to obtain the complete deformation field. The SBAS-InSAR, DInSAR, and UAV can be used to obtain small-scale, mesoscale, and large-scale deformations, respectively. The three types of data were all superimposed by the Kriging interpolation, and the deformation field was integrated using the probability integral model to obtain the complete high-precision deformation field with complete time series in the study area. The study area was in the WangJiata mine in Western China, where mining was carried out from 12 July 2018 to 25 October 2018, on the 2S201 working face. The first observation was made in June 2018, and steady-state observations were made in April 2019, totaling four UAV observations. During this period, the Canadian Earth Observation Satellite of Radarsat-2 (R2) was used to take 10 SAR images, the surface subsidence mapping was undertaken using DInSAR and SBAS-InSAR techniques, and the complete deformation field of the working face during the 106-day mining period was obtained by using the UAV technique. The results showed that the subsidence basin gradually expanded along the mining direction as the working face advanced. When the mining advance was greater than 1.2–1.4 times the coal seam burial depth, the supercritical conditions were reached, and the maximum subsidence stabilized at the value of 2.780 m. The subsidence rate was basically maintained at 0.25 m/d. Finally, the accuracy of the method was tested by the Global Navigation Satellite System (GNSS) data, and the medium error of the strike was 0.103 m. A new method is reached by the fusion of active and passive remote sensing data to construct efficient, complete and high precision time-series subsidence basins with high precision.

CD276 is an immune checkpoint molecule in the epithelial mesenchymal transition (EMT) pathway. In this review, we evaluated the available evidence on the expression and regulation of CD276. We also assessed the role of CD276 within the immune micro-environment, effect on tumor progression, and the potential therapeutic effect of CD276 targeted therapy for malignancies.
CD276 plays an essential role in cell proliferation, invasion, and migration in malignancies. Results from most recent studies indicate CD276 could be a promising therapeutic target for malignant tumors.
In order to satisfy the energy demand of China, it is an effective way to exploit coal resources efficiently in western China. When a coal seam with a shallow burial depth is mined on a large working ...face with width of 300 m in a semi-desert aeolian sand area of western China, the induced subsidence and damage of ground surface are remarkably different from those induced by a traditional mining condition. By taking Working Face 12,406 of Bulianta Coalmine in Shendong Mining Area as an example, this paper, based on actual measurement data, analysed the developmental features and causes of ground surface racks. Research results showed that the shape of the static crack in the peripheral area on the working face was very similar to that contained in the actual measurement results of other areas; specifically, such static crack was arc-shaped and the actually measured static crack angle was 84.5°. However, the dynamic crack above the working face took on uniqueness in two developmental cycles (expansion to restoration). This phenomenon is not available in other research areas. Starting from the structure of rock strata, this paper analysed the two developmental cycles of dynamic cracks according to the periodic fracture theory of key stratum and verified the results of theoretical analysis by employing the similar material model.
Although PEGylation reduces the immunogenicity of protein drugs to some extent, its limitations for highly immunogenic biotherapeutics have been demonstrated. Herein, a proactive strategy to ...alleviate the development of anti‐drug antibodies (ADAs) against protein drugs by immunomodulatory bioconjugation is reported. Rapamycin was conjugated to a PEGylated protein therapeutic via a cleavable disulfide linker. The conjugated rapamycin can be released from the bioconjugate and prevent immune responses once the bioconjugate is uptaken by antigen‐presenting cells. The immunomodulatory bioconjugate significantly reduced the titers of ADAs compared with a PEGylated protein. The inhibition of immune responses was specific to the conjugated antigen, avoiding systemic immune suppression and the risk of increased susceptibility to infections. The reported approach breaks the limitations of PEGylation by the proactive prevention of ADAs.
Immunomodulatory bioconjugation: An immunomodulator rapamycin was conjugated to a PEGylated protein therapeutic via a cleavable disulfide linker. This immunomodulatory bioconjugation effectively reduces anti‐drug antibody (ADA) generation by inducing antigen‐specific immune tolerance. The reported approach breaks the limitations of PEGylation by the proactive prevention of the generation of ADAs.
