Welding process is a nonlinear, multi-parameter coupling and complex process that is difficult to quantify accurately. This paper innovatively combines Industrial Internet technology and neural ...network technology to establish a welding process expert system. Through the use of Industrial Internet, the welding process expert system automatically collects the process data generated by the welding equipment in the process of operation, and uploads these process data to the cloud platform. In the cloud platform, the system uses welding process parameters to conduct neural network learning, establish prediction models, realize high-precision prediction of mechanical properties of welded joints, and achieve the purpose of reducing the number of welding process qualification tests.
Transition Radiation (TR) plays an important role in particle identification in high-energy physics and its characteristics provide a feasible method of energy calibration in the energy range up to ...10 TeV, which is of interest for dark matter searches in cosmic rays. In a Transition Radiation Detector (TRD), the TR signal is superimposed onto the ionization energy loss signal induced by incident charged particles. In order to make the TR signal stand out from the background of ionization energy loss in a significant way, we optimized both the radiators and the detector. We have designed a new prototype of regular radiator optimized for a maximal TR photon yield, combined with the Side-On TRD which is supposed to improve the detection efficiency of TR. We started a test beam experiment with the Side-On TRD at Conseil Europ\'{e}en pour la Recherche Nucl\'{e}aire (CERN), and found that the experimental data is consistent with the simulation results.
The study examines the impact of five major domestic factors on congressional voting in China policy from 1949-1990. These factors are ideology, party, constituency, regionalism and personal social ...and political background. The method for the inquiry is a combination of quantitative roll call analysis with intensive case study. In the House of Representatives, the dominant factor is ideology and the influence of party is secondary. In the period of 1950-1968, there was an ideology consensus of anti-communism in the House. This ideological hegemony suppressed other factors and maintained a unity in House China policy voting. After 1968, the consensus collapsed and was replaced by an ideological cleavage between the liberals and conservatives. Generally, the House liberals were more pro-China than the House conservatives. However, a switch of position took place in the 101st Congress (1989-1990), in the aftermath of 1989 student movement in Beijing. The influence of ideology went far beyond the voting stage in the House and permeated other legislative activities related to China policy. In the Senate, there was a shift from ideological voting to partisan voting. In the 1950s and 1960s, there was also an anti-communist consensus in Senate China policy voting. This ideology unity broke down in the early 1970s, and since then party became the dominant factor and the influence of ideology faded away. The Senate Democrats were generally more pro-China than the Senate Republicans. But on human rights issues, the Senate Democrats took much tougher stands than their Republican counterparts. Higher party pressure was a major cause for the partisan voting in the Senate. Constituency, regionalism and personal characteristics either had no effect or had a weak impact on China policy voting in Congress. Overall, the five factors combined together can explain and predict 50-82% of the China policy votes in the House and 59-83% of the votes in the Senate.
Direct electrical recording and stimulation of neural activity using micro-fabricated silicon and metal micro-wire probes have contributed extensively to basic neuroscience and therapeutic ...applications; however, the dimensional and mechanical mismatch of these probes with the brain tissue limits their stability in chronic implants and decreases the neuron-device contact. Here, we demonstrate the realization of a three-dimensional macroporous nanoelectronic brain probe that combines ultra-flexibility and subcellular feature sizes to overcome these limitations. Built-in strains controlling the local geometry of the macroporous devices are designed to optimize the neuron/probe interface and to promote integration with the brain tissue while introducing minimal mechanical perturbation. The ultra-flexible probes were implanted frozen into rodent brains and used to record multiplexed local field potentials and single-unit action potentials from the somatosensory cortex. Significantly, histology analysis revealed filling-in of neural tissue through the macroporous network and attractive neuron-probe interactions, consistent with long-term biocompatibility of the device.
Mounting evidence suggests that the Hippo coactivator Yes-associated protein 1 (YAP1) is a major mediator of cancer stem cell (CSC) properties, tumor progression, and therapy resistance as well as ...often a terminal node of many oncogenic pathways. Thus, targeting YAP1 may be a novel therapeutic strategy for many types of tumors with high YAP1 expression, including esophageal adenocarcinoma. However, effective YAP1 inhibitors are currently lacking. Here, we identify a small molecule (CA3) that not only has remarkable inhibitory activity on YAP1/Tead transcriptional activity but also demonstrates strong inhibitory effects on esophageal adenocarcinoma cell growth especially on YAP1 high-expressing esophageal adenocarcinoma cells both
and
Remarkably, radiation-resistant cells acquire strong cancer stem cell (CSC) properties and aggressive phenotype, while CA3 can effectively suppress these phenotypes by inhibiting proliferation, inducing apoptosis, reducing tumor sphere formation, and reducing the fraction of ALDH1
cells. Furthermore, CA3, combined with 5-FU, synergistically inhibits esophageal adenocarcinoma cell growth especially in YAP1 high esophageal adenocarcinoma cells. Taken together, these findings demonstrated that CA3 represents a new inhibitor of YAP1 and primarily targets YAP1 high and therapy-resistant esophageal adenocarcinoma cells endowed with CSC properties.

Lung cancer is still the leading cause of cancer-related mortality. Over the past two decades, the management of non-small cell lung cancer (NSCLC) has undergone a significant revolution. Since the ...first identification of activating mutations in the epidermal growth factor receptor (EGFR) gene in 2004, several genetic aberrations, such as anaplastic lymphoma kinase rearrangements (ALK), neurotrophic tropomyosin receptor kinase (NTRK) and hepatocyte growth factor receptor (MET), have been found. With the development of gene sequencing technology, the development of targeted drugs for rare mutations, such as multikinase inhibitors, has provided new strategies for treating lung cancer patients with rare mutations. Patients who harbor this type of oncologic driver might acquire a greater survival benefit from the use of targeted therapy than from the use of chemotherapy and immunotherapy. To date, more new agents and regimens can achieve satisfactory results in patients with NSCLC. In this review, we focus on recent advances and highlight the new approval of molecular targeted therapy for NSCLC patients with rare oncologic drivers.
ABSTRACT
The mechanism of exosomes derived from activated hepatic stellate cells (HSCs) involved in liver fibrosis is poorly understood. We previously reported that hypoxia‐inducible factor 1 (Hif‐1) ...regulated HSC activation, and, therefore, we investigated in current work whether Hif‐1 regulates exosome secretion and the metabolic switch of HSCs, thus affecting the metabolism of liver nonparenchymal cells. In this study, the characteristics of exosomes from HSCs were assessed via electron microscopy, Western blot analysis, and acetylcholinesterase activity. Confocal microscopy was used to measure the uptake of exosomes by quiescent HSCs, Kupffer cells (KCs), and liver sinusoidal endothelial cells (LSECs). Hif‐1α was inhibited via 2‐ME or specific small interfering RNAs to investigate its role in exosomes derived from HSCs. It was determined that glucose transporter 1 and pyruvate kinase M2 were increasingly expressed in fibrotic liver samples, cell lysates, and exosomes derived from activated HSCs. Exosomes released from HSCs were associated with activation and glucose uptake of HSCs. Delivery of exosomes from activated HSCs induced glycolysis of quiescent HSCs, KCs, and LSECs. Disruption of Hif‐1 expression suppressed the glycolysis effect delivered by exosomes. Conclusively, our results demonstrated that exosomes secreted by activated HSCs affect the metabolic switch of liver nonparenchymal cells via delivery of glycolysis‐related proteins. These findings represent a novel mechanism that contributes to liver fibrosis and has significant implications for new diagnosis and treatment of liver diseases.—Wan, L., Xia, T., Du, Y., Liu, J., Xie, Y., Zhang, Y., Guan, F., Wu, J., Wang, X., Shi, C. Fluorescent single crystals that respond to multiple external stimuli are of great interest in molecular machines, sensors, and displays. The integration of photo‐ or acid‐induced fluorescence ...enhancement and bending in one organic crystal, however, has not been reported yet. Herein, we report the interesting plastic photomechanical bending and switching on of the fluorescence of an azine crystal in a single‐crystal transformation, due to extended π‐conjugation and molecular slippage. Moreover, the fluorescent plastic bending driven by multiple volatile acid vapors was firstly observed, and attributed to the synergistic effect of push–pull electronic structure and hydrogen bonding. The single crystal also shows high elasticity under external force. In addition, reversible fluorescence switching can be triggered by grinding and solvent fuming, as well as by the adsorption and desorption of HCl vapor. The integration of plastic, elastic bending and switch‐on fluorescence into one single crystal provides a new strategy for next‐generation smart materials.
A slender single crystal based on azine exhibits controllable plastic bending and fluorescence enhancement upon UV light irradiation or acid vapor fuming, as well as elastic bending under external force and mechanoresponsive fluorescence. This is the first smart single crystal exhibiting fluorescence lit up upon bending triggered by UV light irradiation and exposure to acid.
