Over many years, parts of Panax ginseng (root and rhizome) have been identified and applied for medical purposes as traditional Chinese herbal medicine. Recently, research has indicated that ginseng ...fruit also contains similar compounds and is as rich as the other parts of the ginseng. This discovery may dramatically improve the efficient of outputs derived from ginseng products. Here, a new technique combining high-performance liquid chromatography (HPLC) with electrospray ionization tandem mass spectrometry (ESI-MS) was employed to identify the fingerprint of P. ginseng fruit. Using HPLC, compounds that are important for medical purposes were extracted and purified. Combined with ESI-MS, the characteristic peaks (nine common peaks) of those compounds were identified, and the accuracy was confirmed by analysis using the Chromatographic Fingerprint Similarity Evaluation System (2004A edition). Overall, 15 batches of ginseng fruit had a similarity of more than 0.80, 13 batches of samples had a similarity between 0.97 and 0.99, and two batches had a similarity less than 0.90. The test solution and mobile phase selection was discussed. The HPLC-ESI-MS method can produce repeatable and reliable results and can be applied in the quality control of P. ginseng fruit.
Background. Emerging evidence suggests that T2DM is attributable to the dysfunction of β-cells and the activation of islet stellate cells (ISCs). The wingless-type MMTV integration site family member ...5a (Wnt5a)/frizzled 5 (Fzd5) signalling pathway might take part in this process. Our study is aimed at defining the status of ISCs during β-cell insulin secretion homeostasis by determining the role of the Wnt5a protein in the regulation of insulin production. We examined the effects of the status of ISCs on β-cell insulin secretion in normoglycemic db/m and hyperglycaemic db/db mice. Methods. iTRAQ protein screening and RNA interference were used to determine novel ISC-derived secretory products that may use other mechanisms to influence the function of islets. Results. We showed a significant reduction in insulin secretion by β-cells in vitro when they were cocultured with db/db ISCs compared to when they were cocultured with ISCs isolated from normoglycemic db/m mice; in addition, both Wnt5a and its receptor Fzd5 were more highly expressed by quiescent ISCs than by activated db/db ISCs. Treatment with exogenous Wnt5a increased the secretion of insulin in association with the deactivation of ISCs. Conclusion. Our observations revealed that the Wnt5a protein is a key effector of ISC-mediated improvement in islet function.
Current estimates of the terrestrial carbon fluxes in Asia show large uncertainties particularly in the boreal and mid-latitudes and in China. In this paper, we present an updated carbon flux ...estimate for Asia ("Asia" refers to lands as far west as the Urals and is divided into boreal Eurasia, temperate Eurasia and tropical Asia based on TransCom regions) by introducing aircraft CO2 measurements from the CONTRAIL (Comprehensive Observation Network for Trace gases by Airline) program into an inversion modeling system based on the CarbonTracker framework. We estimated the averaged annual total Asian terrestrial land CO2 sink was about -1.56 Pg C yr-1 over the period 2006–2010, which offsets about one-third of the fossil fuel emission from Asia (+4.15 Pg C yr-1). The uncertainty of the terrestrial uptake estimate was derived from a set of sensitivity tests and ranged from -1.07 to -1.80 Pg C yr-1, comparable to the formal Gaussian error of ±1.18 Pg C yr-1 (1-sigma). The largest sink was found in forests, predominantly in coniferous forests (-0.64 ± 0.70 Pg C yr-1) and mixed forests (-0.14 ± 0.27 Pg C yr-1); and the second and third large carbon sinks were found in grass/shrub lands and croplands, accounting for -0.44 ± 0.48 Pg C yr-1 and -0.20 ± 0.48 Pg C yr-1, respectively. The carbon fluxes per ecosystem type have large a priori Gaussian uncertainties, and the reduction of uncertainty based on assimilation of sparse observations over Asia is modest (8.7–25.5%) for most individual ecosystems. The ecosystem flux adjustments follow the detailed a priori spatial patterns by design, which further increases the reliance on the a priori biosphere exchange model. The peak-to-peak amplitude of inter-annual variability (IAV) was 0.57 Pg C yr-1 ranging from -1.71 Pg C yr-1 to -2.28 Pg C yr-1. The IAV analysis reveals that the Asian CO2 sink was sensitive to climate variations, with the lowest uptake in 2010 concurrent with a summer flood and autumn drought and the largest CO2 sink in 2009 owing to favorable temperature and plentiful precipitation conditions. We also found the inclusion of the CONTRAIL data in the inversion modeling system reduced the uncertainty by 11% over the whole Asian region, with a large reduction in the southeast of boreal Eurasia, southeast of temperate Eurasia and most tropical Asian areas.
Using 448.0×106 ψ(3686) events collected with the BESIII detector, an amplitude analysis is performed for ψ(3686)→γχc1, χc1→ηπ+π− decays. The most dominant two-body structure observed is a0(980)±π∓; ...a0(980)±→ηπ±. The a0(980) line shape is modeled using a dispersion relation, and a significant nonzero a0(980) coupling to the η′π channel is measured. We observe χc1→a2(1700)π production for the first time, with a significance larger than 17σ. The production of mesons with exotic quantum numbers, JPC=1−+, is investigated, and upper limits for the branching fractions χc1→π1(1400)±π∓, χc1→π1(1600)±π∓, and χc1→π1(2015)±π∓, with subsequent π1(X)±→ηπ± decay, are determined.

This disease was characterized by distal limb numbness, some patients with hypoalgesia or allergy, deep sense loss, reduction or disappearance of tendon reflexes, and peeling. One case had muscle weakening and another case had cerebellar ataxia. Examination of electromyography showed only one case had spontaneous potential. Examination of nerve conduction showed that the amplitude decreased by 34 (38.6%) and the velocity decreased by 2 (2.3%) , the percentage of amplitude decreased was significantly higher than that of velocity decreased. PtBi2 with a layered hexagonal crystal structure was recently reported to exhibit an unconventional large linear magnetoresistance, while the mechanism involved is still elusive. Using ...high-resolution angle-resolved photoemission spectroscopy, we present a systematic study on its bulk and surface electronic structure. Through careful comparison with first-principle calculations, our experiment distinguishes the low-lying bulk bands from entangled surface states, allowing the estimation of the real composition of samples. We find significant electron doping in PtBi2, implying a substantial Bi-deficiency-induced disorder therein. Intriguingly, we discover a Dirac-cone-like surface state on the boundary of the Brillouin zone, which is identified as an accidental Dirac band without topological protection. Our findings exclude linear band dispersion in the quantum limit as the cause of the unconventional large linear magnetoresistance but give support to the classical disorder model from the perspective of the electronic structure.

Objective
Previous studies have found that wnt5a promotes β-cell insulin secretion and reduced concentrations in patients with type 2 diabetes. GLP-1RA (Glucagon-like peptide-1 receptor agonists) can ...regulate insulin secretion. However, the evidence that GLP-1RA affect insulin secretion through the Wnt5a is inconclusive. Therefore, this study aimed to evaluate the effect of GLP-1 RA on wnt5a levels in patients with type 2 diabetes.
Methods
A total of 56 onset diabetics were selected our study, 29 of them were treated by GLP-1RAs (1.2mg subcutaneous injection once a day, liraglutide, Novo Nordisk), the rest (27 case) treated by Metformin (0.5 g twice a day, Glucophage, Merck). Individuals who were using medications to manage platelet (Aspirin) and cholesterol (Statins) were enrolled and continued treatment throughout the study.
Results
Our study found that the waist circumference and insulin secretion index in the GLP-1RA intervention group were significantly increased, and the insulin resistance index was lower than that of the control group. More interestingly, the serum Wnt5a protein level increased dramatically after the GLP-1RA intervention, and the level of Secreted frizzled-related protein 5 (Sfrp5) decreased compared with the control group. Multivariate linear regression analysis showed that the change of HOMA-β (Homeostasis model assessment- β) was significantly correlated with the changes of Wnt5a and Sfrp5, and the change of Wnt5a protein was positively correlated with HOMA-β.
Conclusion
Our results confirmed that GLP-1RA may improve HOMA-β in patients with type 2 diabetes by affecting the level of Wnt5a protein.
This article presents a direct and efficient algorithm for simulating high-cycle fatigue failure effects within the framework of a recently proposed elastoplastic model, in which fatigue failure ...effects of metals are incorporated as inherent constitutive features. Toward bypassing very time-consuming procedures in conducting numerical integrations of the elastoplastic rate equations for extremely large number of loading cycles, from the new model it is possible to derive an explicit expression for the fatigue life directly in terms of the stress amplitude. Numerical examples show that the new algorithm thus established is much more efficient than usual direct numerical procedures and that is particularly the case in simulating fatigue failure effects with very high cycle number.
