This study explored the spatio-temporal dynamics and evolution of land use/cover changes and urban expansion in Shanghai metropolitan area, China, during the transitional economy period (1979–2009) ...using multi-temporal satellite images and geographic information systems (GIS). A maximum likelihood supervised classification algorithm was employed to extract information from four landsat images, with the post-classification change detection technique and GIS-based spatial analysis methods used to detect land-use and land-cover (LULC) changes. The overall Kappa indices of land use/cover change maps ranged from 0.79 to 0.89. Results indicated that urbanization has accelerated at an unprecedented scale and rate during the study period, leading to a considerable reduction in the area of farmland and green land. Findings further revealed that water bodies and bare land increased, obviously due to large-scale coastal development after 2000. The direction of urban expansion was along a north-south axis from 1979 to 2000, but after 2000 this growth changed to spread from both the existing urban area and along transport routes in all directions. Urban expansion and subsequent LULC changes in Shanghai have largely been driven by policy reform, population growth, and economic development. Rapid urban expansion through clearing of vegetation has led to a wide range of eco-environmental degradation.
Objective: Severe neurologic complications after spinal anesthesia are rare but highly distressing, especially in pregnant women. Bupivacaine is widely used in spinal anesthesia, but its neurotoxic ...effects have gained attention.
Methods: Furthermore, the etiology of bupivacaine-mediated neurotoxicity in obstetric patients remains unclear. Female C57BL/6 mice were intrathecally injected with 0.75% bupivacaine on the 18th day of pregnancy. We used immunohistochemistry to examine DNA damage after bupivacaine treatment in pregnant mice and measured γ-H2AX (Ser139) and 8-OHdG in the spinal cord. A PARP-1 inhibitor (PJ34) and autophagy inhibitor (3-MA) were administered with bupivacaine in pregnant mice. Parp-1flox/flox mice were crossed with Nes-Cre transgenic mice to obtain neuronal conditional knockdown mice. Then, LC3B and P62 staining were performed to evaluate autophagic flux in the spinal cords of pregnant wild-type (WT) and Parp-1-/- mice. We performed transmission electron microscopy (TEM) to evaluate autophagosomes.
Results: The present study showed that oxidative stress-mediated DNA damage and neuronal injury were increased after bupivacaine treatment in the spinal cords of pregnant mice. Moreover, PARP-1 was significantly activated, and autophagic flux was disrupted. Further studies revealed that PARP-1 knockdown and autophagy inhibitors could alleviate bupivacaine-mediated neurotoxicity in pregnant mice. Conclusion: Bupivacaine may cause neuronal DNA damage and PARP-1 activation in pregnant mice. PARP-1 further obstructed autophagic flux and ultimately led to neurotoxicity.
Yangtze estuary, lying in the subtropical monsoon region of China, is characterized by a unique environmental setting and endemic wetland plant species (Scirpus mariqueter). Methane (CH4) emission ...fluxes were measured at the Yangtze estuarine wetland, Chongming Dongtan (CD), by a static closed chamber technique from May 2004 to April 2005. The results showed that CD is the source of atmospheric CH4, and emission fluxes had significant diurnal and seasonal variation. The annual average CH4 emission flux was 2.06 mg m−2 h−1 at the CD marsh site and 0.04 mg m−2 h−1 at the CD bare tidal flat (nonvegetated). Wetland plant species (S. mariqueter) and temperature were the primary factors controlling the CH4 emission. The results of the light and dark chamber comparison and plant shoot clipping experiment suggest that molecular diffusion and convective gas flow methods were the two main mechanisms of CH4 transported via S. mariqueter plants in July. However, molecular diffusion was believed to be the primary transport mechanism from August to October, with leaf resistance as one of the factors regulating CH4 diffusion. There was significant correlation between CH4 fluxes and temperature, especially the 10 cm depth ground temperature (R2 = 0.7784). Although sediment organic carbon content did not determine CH4 fluxes, net ecosystem production was significantly correlated with CH4 fluxes, suggesting that the photosynthates of S. mariqueter effectively provided the substrate for methanogenic bacteria.
This paper presents a scenario-based study that investigates the interaction between sea-level rise and land subsidence on the storm tides induced fluvial flooding in the Huangpu river floodplain. ...Two projections of relative sea level rise (RSLR) were presented (2030 and 2050). Water level projections at the gauging stations for different return periods were generated using a simplified algebraic summation of the eustatic sea-level rise, land subsidence and storm tide level. Frequency analysis with relative sea level rise taken into account shows that land subsidence contributes to the majority of the RSLR (between 60 % and 70 %). Furthermore, a 1D/2D coupled flood inundation model (FloodMap) was used to predict the river flow and flood inundation, after calibration using the August 1997 flood event. Numerical simulation with projected RSLR suggests that, the combined impact of eustatic sea-level rise and land subsidence would be a significantly reduced flood return period for a given water level, thus effective degradation of the current flood defences. In the absence of adaptation measures, storm flooding will cause up to 40 % more inundation, particularly in the upstream of the river.
In this paper, the influence of long-period stacked ordered (LPSO) phases on the electrochemical impedance spectroscopy (EIS) of a Mg-Gd-Y-Zn-Zr-Ag alloy in 0.9 wt.% NaCl was investigated. The ...Mg-6Gd-3Y-1Zn-0.5Zr-0.3Ag (wt.%) alloy samples with and without LPSO phases in the grain interior (HOMO and LPSO, respectively) were prepared using different heat treatments. The EIS results showed that both the HOMO and LPSO samples' Nyquist diagrams contained two inductive loops. However, in the Nyquist plots of the LPSO samples, the inductive loops at 1.71-0.67 Hz appeared in the first quadrant rather than the fourth quadrant. Analysis of the fitting parameters illustrated that the abnormal shape of the inductive loops is related to greater values of the surface film capacitance
and double layer capacitance
in the LPSO samples. Further investigations through corrosion morphology observation indicated that the greater values of

in the LPSO samples resulted from the existence of intragranular LPSO phases that created more film-free areas. The above results show that a better understanding of the relationship between the inductive impedance and corrosion morphology of a Mg-6Gd-3Y-1Zn-0.5Zr-0.3Ag alloy in 0.9 wt.% NaCl solution was attained.
Pyruvate kinase M2 (PKM2) is a key enzyme of glycolysis, which is highly expressed in many tumor cells, and has emerged as an important player in tumor progression and metastasis. However, the ...functional roles of PKM2 in tumor metastasis remain elusive. Here we showed that PKM2 promoted prostate cancer metastasis via extracellular-regulated protein kinase (ERK)-cyclooxygenase (COX-2) signaling. Based on public databases, we found that PKM2 expression was upregulated in prostate cancer and positively associated with tumor metastasis. Further analysis showed that PKM2 promoted prostate cancer cell migration/invasion and epithelial-mesenchymal transition (EMT) through upregulation of COX-2. Mechanistically, PKM2 interacted with ERK1/2 and regulated its phosphorylation, leading to phosphorylation of transcription factor c-Jun, downstream of ERK1/2, to activate COX-2 transcription by IP and ChIP assay, while inhibition of COX-2 significantly reversed the promotion effect of PKM2 on tumor metastasis
. Taken together, our results suggest that a novel of PKM2-ERK1/2-c-Jun-COX-2 axis is a potential target in controlling prostate cancer metastasis.

Long noncoding RNAs (lncRNAs) have been implicated in neurogenesis. LncRNA WNT5A-AS is upregulated in neural stem cells (NSCs), the proliferation of which is inhibited by sevoflurane. Thus, we hypothesized that knocking down of lncRNA WNT5A-AS may restore the fate of NSCs exposed to sevoflurane. To test this hypothesis, NSCs obtained from postnatal Sprague-Dawley rats were exposed to 2.4% sevoflurane or control gas for 6 h. Bioinformatics analysis, quantitative PCR and RNA interference technology were used to identify the properties of lncRNA WNT5A-AS. Cell proliferation was assessed using counting a Cell Counting Kit-cell 8 assay, a 5-ethynyl-2′-deoxyuridine incorporation assay, and a plate cloning assay. Cell survival was detected by flow cytometry, which was also used to examine the levels of reactive oxygen species (ROS) and the cell cycle. The levels of WNT5A and receptor tyrosine kinase (Ryk) were measured via Western blotting. LncRNA WNT5A-AS was identified to have low coding potency and to be located on the antisense strand of WNT5A. The level of upregulated lncRNA WNT5A-AS was positively correlated with that of WNT5A in response to sevoflurane exposure. The knockdown of lncRNA WNT5A-AS promoted the proliferation and survival of NSCs, whereas it suppressed the WNT5A/Ryk-ROS signaling and drove cell cycle processes. Taken together, findings strongly suggest that the inhibition of lncRNA WNT5A-AS can rescue the fate of NSCs. In addition, WNT5A/Ryk-ROS signaling might be a downstream target of lncRNA WNT5A-AS.
Chronic stress is well known to promote tumor progression, however, little is known whether chronic stress-mediated regulation of osteoblasts contributes to the migration and invasion of metastatic ...cancer cells.
The proliferation, migration and invasion of prostate cancer cells were assessed by CCK-8 and transwell assay. HIF-1α expression of osteoblasts and epithelial-mesenchymal transition (EMT) markers of prostate cancer cells were examined by Western blot. The mRNA level of cytokines associated with bone metastasis in osteoblasts and EMT markers in PC-3 and DU145 cells were performed by qRT-PCR. Functional rescue experiment of cells were performed by using siRNA, plasmid transfection and inhibitor treatment.
Isoproterenol (ISO), a pharmacological surrogate of sympathetic nerve activation induced by chronic stress, exhibited no direct effect on migration and invasion of PC-3 and DU145 prostate cancer cells. Whereas, osteoblasts pretreated with ISO promoted EMT, migration and invasion of PC-3 and DU145 cells, which could be inhibited by β2AR inhibitor. Mechanistically, ISO increased the secretion of CXCL12 via the β2AR-HIF-1α signaling in osteoblasts. Moreover, overexpression of HIF-1α osteoblasts promoted migration and invasion of PC-3 and DU145 cells, which was inhibited by addition of recombinant knockdown of CXCR4 in PC-3 and DU145 cells, and inhibiting CXCL12-CXCR4 signaling with LY2510924 blunted the effects of osteoblasts in response to ISO on EMT and migration as well as invasion of PC-3 and DU145 cells.
These findings demonstrated that β2AR-HIF-1α-CXCL12 signaling in osteoblasts facilitates migration and invasion as well as EMT of prostate cancer cells, and may play a potential role in affecting bone metastasis of prostate cancer.
Ginkgo biloba L. is a valuable medicinal plant known for its high content of flavonoids and terpenoids in the leaves of young trees. Pruning can increase leaf yield in ginkgo plantations; however, it ...is unclear how the intensity of pruning affects leaf yield and quality. In addition, G. biloba exhibits low cutting rooting rates, which limits its efficiency in asexual propagation. In our study, we compared consecutive pruning with varying levels of intensity, including top pruning, light pruning, and heavy pruning, to evaluate the effects of pruning on leaf yield and cutting rooting. The results showed that these three pruning methods all contributed to an increase in the number of new branches, the leaf weight, and the flavonoid content in five-year-old trees. Among them, the effect of light pruning was the best, with a 150% increase in branch number, a 130% increase in leaf weight, and a 40.6% increase in flavonoid content. The secondary pruning further increased leaf area by 22.3%, indicating that secondary pruning further enhanced the rejuvenation of plants and increased leaf yield. At the transcriptional level, pruning can significantly change the expression of genes related to bud sprouting, resulting in a particularly significant increase in SHR expression in the buds. Pruning also promoted the expression of important genes related to flavonoid synthesis, including chalcone synthase (CHS), flavonoid 3′-hydroxylase (F3′H), flavonol synthase (FLS), and dihydroflavonol reductase (DFR). Furthermore, we demonstrated a significant increase in the rooting rate of these second-pruned branch cuttings and screened the optimal hormone ratio for rooting, which is 1.5 μM MeJA + 400 mg/L NAA + 100 mg/L Uniconazole-P. These results suggest that secondary pruning can effectively rejuvenate plants to promote cutting rooting in G. biloba. This method can not only be used to improve the yield and quality of ginkgo leaves, but also for cutting propagation.
