Abstract
Planetesimal formation is a crucial yet poorly understood process in planet formation. It is widely believed that planetesimal formation is the outcome of dust clumping by the streaming ...instability (SI). However, recent analytical and numerical studies have shown that the SI can be damped or suppressed by external turbulence, and at least the outer regions of protoplanetary disks are likely weakly turbulent due to magneto-rotational instability (MRI). We conduct high-resolution local shearing-box simulations of hybrid particle-gas magnetohydrodynamics (MHD), incorporating ambipolar diffusion as the dominant nonideal MHD effect, applicable to outer disk regions. We first show that dust backreaction enhances dust settling toward the midplane by reducing turbulence correlation time. Under modest level of MRI turbulence, we find that dust clumping is in fact easier than the conventional SI case, in the sense that the threshold of solid abundance for clumping is lower. The key to dust clumping includes dust backreaction and the presence of local pressure maxima, which in our work is formed by the MRI zonal flows overcoming background pressure gradient. Overall, our results support planetesimal formation in the MRI-turbulent outer protoplanetary disks, especially in ring-like substructures.
Help-seeking is important to access appropriate care and improve mental health. However, individuals often delay or avoid seeking help for mental health problems. Interventions to improve ...help-seeking have been developed, but their effectiveness is unclear. A systematic review and meta-analysis were therefore conducted to examine the effectiveness of mental health related help-seeking interventions. Nine databases in English, German and Chinese were searched for randomised and non-randomised controlled trials. Effect sizes were calculated for attitudes, intentions and behaviours to seek formal, informal and self-help. Ninety-eight studies with 69 208 participants were included. Interventions yielded significant short-term benefits in terms of formal help-seeking, self-help, as well as mental health literacy and personal stigma. There were also positive long-term effects on formal help-seeking behaviours. The most common intervention types were strategies to increase mental health literacy, destigmatisation (both had positive short-term effects on formal help-seeking behaviours) as well as motivational enhancement (with positive long-term effects on formal help-seeking behaviours). Interventions improved formal help-seeking behaviours if delivered to people with or at risk of mental health problems, but not among children, adolescents or the general public. There was no evidence that interventions increased the use of informal help. Few studies were conducted in low- and middle-income countries (LMICs). This study provides evidence for the effectiveness of help-seeking interventions in terms of improving attitudes, intentions and behaviours to seek formal help for mental health problems among adults. Future research should develop effective interventions to improve informal help-seeking, for specific target groups and in LMICs settings.
It has been realized in recent years that the accretion of pebble-sized dust particles onto planetary cores is an important mode of core growth, which enables the formation of giant planets at large ...distances and assists planet formation in general. The pebble accretion theory is built upon the orbit theory of dust particles in a laminar protoplanetary disk (PPD). For sufficiently large core mass (in the "Hill regime"), essentially all particles of appropriate sizes entering the Hill sphere can be captured. However, the outer regions of PPDs are expected to be weakly turbulent due to the magnetorotational instability (MRI), where turbulent stirring of particle orbits may affect the efficiency of pebble accretion. We conduct shearing-box simulations of pebble accretion with different levels of MRI turbulence (strongly turbulent assuming ideal magnetohydrodynamics, weakly turbulent in the presence of ambipolar diffusion, and laminar) and different core masses to test the efficiency of pebble accretion at a microphysical level. We find that accretion remains efficient for marginally coupled particles (dimensionless stopping time ) even in the presence of strong MRI turbulence. Though more dust particles are brought toward the core by the turbulence, this effect is largely canceled by a reduction in accretion probability. As a result, the overall effect of turbulence on the accretion rate is mainly reflected in the changes in the thickness of the dust layer. On the other hand, we find that the efficiency of pebble accretion for strongly coupled particles (down to ) can be modestly reduced by strong turbulence for low-mass cores.
Plant genome engineering mediated by various CRISPR-based tools requires specific protospacer adjacent motifs (PAMs), such as the well-performed NGG, NG, and NNG, to initiate target recognition, ...which notably restricts the editable range of the plant genome.
In this study, we thoroughly investigate the nuclease activity and the PAM preference of two structurally engineered SpCas9 variants, SpG and SpRY, in transgenic rice. Our study shows that SpG nuclease favors NGD PAMs, albeit less efficiently than the previously described SpCas9-NG, and that SpRY nuclease achieves efficient editing across a wide range of genomic loci, exhibiting a preference of NGD as well as NAN PAMs. Furthermore, SpRY-fused cytidine deaminase hAID*Δ and adenosine deaminase TadA8e are generated, respectively. These constructs efficiently induce C-to-T and A-to-G conversions in the target genes toward various non-canonical PAMs, including non-G PAMs. Remarkably, high-frequency self-editing events (indels and DNA fragments deletion) in the integrated T-DNA fragments as a result of the nuclease activity of SpRY are observed, whereas the self-editing of SpRY nickase-mediated base editor is quite low in transgenic rice lines.
The broad PAM compatibility of SpRY greatly expands the targeting scope of CRISPR-based tools in plant genome engineering.
Timely monitoring of global plant biogeochemical processes demands fast and highly accurate estimation of plant nutrition status, which is often estimated based on hyperspectral data. However, few ...such studies have been conducted on degraded vegetation. In this study, complete combinations of either original reflectance or first-order derivative spectra have been developed to quantify leaf nitrogen (N), phosphorus (P), and potassium (K) contents of tree, shrub, and grass species using hyperspectral datasets from light, moderate, and severely degraded vegetation sites in Helin County, China. Leaf N, P, and K contents were correlated to identify suitable combinations. The most effective combinations were those of reflectance difference (Dij), normalized differences (ND), first-order derivative (FD), and first-order derivative difference (FD(D)). Linear regression analysis was used to further optimize sensitive band-based combinations, which were compared with 43 frequently used empirical spectral indices. The proposed hyperspectral indices were shown to effectively quantify leaf N, P, and K content (R2 > 0.5, p < 0.05), confirming that hyperspectral data can be potentially used for fine scale monitoring of degraded vegetation.
For decades, the therapeutic goal of conventional treatment among inflammatory bowel disease (IBD) patients is alleviating exacerbations in acute phase, maintaining remission, reducing recurrence, ...preventing complications, and increasing quality of life. However, the persistent mucosal/submucosal inflammation tends to cause irreversible changes in the intestinal structure, which can barely be redressed by conventional treatment. In the late 1990s, monoclonal biologics, mainly anti-TNF (tumor necrosis factor) drugs, were proven significantly helpful in inhibiting mucosal inflammation and improving prognosis in clinical trials. Meanwhile, mucosal healing (MH), as a key endoscopic and histological measurement closely associated with the severity of symptoms, has been proposed as primary outcome measures. With deeper comprehension of the mucosal microenvironment, stem cell niche, and underlying mucosal repair mechanisms, diverse potential strategies apart from monoclonal antibodies have been arising or undergoing clinical trials. Herein, we elucidate key steps or targets during the course of MH and review some promising treatment strategies capable of promoting MH in IBD.

The study of neural circuits, which underlies perception, cognition, emotion, and behavior, is essential for understanding the mammalian brain, a complex organ consisting of billions of neurons. To study the structure and function of the brain, in vivo neuronal labeling and imaging techniques are crucial as they provide true physiological information that ex vivo methods cannot offer. In this paper, we present a new strategy for in vivo neuronal labeling and quantification using MRI. We demonstrate the efficacy of this method by delivering the oatp1a1 gene to the target neurons using rAAV2-retro virus. OATP1A1 protein expression on the neuronal membrane increased the uptake of a specific MRI contrast agent (Gd-EOB-DTPA), leading to hyperintense signals on T1W images of labeled neuronal populations. We also used dynamic contrast enhancement-based methods to obtain quantitative information on labeled neuronal populations in vivo.
As an upper-middle-income country, China has a vast territory, a large population and substantial variations in the nature of mental health service provision. Guangzhou, one of the five biggest ...cities in China, is considered as indicative of the current development of mental health services in a more economically developed part of China. In this paper, we first introduce mental health services in Guangzhou and their historical development: from the rising development of mental health care (1949 to the mid-1960s), the stagnant development period of the psychiatric hospitals (1966–1976), the rapid development period of the psychiatric hospitals (1978 to the 1990s), to the integrated development period of community mental health care (2000–2020). This historical trajectory partly reflects the development of mental health services across China. Second, from the public health perspective, we introduce the current mental health care delivery system and resources in Guangzhou, including the psychiatric specialist hospitals, psychiatric units of general hospitals, and community mental health care. Third, based on the current mental health service system, we highlight community mental health care in Guangzhou. We will describe these elements of the mental health system under the following headings: organizational setup, management, and services. We also explore the design of community mental health service models and the challenges in the post-covid-19 era. Finally, we provide considerations and recommendations for mental health service delivery in Guangzhou in the future.

Soft tissue sarcoma is a broad family of mesenchymal malignancies exhibiting remarkable histological diversity. We portray the proteomic landscape of 272 soft tissue sarcomas representing 12 major ...subtypes. Hierarchical classification finds the similarity of proteomic features between angiosarcoma and epithelial sarcoma, and elevated expression of SHC1 in AS and ES is correlated with poor prognosis. Moreover, proteomic clustering classifies patients of soft tissue sarcoma into 3 proteomic clusters with diverse driven pathways and clinical outcomes. In the proteomic cluster featured with the high cell proliferation rate, APEX1 and NPM1 are found to promote cell proliferation and drive the progression of cancer cells. The classification based on immune signatures defines three immune subtypes with distinctive tumor microenvironments. Further analysis illustrates the potential association between immune evasion markers (PD-L1 and CD80) and tumor metastasis in soft tissue sarcoma. Overall, this analysis uncovers sarcoma-type-specific changes in proteins, providing insights about relationships of soft tissue sarcoma.
