Foreign bodies (FB) in the external auditory canal are relative medical emergency. The objective of this study was to describe the types of FB and their complications. Hospital-based descriptive ...study. This study is done at sub-district hospital Nasirabad (district Ajmer) from period 1 January2016 to 31 May 2020 for period of 3 years 5 months included all patients with foreign body ear. A total of 126 patients were included. All cases were attended by ENT specialist. In our study we concluded that most common incidence is children less than 5 years (63.49%), male:female ratio of 1.33:1, most of the patients present within 24 h of presentation, in maximum cases there is no complications, maximum the foreign body is present in right ear. All cases were attended by ENT specialist. Maximum cases are done in local anaesthesia or no anaesthesia at all, 4 case in light anesthesia and 3 cases referred to nearby higher centre. In this we concluded that foreign body is usually present in less than 5 years of age group. Proper instrumentation and proper skill is required to remove foreign body without any complications.

Sesame is the oldest oilseed crop known to humanity, though it contributes a small share in the global vegetable oil production. Sesame oil contains nutrients, including lignans, tocopherols, phytosterols, natural antioxidants, and bioactive compounds. It provides various health benefits such as anti-lipogenic, hypo-cholesterolemic, anti-degenerative, and neural health-promoting properties. Being an under-utilized minor crop, it has not received enough research attention for its food and nutraceutical potential. The sesame crop is a potential candidate to maintain the diversity of food oils and harness its benefits for improving human health. The present review will provide detailed research on sesame oil contents, health effects, nutraceuticals, oil quality, and value addition strategies. Also, the sesame oil nutritional quality was compared with other vegetable oils, highlighting the potential health and nutrition-related benefits. The way forward for further sesame improvement through value addition traits was also discussed.
External stents inhibit saphenous vein graft (SVG) intimal hyperplasia in animal studies. We investigated whether external stenting inhibits SVG diffuse intimal hyperplasia 1 year after coronary ...artery bypass graft surgery.
Thirty patients with multivessel disease undergoing coronary artery bypass graft surgery were enrolled. In addition to an internal mammary artery graft, each patient received one external stent to a single SVG randomly allocated to either the right or left coronary territories; and one or more nonstented SVG served as the control. Graft patency was confirmed at the end of surgery in all patients. The primary endpoint was SVG intimal hyperplasia (mean area) assessed by intravascular ultrasonography at 1 year. Secondary endpoints were SVG failure, ectasia (>50% initial diameter), and overall uniformity as judged by Fitzgibbon classification.
One-year follow-up angiography was completed in 29 patients (96.6%). All internal mammary artery grafts were patent. Overall SVG failure rates did not differ significantly between the two groups (30% stented versus 28.2% nonstented SVG, p = 0.55). The SVG mean intimal hyperplasia area, assessed in 43 SVGs, was significantly reduced in the stented group (4.37 ± 1.40 mm(2)) versus nonstented group (5.12 ± 1.35 mm(2), p = 0.04). In addition, stented SVGs demonstrated marginally significant improvement in lumen uniformity (p = 0.08) and less ectasia (6.7% versus 28.2%, p = 0.05). There was some evidence that ligation of side branches with metallic clips increased SVG failure in the stented group.
External stenting has the potential to improve SVG lumen uniformity and reduce diffuse intimal hyperplasia 1 year after coronary artery bypass graft surgery.
UDP-N-Acetyl glucosamine-2 epimerase/N-acetyl mannosamine kinase (GNE) catalyzes key enzymatic reactions in the biosynthesis of sialic acid. Mutation in GNE gene causes GNE myopathy (GNEM) ...characterized by adult-onset muscle weakness and degeneration. However, recent studies propose alternate roles of GNE in other cellular processes beside sialic acid biosynthesis, particularly interaction of GNE with α-actinin 1 and 2. Lack of appropriate model system limits drug and treatment options for GNEM as GNE knockout was found to be embryonically lethal. In the present study, we have generated L6 rat skeletal muscle myoblast cell-based model system carrying one single Gne allele where GNE gene is knocked out at exon-3 using AAV mediated SEPT homology recombination (SKM-GNEHz). The cell line was heterozygous for GNE gene with one wild type and one truncated allele as confirmed by sequencing. The phenotype showed reduced GNE epimerase activity with little reduction in sialic acid content. In addition, the heterozygous GNE knockout cells revealed altered cytoskeletal organization with disrupted actin filament. Further, we observed increased levels of RhoA leading to reduced cofilin activity and causing reduced F-actin polymerization. The disturbed signaling cascade resulted in reduced migration of SKM-GNEHz cells. Our study indicates possible role of GNE in regulating actin dynamics and cell migration of skeletal muscle cell. The skeletal muscle cell-based system offers great potential in understanding pathomechanism and target identification for GNEM.
Asia continues to suffer from a high prevalence of malnutrition. Persistent malnutrition can be attributed to low dietary diversity, together with low production diversity. Dietary diversity ...represents a more healthy, balanced, and diverse diet, which ensures nutrient adequacy. The principle of dietary diversity is affirmed in all national food-based dietary guidelines. Food-based approaches that address malnutrition, especially micronutrient deficiencies, are embedded in evidence-based healthy diet patterns; however, they are disconnected from the current agricultural production system. Promising neglected and underutilized species (NUS) that are nutrient-dense, climate-resilient, profitable, and locally available/adaptable are fundamental to improving dietary and production diversity. The Future Smart Food Initiative, led by FAO's Regional Initiative on Zero Hunger, aims to harness the enormous benefits of NUS in the fight against hunger and malnutrition. Recognizing that NUS covers crops, livestock, fisheries and aquaculture, and forests, the FAO has set crops as an entry point for NUS to address hunger and malnutrition.
Cell migration is an essential cellular process that requires coordination of cytoskeletal dynamics, reorganization, and signal transduction. The actin cytoskeleton is central in maintaining the ...cellular structure as well as regulating the mechanisms of cell motility. Glycosylation, particularly sialylation of cell surface proteins like integrins, regulates signal transduction from the extracellular matrix to the cytoskeletal network. The activation of integrin by extracellular cues leads to recruitment of different focal adhesion complex proteins (Src, FAK, paxillin, etc.) and activates the signal including Rho GTPases for the regulation of actin assembly and disassembly. During cell migration, the assembly and disassembly of actin filament provides the essential force for the cell to move. Abnormal sialylation can lead to actin signaling dysfunction leading to aberrant cell migration, one of the main characteristics of cancer and myopathies. In the present study, we have reported altered F-actin to G-actin ratios in GNE mutated cells. These cells exhibit pathologically relevant mutations of GNE (UDP N-acetylneuraminic 2-epimerase/N-acetylmannosamine kinase), a key sialic acid biosynthetic enzyme. It was found that GNE neither affects the actin polymerization nor binds directly to actin. However, mutation in GNE resulted in increased binding of α-actinin to actin filaments. Further, through confocal imaging, GNE was found to be localized in focal adhesion complex along with paxillin. We further elucidated that mutation in GNE resulted in upregulation of RhoA protein and Cofilin activity is downregulated, which could be rescued with Rhosin and chlorogenic acid, respectively. Lastly, mutant in GNE reduced cell migration as implicated from wound healing assay. Our study indicates that molecules altering Cofilin function could significantly revert the cell migration defect due to GNE mutation in sialic acid-deficient cells. We propose cytoskeletal proteins to be alternate drug targets for disorders associated with GNE such as GNE myopathy.
Depth of Invasion (DOI) is an important and independent predictor for occult metastasis and recurrence. AJCC staging system (8th edition) has incorporated DOI in the staging of oral cancers. It is an ...essential parameter in deciding elective neck dissection. We hereby conducted a study to determine the accuracy of DOI in the frozen section as compared to the histological section in patients with T1N0 oral squamous cell carcinoma. This study was conducted over a period of 45 months in a tertiary care hospital. 31 patients with diagnosed Oral SCC and with T1N0 were enrolled. The intra-operative frozen section was done DOI was measured as per Cap protocol and compared with the DOI in the histopathological section. Strong correlation was found between DOI measured by frozen section and permanent section (r = 0.998; 95% CI, 0.999–0.997). The paired

test showed a mean difference of 0.0066 mm (95% CI, − 0.0103–0.02346 mm;

= 0.096) between frozen section and permanent section. DOI measured was slightly higher in frozen section however this difference was neither statically nor clinically significant.This study shows the accuracy of frozen specimens in determining tumor DOI in T1N0M0 in oral cavity squamous cell carcinoma. Intraoperative DOI along with other factors may be used to determine the need for END (elective neck dissection) in early-stage oral cavity squamous cell carcinoma and thus avoid a staged operation or over- or under-treatment of the neck.
