Gastric cancer (GC) is a common malignancy and frequent cause of cancer-related death. Long non-coding RNAs (lncRNAs) have emerged as important regulators and tissue-specific biomarkers of multiple ...cancers, including GC. Recent evidence has indicated that the novel lncRNA LINC01133 plays an important role in cancer progression and metastasis. However, its function and molecular mechanism in GC remain largely unknown.
LINC01133 expression was detected in 200 GC and matched non-cancerous tissues by quantitative reverse transcription PCR. Gain- and loss-of-function experiments were conducted to investigate the biological functions of LINC01133 both in vitro and in vivo. Insights into the underlying mechanisms of competitive endogenous RNAs (ceRNAs) were determined by bioinformatics analysis, dual-luciferase reporter assays, quantitative PCR arrays, TOPFlash/FOPFlash reporter assay, luciferase assay, and rescue experiments.
LINC01133 was downregulated in GC tissues and cell lines, and its low expression positively correlated with GC progression and metastasis. Functionally, LINC01133 depletion promoted cell proliferation, migration, and the epithelial-mesenchymal transition (EMT) in GC cells, whereas LINC01133 overexpression resulted in the opposite effects both in vitro and in vivo. Bioinformatics analysis and luciferase assays revealed that miR-106a-3p was a direct target of LINC01133, which functioned as a ceRNA in regulating GC metastasis. Mechanistic analysis demonstrated that miR-106a-3p specifically targeted the adenomatous polyposis coli (APC) gene, and LINC01133/miR-106a-3p suppressed the EMT and metastasis by inactivating the Wnt/β-catenin pathway in an APC-dependent manner.
Our findings suggest that reduced expression of LINC01133 is associated with aggressive tumor phenotypes and poor patient outcomes in GC. LINC01133 inhibits GC progression and metastasis by acting as a ceRNA for miR-106a-3p to regulate APC expression and the Wnt/β-catenin pathway, suggesting that LINC01133 may serve as a potential prognostic biomarker and anti-metastatic therapeutic target for GC.
1H-benzotriazole (BTri) and 5-tolyltriazole (5-TTri) are emerging pollutants; the development of novel materials for their efficient adsorption and removal is thus of great significance in ...environmental sciences. Here, we report the application of zeolitic imidazolate framework-8 (ZIF-8) as a novel adsorbent for fast removal of BTri and 5-TTri in aqueous solution in view of adsorption isotherms, kinetics and thermodynamics, desorption, and adsorbent regeneration. The adsorption of BTri and 5-TTri on ZIF-8 was very fast, and most of BTri and 5-TTri were adsorbed in the first 2 min. The adsorption for BTri and 5-TTri follows a pseudo-second-order kinetics and fits the Langmuir adsorption model with the adsorption capacity of 298.5 and 396.8 mg g–1 for BTri and 5-TTri at 30 °C, respectively. The adsorption was a spontaneous and endothermic process controlled by positive entropy change. No remarkable effects of pH, ionic strength, and dissolved organic matter on the adsorption of BTri and 5-TTri on ZIF-8 were observed. The used ZIF-8 could be regenerated effectively and recycled at least three times without significant loss of adsorption capacity. In addition, ZIF-8 provided much larger adsorption capacity and faster adsorption kinetics than activated carbon and ZIF-7. The hydrophobic and π–π interaction between the aromatic rings of the BTri and 5-TTri and the aromatic imizole rings of the ZIF-8, and the coordination of the nitrogen atoms in BTri and 5-TTri molecules to the Zn2+ ions in the ZIF-8 framework was responsible for the efficient adsorption. The fast adsorption kinetics, large adsorption capacity, excellent reusability as well as the pH, ionic strength, and dissolved organic matter insensitive adsorption create potential for ZIF-8 to be effective at removing benzotriazoles from aqueous solution.

Beclin-1-dependent autophagy in the heart during sepsis and the potential therapeutic benefit of targeting this pathway were investigated in a mouse model of lipopolysaccharide (LPS)-induced sepsis.
LPS induced a dose-dependent increase in autophagy at low doses, followed by a decline that was in conjunction with mammalian target of rapamycin activation at high doses. Cardiac-specific overexpression of Beclin-1 promoted autophagy, suppressed mammalian target of rapamycin signaling, improved cardiac function, and alleviated inflammation and fibrosis after LPS challenge. Haplosufficiency for beclin 1 resulted in opposite effects. Beclin-1 also protected mitochondria, reduced the release of mitochondrial danger-associated molecular patterns, and promoted mitophagy via PTEN-induced putative kinase 1-Parkin but not adaptor proteins in response to LPS. Injection of a cell-permeable Tat-Beclin-1 peptide to activate autophagy improved cardiac function, attenuated inflammation, and rescued the phenotypes caused by beclin 1 deficiency in LPS-challenged mice.
These results suggest that Beclin-1 protects the heart during sepsis and that the targeted induction of Beclin-1 signaling may have important therapeutic potential.
The interfacial flow and mass transfer of multicomponent droplets remain an unsolved basic problem, although multicomponent droplets are widely used in droplet microfluidics, fine chemicals. In this ...paper, Level-set method is used to capture the phase interface, and the evolution of Marangoni convection under different Reynolds numbers is investigated by CFD simulation. The influence of Marangoni effect on the macroscopic characteristics such as droplet velocity and concentration decay rate under different motion states are reported, and the divergence of velocity at the interface is statistically analyzed at microscopic level. The results reveal that the droplet Reynolds number has a great influence on the development of Marangoni convection in life and space; The local divergence can clearly reflect the influence of flow structure and flow velocity on interphase material transport; The Marangoni vortices are multi-scale, and the interphase mass transfer containing the Marangoni effect is non-Gaussian and oscillating, which similar to turbulence. Compared to previous research, this work complements a lot of micro information of the interface Marangoni flow, and deeply discusses the competition between Marangoni and internal circulation flow in droplets under different Reynolds numbers.

We carried out metagenomic shotgun sequencing and a metagenome-wide association study (MGWAS) of fecal, dental and salivary samples from a cohort of individuals with rheumatoid arthritis (RA) and ...healthy controls. Concordance was observed between the gut and oral microbiomes, suggesting overlap in the abundance and function of species at different body sites. Dysbiosis was detected in the gut and oral microbiomes of RA patients, but it was partially resolved after RA treatment. Alterations in the gut, dental or saliva microbiome distinguished individuals with RA from healthy controls, were correlated with clinical measures and could be used to stratify individuals on the basis of their response to therapy. In particular, Haemophilus spp. were depleted in individuals with RA at all three sites and negatively correlated with levels of serum autoantibodies, whereas Lactobacillus salivarius was over-represented in individuals with RA at all three sites and was present in increased amounts in cases of very active RA. Functionally, the redox environment, transport and metabolism of iron, sulfur, zinc and arginine were altered in the microbiota of individuals with RA. Molecular mimicry of human antigens related to RA was also detectable. Our results establish specific alterations in the gut and oral microbiomes in individuals with RA and suggest potential ways of using microbiome composition for prognosis and diagnosis.
Recently, composting cultivation method is widely used in oyster mushroom production. In this study, we focused on the effects of composting processes on nutritional qualities and antioxidant ...activity of Pleurotus floridanus mushroom fruiting bodies. Three treatments of different composting time (2, 4, and 5 days) were performed with an atmospheric sterilization treatment as the control. The results showed that the pH value, total carbon content, and total nitrogen content of substrate were critical parameters which would significantly affect mushroom qualities and bioactivities. Fruiting bodies of the control demonstrated significantly higher crude protein content, total amino acid content, and essential amino acid content than that of composting treatments. Moreover, fruiting bodies of treatment D4 and D5 manifested significantly higher crude polysaccharide contents. Crude polysaccharide of treatment D4 represented the highest scavenging ability toward both radical 3‐ethylbenzthiazoline‐6‐sulfonic acid (ABTS·+) and Hydroxyl radical (OH·). It suggests that composting processes is suitable for oyster mushroom cultivation based on nutritional and antioxidant qualities of fruiting bodies.

The lensing effect of the cosmic microwave background (CMB) is a powerful tool for our study of the distribution of matter in the universe. The quadratic estimator (QE) method, which is ...widely used to reconstruct lensing potential, has been known to be suboptimal for the low noise level polarization data from next-generation CMB experiments. To improve the performance of the reconstruction, other methods, such as the maximum-likelihood estimator and machine-learning algorithms, have been developed. In this work, we present a deep convolutional neural network model named the Residual Dense Local Feature U-net (RDLFUnet) for reconstructing the CMB lensing convergence field. By simulating lensed CMB data with different noise levels to train and test network models, we find that for noise levels less than 5
μ
K-arcmin, RDLFUnet can recover the input gravitational potential with a higher signal-to-noise ratio than the previous deep-learning and traditional QE methods at almost the entire observation scale.

Short-term composting of raw materials for preparing oyster mushroom cultivation media is widely used in China, and its microbial mechanism needs to be further studied. 11-days' peach sawdust-based composting was performed to evaluate material conversion and microbial succession using physicochemical analysis and 16S rRNA and ITS sequencing. Composting bacteria demonstrated much higher abundance than fungi. Firmicutes, Actinobacteriota, and Proteobacteria were the dominant bacterial phyla, while most of fungal species belonged to Ascomycota. Moisture was the key factor at the beginning, while total nitrogen, temperature, and lignin became main influencing factors for composting maturity. Actinobacteriota, Firmicutes, and Proteobacteria of bacterial phyla, Eurotiomycetes and Sordariomycetes of fungal classes involved in lignocellulosic degradation. Bacterial function prediction analysis showed that carbohydrate metabolism and amino acid metabolism were the main metabolic pathways. These results confer a better understanding of material and microbial succession during short-term composting and also provide valuable utilization in mushroom industry.
Agaricus bisporus is in general cultivated on wheat and rice straw in China. However, millet straw is a potential alternative resource for Agaricus bisporus cultivation, but this has hardly been ...studied. In the present study, the feasibility of millet straw based mushroom production was analyzed by three successive trials. Mature compost demonstrated high quality with total nitrogen, pH, and C/N ratio of 2.0%, 7.5, and 18:1 respectively, which was suitable for mushroom mycelia growth. During composting, 47-50% of cellulose, 63-65% of hemicellulose, and 8-17% lignin were degraded, while 22-27% of cellulose, 14-16% of hemicellulose, and 15-21% of lignin were consumed by A. bisporus mycelia during cultivation. The highest FPUase and CMCase were observed during mushroom flushes. Endo-xylanase had the key role in hemicellulose degradation with high enzyme activity during cultivation stages. Laccase participated in lignin degradation with the highest enzyme activity in Pinning stage followed by a sharp decline at the first flush. Yield was up to 20 kg/m

Actinobacteria, Bacteroidetes, Chloroflexi, Deinococcus-Thermus, Firmicutes, and Proteobacteria were the dominant phyla, based on 16S rRNA gene sequencing during composting. The key environmental factors dominating bacterial communities of the samples were determined to be pH value, cellulose content, and hemicellulose content for prewetting and premixed phase of basic mixture (P0); moisture content for phase I (PI); and nitrogen content, lignin content, and ash content for phase II (PII), respectively.
