The geosynthetic-reinforced pile-supported embankment (GRPSE) system has been widely used in road construction on soft soil. However, the application of the GRPSE system is often restricted by its ...high-cost. The reason is that they are designed for bearing control as defined in the past. During the construction process, the pile spacing is reduced to meet the requirements for the embankment bearing capacity and settlement. These factors cause the membrane effect to not be exploited. As a result, the utilization efficiency of the bearing capacity of the soil between the piles is low and the project cost is high. Therefore, in order to solve the problem of insufficient bearing capacity of soil between piles, we established a settlement calculation model of soil between piles based on membrane effect. The model considers the relationship between the geosynthetic reinforcement (GR) and the pile spacing. Based on the obtained model, a method for optimizing the soil bearing capacity of GRPSEs is proposed. By controlling the settlement of soil between piles, the bearing capacity of soil between piles and the membrane effect of embankment can be fully utilized. Therefore, the project cost can be reduced. Finally, the method is applied to field tests for comparison. The results show that the method is reasonable and applicable. This method can effectively exploit the membrane effect and improve the utilization efficiency of the bearing capacity of the soil between piles. An economical and reasonable arrangement scheme for the piles and GR was obtained. This scheme can not only ensure the safety of the project, but also fully utilize the bearing capacity of the soil between the piles and provide a new method for engineering design.
Due to the lack of research on the pathological mechanism of temporomandibular joint osteoarthritis (TMJOA), there are few effective treatment measures in the clinic. In recent years, microRNAs ...(miRs) have been demonstrated to play an important role in the pathogenesis of osteoarthritis (OA) by regulating a variety of target genes, and the latest evidence shows that miR-21-5p is specifically overexpressed in OA. The purpose of this project was to clarify whether miR-21-5p can regulate the TMJOA process by targeting Spry1.
TMJOA was induced by a unilateral anterior crossbite (UAC) model, and the effect of miR-21-5p knockout on TMJOA was evaluated by toluidine blue (TB), immunohistochemical (IHC) staining, Western blotting (WB) and RT-qPCR. Primary mouse condylar chondrocytes (MCCs) were isolated, cultured and transfected with a series of mimics, inhibitors, siRNA-Spry1 or cDNA Spry1. WB, RT-qPCR, IHC and TB were used to detect the effect of miR-21-5p and its target gene Spry1 on the expression of MMP-13, VEGF and p-ERK1/2 in TMJOA. The effect of miR-21-5p on angiogenesis was evaluated by chick embryo chorioallantoic membrane (CAM) assay and WB.
In the UAC model, the cartilage thickness and extracellular matrix of miR-21-5p knockout mice were less damaged, and miR-21-5p and UAC model were shown to affect the expression of Spry1, IL-1β, MMP-13, and VEGF. Luciferase experiments confirmed that Spry1 was the direct target of miR-21-5p. The expression levels of Spry1, MMP-13, VEGF and p-ERK1/2 in MCCs transfected with miR-21-5p mimic were higher than those in the inhibitor group. Under the simulated inflammatory environment of IL-1β, the expression levels of MMP-13, VEGF and p-ERK1/2 were positively correlated with miR-21-5p, while Spry1 was negatively correlated with miR-21-5p. Inhibition of miR-21-5p expression and overexpression of Spry1 enhanced the inhibition of MMP-13, VEGF and p-ERK1/2 expression. MiR-21-5p had a significant role in promoting angiogenesis in the chick embryo CAM assay, and this role was clearly mediated by the ERK-MAPK signalling pathway.
This study verified that miR-21-5p can promote the process of TMJOA by targeting Spry1, which provides a new direction for future research on the treatment of this disease.

Circular RNAs (circRNAs) exert critical functions in tumorigenesis and tumor development, but whether and how circRNAs contribute to laryngeal squamous cell carcinoma (LSCC) is unclear. In this study, we explored the function and mechanisms of circRNA_100290 in LSCC. Tissue samples were obtained from 40 patients with LSCC. The expression of circRNA_100290 and other targets was measured through quantitative reverse transcription-polymerase chain reaction and western blot analysis. Cell proliferation, colony-forming ability, and apoptosis were tested using CCK-8 assay and EdU assay, colony formation assay, and flow cytometry, respectively. Cell migration and invasion were detected by Transwell assay. Moreover, the interactions between circRNA_100290, miR-136-5p, and RAP2C were analyzed by bioinformatics, and verified by dual‐luciferase reporter assays. Here, we found that circRNA_100290 expression was significantly upregulated in LSCC tissues and cell lines compared with the normal controls. Expression of circRNA_100290 positively correlated with advanced TNM stage and lymph node metastasis in LSCC patients. In cell culture, upregulation of circRNA_100290 promoted LSCC cell proliferation, migration, and invasion, while it inhibited cell apoptosis; downregulating circRNA_100290 exerted the opposite effects. In vivo, circRNA_100290 overexpression dramatically promoted tumor growth. Mechanistically, circRNA_100290 may act as a sponge of miR-136-5p, and inhibiting miR-136-5p in LSCC cells indeed reversed the effects of circRNA_100290 downregulation. The RAS oncogene RAP2C was predicted to be a target of miR-136-5p, and downregulating RAP2C in LSCC cells partially reversed the oncogenic effects of circRNA_100290 overexpression or miR-136-5p decrease. Our findings suggest that circRNA_100290 promotes LSCC progression by targeting the miR-136-5p/RAP2C axis, which may lead to the identification of potential therapeutic targets.
Abstract
Multiplex interactions are common and essential in real-world systems. In many cases, we can only obtain aggregated networks without detailed information regarding the type of links ...contained within. Such single-layer networks oversimplify the structural information and lead to misunderstandings of some properties of real systems. In this context, network splitting which aims to correctly separate an aggregated network into multilayer networks, is a meaningful problem to address. To this end, we propose a simulated-annealing-like algorithm based on the link clustering coefficient. We verify the validity of this algorithm with several synthetic networks. Inter-similarities of layers are also taken into consideration, and we can find that the proposed method is valid even if there is a certain proportion of overlapping links between layers. Finally, we apply the algorithm to real international trading networks, which results in accurate splits of different layers.
High energy proton-nucleus (pA) collisions provide an important constraint on the study of the chiral magnetic effect in QCD matter. Naively, in pA collisions one expects no correlation between the ...orientation of event plane as reconstructed from the azimuthal distribution of produced hadrons and the orientation of magnetic field. If this is the case, any charge-dependent hadron correlations can only result from the background. Nevertheless, in this paper we point out that in high multiplicity pA collisions a correlation between the magnetic field and the event plane can appear. This is because triggering on the high hadron multiplicity amounts to selecting Fock components of the incident proton with a large number of partons that are expected to have a transverse size much larger than the average proton size. We introduce the effect of the fluctuating proton size in the Monte Carlo Glauber model and evaluate the resulting correlation between the magnetic field and the second-order event plane in both pA and nucleus-nucleus (AA) collisions. The fluctuating proton size is found to result in a significant correlation between magnetic field and the event plane in pA collisions, even though the magnitude of the correlation is still much smaller than in AA collisions. Here, this result opens a possibility of studying the chiral magnetic effect in small systems.
ATP7B is a copper-transporting protein that contributes to the chemo-resistance of human cancer cells. It remains unclear what the molecular mechanisms behind ATP7B are in cancer, as well as its role ...in human pan-cancer studies.
Our study evaluated the differential expression of ATP7B in cancer and paracancerous tissues based on RNA sequencing data from the GTEx and TCGA. Kaplan-Meier and Cox proportional hazards regressions were used to estimate prognostic factors associated with ATP7B.The correlations between the expression of ATP7B and immune cell infiltration, tumor mutation burden, microsatellite instability and immune checkpoint molecules were analyzed. Co-expression networks and mutations in ATP7B were analyzed using the web tools. An analysis of ATP7B expression difference on drug sensitivity on tumor cells was performed using the CTRP, GDSC and CMap database.
ATP7B expression differed significantly between cancerous and paracancerous tissues. The abnormal expression of ATP7B was linked to prognosis in LGG and KIRC. Infiltration of immune cells, tumor mutation burden, microsatellite instability and immunomodulators had all been linked to certain types of cancer. Cancer cells exhibited a correlation between ATP7B expression and drug sensitivity.
ATP7B might be an immunotherapeutic and prognostic biomarker based on its involvement in cancer occurrence and development.
Ferroptosis is a new form of cell death that is unique and closely related to iron concentration, and reactive oxygen species (ROS) production. We investigated the indicators of ferroptosis between ...vulnerable plaque and stable plaque in atherosclerotic. Quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting were used to detect the expression of the ferroptosis-related genes and proteins and extracellular matrix stability-related genes and proteins (FN, CoL-1). Superoxide dismutase (SOD) activities, glutathione peroxidase (GSH) and malondialdehyde (MDA) were detected by ELISA. The commercially available kit was used to detect Fe2+ concentration in tissue. DCFH-DA fluorescent probe was used to detect the ROS levels. H&E stain, Masson trichrome stain, and Oil Red O stain were used to detect pathological states in vulnerable plaque and stable plaque. Tissue localization and positive rate of GPX4, SLC7A11, COX-2, FN, and COL-1 were evaluated by immunohistochemistry. The results showed a significant increase in the expression of COX2 and a significant decrease in the expression of GPX4 and SLC7A11 in genes related to ferroptosis in vulnerable plaque compared with stable plaque. Pathologic results showed vulnerable plaque with higher levels of inflammatory cell infiltration, more diffuse collagen fibers, and larger particles of lipid droplets. Concentrations of the antioxidant metabolites SOD and GSH were significantly reduced and concentrations of the oxidative metabolites MDA and Fe2+ were significantly increased in vulnerable plaque compared with stable plaque. The expression of FN and CoL-1 was significantly reduced in genes related to extracellular matrix stability in vulnerable plaque. Taken together, these findings indicate that the degree of ferroptosis in vulnerable plaque is higher than that in stable plaque, suggesting that changes in indicators of ferroptosis may affect carotid atherosclerotic plaque stability, target spot in the ferroptosis signaling pathway may provide further theoretical basis for the clinical treatment of carotid atherosclerosis.
Objectives
The purpose of this study was to evaluate whether the clinical outcome of socket shield technique (SST) is superior to that of conventional immediate implantation (CII).
Materials and ...method
Five electronic databases (PubMed, Cochrane, Web of Science, CNKI, and Google Scholar) were searched to identify randomized controlled trials up to June 31, 2021. Five evaluation indexes were extracted, namely, buccal bone resorption at the horizontal and vertical levels (BBH and BBV), the soft tissue recession assessed by pink evaluation scores (PES), patient satisfaction (PS), ISQ, and the success rate of implantation (SRI), to compare the superiority between SST and CII operations. All data analyses were performed using Review Manager (version 5.4).
Results
Ten studies were included in this review. The sample included 388 implants, with 194 in the SST group and 194 in the CII group. Compared with the CII group, the SST group had a lower BBH and BBV (standardized mean difference (SMD), − 1.77; 95% CI, − 2.26 to − 1.28;
P
< 0.00001 and SMD, − 1.85; 95% CI, − 2.16 to 1.54;
P
< 0.00001), higher PES improvement (SMD, 2.27; 95% CI, 1.59 to 2.95;
P
< 0.00001), higher rate of PS (OR, 3.12; 95% CI, 1.08 to 9.04;
P
= 0.04), and slightly higher ISQ (SMD, 0.71; 95% CI, 0.28 to 1.15;
P
= 0.001).
Conclusions
Compared with CII, SST could be a better option for esthetic area implantation, but evaluation of its long-term success is still needed.
Clinical relevance
By comparing and analyzing the operations of immediate implant in esthetic zone, we could choose SST to effectively alleviate the absorption of bone tissue and improve the contouring of soft tissue after anterior teeth extraction, so as to achieve a more stable and superior clinical outcomes of implant in esthetic zone.
The reservoir physical properties, pore types, diagenetic characteristics and reservoir quality controlling effect of the Xishanyao formation coal measure sandstone in the southern margin of the ...Junggar basin were discussed in this study based on thin section observation, high pressure mercury injection, low-temperature nitrogen adsorption and scanning electron microscope observation. The result shows that the porosity and permeability of the sandstone are generally low with a medium-high texture maturity and low compositional maturity. The sandstone storage space is mainly composed of residual intergranular pores, secondary dissolution pores, inter-crystalline pores and micro-fractures. The diagenetic stage of coal measure sandstone is in the mesodiagenesis A1-A2 stage, and their diagenetic interaction types mainly include compaction, cementation and dissolution. The reservoir quality of the coal measure sandstone deteriorates by compaction due to high matrix content and plastic debris content. Because of the large amounts of organic acids generated during the thermal evolution of the coal measure source rock, the coal measure sandstone suffers from strong dissolution. The secondary dissolution pores formed by the massive dissolution of feldspar, lithic fragments and early carbonate cementation in the sandstone significantly improved the reservoir quality. In the coal measure sandstone, clay mineral cementation is the most developed cementation form, followed by quartz cementation and carbonate cementation. Although kaolinite cementation and dolomite cementation can generate a small number of inter-crystalline pores, cementation deteriorates the reservoir quality. The Xishanyao formation coal measure sandstone formed in a lacustrine-delta environment, and its composition and texture make it susceptible to the influence of compaction and dissolution during diagenesis.
Enthesis, the interfacial tissue between a tendon/ligament and bone, exhibits a complex histological transition from soft to hard tissue, which significantly complicates its repair and regeneration ...after injury. Because traditional surgical treatments for enthesis injury are not satisfactory, tissue engineering has emerged as a strategy for improving treatment success. Rapid advances in enthesis tissue engineering have led to the development of several strategies for promoting enthesis tissue regeneration, including biological scaffolds, cells, growth factors, and biophysical modulation. In this review, we discuss recent advances in enthesis tissue engineering, particularly the use of biological scaffolds, as well as perspectives on the future directions in enthesis tissue engineering.
