and Purpose Recent randomized controlled trials have demonstrated consistent effectiveness of endovascular treatment (EVT) for acute ischemic stroke, leading to update on stroke management ...guidelines. We conducted this meta-analysis to assess the efficacy and safety of EVT overall and in subgroups stratified by age, baseline stroke severity, brain imaging feature, and anesthetic type.
Published randomized controlled trials comparing EVT and standard medical care alone were evaluated. The measured outcomes were 90-day functional independence (modified Rankin Scale ≤2), all-cause mortality, and symptomatic intracranial hemorrhage.
Nine trials enrolling 2476 patients were included (1338 EVT, 1138 standard medical care alone). For patients with large vessel occlusions confirmed by noninvasive vessel imaging, EVT yielded improved functional outcome (pooled odds ratio OR, 2.02; 95% confidence interval CI, 1.64-2.50), lower mortality (OR, 0.75; 95% CI, 0.58-0.97), and similar symptomatic intracranial hemorrhage rate (OR, 1.12; 95% CI, 0.72-1.76) compared with standard medical care. A higher proportion of functional independence was seen in patients with terminus intracranial artery occlusion (±M1) (OR, 3.16; 95% CI, 1.64-6.06), baseline Alberta Stroke Program Early CT score of 8-10 (OR, 2.11; 95% CI, 1.25-3.57) and age ≤70 years (OR, 3.01; 95% CI, 1.73-5.24). EVT performed under conscious sedation had better functional outcomes (OR, 2.08; 95% CI, 1.47-2.96) without increased risk of symptomatic intracranial hemorrhage or short-term mortality compared with general anesthesia.
Vessel-imaging proven large vessel occlusion, a favorable scan, and younger age are useful predictors to identify anterior circulation stroke patients who may benefit from EVT. Conscious sedation is feasible and safe in EVT based on available data. However, firm conclusion on the choice of anesthetic types should be drawn from more appropriate randomized controlled trials.
•One new natural product has been isolated from Salicornia herbacea.•Pentadecyl ferulate exhibited strong antioxidant and potent antitumor activities.•Pentadecyl ferulate was the main antioxidant ...agent in the S. herbacea extracts.•Several phytochemicals were responsible for the antioxidant activity of S. herbacea.
An investigation of the chemical constituents of Salicornia herbacea has led to the isolation of one new natural product, pentadecyl ferulate (6), together with 11 known compounds, including phytol (1), stearolic acid (2), γ-linolenic acid (3), (3Z,6Z,9Z)-tricosa-3,6,9-triene (4), linoleic acid (5), stigmasterol (7), ergosterol (8), dioctyl phthalate (9), dibutyl phthalate (10), vanillic aldehyde (11), and scopoletin (12). The chemical structures of these materials were elucidated mainly by spectroscopic analysis. This work represents the first recorded example of the isolation of compounds 1, 2, 3, 4, 9, 10, and 11 from S. herbacea. The antioxidant experiments revealed that compound 6 possessed strong hydroxy radical and superoxide anion scavenging activities and was the principle antioxidant ingredient in the ethyl acetate extract. The antiproliferative results exhibited that compound 1 selectively inhibited HepG2 cells, whereas compounds 3 and 6 showed potent antiproliferative activities against HepG2 and A549 cells.
•Rabbit-derived human Thelazia callipaeda infection occurred in an undergraduate student.•T. callipaeda can be cured through mechanical removal.•Health and safety education of medical students should ...be strengthened.
In November 2021, a 19-year-old medical student was diagnosed with a rabbit-derived Thelazia callipaeda infection in her left eye. She visited her doctor because she removed the white object by herself with a cotton swab 2 days earlier. She explained to the doctor that she rubbed her eyes without washing her hands when rabbit hair got into her eyes during the animal experiment class at school 2 weeks earlier. The other 2 worms were then taken out with ophthalmic forceps and were placed into physiological saline. The worms were sent to the Shandong Academy of Eye Disease Prevention and Therapy for examination. After identification, the worms were regarded as the female T. callipaeda. Ofloxacin eye drops and peranapulin eye drops were administered to control inflammation. No abnormality was found in the follow-up 2 weeks and 1 month later. This report highlights that it is very important to strengthen the health and safety education of medical students during animal experiment classes.
This paper explores whether a Student's choice of major leads to certain personality traits and the reasons for this phenomenon. Specifically, we look at evidence from two Chinese universities, both ...of which specialize in agricultural studies. Using the Sixteen Personality Factor (16PF) questionnaire and the Neuroticism Extraversion Openness Five-Factor Inventory (NEO-FFI) questionnaire, we collected data from two groups of students: those who study agriculture-related majors (ARM), and those who study non-agriculture-related majors (NARM). The surveys all showed no significant change in personality traits during Students' freshman year. However, after 3 years of university study, significant personality trait changes were noted between seniors in the ARM and NARM groups. Whereas ARM seniors tended to be socially shy and lower in communicative competence, NARM seniors were better at expressing themselves and communicating with others. Although a Student's choice of profession has an influence on their personality traits, it is not the only factor. The differences between ARM and NARM training models and curricula are also undoubtedly significant. Moreover, the bias against ARM in Chinese society further magnifies the differences in personality traits among students with different majors.
•Two new noroleanane-type triterpene saponins have been isolated from S. herbacea.•The first reported isolation of several different compounds from S. herbacea.•These two new compounds, Salbige A and ...B, showed potent antiproliferative activities.•Pheophorbide a showed powerful antiproliferative activities.•The phytochemicals in S. herbacea work in synergy for its antiproliferative activity.
Two new noroleanane-type triterpene saponins, Salbige A (1) and Salbige B (2), have been isolated from the aerial parts of Salicornia herbacea together with five other known compounds, including echinocystic acid (3), gypsogenin (4), pheophorbide a (5), (132S)-hydroxy-pheophorbide a (6) and (132S)-hydro-pheophorbide-lactone a (7). The chemical structures of these compounds were elucidated by extensive spectroscopic analysis and on the basis of their chemical reactivity. This work represents the first recorded example of the isolation of these compounds from S. herbacea. Compounds 1 and 2 exhibited potent antiproliferative activities and high levels of selectivity towards A549 cancer cells, with IC50 values of 52.35 and 79.39μM, respectively, whereas compound 5 showed high levels of inhibitory activity against A549 and HepG2 cancer cells with IC50 values of 6.15 and 17.56μM, respectively. None of these compounds exhibited antioxidant activities except for compound 7, which showed weak antioxidant activity.
Sequencing of Ebola virus (EBOV) genomes during the 2014-2016 epidemic identified several naturally occurring, dominant mutations potentially impacting virulence or tropism. In this study, we ...characterized EBOV variants carrying one of the following substitutions: A82V in the glycoprotein (GP), R111C in the nucleoprotein (NP), or D759G in the RNA-dependent RNA polymerase (L). Compared with the wild-type (WT) EBOV C07 isolate, NP and L mutants conferred a replication advantage in monkey Vero E6, human A549, and insectivorous bat Tb1.Lu cells, while L mutants displayed a disadvantage in human Huh7 cells. The replication of the GP mutant was significantly delayed in Tb1.Lu cells and similar to that of the WT in other cells. The L mutant was less virulent, as evidenced by increased survival for mice and a significantly delayed time to death for ferrets, but increased lengths of the period of EBOV shedding may have contributed to the prolonged epidemic. Our results show that single substitutions can have observable impacts on EBOV pathogenicity and provide a framework for the study of other mutations.
During the Ebola virus (EBOV) disease outbreak in West Africa in 2014-2016, it was discovered that several mutations in the virus emerged and became prevalent in the human population. This suggests that these mutations may play a role impacting viral fitness. We investigated three of these previously identified mutations (in the glycoprotein GP, nucleoprotein NP, or RNA-dependent RNA polymerase L) in cell culture, as well as in mice and ferrets, by generating recombinant viruses (based on an early West African EBOV strain) each carrying one of these mutations. The NP and L mutations appear to decrease virulence, whereas the GP mutation slightly increases virulence but mainly impacts viral tropism. Our results show that these single mutations can impact EBOV virulence in animals and have implications for the rational design of efficacious antiviral therapies against these infections.
On 13 December 2012, Chang'e-2 conducted a successful flyby of the near-Earth asteroid 4179 Toutatis at a closest distance of 770 ± 120 meters from the asteroid's surface. The highest-resolution ...image, with a resolution of better than 3 meters, reveals new discoveries on the asteroid, e.g., a giant basin at the big end, a sharply perpendicular silhouette near the neck region, and direct evidence of boulders and regolith, which suggests that Toutatis may bear a rubble-pile structure. Toutatis' maximum physical length and width are (4.75 × 1.95 km) ±10%, respectively, and the direction of the +z axis is estimated to be (250 ± 5°, 63 ± 5°) with respect to the J2000 ecliptic coordinate system. The bifurcated configuration is indicative of a contact binary origin for Toutatis, which is composed of two lobes (head and body). Chang'e-2 observations have significantly improved our understanding of the characteristics, formation, and evolution of asteroids in general.
Posttranslational modifications, such as SUMOylation, play specific roles in the life cycle of invading pathogens. However, the effect of SUMOylation on the adaptation, pathogenesis, and transmission ...of influenza A virus (IAV) remains largely unknown. Here, we found that a conserved lysine residue at position 612 (K612) of the polymerase basic protein 1 (PB1) of IAV is a bona fide SUMOylation site. SUMOylation of PB1 at K612 had no effect on the stability or cellular localization of PB1, but was critical for viral ribonucleoprotein (vRNP) complex activity and virus replication in vitro. When tested in vivo, we found that the virulence of SUMOylation-defective PB1/K612R mutant IAVs was highly attenuated in mice. Moreover, the airborne transmission of a 2009 pandemic H1N1 PB1/K612R mutant virus was impaired in ferrets, resulting in reversion to wild-type PB1 K612. Mechanistically, SUMOylation at K612 was essential for PB1 to act as the enzymatic core of the viral polymerase by preserving its ability to bind viral RNA. Our study reveals an essential role for PB1 K612 SUMOylation in the pathogenesis and transmission of IAVs, which can be targeted for the design of anti-influenza therapies.
A high-reliability SEPIC converter with reconfigurable voltage conversion gain is proposed, which is composed of a basic SEPIC converter and n extended units. Adopting an interleaved parallel ...structure on the input side of the converter not only achieves leg-level hardware redundancy but also reduces the input current ripple. When one branch of the converter suddenly changes or fails, due to the redundancy between branches, the converter can still to work, and the duty cycle of the other branches can be readjusted by using PI control without changing output voltage and power of the converter. This feature undoubtedly presents the reliability of the converter. On the contrary, when the converter works in normal operation, it can also actively control operation mode of each branch to achieve reconfigurable voltage conversion gain function. In addition, since the duty cycles of the switches are not limited, voltage conversion gain of the converter is widely, which makes it more suitable for the occasions where the input side fluctuates greatly. Section 2 details the working principle of the proposed converter and the voltage and current stress on components. Then, the reconfigurable voltage conversion gain of the converter and its high reliability characteristics are analyzed in Section 3. Finally, the correctness of the theoretical analysis is verified by experiments.
