We present hereafter the development of the Liebherr Intelligent Hydraulic Cylinder, in which the hydraulic component is used as smart sensing element providing useful information for the system in ...which the cylinder is operated. The piston position and velocity are the most important signals derived from this new measuring approach. The performance under various load and temperature conditions (measured both on dedicated test facilities and in field in a real machine) will be presented. An integrated control electronics, which is performing the cylinder state processing, additionally allows the synchronized acquisition of external sensors. Providing comprehensive state information, such as temperature and system pressure, advanced control techniques or monitoring functions can be realized with a monolithic device. Further developments, trends and benefits for the system architecture will be briefly analyzed and discussed.
Changing network conditions pose severe problems to video streaming in the Internet. HTTP adaptive streaming (HAS) is a technology employed by numerous video services that relieves these issues by ...adapting the video to the current network conditions. It enables service providers to improve resource utilization and Quality of Experience (QoE) by incorporating information from different layers in order to deliver and adapt a video in its best possible quality. Thereby, it allows taking into account end user device capabilities, available video quality levels, current network conditions, and current server load. For end users, the major benefits of HAS compared to classical HTTP video streaming are reduced interruptions of the video playback and higher bandwidth utilization, which both generally result in a higher QoE. Adaptation is possible by changing the frame rate, resolution, or quantization of the video, which can be done with various adaptation strategies and related client- and server-side actions. The technical development of HAS, existing open standardized solutions, but also proprietary solutions are reviewed in this paper as fundamental to derive the QoE influence factors that emerge as a result of adaptation. The main contribution is a comprehensive survey of QoE related works from human computer interaction and networking domains, which are structured according to the QoE impact of video adaptation. To be more precise, subjective studies that cover QoE aspects of adaptation dimensions and strategies are revisited. As a result, QoE influence factors of HAS and corresponding QoE models are identified, but also open issues and conflicting results are discussed. Furthermore, technical influence factors, which are often ignored in the context of HAS, affect perceptual QoE influence factors and are consequently analyzed. This survey gives the reader an overview of the current state of the art and recent developments. At the same time, it targets networking researchers who develop new solutions for HTTP video streaming or assess video streaming from a user centric point of view. Therefore, this paper is a major step toward truly improving HAS.

HTTP Adaptive Streaming (HAS) technologies, e.g., Apple HLS or MPEG-DASH, automatically adapt the delivered video quality to the available network. This reduces stalling of the video but additionally introduces quality switches, which also influence the user-perceived Quality of Experience (QoE). In this work, we conduct a subjective study to identify the impact of adaptation parameters on QoE. The results indicate that the video quality has to be maximized first, and that the number of quality switches is less important. Based on these results, a method to compute the optimal QoE-optimal adaptation strategy for HAS on a per user basis with mixed-integer linear programming is presented. This QoE-optimal adaptation enables the benchmarking of existing adaptation algorithms for any given network condition. Moreover, the investigated concept is extended to a multi-user IPTV scenario. The question is answered whether video quality, and thereby, the QoE can be shared in a fair manner among the involved users.
Liver injury after COVID-19 vaccination is very rare and shows clinical and histomorphological similarities with autoimmune hepatitis (AIH). Little is known about the pathophysiology of COVID-19 ...vaccine-induced liver injury (VILI) and its relationship to AIH. Therefore, we compared VILI with AIH.
Formalin-fixed and paraffin-embedded liver biopsy samples from patients with VILI (n = 6) and from patients with an initial diagnosis of AIH (n = 9) were included. Both cohorts were compared by histomorphological evaluation, whole-transcriptome and spatial transcriptome sequencing, multiplex immunofluorescence, and immune repertoire sequencing.
Histomorphology was similar in both cohorts but showed more pronounced centrilobular necrosis in VILI. Gene expression profiling showed that mitochondrial metabolism and oxidative stress-related pathways were more and interferon response pathways were less enriched in VILI. Multiplex analysis revealed that inflammation in VILI was dominated by CD8+ effector T cells, similar to drug-induced autoimmune-like hepatitis. In contrast, AIH showed a dominance of CD4+ effector T cells and CD79a+ B and plasma cells. T-cell receptor (TCR) and B-cell receptor sequencing showed that T and B cell clones were more dominant in VILI than in AIH. In addition, many T cell clones detected in the liver were also found in the blood. Interestingly, analysis of TCR beta chain and Ig heavy chain variable-joining gene usage further showed that TRBV6-1, TRBV5-1, TRBV7-6, and IgHV1-24 genes are used differently in VILI than in AIH.
Our analyses support that SARS-CoV-2 vaccination-induced liver injury is related to AIH but also shows distinct differences from AIH in histomorphology, pathway activation, cellular immune infiltrates, and TCR usage. VILI may be a separate entity, which is distinct from AIH and more closely related to drug-induced autoimmune-like hepatitis.
Little is known about the pathophysiology of COVID-19 vaccine-induced liver injury (VILI). Our analysis shows that COVID-19 VILI shares some similarities with autoimmune hepatitis, but also has distinct differences such as increased activation of metabolic pathways, a more prominent CD8+ T cell infiltrate, and an oligoclonal T and B cell response. Our findings suggest that VILI is a distinct disease entity. Therefore, there is a good chance that many patients with COVID-19 VILI will recover completely and will not develop long-term autoimmune hepatitis.

Early specialisation versus early diversification in long-term athlete development remains controversial. Although several parameters of sport specialisation have already been explored, this study ...investigates within-sport specialisation in the careers of young swimmers. In this cohort study, the number of strokes, events and distance categories during childhood and adolescence, as well as entry age were analysed to find potential correlations with success at age 18. Also, national team members were compared to non-members within this cohort. The number of events, strokes or distance categories at a younger age showed a moderate correlation to the best FINA point score at the age of 18, i.e. the greater the diversification, the greater the performance at age 18. The number of events, strokes or distance categories however, showed a stronger correlation when comparing national team members with the non-members. Most of the observed athletes were more likely to have high FINA points at 18 if they had been listed in the top 100 at 11 years of age. Entry age had a strong negative correlation with the FINA point score at 18, i.e. the younger the athlete when entering the top 100, the higher the FINA point score at 18. When national team members were compared to non-national team members, early age of entry into the top 100 showed a greater impact on the national team members. However, being listed in more than one stroke as a child correlates well with success at age 18.

To assess whether IgA autoantibodies target lung-specific proteins and contribute to disease severity.
We collected 147 blood, 9 lung tissue, and 36 BAL fluid samples from three tertiary hospitals in Switzerland and one in Germany. Severe COVID-19 was defined by the need to administer oxygen. We investigated the presence of IgA autoantibodies and their effects on pulmonary surfactant in COVID-19 using the following methods: immunofluorescence on tissue samples, immunoprecipitations followed by mass spectrometry on BAL fluid samples, enzyme-linked immunosorbent assays on blood samples, and surface tension measurements with medical surfactant.
IgA autoantibodies targeting pulmonary surfactant proteins B and C were elevated in patients with severe COVID-19 but not in patients with influenza or bacterial pneumonia. Notably, pulmonary surfactant failed to reduce surface tension after incubation with either plasma or purified IgA from patients with severe COVID-19.
Our data suggest that patients with severe COVID-19 harbor IgA autoantibodies against pulmonary surfactant proteins B and C and that these autoantibodies block the function of lung surfactant, potentially contributing to alveolar collapse and poor oxygenation.
The Internet’s infrastructure shows severe limitations when an optimal end user experience for multimedia applications should be achieved in a resource-efficiently way. In order to realize truly ...user-centric networking, an information exchange between applications and networks is required. To this end, network-application interfaces need to be deployed that enable a better mediation of application data through the Internet. For smart multimedia applications and services, the application and the network should directly communicate with each other and exchange information in order to ensure an optimal Quality of Experience (QoE). In this article, we follow a use-case driven approach towards user-centric network orchestration. We derive user, application, and network requirements for three complementary use cases: HD live TV streaming, video-on-demand streaming and user authentication with high security and privacy demands, as typically required for payed multimedia services. We provide practical guidelines for achieving an optimal QoE efficiently in the context of these use cases. Based on these results, we demonstrate how to overcome one of the main limitations of today’s Internet by introducing the major steps required for user-centric network orchestration. Finally, we show conceptual prospects for realizing these steps by discussing a possible implementation with an inter-network architecture based on functional blocks.
HTTP Adaptive Streaming (HAS) is employed by more and more video streaming services in the Internet. It allows to adapt the downloaded video quality to the current network conditions, and thus, ...avoids stalling (i.e., playback interruptions) to the greatest possible extend. The adaptation of video streams is done by switching between different quality representation levels, which influences the user perceived quality of the video stream. In this work, the influence of several adaptation parameters, namely, switch amplitude (i.e., quality level difference), switching frequency, and recency effects, on Quality of Experience (QoE) is investigated. Therefore, crowdsourcing experiments were conducted in order to collect subjective ratings for different adaptation-related test conditions. The results of these subjective studies indicate the influence of the adaptation parameters, and based on these findings a simplified QoE model for HAS is presented, which only relies on the switch amplitude and the playback time of each layer.
The National Microbial Pathogen Data Resource (NMPDR) (http://www.nmpdr.org) is a National Institute of Allergy and Infections Disease (NIAID)-funded Bioinformatics Resource Center that supports ...research in selected Category B pathogens. NMPDR contains the complete genomes of ~50 strains of pathogenic bacteria that are the focus of our curators, as well as >400 other genomes that provide a broad context for comparative analysis across the three phylogenetic Domains. NMPDR integrates complete, public genomes with expertly curated biological subsystems to provide the most consistent genome annotations. Subsystems are sets of functional roles related by a biologically meaningful organizing principle, which are built over large collections of genomes; they provide researchers with consistent functional assignments in a biologically structured context. Investigators can browse subsystems and reactions to develop accurate reconstructions of the metabolic networks of any sequenced organism. NMPDR provides a comprehensive bioinformatics platform, with tools and viewers for genome analysis. Results of precomputed gene clustering analyses can be retrieved in tabular or graphic format with one-click tools. NMPDR tools include Signature Genes, which finds the set of genes in common or that differentiates two groups of organisms. Essentiality data collated from genome-wide studies have been curated. Drug target identification and high-throughput, in silico, compound screening are in development.
This paper addresses the challenge of assessing and modeling Quality of Experience (QoE) for online video services that are based on TCP-streaming. We present a dedicated QoE model for You Tube that ...takes into account the key influence factors (such as stalling events caused by network bottlenecks) that shape quality perception of this service. As second contribution, we propose a generic subjective QoE assessment methodology for multimedia applications (like online video) that is based on crowd sourcing - a highly cost-efficient, fast and flexible way of conducting user experiments. We demonstrate how our approach successfully leverages the inherent strengths of crowd sourcing while addressing critical aspects such as the reliability of the experimental data obtained. Our results suggest that, crowd sourcing is a highly effective QoE assessment method not only for online video, but also for a wide range of other current and future Internet applications.
