BACKGROUND AND AIMS: Gluconacetobacter diazotrophicus is a plant growth-promoting bacteria (PGPB) that colonizes several plant species. Here, we studied the internal colonization of Arabidopsis ...thaliana tissues by G. diazotrophicus and analyzed its effects on physiology, growth, and activation of plant immune system during such association. METHODS: A. thaliana seedlings were inoculated with G. diazotrophicus and grown in substrate for 50 days. Effects on plant growth were estimated by quantifying number of leaves, leaf area, and fresh and dry weight. Endophytic bacterial population was determined by colony-forming unit (CFU), and its location in plant tissues was assayed by epifluorescence microscopy of red fluorescent protein-labeled bacterium. Whole canopy gas exchange (photosynthesis and transpiration) was determined using a portable photosynthesis system. RESULTS: G. diazotrophicus efficiently promoted A. thaliana plant growth at 50 days after inoculation. Inoculated plants showed higher whole canopy photosynthesis, lower whole plant transpiration, and increased water-use efficiency. The bacterium colonized preferentially root xylem. The inoculation of plants defective in systemic acquired resistance (SAR)-associated defense revealed that plant immune system plays an important role during the early association stages. CONCLUSIONS: G. diazotrophicus endophytically colonizes A. thaliana roots, promotes plant growth, and increases whole canopy photosynthesis. Our results indicate that A. thaliana is useful for molecular studies of the mechanisms involved in the interaction between plants and PGPB, especially those involving G. diazotrophicus.
Objective
An association between bipolar disorder and cognitive impairment has repeatedly been described, even for euthymic patients. Findings are inconsistent both across primary studies and ...previous meta‐analyses. This study reanalysed 31 primary data sets as a single large sample (N = 2876) to provide a more definitive view.
Method
Individual patient and control data were obtained from original authors for 11 measures from four common neuropsychological tests: California or Rey Verbal Learning Task (VLT), Trail Making Test (TMT), Digit Span and/or Wisconsin Card Sorting Task.
Results
Impairments were found for all 11 test‐measures in the bipolar group after controlling for age, IQ and gender (Ps ≤ 0.001, E.S. = 0.26–0.63). Residual mood symptoms confound this result but cannot account for the effect sizes found. Impairments also seem unrelated to drug treatment. Some test‐measures were weakly correlated with illness severity measures suggesting that some impairments may track illness progression.
Conclusion
This reanalysis supports VLT, Digit Span and TMT as robust measures of cognitive impairments in bipolar disorder patients. The heterogeneity of some test results explains previous differences in meta‐analyses. Better controlling for confounds suggests deficits may be smaller than previously reported but should be tracked longitudinally across illness progression and treatment.

This study investigated the potential of selected compounds as inhibitors of SARS-CoV-2 Mpro through pharmacokinetic and toxicological analyses, molecular docking, and molecular dynamics simulations. ...In silico molecular docking simulations revealed promising ligands with favorable binding affinities for Mpro, ranging from −6.2 to −9.5 kcal/mol. Moreover, molecular dynamics simulations demonstrated the stability of protein–ligand complexes over 200 ns, maintaining protein secondary structures. MM-PBSA analysis revealed favorable interactions between ligands and Mpro, with negative binding energy values. Hydrogen bond formation capacity during molecular dynamics was confirmed, indicating consistent interactions with Mpro catalytic residues. Based on these findings, selected ligands show promise for future studies in developing COVID-19 treatments.
Approximately 57% of the Brazilian northeast region is recognized as semi-arid land and has been undergoing intense land use processes in the last decades, which have resulted in severe degradation ...of its natural assets. Therefore, the objective of this study is to identify the areas that are susceptible to desertification in this region based on the 11 influencing factors of desertification (pedology, geology, geomorphology, topography data, land use and land cover change, aridity index, livestock density, rural population density, fire hot spot density, human development index, conservation units) which were simulated for two different periods: 2000 and 2010. Each indicator were assigned weights ranging from 1 to 2 (representing the best and the worst conditions), representing classes indicating low, moderate and high susceptibility to desertification. The results indicate that 94% of the Brazilian northeast region is under moderate to high susceptibility to desertification. The areas that were susceptible to soil desertification increased by approximately 4.6% (83.4 km2) from 2000 to 2010. The implementation of the methodology provides the technical basis for decision-making that involves mitigating actions and the first comprehensive national assessment within the United Nations Convention to Combat Desertification framework.
Context. Open clusters (OCs) are popular tracers of the structure and evolutionary history of the Galactic disc. The OC population is often considered to be complete within 1.8 kpc of the Sun. The ...recent Gaia Data Release 2 (DR2) allows the latter claim to be challenged. Aims. We perform a systematic search for new OCs in the direction of Perseus using precise and accurate astrometry from Gaia DR2. Methods. We implemented a coarse-to-fine search method. First, we exploited spatial proximity using a fast density-aware partitioning of the sky via a k-d tree in the spatial domain of Galactic coordinates, (l, b). Secondly, we employed a Gaussian mixture model in the proper motion space to tag fields quickly around OC candidates. Thirdly, we applied an unsupervised membership assignment method, UPMASK, to scrutinise the candidates. We visually inspected colour-magnitude diagrams to validate the detected objects. Finally, we performed a diagnostic to quantify the significance of each identified over-density in proper motion and in parallax space. Results. We report the discovery of 41 new stellar clusters. This represents an increment of at least 20% of the previously known OC population in this volume of the Milky Way. We also report on the clear identification of NGC 886, an object previously considered an asterism. This study challenges the previous claim of a near-complete sample of OCs up to 1.8 kpc. Our results reveal that this claim requires revision, and a complete census of nearby OCs is yet to be found.

Human kallikreins 5 and 7 (KLK5 and KLK7) exhibit trypsin- and chymotrypsin-like activities and are involved in pathologies related to skin desquamation process. A series of new ...3-acyltetramic acids were developed as a novel class of inhibitors of KLK5, KLK7 and trypsin enzymes. The nature and length of the acyl chain is crucial to the KLK5, KLK7 and trypsin inhibition activities, and the most potent compounds (but not the most selective) 2b, 2c and 2g showed low micromolar IC50 values. While very few of the compounds were selective for KLK5, the selective inhibition of trypsin against chymotrypsin was achieved. Our molecular modelling studies revealed that the double bond in 2g provided the best fit in the binding site of KLK5, while the hydrogen bonding interactions modulated the best fit of 2c in the binding site of KLK7 due to the hydrophobicity of the cavity.
This study was performed to investigate sinus floor augmentation with two different particle sizes of demineralized bovine bone mineral (DBBM) by means of histological and immunohistochemical (IHC) ...analysis. A randomized clinical trial was conducted involving 10 individuals requiring two-stage bilateral maxillary sinus augmentation for implant installation. The patients were randomly divided into two groups following a split-mouth design: the maxillary sinus on one side was filled with small-sized particles (0.25–1mm) and on the contralateral side with large-sized particles (1–2mm). After a healing period of 8 months, 25 implants were placed. During implant site preparation, bone biopsies were obtained from each sinus, perpendicular to the long axis of the implant (buccal–palatal direction), for descriptive and histomorphometric analyses. IHC staining for protein expression of osteocalcin (OCN), vascular endothelial growth factor (VEGF), and tartrate-resistant acid phosphatase (TRAP) was also performed. Histomorphometric analysis revealed no statistically significant difference in the percentage of biomaterial (32.4±8.56% and 38.0±6.92%), newly formed bone (36.1±9.60% and 36.7±5.79%), or connective tissue (30.4±8.63% and 23.8±6.16%) between the small- and large-sized particle groups, respectively. IHC analysis did not reveal differences in the expression of OCN, VEGF, or TRAP. These findings suggest that both particle sizes of DBBM are effective for bone augmentation in the maxillary sinus.
Vascular endothelial growth factor A (VEGFA) is a key factor in the regulation of angiogenesis in adipose tissue. Poor vascularization during adipose tissue proliferation causes fibrosis and local ...inflammation, and is associated with insulin resistance. It is known that 17-beta estradiol (E2) regulates adipose tissue function and VEGFA expression in other tissues; however, the ability of E2 to regulate VEGFA in adipose tissue is currently unknown. In this study, we showed that, in 3T3-L1 cells, E2 and the estrogen receptor 1 (ESR1) agonist PPT induced VEGFA expression, while ESR1 antagonist (MPP), and selective knockdown of ESR1 using siRNA decreased VEGFA and prevented the ability of E2 to modulate its expression. Additionally, we found that E2 and PPT induced the binding of hypoxia inducible factor 1 alpha subunit (HIF1A) in the VEGFA gene promoter. We further found that VEGFA expression was lower in inguinal and gonadal white adipose tissues of ESR1 total body knockout female mice compared to wild type mice. In conclusion, our data provide evidence of an important role for E2/ESR1 in modulating adipose tissue VEGFA, which is potentially important to enhance angiogenesis, reduce inflammation and improve adipose tissue function.
